1.The establishment of a Tet-on transfected human hepatoma HepG2 cell line regulated by doxycycline

Objective To establish human hepatic carcinoma HepG2 cell line in which the expression of exogenous gene could be controlled by doxycycline. Methods The expression plasmid pTet-on was transfected into hepG2 cells by lipofectamine 2000.The stably transfected positive clones were screened with G 418 and subcultured. The expression level of luciferase induced by doxycycline at a concentration of 1 ?g/ml in stably transfected cells was detected one by one by transient transfection of plasmid pTRE-luc into the stably transfected cells. Results The basal expression activity of luciferase in cells of clone No 6 was 87, and the activity was 16764 after induction with doxycycline. Conclusion The HepG2/Tet-on cell line was a regulatory human hepatoma cell line, which is useful for the study of carcinogenesis and gene therapy of hepatic carcinoma.
2.Application of Acupoint Massage Plus Body Massage for Intervention of Premature Pathological Jaundice

Objective To observe the intervention effect of premature pathological jaundice treated with conventional blue light phototherapy combined with acupoint massage plus body massage. Methods Ninety premature infants with pathological jaundice were randomized into control group 1, control group 2 and observation group, 30 cases in each group. The control group 1 was given blue light phototherapy plus oral use of vitamin B2, the control group 2 was given body massage based on the treatment for the control group 1, and observation group was given acupoint massage based on the treatment for the control group 2, the treatment for the three groups lasting for 7 continuous days. Before and after treatment, the levels of transcutaneous bilirubin (TCB), serum total bilirubin (TB), and serum direct bilirubin (DB) were observed, and then the therapeutic effects on jaundice in the three groups were compared. Results After treatment, the overall therapeutic effect and the daily decline of TCB, TB and DB in the observation group were superior to those in the two control groups, the difference being significant (P < 0.05 or P < 0.01). In the control group 2, the post-treatment average decline of TB and the decline of TCB on treatment day 4-7 were superior to those in the control group 1, the difference being significant(P<0.05). Conclusion Acupoint massage plus body massage is effective and safe for the treatment of pathological jaundice in premature infants, and the therapy being simple and practical.
3.10-Item Connor-Davidson Resilience Scale:the relationship between the level of resilience and emotional disturbance stress burden of the clinical nurses

Objective To investigate the relation among clinical nurses′resilience, emotional, the pressure load and self-efficacy with the 10-item Connor-Davidson Resilience Scale. Methods A total of 449 nurse in Guangzhou were recruited to investigate by 10-item Connor-Davidson Resilience Scale (CD-RISC-10), Kessler 10 Scale (K10), General Self-Efficacy Scale (GSES), Chinese Stress Overload Scale (SOS). Results The average score was 23.6±6.7 for CD-RISC-10 , 24.4±8.2 for K10,24.7±5.7 for GSES , 63.3 ±16.6 for SOS ,and the individual vulnerability scoring was (31.2±10.0) points, events load was (32.0±7.5) points. High mental toughness of nurses in general self-efficacy scores higher than that of low mental toughness group (t=-8.53, P<0.01), in the K10 dated, S0S scores were lower than low mental toughness group (t=11.23, 7.70, P<0.01). Nurse relevant regression analysis results showed that gender, general self-efficacy, K10 dated, SOS as the main factors influencing nurses mental toughness, could explain 40%of variances. Conclusions The CD-RISC-10 can better reflect the level of clinical nurses′resilience, and the gender, self-efficacy were the main effects, as well as effectively predict the nurses′emotional distress and stress level. However how to improve the toughness of the clinical nurses need more research.
4.Effect of Five-tone Music Therapy on Anxiety, Depression and Quality of Life of Patients Treated with Chemotherapy After Gastric Cancer Radical Surgery

Objective To observe the effect of five-tone music therapy on relieving anxiety and depression and on improving the quality of life of patients receiving chemotherapy after gastric cancer radical surgery.Methods Ninety anxiety and depression in-patients treated with chemotherapy after gastric cancer radical surgery were randomly divided into observation group and control group,45 cases in each group.Patients of the two groups were treated with the same chemotherapy regimen and nursing,and additionally,the observation group was given fivetone musie therapy and the control group was given conventional music therapy.The intervention effects were evaluated by scoring with Self-rating Anxiety Scale (SAS),Self-rating Depression Scale (SDS) and questionnaire for the quality of life of gastric cancer patients (QLQ-STO22) at admission time and when discharging from hospital.Results The SAS and SDS standard scores and the scores of each dimension of QLQ-STO22 in the observation group were markedly decreased after intervention,the differences being statistically significant (P < 0.01 compared with those before intervention).And the SAS and SDS standard scores of the observation group after intervention were lower than those of the control group (P < 0.01).In the aspects of quality of life,the scores of all dimension of QLQ-STO22 except taste,appearance and hair loss in the observation group were lower than those in the control group (P < O.05).Conclusion Five-tone music therapy is effective on relieving anxiety and depression and on improving the quality of life of patients receiving chemotherapy after gastric cancer radical surgery.
5.Correlation of contrast-enhanced ultrasonography with microvessel density and vascular endothelial growth factor expression of beast tumors

ObjectiveTo determine the correlation of real-time contrast-enhanced ultrasonography (CEUS) patterns and quantitative parameters with microvessel density (MVD) and vascular endothelial growth factor (VEGF) expression of breast tumors.Methods One hundred and five patients with 128 breast lesions(62 benign,66 malignant) underwent CEUS examination.CEUS patterns were analyzed and parameters were obtained by time-intensity curve analysis software.Immunohistochemical staining using anti-factor CD34 was performed to evaluated the MVD and VEGF expression was detected.Results All CEUS patternsweresignificantlyassociatedwithMVDandVEGFexpressionexceptboundary characteristics( P < 0.05).The enhancement parameters showed that the time to peak was significantly faster in malignant tumors than that in benign lesions,and the peak intensity and area under the curve were significantly higher in malignant tumors than those in benign lesions( P <0.05).The time to peak,the peak intensity,area under the time-intensity curve were statistically correlated with MVD (P <0.05),the highest correlation was between the area and MVD( r =0.81,P <0.001).Only the area under the timeintensity curve was significantly associated with VEGF expression (P < 0.05).Conclusions CEUS patterns and parameters of breast lesions are closely correlated with MVD,which can be used to evaluated the angiogenesis in breast tumors.
6.Research on psychological resilience and its influencing factors among parents of children with cancer

Objective To probe into the psychological resilience and its influencing factor among parents of children with cancer.Methods A total of 125 parents were investigated by self-designed general information questionnaire,Connor-Davidson psychological resilience scale (CD-RISC),Chinese version of Parents' Perception Uncertainty in Illness Scale (PPUS),Social Support Rating Scale (SSRS),self-rating depression scale (SDS) and self-rating anxiety scale (SAS).Results The total score of psychological resilience was (54.86±12.88) and the illness uncertainty score was (91.52±14.66).The social support score was moderate with a total score of (39.86±7.99).The anxiety and depression among parents got a total score of (40.08±7.19) and (44.61±7.38) respectively.Psychological resilience was negatively correlated with illness uncertainty and depression,and was positively correlated with the social support.The depression,illness uncertainty,residence,children's age,clinical diagnosis entered into the multiple linear regression equation of psychological resilience,predicting 30.1% of total variation in psychological resilience.Conclusions Social support,illness uncertainty and depression are the important factors which affect psychological resilience among parents of children with cancer.Nurses should assess the social support that parents receive,the level of depression and the disease feeling of the parents,and provide targeted interventions to relieve it,help to establish effective coping style and improve their psychological state.
7.Correlation between caregivers′burden and the coping style among parents of cancer children

Objective To investigate the level of caregivers′burden and the coping style among parents of cancer children. Methods Totally, 229 parents of cancer children participated in the investigation by Chinese version of zarit burden interview (ZBI), Chinese version of coping health inventory for parents (CHIP) and self-designed general information questionnaire. The associations between caregivers′burden and coping styles were tested by Spearman correlation analysis. Results The caregiver′s burden on the parents was in the middle level with a total score of (30.50 ± 12.24). The coping style the parents took most frequently was to unite the family and keep a positive attitude and regard it as most effective. The caregiver′s burden was negatively associated with the positive coping style . Conclusion The parents of cancerous children suffer from middle level of burden . Nurses should assess the caregiver′s burden, provide targeted interventions to relieve it, help the to establish effective coping style and change their psychological and mental state and ultimately improve their quality of life.
8.Expression of AMP-activated protein kinase in subcultured rat endplate chondrocytes

BACKGROUND:Endplate cartilage degeneration initiates intervertebral disc degeneration. AMP-activated protein kinase (AMPK) regulates the formation and degradation of cartilage. OBJECTIVE:To explore the role of AMPK in an in vitro natural degeneration model of chondrocytes derived from endplate of rat intervertebral discs. METHODS:Morphology of in vitro subcultured endplate chondrocytes of rat intervertebral discs at passages 0, 2, and 5 were observed under an inverted microscope fol owing cytoskeleton staining. Chondrocyte phenotype, proliferation, and the cartilage marker genes (type II col agen, proteoglycan, SOX-9, matrix metal oproteinase-3 and-13), and AMPK phosphorylation were determined by toluidine blue staining, MTT assay, real-time PCR analysis, and western blot assay, respectively. RESULTS AND CONCLUSION:The altered morphology, decreased proliferation ability, and phenotype loss were observed in chondrocytes with increased passage number. Gene expression of type II col agen, proteoglycan, SOX-9 was significantly decreased;while gene expression of matrix metal oproteinase-3 and-13 was significantly increased in endplate chondrocytes at passage 5 compared with those at passages 0 and 2. AMPK phosphorylation in endplate chondrocytes at passage 5 was significantly decreased. These findings indicate that AMPK phosphorylation is involved in in vitro natural degeneration of chondrocytes derived from the endplate of rat intervertebral discs, and the degeneration of endplate chondrocytes and intervertebral discs can be inhibited through the regulation of AMPK activity.
9.Role of Estrogen Receptor-alpha in the Regulation of Claudin-6 Expression in Breast Cancer Cells.

PURPOSE: In our previous studies we showed that upregulating claudin-6 (CLDN6) expression may contribute to preventing breast cancer, and that 17beta-estradiol induces a concentration- and time-related effect on CLDN6 mRNA and protein expression in MCF-7 cells. However, the mechanisms of 17beta-estradiol regulation of CLDN6 are still unclear. We determined the role of estrogen receptors in the regulation of CLDN6 expression in human breast cancer tissues and a cell line. METHODS: CLDN6, estrogen receptor alpha (ERalpha) and estrogen receptor beta (ERbeta) expression in breast cancer tissues were examined using immunohistochemistry. The human breast cancer cell line, MCF-7, which expresses ERalpha but not ERbeta was used. CLDN6 and ERalpha expression were measured by reverse transcriptase-PCR, Western blotting and immunofluorescent staining. Treatments with propyl pyrazole triol (PPT) and ICI 182, 780 (ICI) were performed. RESULTS: The results revealed that CLDN6 expression was related to ERalpha in breast cancer tissues (p=0.033). PPT, an ERalpha-selective ligand, upregulated CLDN6 expression at 10-5 mol/L after 24 hours. The effect of PPT on regulating CLDN6 expression in MCF-7 cells was blocked by ICI. CONCLUSION: These findings suggest that Eralpha reulates CLDN6 expression in breast cancer tissues and that 17beta-estradiol induces CLDN6 expression through an ERalpha pathway in MCF-7 cells.

10.Preliminary discussion on the core post competency of nursing students in nursing practice by WeChat official account

Objective:To investigate the application and effects of WeChat official account in improving the nursing trainees' core post competency.Methods:The study included in 105 nursing trainees of Batch 2015 as experimental group, taking the nursing practice WeChat official account based on the routine clinical teaching pattern, and 95 nursing trainees of Batch 2014 as control group, taking traditional teaching mode. At the end of the internship, the scores of special examinations for nursing students and the national examination for nursing qualifications ("nursing qualification examination" for short) of the two group were compared, and the critical thinking ability, ability to acquire information lifelong learning ability, nurse-patient clinical communication ability and teaching satisfaction of the two groups of students were analyzed. SPSS 23.0 software was used for t test. Results:The scores of special examinations, nursing qualification examination and each module at main dimensions of the core post competency of nursing students in the experimental group were higher than those in the control group ( P<0.05). The nursing trainees in the experimental group were satisfied with the teaching effects of the WeChat official account. Conclusion:The WeChat official account in clinical nursing practice helps improve the quality of clinical practice teaching and assist in enhancing core post competency of nursing trainees.
