1. Role of peripheral lymphocytes Sialyl Lewis(X) (CD15s) antigen before and after kidney transplantation

Objective: To investigate the role of peripheral lymphocytes Sialyl Lewis(x) (CD15s) antigen before and after kidney transplantation. Methods: Flow cytometry technique was applied to examine the expression of peripheral lymphoid cell surface CD15s antigen after renal transplantation, and to evaluate various therapeutic regimen. Results: The statistic analysis results of peripheral lymphoid cell surface CD15s antigen expression level showed that there was significant difference among the patients with acute rejection, long-term dialysis and with normal renal function post-transplant; significant difference of CD15s expression level between group of rejection and infection; no significant difference of CD15s expression among the different groups treated by various therapeutic regimens. Conclusion: The different therapeutic regimen has no influence to CD15s expression; Detection of peripheral lymphoid cell surface CD15s antigen expression periodically, intelligently make convenience to understand suitable status of immunosuppression.

Objective To investigate the safety and clinical significance of total thyroidectomy performed for benign thyroid disease. Methods Eighty-eight patients with benign thyroid disease were treated with total thyroidectomy. The postoperative complications were analyzed. 3.Determination of the Content of Meropenem and Observation of the Stability of Meropenem in Mixing With Infusion Fluids

OBJECTIVE:To establish a HPLC method for determining the content of Meropenem and to observe the stability of Meropenem in mixing with 5%GS,GNS and NS.METHODS:The conditions of HPLC were:column,Nova-Pak C18;mobile phase,acetonitrile-0.05mol KH2PO4(4∶96);flow rate,1.0ml/min;detection wavelength,300nm.After Meropenem mixing with 5%GS,GNS and NS,the changes of external apperance were observed in room temperature,the content was detected with HPLC and the pH and insoluble particles were determined.RESULTS:The calibration curve was in good linearity in the range of 12.5~250?g/ml(r=0.9 999)and the recovery was 99.7% with RSD=1.1%.CONCLUSION:This detection method is effective,rapid and accurate.Meropenem is stable in infusion fluids for 4 hr.

By reviewing documents and learning from the Dai medicine teachers, the papertried to show the theory and methods of Dai medicine on diagnosing and treating Rheumatoid arthritis (RA). Dai medicine took a note that RA was caused by the unbalance of the interal Sita and affected by the external Paya Longgananhuang. Those pathogeny can make the partial joint and muscle's function abnormal, which resulted in the RA. RA can be separated into three types which are Longmengshahoufeixiang. They could be treated by boththe internal and external treatments. The etiology and pathogenesis of RA and its diagnosis and treatment methods of traditional Dai medicine were profound, unique and useful. It`s worth of clinical application and further researches.
5.Determination of Ephedrine in KMBYL Capsules by HPLC

[Objective] To establish a method for determination of Ephedrine Hydrochloride in KMBYL capsules by HPLC.[Methods] The DiamonsilTM C18(250mm?4.6mm,5 ?m)column was used.The mobile phase was mobile phase: acetonitrine-0.02 mol?L-1potassium dihydrogen phosphate(0.2% triethylamine,adjusteding pH to 2.7 with phosphoric acid)(5∶95) with a flow rate of 1.0ml?min-1.The detection wavelength was 210 nm.The column temperature was 30℃.[Results] The linear responses range was 0.2148-2.148 ?g?mL-1 of Ephedrine Hydrochloride(r=0?9999).The average recovery was 98.85% and the RSD was 2.95%.[Conclusion] The method is simple accurate reproducible.It can be used for the quality control of KMBYL Capsules.
6.Adenovirue-mediated human p27kip1 gene transfection inhibits neointimal proliferation of rabbit carotid artery after balloon injury

Objective To study the effects of adenovirue-mediated p27kipl gene and its protein product overexpression on neointimal proliferation of rabbit carotid artery after balloon injury. Methods After rabbit arterial carotid injuried, injuried arterial segments were immediately infected by LacZ recombinant adenovirues (AdLacZ) and human p27kipl recombinant adenovirues (Adhp27kip1) in vivo, respectively. Western blot, x-gal stainning, HE stainning, immunochemistry and compute image system were used to analyze the expression of exogenous p27kipl gene and its protein product and the effects on neointimal proliferation. Results Comparison to AdLacZ infected or uninfected arterial segments, Adhp27kip1 infected segments overexpressed p27kipl protein, the peak of expression was in 7-14 d, expression lasted more than 4 weeks. After 4 weeks, in uninfected, AdLacZ infected or Adhp27kipl infected segments, the neointimal area was 1.106 mm2, 0.988 mm2 and 0.278 mm2, respecively; the rate of luminal stenosis was 87.07%, 65.40% and 32.14%, respectively. Conclusion Exogenous p27kipl gene and its protein product overexpression in injuried arterial segments could inhibit neointimal proliferation and luminal stenosis significantly after artery injury.
7.Expression of ICAM-1 and VCAM-1 in human chronic renal allograft rejection

To study the mechanism of human chronic renal allograft rejection, kidney tissues were taken from 16 patients with chronic renal allograft rejection and from 5 healthy subjects, and underwent the frazed section staining for ICAM-1 and VCAM-1 to anti-ICAM-1 and anti-VCAM-1 respectively by using immunohistochemistry(ABC).The results showed that there were differ-ent distribution of ICAM-1 and VCAM-1 expression in nomal kidney and renal allograft during chronic rejection.It was suggested that ICAM-1 and VCAM-1 might play an important role in the pathogenesis of human chronic renal allograft rejection.
8.Clinical research of renal transplant from donation after citizen's death (DCD) donor with acute kidney injury

Objective To explore the clinical effect of renal transplant from donation after citizen's death (DCD) donors with acute kidney injury (AKI).Methods This was an observational retrospective study of 622 patients who underwent renal transplantation from 312 DCD donors' kidneys at the First Affiliated Hospital of Xi'an Jiaotong University from December 2011 to December 2016.The transplant patients were divided into AKI group and non-AKI group according to the Acute Kidney Injury Network (AKIN) criteria based on initial and terminal creatinine values.We evaluated and compared transplant outcomes of these two groups.Results There were 131 donors with AKI,and the incidence of AKI was 42.0 %.AKI group and non-AKI group recipients respectively had DGF in 20.2% and 7.2% of cases (P<0.01),153.6 ± 56.2 and 119.3 ± 40.7 μmol/L of serum creatinine (SCr) levels at 1st month (P<0.01),and 38.5 ± 14.1 and 57.6 ± 23.4 ml· min-1 (1.73 m2)-1 of eGFR at 1st month (P<0.01).There was no significant difference in SCr and eGFR between two groups at 1st year after transplantation.Conclusion Most of kidneys from DCD donors with AKI can be considered for transplantation.Renal transplantation of organs from DCD donors with AKI showed greater DGF but good outcomes.
9.Response of EDAG knockout mice to low-dose radiation-induced damage

Objective To generate the erythroid differentiation associated gene(EDAG) knockout mice and analyze their sensitivity to low dose radiation-induced damage.Methods Zinc finger nuclease technology ( ZFNs ) was used to produce the EDAG knockout mice.The low dose radiation-induced damage was evaluated by peripheral blood cell counts, DNA damage and colony formation of bone marrow cells.Wild-type and EDAG knockout mice were irradiated with 0.31 Gy/min X-ray, one minute per day for seven consecutive days, and the cumulative radiation dose was 2.17 Gy(n=7).The blood cell counts were measured by an automated hemocytometer.DNA damage was detected by immunofluorescence assay with a DNA damage marker p-H2A.x antibody (n=3).The colony formation ability of bone marrow cells was evaluated with a semi-solid culture medium(n=3).Results A model of EDAG knockout mice was established.Compared to wide type mice, white blood cell counts of EDAG knockout mice decreased significantly while the DNA damage marker p-H2A.x expression was increased on the third day after X-ray irradiation.The ability of colony-forming was reduced after 7 days of X-ray irradiation.Conclusion Our present study found that EDAG knockout mice are more sensitive to low dose radiation-induced damage as shown by decreased peripheral blood cells counts, reduced colony-forming ability of bone marrow cells, and increased DNA damage.These results suggest that EDAG knockout mice can serve as a powerful tool for evaluation of the biological effects of low-dose radiation damage.
10.The cell-mediated immune status of patients with cytomegalovirus pneumonia after renal transplantation

Objective To evaluate the clinical value of adenosine triphosphate (ATP) determination in CD4+ cells in cytomegalovirus pneumonia after renal transplantation.Methods The ATP level of CD4+ T cells was measured by ImmuKnowTM kit.The ATP levels were determined in 187 renal transplant recipients before and 30,60,90,180 days after operation,and at the time of CMV pneumonia and 4 weeks after treatment of CMV pneumonia.The associations between ATP levels and CMV pneumonia were analyzed.Analysis of variance (ANOVA),Pearson-Spearman and relative risks were used for data analysis.Results 17 cases out of 187 renal transplant recipients were diagnosed as CMV pneumonia (9.1%),and the onset of CMV pneumonia started on the (2.8 ±1.2)month after renal transplantation.ATP concentrations in CD4+ T cells were significantly lower after operation than those before operation (P<0.01).ATP concentrations reached the lowest on the about postoperative day 90 (P<0.05),then increased gradually.In 17 recipients with CMV pneumonia,the ATP levels before and 30,90 days after operation,at the time of CMV pneumonia and 4th week after treatment of CMV pneumonia were (376 ±182),(283 ± 146),(196 ± 112),(145 ± 102) and (236 ± 117) μg/L respectively.ATP levels at the time of CMV pneumonia were significantly lower than any other time points (P<0.05).There was close correlation between ATP levels and CMV pneumonia.Conclusion The determination of ATP in CD4+ cells could reflect the status of cell-mediated immunity in renal transplant recipients,and could evaluate the severity and prognosis of CMV pneumonia and guide the clinical treatment.
