1.Neuroprotective effect of acteoside against MPTP-induced mouse model of Parkinsons disease
Chinese Pharmacological Bulletin 1987;0(01):-
Aim To study the neuroprotective effect of Acteoside against MPTP-induced mouse model of Parkinsons disease (PD) and its mechanism. Methods The behavioral testing of C57 mice was assessed using spontaneous movement and rotarod test. DA levels in striatum were measured using HPLC-EC. Dopaminergic neurons in the substantia nigra were observed by immunohistochemical staining with an anti-Tyroxine hydroxylase (TH) antibody. Moreover, the mechanism of the neuroprotective effect of acteoside was investigated using Western blot analysis with an anti-?-synuclein antibody in the substantia nigra and striatum. Results ① Compared with control, MPTP lesion significantly reduced the number of spontaneous movement and latent period of mice on the rotating rod (both P

Objective To investigate the change of serum alpha-L-fucosidase(AFU)and its correlation with the blood glucose and lipid level in small for gestational age(SGA)fetuses. Methods 125 SGA fetuses and 128 fetuses in appropriate for gestational age(AGA)with wet lung were treated in our hospital and were investigated as case control study. The serum of AFU ,blood glucose and lipid were measured and compared within 24 hours after birth in these 2 groups. Results Comparing with the AGA infants ,the SGA babies had lower level of serum AFU,high density lipoprotein, apolipoprotein A and apolipoprotein B(P<0.05). The correlation analysis showed that the serum AFU level has positive correlation with blood glucose, total cholesterol, high density lipoprotein, low density lipoproteinand apolipoprotein A(P < 0.05),while it has negative correlation with serum triglycerides in SGA(P<0.05). Conclusions The SGA infants have lower level of serum AFU and lipid metabolic disorders after birth, and its serum AFU level has correlation with its blood glucose and lipid level.

Objective To explore the hearing and cochlear histomorphological changes in APP/PS1 transgenic mice during the process of AD and to determine whether the occurrence and development of AD affect their hearing function. Methods APP/PS1 transgenic mice and wild type littermates were selected at the ages of4,8 and 12 months.Changes to the auditory function of APP/PS1 transgenic mice over time were detected by immunofluorescence staining and auditory brainstem response(ABR)test. Results Compared with the control group, the 4-month-old APP/PS1 transgenic mice had a significantly increased number of Aβ plaques in the hippocampus, indicating that AD symptoms were already present at this time.At the ages of 4,8 and 12 months, there was no significant difference in the hearing threshold between APP/PS1 transgenic mice and wild type mice.Histomorphological examination of the cochlea showed no significant difference in key cells of he inner ear, such as cochlear hair cells and spiral ganglion, between the two groups of mice.An ABR test showed that the hearing threshold of both APP/PS1 transgenic mice and wild type littermates increased significantly with age,and both groups showed age-related hearing loss. Conclusions There was age-related hearing loss in APP/PS1 transgenic mice, but the occurrence and development of AD had little effect on their auditory functions.
