1.Study on the immune status of T helper 17 cells in mice with periodontitis.

OBJECTIVEThis study investigates the immune status of T helper (Th) 17 cells in mice with periodontitis.
METHODSSeven-week-old C57BL/6 female mice were randomly divided into the control and periodontitis groups. Each group comprisedfour mice. Experimental periodontitis was induced in mice by oral infection with Porphyromonas gingivalis in the periodontitis group. Phosphate-buffered saline solution was used in the control group. All mice were sacrificed 4 weeks after the last P. gingivalis infection. CD4⁺retinoid-related orphan receptor (ROR) γτ⁺(Th17) cells were analyzed by flow cytometry. The protein expression of Th17 cell-related cytokine interleukin (IL)-17A was detected by enzyme-linked immunosorbentassay (ELISA).
RESULTSFlow cytometry showed that the percentage of CD4⁺RORγτ⁺(Thl7) cells in all CD4⁺ cells and the cell number in gingival tissues, cervical lymph nodes (CLNs), and the peripheral blood obviously increased (P < 0.01) in the periodontitis group. ELISA showed that compared with the control group, the periodontitis group exhibited an obvious increase in the protein expression of IL-17A (P < 0.05).
CONCLUSIONTh17 cell-mediated cell response is enhanced, and the gingival tissues, CLNs, and the peripheral blood are probably the main locations of Thl7 cell-mediated cell response during the development of periodontitis.

2.Methodological study of preparing iris pigment epithelial cells for autologous transplantation in rabbits

Objective To explore a better method in obtaining iris pigment epithelium (IPE) specimen for autologous transplantation in rabbits. Methods IPE was obtained from 20 black rabbits with method A, i.e., surgical peripheral iridectomy at 12:00 position obtaining a triangle iris tissue with the hemline of 4-5 mm in left eyes,and method B,i.e., surgical peripheral iridectomy at 11:00 and 1:00 positions obtaining two triangle iris tissues with the hemlines of 2-2.5 mm in right eyes . The IPE cells were isolated precisely with enzyme microdissection-enzyme isolation method, cultured in vitro, observed with light and electronic microscope, and identified with immunocytochemical staining. Results The success rate of cells culture were 65% for method A and 95% for method B. After 3-4 generations of culturing,the amount of IPE cells was enough for transplantation, and most of the functions of primary clutured IPE cells were kept still. Viability of IPE cells was 85%-93%. Conclusion The success rate of cells culture for method B is higher than that for method A. The third generation of cultured cells is available for autologous transplantation.
3.The study on Treg cell-mediated immune response in mice with periodontitis

Objective:To investigate regulatory T(Treg)cell-mediated immune response in mice with periodontitis.Methods:8 sev-en-week old C57BL/6 female mice were randomly divided into 2 groups(n =4).Periodontitis was induced in mice by oral infection with Porphyromonas gingivalis(P.gingivalis).Carboxymethyl cellulose(CMC)was used for the sham control mice.All mice were sacrificed 4 weeks after the last P.gingivalis infection.CD4 +forkhead box P3 (Foxp3)+ cells were analyzed by flow cytometry.The protein ex-pression of TGF-1 and IL-1 0 was detected by ELISA.Results:Periodontitis mice showed an obvious decrease in the percentage of CD4 +Foxp3 + cells in CD4 + T cells in gingival tissues,cervical lymph nodes(CLNs)and peripheral blood(P <0.01 ).Moreover,peri-odontitis mice had an obvious increase in the expression of TGF-1 and IL-1 0 in gingival tissues,CLNs and serum(P <0.05).Conclu-sion:Treg cell-mediated immune response is weakened in the mice with periodontitis.Gingival tissues,CLNs and the peripheral blood are probably the main source of Treg cells during periodontitis.
4.Immune -mediated liver failure

The primary causative factors of liver failure include direct damage and immune -mediated liver injury.Increasing evidence sug-gests that immune -mediated injury plays a pivotal role in the pathogenesis of liver failure.The new concepts concerning the mechanisms of immune -mediated liver injury in liver failure are reviewed with relevant basic and clinical studies in both humans and animals.The innate and adaptive immunity,particularly the interaction of various immune cells and molecules,as well as apoptosis -related molecules,are dis-cussed in detail.
5.A New Class Model Based on Partial Least Square Regression and Its Applications for Identifying Authenticity of Bezoar Samples

SIMCA(self independent modeling of class analogy) is a classical class modeling method for chemical) pattern recognition. Although widely used, SIMCA suffers difficulties in selecting a proper number of principal components and determining the decision region. A new class modeling technique based on partial least squares regression, partial least squares class model(PLSCM) is proposed, where the number of latent variables) and decision region can be readily estimated by the routine methods in multivariate calibration, e.g. Monte Carlo cross validation. PLSCM is successfully applied to identify trueborn bezoar samples from artificial and adulterated bezoar samples based on infrared spectra measured in the range of 4000-9000 cm~(-1). It is demonstrated that PLSCM outperforms SIMCA in terms of both maneuverability and identification accuracy. For the raw spectra, both the training and prediction accuracy of PLSCM are 100%. For the standard normal variate-processed data, the training and prediction accuracy of PLSCM is 99% and 100%, respectively.
6.Relationship among HLA-A alleles, supertype and the susceptibility of cervical cancer

Objective To study the relationship among HLA-A alleles, supertype, HPV infection and cervical cancer in Tu Nationality of Hubei province. Methods As a case-control surevy. The comparisons included the comparison between HPV positive cases and HPV positive women in control group, and the comparison between HPV positive cases and HPV negative women in control group. Number of cases was 100 ( HPV positive in 86) , and control was 187 ( HPV positive in 95 and HPV negative in 92). The most polymorphism of 2 and 3 exons of the HLA-A alleles were analyzed by the high-resolution typing method-sequence-based typing( SBT). Results Compar-ison between HPV positive cases and HPV positive control women. Supertype HLA-A3 (P_(corrected) = 0. 005, OR = 2. 36, 95% CI = 1. 45~3. 85) was risk factors. Comparison between HPV positive cases and HPV negative control women, HLA-A * 0206 alleles (P_(corrected)=0. 025,OR =0. 20,95% CI =0. 07~ 0. 58 ) supertype HLA-A2 ( P_(corrected) = 0.005 , OR = 0. 57 ,95% CI = 0. 37 ~ 0. 88 ) was protective factor. Supertype HLA-A3 ( P_(corrected) = 0. 005 , OR = 2. 36, 95% CI = 1. 45~3. 85) was also related to the susceptibility of cervical carcinoma. Conclusion Supertype HLA-A3 is a risk factor of cervical cancer.
7.Correlation of fgl2 prothrombinase gene expression with pulmonary impairment in the murine model of severe acute respiratory syndrome

Objective: To investigate the relationship of murine fibrinogen-like protein 2(mfgl2) / fibroleukin with pulmonary impairment in the murine model of severe acute respiratory syndrome(SARS).Methods: The Balb/cJ mice infected with murine hepatitis virus strain 3(MHV-3) through the trachea were observed for the pathological features and virus distribution in different organs.The expressions of both mfgl2 and fibrino in the lungs were determined by in situ hybridization and immunohistochemistry.Results: The MHV-3 infected mice developed typical interstitial pneumonia with extensive hyaline membrane formation in the alveoli,presented micro-vascular thrombosis in the pulmonary vessels and died within 5 days.MHV-3 virus replication was identified in all the organs observed.The specific expression of mfgl2 prothrombinase was noted in the terminal and respiratory bronchioles,alveolar epithelia and infiltrating cells.Conclusion: The characteristics of the pulmonary impairment of SARS in human can be properly simulated by the MHV-3 induced murine model of SARS.The up-regulation of the specific gene mfgl2 in the lungs involved in fibrino deposition and microvascular thrombosis may be largely responsible for SARS-associated pulmonary damages.
8.Relationship among HLA-A alleles,supertype and the susceptibility of cervical cancer

Supertype HLA-A3 (Pcorrected=0.005,OR=2.36,95% CI=1.45~3.85) was also related to the susceptibility of cervical carcinoma.Conclusion Supertype HLA-A3 is a risk factor of cervical cancer.
9.MicroRNA-34a inhibits human brain glioma cell growth by down-regulation of notch1.

The effects of microRNA-34a (miR-34a)-regulated Notch1 gene on the proliferation and apoptosis of the human glioma cell line U87 were investigated in this study. The U87 cells were divided into miR-34a mimics, negative control, mock transfection and blank control groups in terms of different treatments. In miR-34a mimics group, human U87 glioma cells were transfected with miR-34a mimics by using lipofectamine 2000. The cells transfected with nonsense microRNA were set up as negative control group. Those treated with lipofectamine 2000 only were designated to the mock tranfection group. In the blank control group, the cells were cultured routinely and no treatment was given. The expression of miR-34a and Notch1 was detected by using real-time RT-PCR. Western blotting was employed to monitor the change in Notch1 protein. Cell proliferation and apoptosis were measured by CCK-8 and flow cytometry. The results showed that the proliferative ability of U87 cells was significantly reduced and the apoptotic cells increased in miR-34a mimics group relative to control groups. The expression of miR-34a was significantly up-regulated in mimics group as compared with control groups (P<0.05). Furthermore, Notch1 protein levels were significantly decreased in miR-34a mimics group when compared with control groups (P<0.05), but the mRNA expression of Notch1 showed no significant difference among these groups. It was concluded that miR-34a may suppress the proliferation and induce apoptosis of U87 cells by decreasing the expression of target gene Notch1, suggesting that miR-34a may become a promising gene therapeutic target for brain glioma.
10.Effect of Qiangxin Oral Liquid on Experimental Left Ventricular Hypertrophy Induced by Isoproterenol and Ultrastructure in Rats

Objective To observe the protective effect of Qiangxin oral liquid on experimental left ventricular hypertrophy induced by isoproterenol and ultrastructure in rats.MethodsMyocardial hypertrophy model of rats were established by injection of isoproterenol (5 mg/kg/d) for 7 days. Thirty rats were divided into the control group, model group and treatment group (treated with Qiangxin oral liquid from second day after myocardial hypertrophy model made and continued for 12 weeks). Cardiac structure and function were detected in all groups by ultrasonography on 2nd, 6th and 12th week after model made respectively. After 12 weeks, the left ventricular weight/body weight index (LVW/BW) of all animals was tested and myocardial ultrastructure was examined.ResultsCompared with the model group, the interventricular septal end systolic thickness, left ventricular posterior wall thickness, left ventricular diameter, and left ventricular mass index of the treatment group decreased significantly ( P<0.05~0.01), while the damage of myocardial ultrastructrue lightened.ConclusionQiangxin oral liquid can improve the experimental myocardial hypertrophy of rats induced by isoproterenol.
