1.Content Determination of Triptolide in Tripterygium Wilfordii Cut Crude Drugs by HPLC

OBJECTIVE:To develop a RP-HPLC method for the determination of triptolide in tripterygium wilfordii cut crude drugs.METHODS:Samples were extracted with chloroform and acetic ether by ultrasound and underwent neutral alu-mina column elution and isolation,the content determination was performed with acetonitrile-water(23∶77)as mobile phase at a flow rate of1.0ml/min and detection wavelength at218nm.RESULTS:The linear detection concentration range of trip-tolide alcohol was2.74~96?g/ml(r=0.9999),and the average recovery were94.78%(RSD=1.57%)and98.2%(RSD=1.7%),respectively.CONCLUSION:Both methods are applicable for the quality control of triptolide alcohol,yet the separation and determination outcome were better for the sample treated with acetic ether.
2.Changes and Significance of Patients′ Blood Routine and T lymphocyte Subsets with Lymphoma and EB Virus Infection

Objective To investigate the changes and significance of patients′ T lymphocyte subsets with lymphoma and EB virus infection.Methods 57 patients with malignant lymphoma who were admitted to our hospital between January 2015 and January 2017 were divided into A group (with EB infection, n=31) and B group (without EB infection, n=26).The other 30 healthy cases were selected as control group (group C) at the same time.The fasting venous blood of all subjects was collected, and the levels of T lymphocyte subsets were measured by flow cytometry.Results Compared with group C, the levels of CD3+, CD4+ and CD4+/CD8+ in A group and B group were significantly lower while the levels of CD8+ were significantly higher (P<0.05).Compared with B group, the levels of CD3+, CD4+ and CD4+/CD8+ in A group were significantly lower while the levels of CD8+ were significantly higher (P<0.05).Conclusion The levels of T lymphocyte subsets can be used as important indexes to judge the severity of lymphoma.When there is severe T lymphocytes immune dysfunction, EB virus infection should be considered, and timely diagnosis and early treatment should be carried out.
3.Glaucoma gene therapy:obstacles and challenges

Trabecular meshwork,ciliary body,retina and optic nerve will be the targets for glaucoma gene therapy.The strategies include lowering intraocular pressure,neuroprotection and preventing wound healing in glaucoma filtration surgery. Overexpression of certain gene(s)in trabecular meshwork to increase the resistance to aqueous humor outflow would be an ideal method to create an animal model of glaucoma.Even though great progress has been made in this field,significant challenges and obstacles, including lack of tissue specific promoter,problems in long-term gene expression and side effects,still remain to be resolved.
4.The influence of cold provocation on retinal light sensitivity of patients with primary open-angle glaucoma

Objective To evaluate the effects of cold provocation on optic dise blood flow and retinal light sensitivity of primary open-angle glaucoma (POAG) patients,and explore the relationship between the changes of optic disc blood flow and retinal light sensitivity. Methods A total 33 POAG patients (33 eyes) and 13 normal controls (13 eyes) were tested by using the Heidelberg retinal flowmetry (HRF) and Topcon automatic perimeter,and the optic disc blood flow and retinal light sensitivity of POAG patients and normal controls were measured at normal conditions and after cold provocation. Results The mean optic disc blood flow,volume and the mean retinal light sensitivity of POAG patients decreased significantly (P<0.05) after cold provocation.There was a linear and significant relationship between the decrease of mean optic disc blood flow and mean retinal light sensitivity of POAG patients (r=0.615,P<0.001).The optic disc blood flow of POAG patients with a history of migraine were more likely to reduce in response to cold provocation and reduced much more than those without such history (P<0.05). Conclusion Cold provocation can significantly reduce both the optic disc blood flow and retinal light sensitivity in POAG patients.A close correlation was found between the amount of mean optic disc blood flow decrease and the volume of mean retinal light sensitivity decline.
5.Influence of thymus transplantation on T cell subsets of peripheral blood in patients with allergic rhinitis

Cultured human embryonal thymus graft was performed on 80 patients with allergic rhinitis to observe the immune function of the patients and the treatment efficiency. The thymus from the fetus of intermediate abortion was cultured for 7 to 10 days. The T cell subsets and serum antigen special IgE(sas IgE) level of the patients were determined before and after the thymus transplantation. The gross treatment efficiency is up to 92.4% among the trial patients after graft for 3 years and CD8 positive cells were increased with a significantly statistical difference (P<0.01), and sas IgE level was much lower than before thymus transplantation (P<0.01). It was concluded that the thymus transplantation might regulate the immune function of the patients with allergic rhinitis, including increases in the number and the function of Ts cells, and a decrease of IgE level.

Object To develop a method for the separation and purification of polyprenyl acetates from the leaves of Ginkgo biloba L Methods Lipids, including polyprenyl acetates were extracted by petroleum ether, and adsorbed by silica gel to remove most of the polar impurities After removing wax from the partially purified extract, it was further purified with silica gel chromatography Results Finally, a mixture of homologues polyprenyl acetate was obtained The purity of which as determined by HPLC external standard method was 84% Conclusion This procedure is simple and relatively effective
7.Pathological study of uterine fibroids after transcatheter uterine artery embolization

Pathological changes of uterine fibroide after transcatheter uterine artery embolization(TUAE) has been proven with regular pathological change during the elapse time after TUAE resulting in fibrotic proliferation. Inflammation took place first, followed by coagulation necrosis and hyaline degeneration but incomplete in most cases and finally with proliferative fibrous tissue formation. These dynamic changes of pathology are basis for fibroid volume reduction and recurrences. The factors affecting the extent of fibroid necrosis included tumor size,position and elapse of duration after TUAE. Inflammation was found in normal uterine tissue after TUAE but no necrosis occurred. The finding of embolization agent in peripheral vessels of adnexa may suggest future possible damage of function in ovary and fallopian tube. (J Intervent Radiol, 2006, 15: 507-509)
8.Current status, questions and challenges of transcatheter uterine artery embolization for the treatment of uterine fibroids

Current status, questions and challenges of transcatheter uterine artery embolization(UAE)in the treatment of uterine fibroids were summarized and analysed. It has been proved that UAE presents a good effectiveness in controlling the symptoms and shrinkage of fibroid and uterine volumes during follow-up of 4 to 6.9 years domestically and abroad, but relapse of the fibroid may however occur in 2 years or longer after UAE. Generally speaking,UAE is safe in the treatment of uterine fibroids but has a possibility of serious complications. UAE has no damage on normal uterine tissues but may affect pregnancy and delivery of patients significantly later on the cause of hypoxia and inertia of uterus. UAE may cause amenorrhea in the minority of women with ovarian failure and endometrium atrophy. The current questions are how to improve long-term efficiency to reduce relapse of tumor and to insure the safety of UAE. It is our further task to exploit more new effective and safe embolic agents by using animal and clinical study on the basic knowledge of pathology, pharmacology, biochemistry, endocrinology and molecular biology. 9.Applications and advances of hepatic arterial infusion chemotherapy in surgical treatment of hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is one of the most common malignancies in China, which serious threat to life and health. Surgical treatment of HCC is one of the most important therapeutic options for long-term survival of HCC patients, including hepatectomy and liver transplantation. Hepatic arterial infusion chemotherapy (HAIC) infused the therapeutic agents directly into the hepatic artery, the main blood supply in HCC, providing higher concentration and reaction rate. HAIC is an effective treatment for hepatectomy of downstaging and adjuvant therapy, which can improve the feasibility of hepatectomy and decrease the recurrence rates after resection. In this paper, we will review the applications and advances of HAIC in surgical treatment of HCC.
10.Expression of TrkA in Wistar rat brain during embryonic phase

Objective To detect the expression of TrkA in the brain of the Wistar rats during the embryonic phase.Methods Adult Wistar rats were fed in one cage.It was E0 day when sepermia were found in vaginal suppository and vaginal smear.The expressions of TrkA in the brain of Wistar rats during various brain regions were observed by immunohistochemistry on E13,E15,E17,E19 and E21 day. Results On E13 day no obvious expression of TrkA was observed in the embryonic brain.The expression of TrkA had an obviously increasing tendency in the cortex of telencephalon from E15 day to E17 day.And on E17 day,the positive rate of the expression of TrkA reached the second peak.While on E19 day,the expression of TrkA had a temporary decreasing.Then on E21 day the expression of TrkA reached the highest peak.The striatum and hippocampus developed later than the telencephalon till E17 day,and the expressions of TrkA during these stages were same as the cerebrum.Conclusion From E15 to E17 day,obvious expressions of TrkA are observed in the embryonic telencephalon,and it accords with embryonic induction.
