2.Exploration of Strengthening Quality Management of Drug Microbiology Laboratories

OBJECTIVE:To strengthen quality management of drug microbiology laboratory and provide reference for relevant inspection institutions. METHODS:Taking measures from the laboratory document management,personnel training,quality con-trol,quality supervision,process control and biological safety,the elements of laboratory quality management were analyzed,and effectively measures were put forward to improve the management level. RESULTS & CONCLUSIONS:The laboratory should es-tablish strict,standardized,systematic laboratory management rules and regulations,quality documents,operating procedures, work guide,etc. to achieve documented management;develop various forms of annual training and assessment programs to con-struct a professional team;strengthen internal(developing an annual internal quality monitoring plan,conducting quality control ac-tivities,regularly checking the suitability of the medium) and external (capability testing,inter-laboratory comparison) manage-ment control level,and reasonably formulate the contents and frequency by combing with actual situation. Besides,the laboratory should strengthen inspection process control,attach great importance to the biological safety management to reduce the risk of labo-ratory quality to a minimum.
3.Pectoralis major myocutaneous flap in the application of the head and neck surgery

Objective To summarize experience in pectoralis major myocutaneous (PMMC) flap reconstruction for patients undergoing head and neck surgeries. Methods We conducted a retrospective study to analyse the clinical data of 20 patients who had PMMC flap reconstruction in a head and neck surgery from March 2011 to June 2016. All patients were male,with the average age of 54 years(ranging from 27 to 71 years). The PMMC flaps of all patients survived after the surgery. Only one patient developed pharyngocutaneous fistula ,and was recovered after treatment with incision dressing. Among follow?ups for 2 to 24 months,all patients got well recovered,the swallowing and communication functions normal communication and the appearance a satisfactory. good looking. Conclusion PMMC renders effective flathe flap reconstruction with broad clinical application value is an optimal choice for patients underwent head and neck surgery with muscle or skin defect,especially for those who had postoperative radiotherapy because of its rich blood supply and it has a lot of affluent tissues,fmore flexibility inle application and higher survival rate. So it is highly applicable,especially for grass?roots hospitals.
4.Expression and purification of truncated human recombinant nuclear apoptosis-inducing factor 1 in E.coli

Objective:Nuclear apoptosis-inducing factor 1(NAIF1) is a novel apoptosis gene cloned in laboratory. To analyze the binding proteins of NAIF1 by pulldown method, the fusion expression vector of truncated human nuclear apoptosis-inducing factor 1〔NAIF1(73-327)〕 was constructed, were expressed and purified the recombinant GST-NAIF1(73-327) fusion protein in E.coli.Methods:The cDNA encoding human NAIF1(73-327) was amplified by PCR and cloned into pGEX-KG vector. The GST-NAIF1(73-327) fusion protein was expressed in E.coli and purified by affinity chromatography. The purified protein was detected by SDS-PAGE, Western blot and ESI-Q-TOF-MS/MS.Results:A prokaryotic expression vector of GST-NAIF1(73-327) was constructed and the GST-NAIF1(73-327) fusion protein was expressed in E.coli at high level. SDS-PAGE analyses indicated that the purified protein was about 53 kD. Western blot and MS/MS analyses verified the recombinant fusion protein.Conclusion:An efficient method for obtaining recombinant GST-NAIF1(73-327) fusion protein had been established and it could be used for further studies on the structure and function of NAIF1.
