1.Research on Reprimo,hMLH1 Gene Methylation in Early Diagnosis Value of Gastric Cancer

Objective To investigate the Reprimo and hMLH1 gene promoter methylation detection value in the diagnosis of early gastric cancer.Methods Chose patints in Shaanxi Provincial People’s Hospital from September 2013 to April 2014,50 cases of patients with chronic atrophicgastritis with intestinal metaplasia,50 cases of patients with gastricmucosal atypical hyperplasia,50 patients with gastric cancer,endoscopic gastric biopsy samples,and 30 cases of normal gastric mucosa biopsy tissues as control group.Analysis abnormal expression in Reprimo gene and hMLH1 genes promoter methylation,compared the differences.between groups of patients.Results The patients with gastric mucosatissues Reprimo and hMLH1 genes promoter methylation positive rate was significantly higher than that of normal group,the difference wasstatistically signifi-cant (P<0.05).Reprimo gene promoter methylation were:patients with chronic atrophic gastritis with intestinal metaplasia 28% (χ2 =10.18,P < 0.05).Patients with gastric mucosalatypical hyperplasia 56% (χ2 = 25.84,P < 0.05)and patients with gastric cancer 62% (χ2 = 30.36,P < 0.05).hMLH1 gene promoter methylation were:patients with chronic atrophic gastritis with intestinal metaplasia 20% (χ2 =4.39,P <0.05),patients with gastric mucosal atypical hyperplasia 44% (χ2 =15.13,P <0.05)and patients with gastric cancer 48% (χ2 = 17.41,P <0.05),high specificity of detection.Conclusion Reprimo and hMLH1 gene’s detect value in the diagnosis of early gastric cancer is very high,high specificity,it is an effec-tive way of diagnosis,treatment in clinical diagnosis of patients with broad prospects.
2.Effect of transplanted mesenchymal stem cells transfected with adenoviral ventor mediated HGF gene on snrvival volume of grain fat grafts

ObjectiveTo investigate the effect of mesenchymal stem cells (MSCs) transfected with hepatic growth factor (HGF) gene on the survival volume of free grain fat grafts.Methods MSCs of male Wistar rats were transfected with Ad-GFP or Ad-HGF.The transfection infectivity of Ad-GFP to MSCs,the expression of HGF were measured using ELISA assay.150 rats were randomly divided into 5 groups:control group (group A),MSCs group (group B),Ad HGF group (group C),MSCs transplanted with Ad-GFP group (group D),and MSCs transplanted with Ad-HGF (MSCsHGF) group (group E).Then,the same volume of frain fat graft,mixed with DMEM LG andMSCs,Ad HGF,MSCs-GFP,MSCs HGF respectively,were transplanted to rats back,but control group were only mixed with DMEM-LG.Fat graft was obtained on days 3,5,7,14,28,and 60 after implantation.The volume of fat graft was measured by messcylinder,and the expression of HGF and CD34 in transplanted fat tissue were detected by immunohistochemistry.ResultsThe transfection infectivity of Ad-GFP to MSCs was 89.6 % at 100 MOI,the expression of HGF in MSCs culture medium reached to the level after being transfected with Ad-HGF for 48 h.Compared with other 4 groups,at days 3,5,7,and 14 post-transplantation,the expression of HGF in E group transplanted fat of group E had statistics significance (P<0.05).The persentage of survival volume of fat graft in group E was significantly higher than that of other group ( P<0.05) at days 28,and 60 post transplantation.ConclusionsMSCs transplanted with Ad-HGF could secrete HGF and increase the survival volume of fat grafts.
3.Intracellular calcium and growth suppression of human breast cancer cells exposed by high intensity ultrasound

Objective Study on the intracellular calcium and growth suppression of human breast cancer cells exposed by high intensity ultrasound (HIU). Methods Exposed human breast cancer cells MDA-MB-231 and MCF-7 in vitro with HIU (50 W/cm2). Examined the intracellular calcium from exposed cells with Fura-2 fluorescence probe. The cell viabilities were measured by MTT assay. The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. Results The lever of intracellular calcium went up in pace with exposed time of HIU, MCF-7 cells were (572±20.1), (670±18.9), (815± 16.3) nmol/L (F = 663.65, P<0.001), MDA-MB-231 cell was (582±16.3), (687±19.7), (843± 14.8) nmol/L (F = 863.06, P<0.001), and the distribution of cell cycle waved to G0-G1, the ratios of G0-G1 in MCF-7 and MDA-MB-231 were (60.5±5.5)%, (66.3±7.0)%, (74.5±8.2)% (F=8.17, P = 0.002) and (58.5± 6.3) %, (66.1±6.3)%, (71.2±7.9) % (F=7.51, F= 0.003). Apoptotic rate upgraded gradually, the apoptotic rates of MCF-7 and MDA-MB-231 were (7.3±1.7)%, (13.2±3.5) %, (19.3±3.7)% (F= 18.73, P<0.001) and (6.3±1.8)%, (11.4±2.31)%, (16.4±3.3)% (F = 19.26, P<0.001). Under MTT assay, the rate of cell growth suppression increased significantly, the rates of cell growth suppression in MCF-7 and MDA-MB-231 were (9.2±2.2) %, (24.3±3.9)%, (48.6±5.5)%(F=117.16, P <0.001) and (9.0±1.7)%, (22.3±3.5) %, (416± 6.4)% (F =71.25, P<0.001). Conclusion HIU enhanced the intracellular calcium of human breast cancer cells within given time and promoted the distribution of cell cycle to G0-G1. The rate of cell apoptosis and the cell's death rate increased evidently.
4.Exploration and research on artificial intelligence courses in medical colleges—taking the Army Medical University as an example

The implementation of artificial intelligence courses in medical colleges is of great significance to the cross-integration of artificial intelligence and medical fields. However, there are problems such as the lack of basic knowledge of artificial intelligence, the lack of closely related teaching materials, and the difficulty in copying the teaching mode of medical courses. Based on the above problems, through the establishment of artificial intelligence course in our school, the teaching implementation process and teaching effects have been summarized and analyzed, aiming at exploring the artificial intelligence curriculum system which is suitable for medical undergraduate students, so as to improve the students' ability of using artificial intelligence technology for medical project practice, promote the combination of medical and engineering, and provide reference for the teaching reform of undergraduate artificial intelligence courses in medical colleges at home and abroad.
5.Study of influential factors on mental health of occupational females

Objective To study the mental health condition of occupational female and influencing factors of mental health. Methods 827 occupational females in Dongguan city were investigated using the questionnaires, the contents included the basic personal information, mental health condition and influencing factors of mental health.Results The results showed that there existed higher ratio of mental disorder in people of age over 50 years, post-graduate educational background, monthly income levels of 4001 to 5000 yuan and divorced occupational females.Moreover, the influential factors on mental health of occupational females were correlated with age, culture degree,family population and society. Conclusion We should pay more attention to occupational females and take effective measure to relieve their mental stress. This is the demands of woman as well as society.
6.Inhibition of α-crystallin on proliferation of rat retinal microglia following optic nerve injury

Objective To investigate the effect of α-crystallin on the proliferation of rat retinal microglia after optic nerve injury. Methods The effect of α-crystallin on number and proliferation of microglia were analyzed by MTT assay.After the rat model with optic nerve injury was established,α-crystallin was iniected into vitreous cavity and the microglia cell number were counted and compared by retinal fiat counting and immunofluorescence labeling in different groups. Results The proliferation and activation of microglia cells could be stimulated by LPS at 10-6g/L to 10-2g/L.α-crystallin at 10-4g/L and 10-6g/L could inhibit proliferation and activation of microglia cells.Compared to BSA iniection group,α-crystallin could inhibit more significantly the number of microglia cells 1-3 weeks after injury (P<0.05). Conclusions α-crystallin can inhibit proliferation and activation of retinal microglia and alleviate overphagocytosis and secondary damage of retinal microglia to retinal ganglion cells(RGCs),which may be another mechanism that α-crystallin contributes to indirect protection of RGCs.
7.Selection and Identification of the Biological Characteristics of a Cold-adapted Genotype G1P8 ZTR-68 Rotavirus by Serial Cold-adapted Passaging.

We wished to select a cold-adapted genotype G1P[8] ZTR-68 rotavirus (China southwest strain) in MA104 cells for possible use as a live vaccine. ZTR-68 was recovered originally from children with diarrhea. The virus was cultivated at 37 degrees C at the first passage. Then, the cultivation temperature was decreased stepwise by 3 degrees C per eight passages. In total, the virus was passaged 32 times, and cultivation was terminated at 28 degrees C. Biological characteristics of the virus were analyzed during serial passages. There was no difference between the migration patterns of genomic dsRNA segments according to polyacrylamide gel electrophoresis of original and cold-adapted viruses. Infectious and red cell-agglutination titers of cold-adapted virus were lower than those of the parent virus. Also, the virus formed small-size plaques with irregular shapes at 31 degrees C and 28 degrees C. These results suggested that a genetically stable attenuated virus can be obtained through serial cold-adapted passages. Thus, an alternative strategy is provided by cold-adaption for development of attenuated live rotavirus vaccines.

8.Evaluation of conventional ultrasound in the detection of subclinical hemorrhage of hemophilia joint

Objective:To explore the application value of conventional ultrasound(US) in detecting occult bleeding lesions in joints of patients with hemophilia.Methods:Twenty-seven children with severe hemophilia A who received factor Ⅷ prophylactic treatment from March 2017 to April 2018 in Nanfang Hospital, Southern Medical University were enrolled. Clinical data were collected from routine practice accompanied with US examination, hemophilia joint health score (HJHS) 2.1 assessment and the history of hemarthrosis of both ankles, elbows and knees at baseline and at 1 year follow-up.Results:A total of 157 joints of 27 patients were followed up for one year. During the treatments, 74 joints were found without history of hemarthrosis, among which the total US score of 14(14/74, 18.9%) joints increased, and HJHS 2.1 score of 11(11/74, 14.9%) joints developed. There was no significant change in the HJHS 2.1 score of 108 joints, among which 25 joints (25/108, 23.1%) had an increase in the total US score.However, there was no significant change in the total US score for 93 joints, of which 9(9/93, 9.7%) had an increase in HJHS 2.1 score.Conclusions:US examination can sensitively detect hemophilia arthropathy in joints without history of hemarthrosis and without obvious progression of arthropathy by HJHS 2.1 examination, and provide clinicians with more accurate joint information.
9.The polymorphism of HLA-DRB1 and -DQB1 genes in Bai ethnic group in Yunnan province

Objective:To investigate polymorphism of human leukocyte antigen (HLA)-DRB1 and -DQB1 genes in Bai ethnic group in Dali,Yunnan province.Methods:Polymerase chain reaction-sequence specific primers (PCR-SSP) were used to determine HLA-DRB1 and -DQB1 alleles in 124 unrelated healthy Bai ethnic individuals living in Eryuan County of the Dali Bai autonomous prefecture,Yunnan province.Results:Among all the 21 DRB1 alleles and 15 DQB1 alleles were identified,the predominant alleles were DRB1*1202(26.61%),DRB1*0901(13.89%) and DRB1*0803(9.92%) on DRB1 locus and DQB1*0301(31.45%),DQB1*0601(10.08%),DQB1*0401(8.06%)and DQB1*0502(8.06%)on DQB1 locus.The most common haplotypes were DRB1*1202-DQB1*0301(20.08%)and DRB1*0803-DQB1*0601(7.19%).Conclusion:The phylogenetic tree constructed according to the HLA-DRB1,-DQB1 allele frequencies of Bais with those of other 10 populations suggests that the Bai ethnic group belongs to the southern group of China,but it keeps genetic distance from others and the HLA genes exhibits a unique profile.This study would provide HLA polymorphism information of Bai for the future investigation on the disease related to the genetic polymorphism.
10.Progress in magnetic resonance imaging study of anhedonia and suicidal behavior in depression

Many studies have found a correlation between suicidal behavior (SB) and anhedonia, the main symptom of depression, in terms of both psychological and neurophysiological findings. The purpose of this review is to find the relationship between the two neuroimaging mechanisms, and to provide help for the future study of how the brain imaging changes can promote the mechanism of SB in depression patients with anhedonia symptoms. This review also emphasizes the necessity of intervention for the symptoms of anhedonia when preventing depression from committing suicide. The latest research results were reviewed about anhedonia in depression and magnetic resonance imaging of SB.The results showed that the default network, insula, lateral orbitofrontal gyrus, anterior cingulate gyrus, ventral striatum gyrus, ventral lateral and dorsolateral prefrontal gyrus, thalamus and habenula nucleus were dysfunction in depression with state anhedonia symptoms, affecting SB in terms of mood, execution, reward and aversion processing, especially the low lethal SB.
