Objective: To establish an ultra performance liquid chromatography-tandem quadrupole mass spectrometry ( UPLC-MS/MS) method to determine the concentration of carbamazepine and phenobarbital in human plasma, and apply in the clinical moni-toring. Methods:Diazepam was used as the internal standard, and the samples were precipitated by acetonitrile. An ACQUITY UPL-C? BEH C18 (50 mm × 2. 1 mm, 1. 7 μm) column was used as the stationary phase at 40℃ with a Waters XEVO TQD. The mobile phase consisted of acetonitrile (containing 10 mmol·L-1 ammonium formate) and water (containing 10 mmol·L-1 ammonium for-mate and 0. 1% formic acid) with gradient elution pumped at a flow rate of 0. 4 ml·min-1 . ESI was applied and the samples were scanning analyzed by positive ion multi-reaction monitoring mode. The plasma was precipitated by 200 μl acetonitrile and centrifugated at 12 000 × g for 10 min and tranfer it into an Ep tube. The sample size was 20 μl. Results:The retention time of carbamazepine was 1. 23 min. Excellent linear calibration curves of carbamazepine were obtained within the concentration range of 0. 25-25μg·ml-1(r=0. 999 7). The lower limit of quantification of carbamazepine was 0. 01 μg·ml-1. The retention time of phenobarbital was 1. 11 min. Excellent linear calibration curves of carbamazepine were obtained within the concentration range of 0. 5-50 μg·ml-1(r=0. 999 6). The lower limit of quantification of carbamazepine was 0. 05 μg·ml-1. The recovery of three concentrations of carbamazepine was (82. 1 ± 6. 83)%, (82. 91 ± 4. 3)% and (84. 35 ± 3. 09)%, and the recovery of three concentrations of phenobarbital was (84. 27 ± 6. 91)%, (84. 32 ± 7. 74)% and (89. 07 ± 6. 24)%, respectively. The intra- and inter-day RSDs were all less than 10%. There were no endogenous substances existing in the incubation system, therefore, there was no interference with the determination. Conclu-sion:The simple, accurate and rapid method is suitable for the determination of carbamazepine and phenobarbital in human plasma, which can contribute greatly to the therapeutic drug monitoring service for patients.

Objective To study the effect of CsA in a therapeutic dose on the hepatic ultrastructure in rats.Methods Twenty-four SD rats were divided into 3 groups.The control group was orally administered olive oil,and the other 2 groups were orally given CsA 25 mg/kg and 50 mg/kg daily respectively.After 2 weeks.the liver samples were taken for ultrathin sections.The stereological method was used to quantify some hepatic ultrastructural parameters including volume density (Vv),surface density(Sv),numerical density(Nv) and specific surface(δ).Results CsA caused an increases of Vv and Sv and a decrease of δ in endoplasmic reticulum;a decreases of Vv and Nv in microbody;decreases of Vv in Golgi complex and glycogen inclusions;an increases of Vv and Nv in lipid inclusions.Conclusion CsA in a therapeutic dose could cause the change of hepatic ultrastructure of rat.

To explore the change in gene expressions of p38 mitogen-activated protein kinase (p38MAPK) and its upstream signal-regulated molecule (mkk3 and mkk6) in normal skin versus hypertrophic scars underlying its potentially biological significance. Total RNAs were isolated from 8 specimens of hypertrophic scars and 8 specimens of normal skin, then they were purified to mRNAs, and the gene expression of mkk3, mkk6 and p38MAPK was examined with reverse transcription-polymerase chain reaction (RT-PCR). The results showed that the intensities of gene expression of mkk6 and p38MAPK were weak in normal skins, while the expression of these 2 genes was significantly elevated in hypertrophic scars compared with normal skins (P0.05). In hypertrophic scar, the elevation of mkk6 and p38MAPK gene expression, which plays pilot roles in cell proliferation, may be one of the mechanisms controlling the formation of hypertrophic scars.

To investigate the effect of FGF-10 on the secretion of GM-CSF by adult keratinocytes in vitro and to understand the mechanisms involved in the stimulation of granulation tissue formation by FGF-10 during wound healing. Concentrations of FGF-10 used were 4, 16, 125 and 500 ng/ml. Cells were seeded in the amount of 2 500 cells/cm 2 or 5 000 cells/cm 2 in dishes in serum-free medium, and supernatants were collected at 24, 48 and 72 hours after culture. The amounts of GM-CSF in cell culture supernatants were determined using GM-CSF ELISA kits, and cell numbers were counted by haemocytometer. For cells seeded in low density (2 500 cells/cm 2), GM-CSF was not detected at 24 hours. At 48 hours, both in absolute concentrations and on a per-cell basis, the amounts of GM-CSF secreted in cultures with 125 and 500ng/ml FGF-10 were significantly higher than that in negative control (P0.05). At 48 hours, the keratinocytes in the middle area were confluent, and a number of cornified cells were observed, while the productions of GM-CSF in FGF-10 cultures were not higher than that in negative control. Objective To study the role of bcl 2 gene expression in human prostate. Methods The level of bcl 2 gene mRNA expression were investigated by in situ hybridization in 11 cases of fetal prostates, 27 cases of adult (specimens obtained from autopsy)and 60 cases of hyperplastic prostates. Results In fetal prostates, positive signals were detected in all epithelial cells of the prostate buds, while in adult and hyperplastic specimens, positive signals were predominantly concentrated to the basal cell layer of the epithelium, with little involvement of the secretory epithelial cells. The expression in hyperplastic prostates was significantly higher than in normal ones and that of the basal epithelial cells was significantly higher than the secretory ones in both the normal prostates and the hyperplastic ones. Conclusions Bcl 2 gene may play an important role in regulating the apoptosis of the basal cells while if plays a secondary role in the secretory cells of the prostate.

Objective To discuss the value of vacuum sealing drainage combined with continuous fluid irrigation in deep second degree burn.Methods 33 patients with deep second degree burns in burn department of the Third Affiliated Hospital of Qiqihar Medical College were selected as study object.The observation group used vacuum sealing drainage combined with continuous fluid irrigation,while the control group used the traditional method of treatment.The rate of wound healing,wound healing time and pain conditions were compared between the two groups of patients.Results After 3 days in the observation group,there was dissolved necrotic tissue which were drained into the negative pressure bottles,wound necrosis dropped off and dissolved when changing VSD dressings after seven days,a large number of newborn epithelial tissue appeared on the wound after 14 days of treatment.The healing rate after 7 days and 14 days,total healing time and wound pain showed statistically significant differences between two groups.Conclusions VSD combined with continuous fluid irrigation can effectively improve local microcirculation of burn wounds,increase local blood flow,keep the wound stay at a moist environment,accelerate the exudate drainage,which will help reduce the damage caused by local inflammation,promote epithelial cell regeneration from multiple perspectives,promote wound healing,and improve local pain symptoms and the quality of life of patients.

Objective To investigate the effects of regional myocardial stunning on systemic hemodynamics and myocardial oxygen balance Methods Thirty three open chest anesthetized dogs were subjected to 15min left anterior descending (LAD) coronary artery occlusion under normal temperature, the systemic hemodynamics, coronary blood flow (CBF), myocardial oxygen delivery(MDO 2) and consumption(MVO 2)were determined following reperfusion Results MAP decreased slightly during LAD occlusion, but returned to baseline following reperfusion CO, SV and LVSWI reduced rapidly after LAD occlusion, increased slightly 5min following reperfusion, followed by a more severe reduction , with the progressive increases of SVR and PVR during whole periods At the early stage of reperfusion (5min), CBF, MDO 2 and MVO 2 increased significantly, then decreased rapidly 10min following reperfusion Myocardial oxygen extraction rate remained stable during whole procedures Histological changes of ischemic area 30min following reperfusion showed interstitial and intracellular edema, mitochondrial vacuolization, and capillary stasis under electronic microscopy Conclusions Regional myocardial stunning may resulte in sinificant changes of systemic hemodynamics; at the early stage of reperfusion, oxygen consumption paradox occurres ,but MDO 2 matches MVO 2 10min following myocardial reperfusion
