Objective To explore the mechanism of protection role of ambroxol against acute lung injury (ALI)by studying the change of cytokine mRNA expression during the course of ALI.Method The study was composed of experiments both in vivo and in vitro. (1)Experiments in vivo were as follows.Twenty-four sprague-Dawley rats were randomly divide into 3 groups, namely, control group(n=8),acute lung injury group(LPS group,n=8)and ambroxol group(LPS+A group,n=8).The rat model of acute lung injury was induced by intraperitoneal injection of 10 mmol/L lipoopolysaccharide(LPS).The pathological alteration of lung tissue and arterial partial pressure of oxygen(PaO2)were observed.Expressions of TNF-α,IL-10 and IL-24 mRNA were determined by using RT-PCR assay. (2)Experiments in vitro were the followings.Alveolar macrophage cells were collected and divided into 3 groups as above mentioned.Cells were treated with normal saline, with LPS,and with LPS plus ambroxolin dose of 180 μmol/L at 0,6,12 and 24 hours after exposure of LPS,respectively,in 3 groups as above stated. The expressions of TNF-α,IL-10 and IL-24 mRNA were also determine by using RT-PCR.Results The pathological changes of could be parially ameliorated by giving ambroxol.Massive hemorrhage along with vascular edema and infiltration occurred in the lungs of ALT rats. The pathological alterations in ALI rats treated with ambroxol were less severe. The expressions of TNF-α,IL-10 and IL-24 mRNA were dramatically increased in ALI rats, and were partially attenuated after treatment of ambroxol.Macrophages oxposed to LPS for 6 hours showed dramatical increase in expressions of TNF-α,IL-10 and IL-24mRNA those remained at high levels afterwards. The expressions of TNF-α,IL-10 and IL-24 mRNA in macrophages after exposure to both LPS and ambroxol were increased less than those exposed to LPS alone. Conclusions Ambroxol can partially ameliorate the expressions of TNF-α,IL-10 and IL-24 mRNA in alveolar macrophage induced by LPS.

Objective To evaluate the proliferation and invasion potential of human colon cancer SW480 cells transfected by AP-2α gene in vitro. Methods pcDNA3.1 (+)-AP-2α was created by cloning AP-2α eDNA into the EcoRI site of poDNA3.1 (+). LipofectamineTM 2000 Reagent was used to mediate the transfection of pcDNA3.1(+)-AP-2α and pcDNA3.1 (+), and normal SW480 celia were cultured as a negative control. The mRNA and protein level of AP-2α in the cells of each group were detected 48 h after being transfected with plasmids above by RT-PCR and Western blotting analysis. The cell proliferation and invasion potential were examined by colony formation assay and Transwell invasion assay respectively. Results Lack of AP-2α protein expression in SW480 cells was verified by western blot analysis. After being transfected with AP-2α gene, the mRNA amount and protein expression increased dramatically, while the colony formation efficiency decreased(P <0.05), the cell proliferation in soft agar was inhibited, and the ability of its invasion dropped off(P <0.05) in vitro. Conclusion AP-2α gene suppresses the proliferation and invasion potential of human colon cancer SW480 cells in vitro.

Objective To investigate whether prostaglandin E2(PGE2) can promote the ability of adhesion.migration and invasion of colorectal cancer cells SW480 and its mechanism. Methods Extrinsic source PGE2 and the antagonist of EP1 SC19220 were added to the culture media. MTr assay was used to identify the adhesion ability of SW480 cells. Migration ability was tested by transwell plate. The invasion ability was tested bv ECM gel coated transweU plate. RT-PCR and western blotting were used to detect the mRNA and protein level of vascular endothelial growth factor (VEGF). Results The adhesion.invasion and migration ratio of SW480 ceils were all increased significantly after treated with PGEh the A values of adhesion ceils increased from 0.207±0.009 to 0.417±0.088. Migration cells increased from 6.33±0.33 to 43.33±0.88.invasion cells increased from 3.67±0.34 to 26.33±0.89(P<0.05).The adhesion.migration and invasion cells of PGE2+SC 19220 group decreased significantly compared to PGE2 group. The A values of adhesion cells decreased from 0.417±0.088 to 0.140±0.006. Migration cells decreased from 43.33±0.88 to 28.00±0.58.invasion cells decreased from 26.33±0.89 to 5.67±0.33 (P<0.05).The results of RT-PCR and western blotting showed that the expression of VEGF mRNA and protein increased in a dose dependent manner after PGE2 treatment. Conclusion The ability of adhesion. Migration and invasion of SW480increased after PGE2 was added to the culture media. It may be related to the upregulation of VEGF.

Objective The expression and impaired function of ion channels might be one of the pathophysiological mecha -nisms responsible for diarrhea in inflammatory bowel disease ( IBD) .Proper animal model is the key to explore detailed pathophysiolog-ical process. The purpose of this study was to build a rat model of acute colitis induced by dextran sodium sulfate (DSS) in C57BL/6 mice and evaluate diarrhea-associated clinical , histological , pathological parameters and expressions of ion channel protein . Methods C57BL/6J mice of model group were treated with 4%DSS solution for 7 days to induce acute colitis.Mice body weight, stool moisture, stool consistency and the degree of hematochezia were recorded .The histopathological changes of mice colon specimens were observed visually and microcosmically, and the ion channel SLC26A3 protein was detected by Western Blot . Results All experimental mice survived. In the experiment, compared with control group , bloody diarrhea and weight lose occurred in model group , along with increased stool moisture ([73.30 ±8.31]% after experiment vs [44.32 ±6.42]% before experiment, P=0.004), and rapidly in-creased disease activity index (DAI) of acute colitis ([3.50 ±0.87] after experiment vs [1.0 ±0.00] before experiment, P=0.000).At the end of this experiment , compared with control group , the model group resulted in higher colonic damage score and pathological inflammation score (P=0.00, P=0.002), significantly shortened co-lon (P=0.00) and decreased expression of SLC26A3. Conclusion The intestinal mucosal injury and phenotypic features of 4%DSS-induced acute colitis are very similar to those of human ulcerative colitis .Impaired expression of intestinal ion transporter SLC26 A3 coexists with diarrhea in model group mice , and this model can support the research on mechanism of functional changes of ion channels in inflammatory diarrhea .

Journal of Clinical Hepatology 2020;36(9):2005-2009
ObjectiveTo investigate the effect of artificial liver support therapy on the short-term (28- and 90-day) mortality rate of patients with liver failure in the plateau stage through a stratified analysis based on Model for End-Stage Liver Disease (MELD) score. MethodsA retrospective analysis was performed for 187 patients with liver failure who were admitted to Nanfang Hospital, Southern Medical University, from January 2015 to April 2019, with 73 patients in the artificial liver group and 114 in the non-artificial liver group. The stratified analysis based on MELD score in the plateau stage was performed to investigate the differences in 28- and 90-day mortality rates, hospital costs and length of hospital stay of surviving patients, and incidence rate of adverse reactions of artificial liver support therapy between the two groups. The t-test was used for comparison of continuous data between the two groups, and the chi-square test or the Fisher’s exact test was used for comparison of categorical data between the two groups. ResultsCompared with the non-artificial liver group, the artificial liver group had a significant reduction in the 28-day mortality rate of the patients with an MELD score of 30-39 (5.9% vs 39.6%, P<0.001) or those with an MELD score of 40 (25.0% vs 72.7%, P<0.05). Compared with the non-artificial liver group, the artificial liver group had a significant reduction in the 90-day mortality rate of the patients with an MELD score of 30-39 (23.5% vs 623%, P<0.001). Artificial liver support therapy did not significantly shorten the mean hospital stay of the surviving patients (P>0.05) and had no significant influence on the total hospital costs of the surviving patients within 90 days (P>0.05). The incidence rate of adverse reactions related to artificial liver support therapy was 29.1%, but the symptoms were mild and were relieved after symptomatic treatment. ConclusionPatients with an MELD score of <30 in the plateau stage tend to have low 28- and 90-day mortality rates, and artificial liver support therapy can be reasonably selected according to the patient’s economic conditions and willingness. Artificial liver support therapy is recommended for patients with an MELD score of 30-39 in the plateau stage if there is no obvious contraindication. For patients with an MELD score of 40 in the plateau stage, artificial liver support therapy is recommended within 28 days if there is no obvious contraindication, and liver transplantation is recommended as soon as possible. Artificial liver support therapy has no significant influence on the total hospital costs and mean hospital stay of the surviving patients within 90 days and does not increase the economic burden of patients.
