Abstract Objective: To investigate the regulatory effect of PAF on early events in signal transduction during T-cell activation. Methods:T-cell activation was achieved by stimulation of human T cells with CD3 mAb or PMA/ionomycin in the presence or absence of PAF. T cell Pro-liferatilon was determined by 3 H-thymidine incorporation, IL-2 production was measured by MTT method, IL-2R(CD25)expression was evaluatedby flow cytometry and,PKC activity was assayed by the method described by Hauschildt. Results:PAF inhibited CD3 mAb-induced T-cell pro-liferation, IL-2 production and CD25 expression. AIthough it inhibited T-cell proliferation and IL-2 production induced by PMA/ionomycin, PAFfailed to inhibit IL-2 production induced by PMA/ionomycin.The translocation of PKC was also inhibited by PAF if T cells were activated byCD3mAb. Conclusion: PAF inhibits T-cell activation via its inhibitory effect on PKC activation. Its differential effect on IL-2 production suggeststhat PAF regulatory more early events in signal transduction such as the activation of phospholipase C ?-1.

AIM: To study the expression of type Ⅰtransforming growth factor ?(T?R-Ⅰ)in renal cortex in streptozotocin-induced type Ⅱ diabetic mellitus and the regulation of valsartan. METHODS: The rat models of type Ⅱ diabetic rats were made. At the end of the 20th weeks, the kidneys were taken out to measure the expression of T?R-ⅠmRNA by RT-PCR. RESULTS: The expression of T?R-ⅠmRNA in diabetic rats without any therapy ( 0.72? 0.14) was higher than that in control ( 0.26? 0.12) (P

Objective To study the quality control of urinary arsenic detection .Methods Quality control before ,during and after analysis were performed to avoid the random error and control the system errors .Results The low detection value of self-made quality control material of urinary arsenic and coefficient of variation (CV) were (0 .644 ± 0 .024) mg/L and 3 .72% ,respectively , while the high detection value and CV were (1 .353 ± 0 .042) mg/L and 3 .14% ,respectively .Conclusion Quality control before , during and after analysis may effectively reduce the incidence of random error and control the system error of urinary arsenic detec-tion and ensure the accuracy of test results .

Purpose To investigate the clinicopathologic changes of the lung epithelial tumor with epithelioid granuloma. Methods E-leven cases of lung malignant tumor with epithelial epithelioid granuloma reaction were studied according to the histological changes of lung epithelial tumor, immunophenotype, acid-fast staining, and tuberculosis PCR test results combined with follow-up data. Results The mean age was 62. 3 years, and male to female ratio was 10∶ 1. The main clinical manifestations displayed a cough, chest pain, fever and hemoptysis or bloody sputum. Nine cases were malignant tumors, tuberculosis or inflammatory lesions was 1 case each in pre-vious surgery. The tumor with epithelioid granuloma formation of the 11 cases displayed in the same site ( mixed) . The histopathologic changes of the lesions showed a epithelioid or typical tuberculous granulomas formation, including 4 cases of adenocarcinoma, 5 cases of squamous cell carcinoma and 2 cases of neuroendocrine tumors. The acid-fast bacilli staining was positive in 3 cases;6 cases were positive for PCR detection of mycobacterium tuberculosis. 5 patients died within 2 years due to various causes. Conclusions The lung epithelial tumors with epithelioid granuloma is more commonly found in the middle-aged, The signs and symptoms of the patients are similar to primary epithelial malignant tumors of the lung . The biological behavior was progressive with poor prognosis. The diagnosis of the lesions depends on pathological examination. It is very important to pay more attention to the pathological examination, especially in lung puncture samples, and the differential diagnosis for a definate diagnosis and treatment.

Objective To evaluate the effect and safety of catheter constant drainage combined with pericardial perfusion lentinan in treatment of patients with malignant pericardial effusion caused by lung cancer. Methods One hundred and eighteen patients with malignant pericardial effusion caused by lung cancer were divided into 2 groups by table of random digit, the treatment group (56 cases) was treated by pericardial perfusion lentinan after catheter constant drainage by central venous catheter, and the control group (62 cases) was treated by pericardial perfusion cisplatin after catheter constant drainage by central venous catheter. The therapeutic effect and adverse reaction were contrasted between the two groups. Results There was no significant difference in effective rate between treatment group and control group [89.3%(50/56) vs.83.9% (52/62),x2 =0.736,P =0.391].The adverse reaction rate of treatment group was significantly lower than that of control group [8.9% (5/56) vs.24.2% (15/62),x2 =4.871,P =0.027].There was no significant difference in recurrence rate between treatment group and control group [14.3%(8/56) vs.11.3%(7/62),x2 =0.004,P =0.948].Conclusions Catheter constant drainage combined with pericardial perfusion lentinan for malignant pericardial effusion caused by lung cancer is safety and effective, and can be widely applied to clinical application.

Aim To investigate metabolomic profiles of acute myocardial ischemia (AMI) in rat plasma and explore the intervention effects and its mechanism of leonurine using a metabolomics approach based on nuclear magnetic resonance (NMR).Methods The plasma metabolomic characteristics in rats of sham group,AMI model group, and leonurine-treated group were detected by 1H NMR,and the different metabolites between AMI group and sham group or leonurine-treated group were analyzed by pattern recognition and multivariate data analysis. Results Orthogonal partial least squares discriminant analysis (OPLS-DA) demonstrated that six metabolites related to AMI were screened out, including alanine,lysine,glycine,creatine,N-acetyl glycoprotein, and O-acetyl glycoprotein and all their levels were elevated in AMI group compared to sham group.Treatment of leonurine decreased the levels of alanine, lysine, and glycine, and increased the levels of choline, phosphocholine, and scyllo-inositol compared with the model group. Conclusions Leonurine can improve amino acid metabolism disorder under AMI conditions and enhance the function of choline and inositol pathway, which may explain its cardioprotective effect. The developed metabolomics approach in this study is a powerful tool for the investigation of the cardioprotective effect of leonurine and provide a new insight to understand its pharmacological mechanism.
