Objective To study time-dependent degradation of DNA in lymphocytes in the human bloodstain in order to estimate aging of bloodstain.Methods Lymphocytes were isolated from the human bloodstain placed under room temperature and were analyzed by single-cell gel electrophoresis(SCGE).The percentage of DNA-degraded lymphocytes were calculated in the cloodstain placed at different time intervals from 0 to 72h under fluorescent microscope connected with auto-analysis-image system.Results The percentage of DNA degraded lymphocytes was closely correlated with time lapse within 72h.A regression equation was formulated with R=0.9522(P

Objective To evaluate the clinical curative effect of Salvia miltiorrhiza Ligustrazine combined with Butylphthalide in cerebral infarction of children. Methods Sixty-two children with acute cerebral infarction were randomly divided into treatment group (33 patients) and control group (29 patients). The patients in two groups received the same basic treatment. The control group used Salvia Ligustrazine injection, and the treatment group used Salvia Ligustrazine injection combined with Butylphthalide. The total efficiency, the degree of recovery of muscle strength and the scores of nerve defect 2 weeks after treatment were compared. Results The total efficiency in treatment group was 87.9%(29/33), in control group was 75.9%(22/29), and there was significant difference (P<0.05). Two weeks after treatment, the muscle strength scores in treatment group was (4.00 ± 0.47) scores , in control group was (3.59 ± 0.98) scores, and there was significant difference (P<0.05). Two weeks after treatment, the scores of nerve defect in treatment group was (5.42 ± 2.18) scores, in control group was (6.86 ± 2.23) scores, and there was significant difference (P<0.05). Conclusions Salvia Ligustrazine combined with Butylphthalide in treatment of cerebral infarction of children has obvious effect, and it is worthy of spreading.
3.Clinical efficacy on mental retardation in the children treated with JIN's three scalp needling therapy and the training for cognitive and perceptual disturbance.

OBJECTIVETo explore the impacts on the cognitive level of the children with mental retardation (MR) treated with JIN's three scalp needling therapy and the training for cognitive and perceptual disturbance so as to seek the more effective therapeutic method for the treatment of MR in children. Methods Sixty-six MR children were randomized into a head-point needle-retaining therapy + training group (group A) and a training after acupuncture group (group B). Seven cases and 12 cases were dropped in the two groups respectively. Twenty-six cases accomplished the treatment in the group A and 21 cases in the group B. In the group A, the points on the head were stimulated and the needles were retained (30 min after the general acupuncture, the needles on the body points were removed; and those on the head points were retained for 1 h, including Sishenzhen, Naosanzhen, Zhisanzhen and Niesanzhen). Simultaneously, the training for the cognitive perceptual disturbance was combined. In the group B, 1 h after needle retaining at the body points and head points, all the needles were removed; the training for the cognitive perceptual disturbance was followed. The treatment was given once a day, and the treatment for 3 months was required. Before and after treatment, the Wechsler intelligence scale for children (WISC) was used for evaluation and observation of verbal intelligence quotient (VIQ) , performance intelligence quotient (PIQ) and full-scale intelligence quotient (TFIQ) and score changes of 11 items such as information, picture vocabu; lary, arithmetic, picture generalization, comprehension, etc.
RESULTSAfter treatment, FIQ, VIQ and PIQ scores were different significantly as compared with those before treatment (all P<0. 01). In the group A, the results of picture vocabulary, animal egg laying, maze, block design and geometric figure were all improved significantly (all P<0. 05). In the group B, the results of information, comprehension, block design and geometric figure were all improved significantly (all P<0. 05). After treatment, concerning the value difference in FIQ and PIQ of the two groups; the changes in the group A were more significant (both P<0. 01). After treatment, the results of picture vocabulary and maze were improved significantly in the group A as compared with the group B (both P<. 01). The IQ categories changed apparently after treatment in the two groups, toward the higher level in tendency generally, but without significant difference (both P>0. 05).
CONCLUSIONThe simultaneous treatment with head point retaining of JIN's three needling therapy and the training for cognitive and perceptual disturbance obviously improves children patients' verbal comprehension, expression ability, hand-eye coordination ability, attention, logical reasoning ability and visual perception. The efficacy is better than that in the treatment of the training after acupuncture.

4.Impact of acute renal artery stenosis on tissue elasticity of the kidney: an animal study

Objective To study the changes of Young's modulus value of the renal cortex,vertebral body and sinus when there was a different degree of acute renal artery stenosis.Methods 10 Japanese white rabbits were used for this study.The left renal artery was dissected.An ultrasound probe was placed on the left kidney,Young's modulus of the renal cortex,vertebral body and sinus were recorded using shear wave ultrasound elastic imaging technique during three stages:when the renal artery was intact (0% stenosis),tied with a suture (> 50% stenosis),and ligated (100% stenosis).Renal tissue elasticity with different degrees of renal artery stenosis were compared.Pathological study was carried out in the kidneys,each with either 0%,> 50% or 100% renal artery stenosis.Results The mean values of Young's modulus from the renal cortex,vertebral body and sinus with different degrees of renal artery stenosis were as follows:0% stenosis,(16.31 ±1.70) kPa,(13.38± 1.63) kPa,and (12.75±2.26) kPa;> 50% stenosis,(14.16±2.34) kPa,(11.49±2.70) kPa,and (10.72±2.56) kPa;100% stenosis,(11.35 ± 1.48) kPa,(8.39± 1.29) kPa,and (7.08 ± 1.52) kPa.The mean value of Young's modulus from the renal cortex was significantly higher than those from the renal vertebral body and sinus (P < 0.05).The mean values of Young's modulus from different parts of the kidney decreased progressively with an increase in renal artery stenosis (P < 0.05).The pathological study demonstrated that with the increase in the severity of renal artery stenosis,glomerular volume,renal interstitial capillary diameter and red blood cells decreased.When renal artery stenosis reached 100%,there was patch degeneration and inflammatory cell infiltration within the renal interstitium,and mild degeneration of tubular epithelial cells with obscure cell borders.Conclusions The value of Young's modulus from the renal cortex was higher than those from the renal pyramid and sinus in rabbits.The values of Young's modulus from the renal cortex,pyramid and sinus decreased with an increase in the severity of renal artery stenosis,which suggest that,in the study of the elasticity of the kidney,the renal artery blood flow dynamic state can obviously affect the value of the elasticity of the kidney.
5.Drug Resistance Surveillance of Bacteria for 8 Years in Succession and Its Results

OBJECTIVE To survey the drug resistance variation of pathogens.METHODS We analyzed all of the pathogens isolated from Jan 1998 to Dec 2005 and the drug resistance results.WHONET5 software was used to analyze the data.RESULTS A total of 19 832 strains of pathogens were detected for eight years.There was ascending tendency in ESBLs and MRS.The ratio of strains and resistance also varied.The general trend was that the order of Staphylococcus was going up and the order of non-fermented and fermented bacteria was ascending alternately.CONCLUSIONS The resistance variation of pathogens in our hospital consists with the global tendency of bacteria resistance.Of course it has its own characteristics.The resistance surveillance should be continued especially for ESBLs,MRS,VISA and VRSA.
6.Comparative Study on Spinal Metastasis Between MRI and Radionuclide Bone Scan

Objective To evaluate the reasons of positive MR findings and negative radionuclide bone scan in the spinal metastasis.Methods 68 patients with spinal metastasis were undergone MR scan and radionuclide bone scan.MRI appearances including the site,location and their relation to the cortex of bone were analyzed. The relationship between MRI and radionuclide bone scan appearances were also analyzed.Results 561 lesions were detected by MRI,and only 199 lesions were found by raionuclide(199/561=35.47%).133 lesions located in the bone marrow without cortex erosion were not detected by radionuclide bone scan. 7. Qualitative study on the experience of young women with breast cancer when disclosing the disease to children

Objective:
To understand the experience of young breast cancer patients when disclosing the disease to children.
Methods:
The phenomenological method was adopted in the study. Semi-structured interviews were performed in 13 young breast cancer patients. The date were analyzed by Colaizzi analysis method.
Results:
Four themes were found: disclosing decision, professional information needs, feel guilty and worried about children, positive experiences with children.
Conclusions
The medical staff should reduce patients′ stress of disclosure and make intervention strategy to implement individual guidance.
8.Development of referral indications for low back pain with RAND-UCLA consensus panel method

Objective To study the validity of RAND-UCLA (Rand Corporation and University of California at Los Angeles) consensus panel method in developing guidelines of referral indications for low back pain (LBP).Methods Evidence-based clinical guidelines for LBP management at community level and its referral guidelines published since 2001 and other tools were retrieved with varied tools.All clinical guidelines met inclusion criteria were evaluated with clinical studies and evaluation tools (AGREE).An pool of indication items was established based on evidence for developing referral indications for LBP, which were added by RAND-UCLA consensus panel method, and alternative referral indications were selected and clinical guidelines for LBP referral were established.Results A total of 15 copies of clinical guidelines from nine countries or regions were included in it after critical appraisal.Four copies of referral guidelines from two countries were included.Referral indications for LBP were derived directly from the RAND-UCLA consensus panel process, consisting of 44 referral indications for three groups (immediate, urgent and routine referral).Conclusions The RAND-UCLA consensus panel method is a more useful and practical tool in developing clinical guidelines, referral guidelines, which is worthwhile being recommended and spread.
9.The effect of heterogeneity in different parts of deep venous thrombus on the ultrasound shear wave elasticity in rabbit

Objective To study the effect of heterogeneity in different parts of acute and chronic deep venous thrombus on the ultrasound shear wave elasticity value.Methods Fifteen Japanese rabbits were used to create an inferior vena cava (IVC) thrombus model via IVC ligation.Young's modulus of the thrombus head,body and tail were measured using ultrasonic shear wave elasticity technique on the fifth day (acute phase) and 14th day (chronic phase) of thrombus formation.Pathological sections were obtained from the IVC thrombus in five rabbits at the two points of time mentioned above.Pair t-test and one-way analysis of variance (ANOVA) were used to compare the data between groups.Results On the fifth day,Young's modulus values of the thrombus head,body and tail were (5.73 ±0.47)kPa,(7.82±0.63)kPa and (4.76±0.45)kPa respectively.ANOVA showed significant difference among three parts (F=134.468,P < 0.01).The value of the thrombus body was significantly higher than that of the head and tail (both P < 0.01),and the value of the head was significantly higher than that of the tail (P < 0.01).On the 14th day,Young's modulus values of the thrombus head,body and tail were (12.46 ± 2.59)kPa,(15.08 ±2.71)kPa and (10.03 ± 2.02)kPa,respectively.ANOVA analysis also showed significant difference among three parts (F=10.539,P < 0.01).The value of the thrombus body was significantly higher than that of the tail (P < 0.01),and also higher than that of the head,although the difference was not significant (P > 0.05).There was no significant difference between the head and tail (P > 0.05).Young's modulus values of the thrombus head,body and tail in the chronic phase were significantly higher than those in the acute phase (t=-7.456,-7.989 and-8.159,respectively,all P < 0.01).Pathological results showed that there was significant difference in thrombus structure and composition among different parts of the thrombus and among the thrombi in different individuals at corresponding points of time following thrombus formation.Conclusion There is significant heterogeneity among different parts of the same thrombus and among different thrombi at corresponding points of time after thrombus formation.Continual monitoring of the thrombus with ultrasound elasticity imaging may help to improve the accuracy of thrombosis staging.
10.Clinical phenotype and genetic analysis of pyridoxine dependent epilepsy induced by aldehyde dehydrogenase 7 family member A1 gene mutation

Objective:To investigate the clinical phenotypes, therapy and genetic features of aldehyde dehydrogenase 7 family member A1 (ALDH7A1) gene mutations in five cases of pyridoxine dependent epilepsy (PDE) with diagnosis confirmed by next generation sequencing.Methods:Retrospective analysis was carried out on clinical data of five cases of PDE children with early epilepsy onset who were treated in the Department of Neurology of Children′s Hospital Affiliated to Zhengzhou University from February 2018 to November 2019. Next generation sequencing approach was used for genetic sequencing of proband ALDH7A1 gene and the first generation Sanger was used for validation of family members. And the characteristics of gene mutations were analyzed.Results:Among the five children diagnosed with PDE, the male to female ratio was 4 ∶ 1 and ages at clinic visit ranged from two months to 10 months old. In clinical phenotypes, all five cases experienced onset in neonatal period, with repeated seizures, manifested as myoclonus, spasms or focal paroxysm. The administration of antiepileptic drugs performed poorly in seizure control while long term oral intake of large dose pyridoxine showed better efficacy. All the five cases of children came from compound heterozygous mutations of father and mother, i.e. slicing homozygous mutation c.247-2(IVS2)A>T, missense mutation c.584A>G (p.N195S) and nonsense mutation c.1003C>T(p.R335 *), missense mutation c.1553G>C(p.R518T) and c.1547A>G(p.Y516C), missense mutation c.1547A>G(p.Y516C) and frameshift mutation c.1566_1568delTAC, missense mutation c.1061A>G(p.Y354C) and nonsense mutation c.841C>T(p.Q281X, 259), among which c.247-2(IVS2)A>T was novel splicing site mutation not reported before. Conclusions:PDE is induced by ALDH7A gene mutation. Early clinical manifestations are mostly onset of refractory epilepsy in neonatal period. Antiepileptic drugs perform poorly in terms of efficacy while pyridoxine can control seizure effectively. Gene analysis should be conducted on such patients for confirmed diagnosis.
