1.Expression and clinical application of the mRNA of PI3K and Akt mRNA in leukemia

Objective To study the expression of PI3K and Akt mRNA in acute leukemia(AL) and chronic myelocytic leukemia (CML) and try to find out a relationship between the prognostic significance of acute leukemia and the expression level of PI3K and Akt. Methods The samples were collected from 63 ALpatients, 7 relapsed AL patients, 14 CML patients and 10 AL patients in complete remission(CR) patients, 11samples of normal controls(NC). The expression of PI3K, Akt mRNA were measured by semi-quantity reverse transcription polymerase chain reaction(RT-PCR). Results The expression of PI3K mRNA in AL and relapse group were significantly higher than that in normal control. In CML, the P13K mRNA expression level was higher than that in NC and CR. The similar results of the expression of Akt mRNA were observed in CML, NC and CR. At the same time, the expression level of Akt mRNA in relapse group was significantly higher than that in NC. The complete remission rate in PI3K (+) was lower than that in PI3K (-). Similar result was obtained in Akt (+) and Akt (-). Conclusion The PI3K/Akt pathway may contribute to the occurrence of leukemia. The patients with positive expression of the members of this pathway had a lower remission rate.

AIM: To investigate the effect of IL-10 on IL-1?-induced prostaglandin E 2(PGE 2) release and cyclooxygenase-2(COX-2) expression in human mesangial cells and to examine whether IL-10 has effect on the biological function of IL-1?.METHODS: The PGE 2 concentration in supernatants of HMC was measured by radioimmunoassay. The COX-2 mRNA and protein were measured by RT-PCR and Western blot, respectively. RESULTS: PGE 2 and COX-2 were significantly increased after treatment with IL-1?( P0.05 , respectively), while it inhibited IL-1?-elicited PGE 2 production, as well as COX-2 mRNA and protein expression in a concentration-dependent fashion. CONCLUSIONS: These results indicated that IL-10 depressed the IL-1?-induced release of PGE 2 and expression of COX-2. These data suggested that IL-10 could exert anti-inflammatory actions at several levels, not only by inhibiting the production of pro-inflammatory cytokines but also by suppressing their biological function.
4.The blood and chest fluid expression levels and significance of Th17 cell and IL-17 in patients with tuberculous pleural effusion

Objective To investigate the blood and chest fluid level of Th17 cell and IL-17 in patients with tuberculous pleural effusion and its pathological role.Methods Flow cytometry and enzyme linked immunosorbent assay (ELISA) were used to measure the blood and chest fluid level of Th17 cell and IL-17 from 30 patients with tuberculous pleural effusion,20 patients without tuberculous pleural effusion,and 20 healthy persons.Results The blood level of Th17 cell and IL-17 wwere higher in tuberculous pleural effusion than in the other two groups(P <0.05).The chest fluid level of Th17 cell and IL-17 in patients with tuberculous pleural effusion were significantly higher than those in patients without tuberculous pleural effusion (P < 0.05 ).The chest fluid level of Th17 cell and IL-17 in patients with and without tuberculous pleural effusion were significantly higher than that of the blood serum level.After treatment for 1,3,7 and 14 days,tbe blood serum and chest fluid level of Th17 cell and IL-17 were obviously lower.( P < 0.01 ).After treatment for 1 day,the blood level of Th17 cell was obviously lower than before treatment( P < 0.01 ).After treatment for 3 days,the chest fluid level of Th17 cell was obviously lower than before treatment( P <0.01 ).After treatment for 3 days,the blood serum level of IL-17 was obviously lower than before treatment (P < 0.01 ).After treatment for 7 days,the chest fluid level of IL-17 was obviously lower than before treatment ( P <0.01 ).Conclusion Th17 cell and IL-17 might play an important role in the pathogenesis of tuberculous pleural effusion and they were correlated with disease progression and the therapeutic effect.
5.Effects of endogenous sulfur dioxide on the oxidative stress induced by cobalt chloride in the rat pulmonary artery smooth muscle cells

Objective To investigate the effects of endogenous sulfur dioxide (SO2) on the oxidative stress induced by cobalt chloride (CoCl2) in the rat pulmonary artery smooth muscle cells (PASMCs).Methods Rat PASMCs were treated with 200 μ mol/L CoCl2 to mimic the hypoxia insult.Endogenous SO2 generating enzyme aspartate aminotransferase 1 (AAT1) expression was upregulated or downregulated (AAT1 sh) by transfection with lentivirus.Rat PASMCs were randomly divided into 8 groups:vehicle group,vehicle + CoCl2 group,AAT1 group,AAT1 + CoCl2 group,scramble group,scramble + SO2 group,AAT1 sh group and AAT1 sh + SO2 group.SO2 donor Na2 SO3/NaHSO3 at concentration of 100 μ mol/L were added in scramble + SO2 group and AAT1sh + SO2 group.The expressions of AAT1,superoxide dismutase 1 (SOD1) and SOD2 in PASMCs were detected by Western blot method.In situ SO2 content in PASMCs was detected by fluorescent probe.The superoxide anions in PASMCs were labeled by dihydroethidium (DHE) probe under fluorescent microscope.Results Compared with the vehicle group,the levels of SO2 and the expressions of AAT1 (0.221 ± 0.002 vs.0.446 ± 0.004),SOD1 (0.076 ± 0.028 vs.0.171 ± 0.019) and SOD2 (0.080 ± 0.031 vs.0.196 ± 0.018) significantly decreased (all P < 0.01),and superoxide anion increased in rat PASMCs of vehicle + CoCl2 group.Meanwhile,compared with vehicle + CoCl2 group,the levels of SO2 and the expressions of AAT1 (0.839 ± 0.056 vs.0.221 ± 0.002),SOD1 (0.177 ± 0.020 vs.0.076 ± 0.028) and SOD2 (0.195 ±0.018 vs.0.080-± 0.031) markedly increased (all P < 0.01),and superoxide anion decreased in rat PASMCs of AAT1 + CoCl2 group.On the contrary,compared with the scramble group,the levels of SO2 and the expressions of AAT1 (0.062 ±0.017 vs.0.354 ±0.034),SOD1 (0.054 ±0.029 vs.0.157 ±0.023) and SOD2(0.180 ±0.100 vs.0.586 ± 0.176)significantly decreased (all P < 0.01),and superoxide anion increased in rat PASMCs of AAT1sh group.Furthermore,compared with the AAT1 sh group,the levels of SO2 and the expressions of SOD1 (0.155 ± 0.022vs.0.054 ± 0.029) and SOD2 (0.578 ± 0.200 vs.0.180 ± 0.100) significantly increased (all P < 0.01),and superoxide anion decreased in rats PASMCs of AAT1sh + SO2 group.Conclusion Endogenous SO2/AAT1 inhibits CoCl2-induced oxidative stress in rat PASMCs.

Objective To investigate the breakfast behaviors and nutritional quality among primary and secondary students,and to provide scientific evidence for developing intervention strategies.Method Random three-stage clustering sampling method was employed in the study.A questionnaire survey was conducted among 9194 primary and secondary students aged 6-17y,from seven cities in China.Results The rates of primary and secondary students having breakfast everyday were 97.0%,92.4%,and 93.4%,Most of the students had their breakfast within 30 min after getting up,the rates being 75.4%,74.7%,and 68.6% respectively.Mothers primarily cook breakfast for the students,the rates being 53.1%,49.9%,and 49.7% respectively.In most cases,they had breakfast at home,the rates being 71.8%,67.0%,and 63.8% respectively.The rates of students having poor quality breakfast were above 80%.Conclusion Primary and secondary students are one of the focal target people for preventing unhealthy dietary behaviors due to shortcomings of eating breakfast.It is necessary to improve the nutritional quality of breakfast for better health of students.
7.The antagonistic effect and mechanism of N-acetylcysteine on acrylamide-induced hepatic and renal toxicity.

OBJECTIVEThe aim of this study is to investigate hepatic and renal toxicity of acrylamide (ACR) , the antagonistic effect and possible mechanism of N-acetylcysteine (NAC) on the toxicity.
METHODSForty female SD rats were randomly divided into four groups. All the rats were administrated by intraperitoneal(i.p.) injection and 1.5 hours later by gavage. The control group was administrated with 0.9% NaCl by i.p. injection and gavaged with 0.9% NaCl. The NAC group was administrated with 200 mg/kg NAC by injection and gavaged with 0.9% NaCl. The ACR group was administrated with 0.9% NaCl by injection and gavaged with 40 mg/kg ACR. The combined treatment group was administrated with 200 mg/kg NAC by i.p. injection and gavaged with 40 mg/kg ACR. The rats were administrated once a day for 2 weeks. After 24 hours of the last administration, the rats were decapitated. The blood was collected, the liver and kidney were separated. The body weight, organ coefficient and serum biochemical parameters were measured, and the pathological changes of the tissues were examined with a microscope. Then the expression of NF-κB p65, IκB-α and COX-2 were detected by Western blot.
RESULTSFrom the second day to the end of the exposure, the body weight of rats in the ACR group was statistically lower than that in the control group (P<0.05) . Compared with the combined treatment group, the body weight in the ACR group statistically decreased in the second and third days (P < 0.05) . The liver and kidney organ coefficients in the ACR group were (4.159%±.371%) and (0.764%±0.068%) respectively, which increased statistically when compared with the control group (P < 0.05) . The contents of ALT, AST and Cr in the serum in the ACR group were (77.370±16.397) U/L、(379.410±57.817) U/L and (77.812±6.391) μmol/L respectively, which were not significantly different with those in the control group and the combined treatment group (P>0.05) . The content of BUN in the serum in the ACR group was (7.005±1.009) mmol/L, which was statistically higher than that in the control group (P<0.05) . Histopathology results showed unclear boundary and nucleus pyknosis in hepatocytes, loose and disordered structures of hepatic cords in the ACR group, but no obvious pathology changes were observed in the kidneys of each group. In the Western blot results, the expression of nuclear NF-κB p65 and COX-2 in the liver in the ACR group was statistically higher than that in the control group and the combined treatment group (P<0.05) , and the expression of IκB-α in the liver in the ACR group statistically decreased compared with the control group and the combined treatment group (P<0.05) . The expression of total NF-κB p65 in the liver in the ACR group was statistically higher than that in the control group (P<0.05) .
CONCLUSIONUnder the conditions of this experiment, ACR may induce hepatic toxicity through the activation of NF-κB signaling pathway, and NAC could antagonize the hepatic toxicity of ACR by inhibiting the NF-κB signaling pathway, whereas the toxic effect of ACR on kidney needs to be further studied.

8.Effects of egg and milk supplementation on growth and development among children in poor rural area.

OBJECTIVETo evaluate the effects of egg and milk supplementation on growth and development and body composition among children in poor rural area in Tianyang County of Guangxi province.
METHODSTotal four schools were randomly selected from four towns in Tianyang County of Guangxi province as intervention group in April, 2013. The intervention measures included that these students were given salty egg (net weight: 50 g) and ultra-high-temperature-sterilization school milk (net weight: 200 g) every school day and these schools were equipped with standard kitchens. Another four schools of familiar socio-economic level, teaching quality and size from the same town were randomly chosen as control group and none of the intervention measures were implemented. About 25 students were randomly selected and stratified by grades from grade one to grade five. The height, weight, and body composition of all students were measured in April, 2013 and one year after the intervention. A total of 978 students were measured at baseline from age 6 to 13, 552 students as intervention group and 426 as control group. t-test was used to compare the differences between groups and multivariate unconditional logistic regression was used to analyze the factors of malnutrition.
RESULTSAfter one year intervention, 892 students were measured randomly, with 515 students in intervention group and 377 in control one. The average weight of boys in intervention group increased (3.6 ± 1.7) kg compared with baseline. It was significantly higher than that of control group ((2.9 ± 1.5) kg) (t = 4.40, P < 0.001). The boy's lean body mass of intervention group increased (2.6 ± 1.4) kg, higher than the control group ((2.0 ± 1.2) kg) (t = 3.95, P < 0.001). The decrease of malnutrition rate of intervention schools (11.8%) was significantly higher than that of the control schools (4.7%, χ² = 16.90, P < 0.001), and the odds ratio was 0.37 (95% CI: 0.23-0.59). The risk difference of overweight and obesity was not statistically significant between the two groups (OR = 1.68, 95% CI: 0.57-4.94).
CONCLUSIONAfter supplementing milk and egg, the nutritional status of the poor rural pupils was improved.

9. Anemia status and related factors among left-behind children in poverty-stricken rural areas, in China, 2016

Objective:
To explore the prevalence of anemia and related factors among left-behind children in poverty-stricken rural areas in China, to provide basic information for anemia prevention and treatment for children.
Methods:
Data from the National Nutrition Improvement Program for Rural Compulsory Education Students in 2016 were used to express the prevalence of anemia among left-behind children. By using the multistage stratified random cluster sampling method, left-behind children of the 6-17-years-old in poverty-stricken rural areas were recruited in this study. Blood hemoglobin concentration of these students was determined by cyanmethemoglobin method. Anemia was judged by the WHO recommended standard and combined with the sea level elevation correction standard. Frequencies of food intake were collected through questionnaires. Data was then available for analysis including the level of hemoglobin and anemia rates. Relationship between anemia and food intakes as well as other influential factors was also analyzed.

10.Height changes among students under the National Nutrition Improvement Program for Rural Compulsory Education Students during 2012-2017

Objective:
To analyze the height changes among children who had received subsidies of the National Nutrition Improvement Program for Rural Compulsory Education Students (NNIPRCES) by comparing data from the year of 2012 and 2017.
Methods:
Among 699 counties where NNIPRCES was implemented, at least 10% of elementary schools and primary middle schools with different food supply modes (school canteen meals, company meals, and mix meals) in the county were selected randomly. Forty students from one or two classes from each grade (grade 1 to grade 9) were randomly selected, to ensure equal proportion of boys and girls in each grade were surveyed, data on height from 1 655 793 students measured in 2012 were compared with 1 419 281 students measured in 2017. Height was measured by metal height pole within 0.1 cm accuracy.
Results:
From 2012 to 2017, the average increase of height in boys and girls aged 6-15 years was 1.9 and 2.0 cm, respectively. Among them, maximum increment was found in boys at the age of 13 years and in girls at the age of 12 years, respectively, which was 3.0 and 2.8 cm. The average increase in boys from the central and western region was 2.1 and 2.2 cm, respectively, while increase in girls from the central and western region was both 2.2 cm. Compared with the results from 2012, height of boys with the 5th, 50th and 95th height percentiles in 2012 increased by 1.7, 2.0 and 2.1 cm, respectively in 2017, while the increase in girls was 2.1, 2.0 and 2.2 cm, respectively. The average increment of western students with the 5th and 95th height percentiles was higher than those of students from the central region.
Conclusion
The average height of students has increased after the implementation of NNIPRCES. However, the development was unbalanced between the central and western region, which requires more targeted intervening strategies to improve the nutritional status of students.
