1.A study on the expression of VEGF in ischemic rabbit hearts and its postmortem stability

Objective To observe the stability and the change pattern of VEGF expression in myocardium in varied postmortem periods,and to evaluate its usefulness in diagnosis of early myocardial ischemia after death.Methods An animal model of early myocardial ischemia was established by ligating coronary artery of rabbit,immunohistochemistry(SP protocol),image analysis technique and statistical analysis system are applied to detect and compare the area and intensity of expression of VEGF in cardiocytes,and its stability in different postmortem perieds.ResultsIn ischemic myocardium,VEGF shows foci or sheets of intense positive staining in cardiocytes,with negative or weak staining in some cells.There was weak staining in vascular endothelial cells and smooth muscle cells,but no staining in interstitium.When ischemic myocardium stored at 4℃,the staining of VEGF became weaker with time delay,but there was weak staining in a fraction of cardiocytes even 10d after death.There was significant difference in the intensity of VEGF staining between ischemic and normal postmortem myocardium from 1 to 10d.Conclusion VEGF can withstand a certain degree of autolysis or putrefaction,and the immunohistochemical staining of it is valuable in the diagnosis of early myocardial ischemia in corpses stored at 4℃ for 1~10d after death.

3.Effects of intravenous anesthetics on endotoxin-induced inflammatory responses in rabbits undergoing mechnical ventilation

Objective: To determine the effects of intravenous anesthetics (fentanyl,midazolam and propofol )on inflammatory responses. Method: We employed carrageenan-sensitized endotoxemic rabbit model. Thirty-two New Zealand rabbits were randomly assigned to one of four groups,control group(no anesthetic was used), fentanyl group, midazolam group and propofol group. Different anesthetics were used intravenously for anesthesia induction and maintenance according to the group classification. Meanwhile, all animals were mechanically ventilated, then, 2?g?kg~(-1) lipopolysaccharide(LPS) were injected for endotoxin challenge. Result:In all groups, serum tumor necrosis factor (TNF)activity began to increase at 30 min,reached its maximal level at 60 min,and tended to decrease at 120 min after (LPS) challenge,and the significant increase of TNF activity was associated with development of hypotension. Meanwhile,serum phospholipase A_2(PLA_2)activity began to increase at 60 min and persisted in going up within 2h. Those animals,with continuous infusion of these three different anesthetics and mechanical ventilation,had much lower peak level of TNF and higher mean arterial pressure (MAP)than control levels, but their changes of serum PLA_2 activity had no significant difference from control level. Conclusion: With mechanical ventilation, intravenous infusion of these anesthetics can reduce LPS induced injury in the acute phase of endotoxemia, probably by the inhibition of TNF production.
4.Study on the Correlation among the p53 Protein Expression, Tumor Biological Behavior, p53 Gene Mutation and EBV Infection in NPC

Objective To investigate p53 protein expression in nasopharyngeal carcinoma (NPC) and its correlation with biological behavior, p53 gene mutation and EBV infection in 43 cases of NPC. Methods Immunohistochemical staining was applied to detect p53 protein,PCR-SSCP was used to detect p53 gene mutation, and EBV infection was determined with PCR. Results The positive rate of p53 protein expression was 76 7%(33/43)in the NPC. It showed that the distribution of examination score was Gaussian distribution,the average score was 74.4,the paper reliability was 0.81,the difficult coefficient was 0.74 by analyzing and evaluating the quality of the examination paper of pharmacology from 2002 seven-year medical students in Central South University.Some objective questions are too difficult,while some subjective questions are too easy.According to the results of analysis,we think it better to culture the student's multi-ability in analyzing and solving questions by increasing the proportion of subjective question with some difficult coefficient and improving the teaching methods in future teaching.

7.Mitochondria respiratory chain related genes expression in post ischemia/reperfusion rat

AIM: Apply the newly invented method of suppression subtractive hybridization to scan the mitochondria genes expression of hippocampus neurons in post ischemia/reperfusion rats. METHODS: Decapitate the rats suffering 10 min of whole brain ischemia and 24 h of reperfusion in Pulsinelli’s 4 vessels occlusion ischemia/reperfusion model. Dissociated hippocampus and isolated mRNA. After reversed transcription, use SSH to subtract the common sequences, then use suppressed PCR to amplify the differentially expressed sequences. After cloning and DNA sequencing, the dot hybridization was used to finally identify the differential gene expression. RESULTS: In the 78 positive clones acquired from cloning, 7 clones represent differential gene expression after confirmed by clone PCR, dot blot and DNA sequencing. The DNA sequencing data indicate these clones are ESTs of Cytochrome c oxidase subunit and NADH-ubiquinone oxidoreductase chain 4 gene. CONCLUSION: Cytochrome C oxidase mRNA level changs during brain ischemia /reperfusion. The differential expression of NADH-ubiquinone oxidoreductase chain 4, one of those 13 subunits of subcomplex ?, suggests that subcomplex ? may have important function associated with pathophysiologic changs in reperfusion injury.
8.Research progress of bone filling materials in percutaneous vertebroplasty

BACKGROUND: As a minimally invasive technology, percutaneous vertebroplasty is a safe and effective treatment for osteoporotic vertebral compression fractures.OBJECTIVE: To overview the research progress concerning the biomechanical properties, bone strength maintenance, bone absorption and degradation of bone filling materials used in percutaneous vertebroplasty.METHODS: The first author conducted a computer-based retrieval of CNKI, PubMed and Medline databases for relevant articles published from January 2005 to May 2016. The keywords were bone cement, bone filling materials, percutaneous vertebroplasty in English and Chinese, respectively.RESULTS AND CONCLUSION: Polymethyl methacrylate is not an ideal material for osteoporotic vertebral compression fractures. Calcium phosphate cement and calcium sulfate cement can replace the traditional polymethyl methacrylate; however, some problems still exist, such as poor effect of venography, incontrollable biological degradation rate, and lack of the evidence-based medicine about its long-term effect. Composite bone cement, as a good bone repair material, holds the advantages of various bone cements. As the composite bone cement has just been introduced in clinical practice, its long-term curative efficacy needs to be further studied.
9.A Design of Multi-parameter Smartphone Monitoring Interface Based on J2ME

Purpose: This paper introduces a design of wearable multi-parameter emluator experiment smartphone monitoring interface based on J2ME. Methods: The system carries in simulated environment of Wireless Toolkit, and the remote monitoring of multiple physiological parameters is implemented with wearable detecting technology and GPRS. Results: The system can measure real-time data such as ECG, heart rate, blood pressure, body temperature and oxygen saturation, which can also analyse the data automatically and possess warning functions. At last, the system can send parameters to intelligent handheld devices, and can establish communication with medical service center. Conclusions: The experimental result of simulation shows that the system has the excellence of transplantable, simpleness and quick speed of response. It shows that the system is expected to realize the important application of medicine in 3G era and to provide a basis for further research in telemedicine.
10.Gene expression profile of neutrophils in system inflammatory response syndrome patients

AIM: To study the difference in gene expression of peripheral blood neutrophils between system inflammatory response syndrome(SIRS) patients and the health volunteers.METHODS: cDNA microarray was used to detect the mRNA from peripheral blood neutrophils in both 4 patients with SIRS patients and 6 health volunteers.RESULTS: Among 8 400 genes,there were significant differences in 382 gene expression,of which 122 expressed higher and 260 lower in SIRS patients than those in healthy volunteers.The differently expressed genes consisted of intracellular signaling transduction genes(35%),cell receptor genes(22%),genes of DNA binding or transcriptional factors(23%)and genes of cytokines(11%).CONCLUSION: Our results suggests the changes in gene expression of polymorphonuclear neutrophils in SIRS may be more complicated than those in vitro experiments,and associated with the effect produced by pro-inflammatory and anti-inflammatory factors.
