Objective To investigate the lentivirus-mediated RNA interference(RNAi) reduced human epidermal growth factor 2 (HER2) gene expression in ovarian cancer chemosensitivity.Methods Specifically targeted short hairpin RNA HER2 gene ( shRNA) lentiviral expression vector and negative control sequences lentiviral vector were constructed.stable expression of HER2-shRNA in SKOV3 cell line ( KD) and negative control cell line (NC) were established.The uninfected SKOV3cell lineserved as blank control(CON)group, sensitivity to changes in three kinds of cells to cisplatin chemotherapy in the HER2 gene expression was detected by RT-PCR and Western blot technique, CCK-8 assay CON and KD cells strains were established xenograft model in nude mice in each group to detect the expression of the HER2 protein, as well as by cisplatin chemotherapy tumor size. Results Expression results showed that a stable cell line HER2-shRNA and negative control cell lines had been successfully establish by fluorescence and flow cytometry.RT-PCR results showed KD cells relative HER2 mRNA expression levels compared with the NC group, the difference was statistically significant (P<0.01), whereas no significant difference when compared with the CON group, HER2 mRNA KD Group inhibition rate was (95.5 ± 0.12)%.After transduction specific HER2-shRNA ovarian cancer tumor cells, HER2 protein expression was significantly reduced, growth had slowed down significantly after cisplatin treatment ( P<0.01 ) .Conclusion Lentivirus-mediated HER2-shRNA can effectively inhibit the expression of the HER2 gene, increase chemosensitivity to cisplatin in ovarian cancer.

Objective To study the role of calcitonin-gene-related peptide (CGRP) and substance P (SP) in the pathogenesis of prurigo nodularis. Methods SABC immunohistochemical technique was used to examine the expression of CGRP and SP in lesional skin and uninvolved skin of 30 cases of prurigo nodularis and in skin of 15 normal controls. Results The strong expression of CGRP and SP was observed in the whole epidermis of lesional skin in 30 cases, with the expression surrounding cell membranes. The weak expression of CGRP and SP was observed in nonlesional and normal epidermis. The study was based on the maths model of waves which were reflected by a reflecting ellipsoid and whose distributing of energy in the ellipsoid. We developed an equipment by ellipsoidal focus to substitute for energy based on the research. The equipment has the characteristics of revolving therapy as the X-knife and the γ-knife which can be used in 3D therapy. We call the system ″Ultrasonic-knife″ which were used in 3D hyperthermia. The equipment simulates an source of energy by the ellipsoidal focus. The source can be put in the tumor in deep organ． It also avoids some shortcomings, just as heat is in limit area and temperature is too high in the focus. We could do an ideal conformal therapy by a reasonable treatment planning.

Objective To research a computer assisted 3D puncture system. Methods The system drew lessons from the method of CAS. MITK and the frame of X-knife were used to design the machine arm independently. Result The system came to the designing standard by experiments. Conclusion The clinical demanded product is developed with lower price in shorter time by integrating the mature techniques.

Objective To summarize the mid-term result and clinical experience of individual treatment for Stanford type B aortic dissection.Method 213 patients (172 male,41 female) with Stanford type B aortic dissection were admitted between Apr 2008 and Jan 2013.Age ranged from 23 to 83 averaging 54 ± 9.63 patients were treated conservatively and 47 patients underwent open surgery,103 patients by endovascular repair.Result Patients in conservative group were older than those in open surgery group.Preoperative waiting time was the longest in open surgery group.Concomitant procedures adopted in open surgery group included replacement of ascending aorta and aortic arch in 3 patients and fenestration of stented elephant trunk in 19 patients; 31 patients received bypass graft of cervical vessels during endovascular repair.Mortality was 11.1％,4.3％,3.9％ in conservative group,open surgery group and endovascular group,respectively.Compared with open surgery,endovascular repair had a lower incidence of perioperative complications (P ＜ 0.05).186 patients were followed-up with a mean follow-up duration of (26± 17) months.There were no difference in mortality and complications between the three groups.Conclusions In Stanford B aortic dissection,individualized therapy based on patient's condition achieves a favorable mid-term result with low mortality and complication rate.

Objective:To study the analgesic effect of dezocine combined with sufentanil in hip arthroplasty. Methods:Totally 60 patients with hip arthroplasty were randomly divided into the observation group and the control group. The 30 patients in the observation group were treated with dezocine combined with sufentanil, and those in the control group were treated with sufentanil. The analgesic efficacy and adverse reactions in the two groups were investigated and analyzed. Results:The visual analog scale ( VAS) in the obser-vation group in 3h, 6h, 12h, 24h and 48h after the surgery was significantly lower than that in the control group (P<0. 05). MMSE score in the observation group in 1h, 3h and 5h after the surgery was higher than that in the control group (P<0. 05). The total effec-tive rate of the observation group was 100. 00%, which was much higher than that of the control group (70. 00%, P<0. 05). There was significant difference in the adverse reactions between the two groups(P<0. 05). Conclusion:The efficacy of dezocine combined with sufentanil used in hip arthroplasty is significant with lower incidence of adverse reactions, which is worthy of popularized applica-tion in clinical practice.

Purpose:To study the effects of hydroxycamptothecin with thermotherapy on anti-angiogenesis in vitro and in vivo. Methods:We chose human microvascular endothelial cell (HMVEC) culture and chick embryo choriallantoic membrane (CAM) model and used MTT and number-calculating methods to observe hydroxycamptothecine on HMVEC’s proliferation , sprouts and CAM blood vessels’ formation.Results:The survival rate of endothelial cells was in the range of (68.2%)-44.7% within the dose of 20-40 ng/ml and 5-80 ng/ml and was negatively correlated with the concentration (correlation coefficient was -0.906,-0.469,P=0.00003,0.0051). Hydroxycamptothecine could significantly suppress the endothelial cells’ proliferation and the sprouts. Hydroxycamptothecine could significantly suppress CAM vessels. The survival rate of HepG II cells is in the range 100%-90% within the dose of 5-80 ng/ml. There was no cytotoxicity.There was a synergestic anti-angiogenetic effect when hydroxycamptothecin (20 ng/ml) was combined with thermotherapy in vitro while there was additive effect when hydroxycamptothecin (40 ng/ml) was combined with thermotherapy in vitro.Conclusions:This experiment shows that small doses of hydroxycamptothecine (20-40 ng/ml) with thermotherapy has anti-angiogenetic synergestic or additive effect on proliferation and migration both in vivo and in vitro.

Objective To investigate the expression of Skp2 and C-myc in hepatocellular carcinoma and its relationship with clinicopathologic characteristics.Methods The expression of skp2 and C-myc was evaluated in hepatocellular carcinoma tissues of 48 patients,liver cirrhosis tissues of 20 cases,and normal liver tissues of 16 patients by immunohistochemistry(PV9000 two steps methods).Results The expression rate of skp2(33.3%) in hepatocellular carcinoma tissues was significantly higher than that in liver cirrhosis tissues((negative) expression)(P=0.008)and normal liver tissues(negative expression)(P=0.020).The(expression) of skp2 was correlated with histological differentiation and metastasis(P﹤0.001 and P=(0.017)),but not with tumour size(P=0.058),and it was not expressed in highly differentiated(hepatocellular) carcinomas.The expression rate of C-myc protein in hepatocellular carcinoma tissues((58.3)%) was significantly higher than that in liver cirrhosis tissues(15%)(P=0.001) and normal tissues((negativ)e expression)(P

Cardiovascular and cerebrovascular diseases have be-come a great health threat.Increasing evidence has shown that both asymmetric dimethylarginine (ADMA)and symmetric dim-ethylarginine (SDMA)play important and independent roles in the development of cardiovascular and cerebrovascular diseases as well as in the prediction of cardiovascular and cerebrovascular events.Alanine-glyoxylate aminotransferase 2 (AGXT2 )is re-cently observed to be involved in ADMA metabolism.Deficiency in the expression and activity of AGXT2 may thus play a role in the development of cardiovascular and cerebrovascular diseases by affecting ADMA levels in vivo.Several single-nucleotide poly-morphisms at AGXT2 locus are observed to be associated with plasma SDMA level.This review summarizes recent advances in AGXT2 and its role in ADMA metabolism and the clinical rele-vance.

ObjectiveTo discuss the causes and the managem ent of hemorrhagic c omplications after sterotactic surgery for Parkinson's disease (PD).Me thodsA total of 350 PD patients unresponsive to drug therapy from April 1999 to Decemb er 2001 underwent sterotactic surgery. Intracranial hemorrhage occurred in 6 cas es, consisting of 3 cases of lesion hemorrhage and 3 cases of puncture path hemo rrhage.ResultsOf the 6 cases, 4 were treated by surgical ope rations, with re covery of consciousness but hemiplegia left, and 2 were treated conservatively w ithout dysfunction left. No fatal cases were seen in the study.Conclus ionsHe morrhagic complications are often in association with repeated puncture, excessi ve heat, operative skills, patient's general condition and so on. Accurate targe t location by MRI combined with microelectrode guidance, reduce of microelectrod e recording times, lowering of the lesion temperature, and sufficient perioperat ive care contribute to the prevention of intracranial hemorrhagic complications.
