Objective:To investigate the effect on uterine involution in caesarean section.Methods:1230 cases of primipara were divided into group A(cesarean section,n=650),group B(spontaneous delivery,n=580),measuring the level of uterine fundus,descending speed of uterine fundus,observing the duration of lochia,routinely screening of uterine involution.Results:There was significant t difference between the two groups with regard to the average decending speed of uterine fundus within 5 days after delivery,group B was superior to group A;After 42 days,The cases whose lochia disappeared were 61.8%and 92.4%in group A and group B in turn;The day 5,30 and 42 ultrasound measurement of uterine volum,showing significant difference(P

Objective:To inhibit GR expression in macrophage RAW264.7 cells of mouse by RNAi and Cre-loxP system.Methods:Two pairs of RNAi target sequence against GR gene were designed and inserted into pBS/U6-Neo vehicle.Recombinant plasmid constructed by Cre-loxP system and RNAi was identified by PCR and sequencing.The recombinant plasmid and Cre-ERT2 plasmid was transfected into the mouse macrophage RAW264.7 cells,in which GR expression was measured on mRNA and protein level respectively by RT-PCR and Western blot.Results:GR expression on mRNA and protein level was significantly inhibited by the two recombinant plasmid against GR gene.Conclusion:Combination using of RNAi and Cre-loxP system could inhabit GR expression in mouse macrophage RAW264.7 cells.
3.The clinical features and risk factors of children with post-infectious bronchiolitis obliterans treated with invasive mechanical ventilation because of severe pneumonia

Objective To summarize the clinical features and risk factors of children with post-infectious bronchiolitis obliterans (PBO), who were treated with invasive mechanical ventilation because of severe pneumonia, and to provide clinical reference for early diagnosis, early treatment and improvement of prognosis of pediatric patients with PBO. Methods The pediatric patients treated with invasive mechanical ventilation because of severe pneumonia, who were hospitalized in PICU of Tianjin Children's Hospital from October 2010 to December 2015 were included in this study. The pediatric patients who were diagnosed with PBO were used as the observation group. The general characteristics, clinical presentation, treatment time, treatment results, distribution of pathogens and relevant laboratory examination results were analyzed by using SAS 9.3 statistical software. Results A total of 73 pediatric patients were included in this study, including 49 male and 24 female, and their mean age was 8 months (P25-P75:4-12), the minimum age was 1 month and the maximum age was 36 months. They were divided into PBO group (n=30) and control group (n=43). It was found that wheezing (27 cases, 90.0%), persistent cough (22 cases, 73.3%) and exercise intolerance (22 cases, 73.3%) were the main clinical symptoms in PBO group. Results of physical examination showed wheeze (24 cases, 80.0%) and crackles (21 cases, 70.0%). Mosaic perfusion (25 cases, 83.3%) was the most prominent sign in lung CT scan. More than 2/3 of the pediatric patients were found moderate and severe pulmonary dysfunction. Pathogen detection rate was 50%in PBO group, and virus was prevalent (respiratory syncytial virus 4 cases, influenza virus 1 case, measles virus 2 cases and adenovirus 4 cases). The results of Logistic multivariate regression analysis showed that the risk factors included using corticosteroids at the beginning of disease (OR=8.515, 95%CI:1.948-37.224), prolonged mechanical ventilation (OR=1.116, 95%CI:1.031-1.327) and postponing clinic time (OR=1.114, 95%CI:1.010-1.227). Conclusion Once the severe pneumonia children, with the risk factors of using corticosteroids at the beginning of disease, prolonged invasive mechanical ventilation and postponing clinic time, are found the wheezing, persistent cough and exercise intolerance, and persisting wheeze and/or crackles, the PBO relevant examination should be executed as soon as possible to diagnosis and treatment.
4.The clinical significance of a new model with emergency triage information system combined with pre-registration during the emergency process

Objective To explore the clinical significance of a new model with emergency triage information system (ETIS) combined with pre-registration during the emergency process.Methods A total of 1 000 patients who applied the new model with ETIS combined with pre-registration from January to April 2016 were assigned as the observation group,while another 1 000 patients from January to April 2015 who applied the conventional model with triages were assigned as the control group.The used time of triage,used time of registration,collection conditions of vital signs,and accurate rate of triage were compared between these two groups.Meanwhile,mortality,rescue success rate,omission rate,excessive-detection rate,transferring surgical duration,as well as door-to-needle time (DNT) and percutaneous coronary intervention (PCI) time in red region,were compared between these two groups.Results Subjects in the observation group had notably shorter used time of triage and registration,prominently higher accurate rate of triage,markedly better collection conditions of vital signs (P < 0.01),obviously lower mortality,omission rate,excessive-detection rate,transferring surgical duration,DNT and PCI time (P < 0.05),and significantly higher rescue success rate (P < 0.01),compared with patients in the control group.Conclusions The application of the new model with ETIS combined with pre-registration has high clinical significance.It greatly optimizes the emergency process,significantly shortens patient's triage and registration time,promotes the accurate rate of triage,allows the rapid and complete collection of vital signs,and more importantly,it also improves the time for rescuing critical patients,tremendously raises the rescue success rate and reduces mortality.
5.Clinical Observation on Treatment of Women Climacteric Insomnia by Plum-Blossom Needle: A Report of 50 Cases

To observe the treatment of 50 female patients with climacteric insomnia by plum-blossom needle therapy. According to differentiation of symptoms and signs the head, back, belly and other location were tapping. Besides 50 patients were treated with Western medicine as a control group. The result showed the immediate and long-term effect in plum-blossom needle group was better than that in control group (P<0.01).
6.Signaling pathways of epithelial-mesenchymal transition in colon cancer

Epithelial-mesenchymal transition (EMT) plays a critical role in the carcinogenesis of colon cancer,and promotes the tumor invasion and metastasis.Studies have shown several signals are involved including transforming growth factor-β (TGF-β),Wnt/β-catein,Notch,nuclear factor-κB (NF-κB) and PI3K/Akt signal pathway.It is important to elucidate EMT-related signal pathway thereby providing new insights into possible therapeutic interventions of colon cancer.
7.Recent advances of thymic stromal lymphopoietin in respiratory tract infection

Thymic stromal lymphopoietin (TSLP) is an epithelial cell-derived cytokine. TSLP exerts a profound influence on the polarization of dendritic cells and T helper2 cytokine production. TSLP also directly promotes T-cell proliferation in response to T-cell receptor activation and Th2 cytokine production. TSLP affects different populations of cells,indicative of the broad role of TSLP in the regulation of respiratory tract infected and inflammatory processes.
8.Establishment of Mouse Models with Kidney-positive Deficiency with Antisense Oligonucleotide-loaded Bio-degradable Polylactic Acid Microspheres

OBJECTIVE:To investigate the feasibility of mouse models establishment with kidney-positive deficiency with anti-sense oligonucleotide-loaded biodegradable polylactic acid microspheres. METHODS:Three-week old mice were randomized into groups A,B,C,D and E,with 6 mice in each group,where the mice in group A were given subcutaneous injection 0.2 ml nor-mal saline,and those in groups B-E were given subcutaneous injection 0.1,0.2,0.4 and 0.8 mg each mice of antisense oligonucle-otide-loaded biodegradable polylactic acid microspheres. After the administration,the weights and activities of mice in all groups were observed;and one month later,their livers,adrenal glands and brains were collected to detect the expression of glucocorti-coid receptor (GR) by real-time quantitative polymerase chain reaction (RT-PCR). RESULTS:All mice except those in group A were found to have kidney-positive deficiency symptoms such as developmental delay,light weight,less activities and gathering to-gether. Compared to group A,other groups showed lower GR expressions in liver,adrenal gland and brain. There was statistically difference (P<0.05). CONCLUSIONS:Antisense oligonucleotide-loaded biodegradable polylactic acid microspheres can inhibit GR expression,and the mice whose GR expression is inhibited show symptoms of kidney-positive deficiency,which provides a re-ference for the establishment of mouse models with kidney-positive deficiency.
9.Effect of Rutaecarpine on the Expression of Angiotensin C in Thoracic Artery of Hypertensive Rats

OBJECTIVE:To explore the effect of rutaecarpine(Rut)on the expression of angiotensin C in the thoracic artery of hypertensive rats.METHODS:Two-kidney and one-clip goldblatt(2K1C)hypertensive model rats were enrolled in our study.The blood pressure of the model rats was measured by tail-clipping method.The model rats were assigned to receive losartan(20 mg?kg-1?d-1)or Rut(10,40 mg?kg-1?d-1)with their blood pressure was followed every week.The morphology of aorta was analyzed and the content of angiotensin C in the thoracic artery was detected by Western blotting.RESULTS:After treatment with Rut for 4 weeks,the systolic arterial pressure(SAP)in the treatment group decreased significantly;as compared with model group,the luminal diameter dilated significantly,the medium thickness of the artery decreased,angiotensin Ⅱ level in plasma and arteries decreased significantly,while the expression of angiotensin C increased significantly.CONCLUSION:Rut can effectively decrease blood pressure and improve arterial remodeling in model rats.The mechanism might be associated to the deactivation of angiotensin Ⅱ and the activation of kallikrein medicated by the enhanced expression of angiotensin C.
