1.Effects of sodium arsenite on cell apoptosis in rat cerebellar granule and nerve globin mRNA expression

Objective To observe the effects of sodium arsenite (NaAsO2) on apoptosis in primary cultured rat cerebellar granule cell and nerve globin mRNA expression.Methods Twenty 7-8 days postnatal Wistar rats were selected and decapitated cerebellar cortex, granular cell suspension was prepared and 4 × 109/m2 cells were seeded in the culture flask.Cells were exposed to different concentrations NaAsO2 [0 (control), 2, 5 and 10 μmol/L] for 24 h and intracellular reactive oxygen species (ROS) content and apoptosis were determined by laser confocal,while real-time quantitative PCR was used to detect mRNA expression in cultured cells after 24 and 48 h cell culture.Results Fluorescence intensity of ROS of 0, 2, 5, 10 μmol/L fluoride groups were 421.32 ± 53.52,1 082.09 ± 321.58, 2 223.02 ± 1 011.37 and 1 366.87 ± 102.14, the difference was statistically significant (F =5.873,P < 0.05);Apoptosis rates were (8.91 ± 5.63)%, (26.46 ± 1.77)%, (43.65 ± 14.45)% and (56.04 ± 6.95)%, the difference was statistically significant (F =18.369, P < 0.05);24 h nerve globin mRNA expression were 1.00 ± 0.06,0.77 ± 0.22, 0.43 ± 0.18 and 0.42 ± 0.14, the difference was statistically significant (F =18.235, P < 0.05);48 h nerve globin mRNA expression were 1.00 ± 0.01, 0.89 ± 0.09, 0.60 ± 0.16 and 0.08 ± 0.02, the difference was statistically significant (F =80.843, P < 0.05).Conclusion With increased NaAsO2 concentration, ROS and apoptosis levels are increased, while nerve globin mRNA expression is significantly reduced, suggesting that after the body is stimulated, protective proteins might be consumed, further suggesting nerve globin expression might be involved in nerve damage caused by arsenic.
2.Influence of clinic design of the prostheses with ERA attachment for distal extension defect: A three-dimensional finite dement analysis

Objective:To evaluate the stress distribution and displacement pattern in the restoration of Stern-ERA attachment mandible removable partial denture (RPD) with different designs through three-dimensional (3D) finite element (FE) analysis.Methods:3D-FE models were established based on human CT data and manufacturer's catalog.Vertical and 45° lingual oblique forces of 100 N were applied on the artificial teeth of the RPD.Three ERA attachment partial dentures with different bracing arms were designed,namely the basic design,rest bracing arm design and cross-arch bracing arm design.The von Mises stresses and displacements of abutment teeth and mucosa under denture base were analysed.The displacement of denture bases was examined.Results:Under the vertical and oblique loading,the maximum yon Mises stress was found at the distal shoulder on the distal abutment.The maximun displacement was observed at the buccal and distal occlusal surfaces of the distal abutment.The maximum displacement on the abutment was found to be the highest in rest bracing arm design group (14.91 μm and 63.09 μm),followed by the basic design group (9.38 μm and 50.56 μm) and the cross-arch bracing arm design group exhibited the smallest displacement (8.85 μm and 47.31 μm).Conclusion:The basic design Stern_ERA attachment for unilateral mandible distal extension defect appears to be more reasonable compared with rest arm and crossing-arch arm design from biomechanical point of view.
3.Research Progress in New Dosage Forms of Nevirapine

Nevirapine ( NVP) is the first-generation non-nucleoside reverse transcriptase inhibitor, which is a front-line drug for the treatment of AIDS and the prevention of mother-to-child transmission of HIV-1. NVP is with poor solubility and low bioavailability. With the development of new dosage forms, such as solid dispersion, self-emulsifying, microparticles, micelles, sustained-release tab-lets and floating microspheres, the bioavailability of NVP can be increased and the adverse reactions can be decreased. Research pro-gress in the new dosage forms of NVP at home and abroad was reviewed through searching literatures in order to provide basis for the development of new dosage forms of NVP.

5.Observation on early clinical efficacy of fixed platform and rotating platform in knee prosthesis arthroplasty for osteoarthritis of the knee

Objective To observe the early clinical efficacy of PFC fixed platform and RP rotating platform prosthesis two kinds of artificial knee replacement system for the treatment of knee osteoarthritis.Methods 39 cases (39 knees)with knee osteoarthritis were treated with total knee arthroplasty(TKA).According to intraoperative use of different types of prosthesis,the cases were divided into two groups:fixed platforms group 16 cases and rotating platform group 23 cases.The wound healing,patients with gait and knee activity after operation were observed.Results Three cases took place postoperative deep vein thrombosis,after conservative treatment,the symptoms improved and discharged,two cases of postoperative wound fat liquefaction,by dressing wound healing,time stitches discharged.3 to 12 months follow-up visit(average 10 months),all patients could walk uptight without noticeable limp.Last follow-up group,fixed platforms,rotating platform group Knee Score (HSS) and knee activity (ROM) were (83.17 ± 7.12),(81.32 ± 5.39) points and (109.73 ± 6.35) °,(103.12 ± 4.28) °,compared with the preoperative differencewasstatisticallysignificant (AgroupHSS:t =22.10,ROM:t =9.21 ; BgroupHSS:t =30.69,ROM:t =9.54,all P < 0.01),but the difference between groups was not statistically significant(HSS:t =0.88,ROM:t =3.63,all P > 0.05).Conclusion Using PFC and RP prosthesis after total knee replacement system in early has a good effect,both of which improve joint function early with no significant difference,but long-term effects need further observation.
6.Ultrashortwave irradiation for the repair of sciatic nerve injury and its mechanism

Objective To observe the effect of low doses of ultrashortwave therapy (USW) on sciatic nerve injury and to deduce its possible mechanism.Methods Fifty-four Sprague-Dawley rats were randomly divided into a USW group,a control group and a normal group with 18 rats in each.Each group was then sub-divided into 1 week,2 week and 3 week subgroups with 6 rats in each.A model of peripheral nerve injury was established by forceps clipping of the sciatic nerve in the USW and control groups.The USW group was then treated with USW exposure.Rats from the appropriate subgroups were sacrificed after 1,2 and 3 weeks of treatment.Sciatic nerve samples were stained using hematoxylin-eosin and toluidin blue.Expression of basic fibroblast growth factor (bFGF) was detected by immunohistochemical methods.Results Degeneration of axons was observed in both the therapy and control groups after 1 week,and regeneration at the end of the 2nd and 3rd weeks.The number of axons with myelin sheaths was significantly higher in the therapy group than in the control group at the end of the 2nd and 3rd weeks.The expression of bFGF was significantly higher in the USW group compared with the control group at all observation time points.Conclusion USW can obviously accelerate the regeneration of the sciatic nerve,probably through increased expression of bFGF.
7.Generation and application of three-dimensional cell traction force

BACKGROUND: Traction force generated by adherent cells exerts on the extracellular matrix or cell substrate through the interaction between actin and myosin, which involves a variety of physiological and pathological processes.OBJECTIVE: To review the fundamental principles and application of several traction force techniques.METHODS: The first author retrieved the databases of CNKI, WanFang, PubMed and SpringerLink for the research addressing the measurements of traction force using the keywords of cell traction force, biomechanics in Chinese and English, respectively. Repetitive articles were excluded, and 30 literatures were enrolled for analysis, including 1 Chinese and 29 English articles.RESULTS AND CONCLUSION: There are three kinds of measurements of traction force, such as silica gel membrane, traction force microscopy, and micromachined levers. The precise measurement of traction force contributes to further understand the cell-cell and cell-microenvironment mechanical interactions.

9.Research on efficiency by integrating method for international students' basic chemistry experiment teaching in medical colleges

Objective This study discusses the effect of integrating method in the international students' basic chemistry class of medical colleges.Methods The integrating method was composed of the new teaching methods,such as PBL and micro-learning resources,and traditional method.The experimental group was composed of 76 students from the Class 1 of international students of Grade 2015 in clinical medicine of China Medical University,where integrating method was used,namely the teaching methods and means of problem based learning and micro class were integrated into the experimental courses,while the control group was composed of 80 students in Class 2 from the same major and grade,which was using the traditional method of teaching and practicing.A unified examination was applied to evaluate the teaching efficiency in both two groups and the teaching effect of the experimental group was investigated by questionnaire.The data was entered into SPSS 13.0 for two independent samples t-test.Results In the exams the students in the test group got higher scores than that in the control group.The average scores were (17.6 ± 4.5) and (16.0 ± 4.8) respectively (t=4.42,P=0.00),the differences were statistically significant.The effective recovery rate of the experimental group was 100%,and among them,97.4% (74) of the students thought that they should continue to use the integrating teaching.Conclusion The application of integrating teaching in the course of basic chemistry experiment can improve students' comprehensive ability,and the effect is better than the traditional teaching.
10.Effect of four 5-hydroxytryptamine type 4 receptor agonists on rat cardiac Iκ1 channels and proarrhythmic risk stratification

OBJECTIVE To compare the effect of four 5-hydroxytryptamine type 4 (5-HT4) receptor agonists:cisapride,zacopride,macopride and 2-[1-(4-piperonyl) piperazinyl]-benzothiazole (BZTZ),on rat cardiac inward rectifier potassium channel (IK1)and heart rhythm.METHODS The whole-cell configuration of patch-clamp technique was used to record effects of 5-HT4 receptor agonists onIk1 in enzymatic dissociated rat ventricular myocytes or Kir2.1 transfected HEK 293 cells.Western blotting was used to observe the expression of Kir2.1 channel exposed 24 h to agents in ventricular myocytes.Langendorff-perfused hearts were perfused with four agents respectively for 30 min.The electrocardiogram was recorded simultaneously.RESULTS BZTZ,cisapride and mosapride 0.1-10 μmol· L-1 decreasedIk1 in a concentrationdependent manner.At the same concentration (1 μmol· L-1),BZTZ showed the most potent inhibition onIκ1 (P<0.01),followed by cisapride.Mosapride showed slight inhibition efficiency.However,zacopride enhanced Iκ1 (P<0.01).In Kir2.1 heterologous expression systems,zacopride activated Kir2.1 current (P<0.01) while mosapride had no effect.In ex vivo Langendorff-perfused hearts,BZTZ and cisapride 1μmol· L-1 elicited singnificant rhythm disturbances,and the total of premature ventricular beats (PVB) were 159±28 and 61±13.50% (4/8) (P<0.05) and 25% (1/8) of the hearts exhibited ventricular tachycardia (VT),while 37.5% (3/8) and 12.5% (1/8) of the hearts exhibited ventricular fibrillation (VF),respectively.Mosapride and zacopride had no side effects on heart rhythm.Zacopride also suppressed BZTZ-or cisapride-induced arrhythmias.BZFZ had the strongest proarryhthmic potency among the 5-HT4 agonists,followed by cisapride,mosapride and zacopride.CONCLUSION Iκ1 might be an independent risk factor for arrhythmogenesis and a new target for screening safe 5-HT4 receptor agonists and gastrointestinal prokinetic agents.
