3.In Vivo Suceptibilities of Ascitic Sarcoma 180(S180)Cells to Natural killer (Nk)C8ll-Mediated Binding and Killing During Their Development
Chinese Journal of Immunology 1985;0(06):-
The suceptibilities of ascitic S180 cells to NK cell-mediated binding and killing during their development in vivo were studied at a single cell level, usingsingle cell cytotoxic and binding assaies. The results showed that the NK binding and killing suceptibilities of ascitic S180 cells changed sequentialy during their growth in vivo, the kinetics of which had an appearence of reversed S with both NK binding and killingsuceptibilities being the highest at the fifth day and the lowest at the second day and the tenth day after inoculation of the tumors. However, the changes were seen to be similar when the tumors were inoculated in either adult(6-7-week-old) or yaung(3-week-old) mice, or in different strains of adult mice. Analysis at a single cell level revealed that the change in the NK suceptibilitg of the tumor cells at different growth stages were concomitant with the changes in the number of the tumor target cells capable of being bound by NK cells as shown with an NK-target binding assay, which was consistant to the flactuation of the capability on which the tumor target cells could bind more than one NK cell. Our results indicate the NK-suceptibility of ascitic S180 cells changes uring their growth in vivo, due to the changes of the ratio of NK-suceptible and-nonsuceptible target cells in the tumor cells at different growth stages. The possible cause(s) for the changes is discussed here.
4.Expression and clinical significance of extracellular matrix metalloproteinase inducer molecule CD1 4 7 in bladder urothelial carcinoma tissue
Chongqing Medicine 2014;(34):4574-4578
Objective To investigate the expression of extracellular matrix metalloproteinase inducer CD147 in patients with bladder urothelial carcinoma and its relationship with clinicopathologic features and prognosis. Methods RT-PCR and Western blot were used to evaluate the mRNA and protein expression of CD147 in cancer tissues and corresponding paracanerous tissues from 20 patients with bladder urothelial carcinoma;tissue microarray containing specimens from 108 patients with bladder urothelial carcino-ma and 20 normal controls was constructed and immunohistochemistry was applied to examine the expression of CD147,and the correlations between CD147 and clinicopathological characteristics and prognosis were also analyzed. Results Expression level of CD147 mRNA and protein in cancer tissues from 20 bladder urothelial carcinoma patients was significantly higher than that of cor-responding adjacent tissues(P<0.01).Immunohistochemical detection showed normal bladder urothelium from 20 control cases were either no or weak protein expression of CD147,positive expression rate of CD147 in 108 cases of bladder urothelial carcinoma was 73.1%.the CD147 expression level had positive correlation with lymph nodes metastasis, histological grade, tumor stage and tumor size of patients with bladder urothelial carcinoma(P<0.05)and had no relationship with age, sex, the number of tumor(P>0.05).Kaplan-Meier survival curves analysis showed that the survival time of patients with bladder urothelial carcinoma whose ex-pression of CD147 were high was significant lower than that of patients with bladder urothelial carcinoma whose expression of CD147 were low(P<0.01).Univariate analysis showed that histological grade, tumor stage, lymph node metastasis, tumor size and CD147 expression were all positively correlated with a poor prognosis(P<0.05).Cox-multivariate analysis revealed that histologi-cal grade, tumor stage, lymph node metastasis, and CD147 expression level were independent predictors of an unfavorable prognosis (P<0.05).Conclusion High expression of CD147 in bladder urothelial carcinoma may be correlate with the its progress and its expression state is an independent prognosis factor.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2014;28(15):1154-1157
OBJECTIVE:
To observe the effects of different maintain doses of Dexmedetomidine on plasma cortisol and glucose during anesthesia recovery period in patients undergoing uvulopalatopharyngoplasty under sevoflurane inhalation anesthesia.
METHOD:
In this prospective, randomized, double-blind study, 120 ASA I and II patients undergoing selective uvulopalatopharyngoplasty under general anesthesia were included. The patients were randomly allocated to three groups (n = 40): Dexmedetomidine low maintain dose group (D1), Dexmedetomidine high maintain dose group (group D2) and control group (group C). The Dexmedetomidine groups and control group were given Dexmedetomidine 1 microg/kg and normal saline in 20 ml within 15 min just before induction of anesthesia. Then Dexmedetomidine were maintained at 0.2 microg x kg(-1) x h(-1) and 0.7 microg x kg(-1) x h(-1) in group D1 and group D2 and were withdrawed 5 min before the end of operation, the same maintained speed of normal saline was given in group C. BIS value was maintained at 40-60 by adjusting the inhaled concentration of sevoflurane. Anesthetic was withdrawed 10 min before the end of operation. Thus, plasma cortisol concentration and blood glucose was needed to be detected just before anesthesia (T0), tracheal extubation (T1), 5 min after extubation (T2) and 15 min after extubation (T3). Duration of operation and anesthesia, consumption of sevoflurane, emergence time, extubation time, the occurrence of dysphoria, bucking and hypoxemia (SpO2 < 90%) during extubation were recorded.
RESULT:
Compared with group C, MAP and HR at T1, plasma cortisol concentration and blood glucose at T1 - T3 were all significantly lower in group D1 and group D2 (P < 0.05), and so were the consumption of sevoflurane and the occurrence of dysphoria (P < 0.05). The emergence time and extubation time were significantly prolonged in group D2 compared with group D1 and group C (P < 0.05). There was no significant difference in the occurrence of bucking and hypoxemia in three groups (P > 0.05).
CONCLUSION
In the patients undergoing UPPP under sevoflurane inhalation anesthesia, Dexmedetomidine infused at 0.2 microg x kg(-1) x h(-1) maintains a stable hemodynamics without respiratory depression, alleviates stress response during extubation and reduces both the consumption of sevoflurane and the occurrence of dysphoria without prolonging emergence time and extubation time.
