3.Expression and clinical significance of extracellular matrix metalloproteinase inducer molecule CD1 4 7 in bladder urothelial carcinoma tissue
Chongqing Medicine 2014;(34):4574-4578
Objective To investigate the expression of extracellular matrix metalloproteinase inducer CD147 in patients with bladder urothelial carcinoma and its relationship with clinicopathologic features and prognosis. Methods RT-PCR and Western blot were used to evaluate the mRNA and protein expression of CD147 in cancer tissues and corresponding paracanerous tissues from 20 patients with bladder urothelial carcinoma;tissue microarray containing specimens from 108 patients with bladder urothelial carcino-ma and 20 normal controls was constructed and immunohistochemistry was applied to examine the expression of CD147,and the correlations between CD147 and clinicopathological characteristics and prognosis were also analyzed. Results Expression level of CD147 mRNA and protein in cancer tissues from 20 bladder urothelial carcinoma patients was significantly higher than that of cor-responding adjacent tissues(P<0.01).Immunohistochemical detection showed normal bladder urothelium from 20 control cases were either no or weak protein expression of CD147,positive expression rate of CD147 in 108 cases of bladder urothelial carcinoma was 73.1%.the CD147 expression level had positive correlation with lymph nodes metastasis, histological grade, tumor stage and tumor size of patients with bladder urothelial carcinoma(P<0.05)and had no relationship with age, sex, the number of tumor(P>0.05).Kaplan-Meier survival curves analysis showed that the survival time of patients with bladder urothelial carcinoma whose ex-pression of CD147 were high was significant lower than that of patients with bladder urothelial carcinoma whose expression of CD147 were low(P<0.01).Univariate analysis showed that histological grade, tumor stage, lymph node metastasis, tumor size and CD147 expression were all positively correlated with a poor prognosis(P<0.05).Cox-multivariate analysis revealed that histologi-cal grade, tumor stage, lymph node metastasis, and CD147 expression level were independent predictors of an unfavorable prognosis (P<0.05).Conclusion High expression of CD147 in bladder urothelial carcinoma may be correlate with the its progress and its expression state is an independent prognosis factor.
4.In Vivo Suceptibilities of Ascitic Sarcoma 180(S180)Cells to Natural killer (Nk)C8ll-Mediated Binding and Killing During Their Development
Chinese Journal of Immunology 1985;0(06):-
The suceptibilities of ascitic S180 cells to NK cell-mediated binding and killing during their development in vivo were studied at a single cell level, usingsingle cell cytotoxic and binding assaies. The results showed that the NK binding and killing suceptibilities of ascitic S180 cells changed sequentialy during their growth in vivo, the kinetics of which had an appearence of reversed S with both NK binding and killingsuceptibilities being the highest at the fifth day and the lowest at the second day and the tenth day after inoculation of the tumors. However, the changes were seen to be similar when the tumors were inoculated in either adult(6-7-week-old) or yaung(3-week-old) mice, or in different strains of adult mice. Analysis at a single cell level revealed that the change in the NK suceptibilitg of the tumor cells at different growth stages were concomitant with the changes in the number of the tumor target cells capable of being bound by NK cells as shown with an NK-target binding assay, which was consistant to the flactuation of the capability on which the tumor target cells could bind more than one NK cell. Our results indicate the NK-suceptibility of ascitic S180 cells changes uring their growth in vivo, due to the changes of the ratio of NK-suceptible and-nonsuceptible target cells in the tumor cells at different growth stages. The possible cause(s) for the changes is discussed here.

China Pharmacist 2017;20(2):225-228
Objective:To synthesize poly (2-ethyl-2-oxazoline)-poly(lactic acid) copolymer (PEOz-PLA), prepare pH-sensitive paclitaxel-loaded polymeric micelles, and evaluate the in vitro properties. Methods: The structures of all the synthesized copolymers were confirmed by 1 HNMR and Fourier transform infrared spectroscopy ( FTIR) . The drug-loaded micelles were prepared by a dialysis method. The micelles solution was freeze-dried and the lyoprotectants were screened. The critical micelle concentrations ( CMC) of the copolymers were measured by pyrene fluorescent probe technique. The distribution of particle size was measured by dynamic light scat-tering ( DLS) . The in vitro release behavior of the loaded micelles at different pH conditions was studied by a dialysis method. Re-sults:The CMC of micelles was 25. 63 mg·ml-1 . When 10% polyethylene glycol 4000 was used as the lyoprotectant, the re-solubili-ty of micelles was good and the distribution of particle size was narrow. The drug loading rate of micelles was 8. 12% and the encapsu-lation efficiency was 69. 33%. The average particle size of freeze-dried micelles was 183. 7 nm. The in vitro release of the drug-loaded micelles showed that the drug released slowly at pH 7. 4 and fast at pH 5. 0, which indicated that the change of pH could affect the drug release. Conclusion:The preparation of PEOz-PLA polymer micelles is simple. The particle size, entrapment efficiency and drug loading are controllable. The micelles have a certain sustained-release effect, which provides basis for further pharmacological study and clinical application.

Chinese Journal of Urology 1994;0(02):-
Objective To highlight the diagnostic and t herapeutic procedures for extra-adrenal pheochromocytoma. Methods Thirty-eight cases of extra-adrenal pheochromocytoma proven patholo gically from 1998 to 2002 were reviewed and studied.Hypertension was observed in 34 cases and intermittent macrohematuria in 4 patients.Another 2 cases were adm itted to hospital with abdomen masses through routinely physical examination.Uri nary VMA,serum and urinary catecholamine were measured in 33 cases.38 cases had B-model ultrasound examination and 34 cases took CT scan. The level of urine VMA elevated in 30 cases(91%) and serum or urinary catecholamine were elevated in 2 8 (85%).The positive rates of localization of B-ultrasound and CT were 92% and 100%,respectively.29 cases took ?-receptor blocker for at least 2 weeks preope ratively. There were 6 cases operated under epidural anesthesia and 32 general an esthesia.Six cases were operated transvesical.10 cases with lumber incision and 19 with abdominal incision. The other 3 cases operated laparoscopically. Results Tumor location,6 cases in bladder wall,7 in renal hilum, 9 in upper pole of kidney,4 in low pole,10 in anterio-lateral to the adrenal gl and,and 2 cases of multifoci. The specimens were from 2 to 9 cm in size.Among 34 cases with hypertension, the blood pressure of 26 patients return to normal and 2 cases were still hypertensive.10 cases were assayed as malignant pheochromocyto ma.There were 9 cases of recurrence or metastases and 5 patients died during fol lowed-up from 13 months to 10 years. Conclusions Urine VMA level is a useful marker to detect extra-adrenal pheochromocytoma and CT sc an is the most reliable in localizing the lesions. 131I-MIBG scintigra phy is of great value for the localization and quantitative for extra-renal phe ochromocytoma with high sensitivity and accuracy. It may be also used as a therap eutic method.Surgical manipulation of the tumor is the best management and metic ulous perioperative management is extremely important.Measuring the level of uri nary catecholamine can monitor the recurrence or metastasis of the tumor.
