objective: To develop injectable tissue engineered bone through injection of osteoblasts/alginate composite in rabbits. Methods: Bone marrow cells isolated from iliac bone of New Zealand rabbits were cultured and induced to differentiate into osteoblasts. The osteoblasts were mixed with 25 g/L sodium alginate solution to generate osteoblasts/alginate composite with final cellular density of 5?10 6/ml. 0.17 g of sterilized CaSO 4 powder was then added to 2 ml osteoblasts/alginate. The mixture was injected into the dorsal subcutaneous tissue at left side of 6 New Zealand rabbits. The alginate composite without osteoblasts was injected into the right side as the control. 4 and 8 weeks after implantation, the bone formation was evaluated by means of gross, X ray and histological observation. Results: 4 weeks after implantation, cartilage formation was observed and 8 weeks after implantation, new mature bone was observed in the osteoblasts/alginate composites. No new bone formation was observed in all of the control specimens. Conclusion: Calcium alginate can be used as a carrier in injectable bone tissue engineering, and new bone can be created through injection of osteoblasts/alginate composites in immune animals.

Objectives: To study the treatment outcome of mucoepidermoid carcinoma(MEC).Methods:128 cases of MEC with definite pathologic diagnosis treated in our hospital during the past 15 years were investigated. The relationship between the effects of the therapy and the prognosis was analysed.Result:Treatment with surgical operation only was used in 99 cases, radiotherapy after operation was applied in 26 cases and follow-up was carried out in 76 cases. 5-year survival rate was 90.7%.5 patients who died within 5 years after operation were all with the diagnosis of MEC in clinical stage Ⅳand poor differentiaton. Conclusion:The local extensive excision for the well differentiated MEC,the radiotherapy after operation for poorly differentiated or that of clinical stage Ⅳ,especially in maxilla, may increase 5-year survival rate.Since there were cases of recurrences of MEC 13 years after surgical treatment,long-term follow-up is necessary.

Objective To improve the diagnosis and differential diagnosis of adrenal incidentalomas for the optimal management of adrenal masses. Methods The clinical data of 52 cases (35 men and 17 women;mean age,42 years;age range,23-65 years) of adrenal incidentalomas was reviewed.Among them, 33 cases had tumors ≥ 3.0 cm in diameter.None of 52 cases had clinical symptoms and signs of hypertension and endocrine dysfunction. All of the tumors were incidentally found by ultrasonography or CT on regular physical examination.A series of blood chemical examinations (including electrolytes, adrenal cortical and medullary metabolic biochemistry),abdominal ultrasound and CT were performed in all 52 cases;and MRI,PET and 131 I-MIBG imaging were performed in 6,2 and 1 cases, respectively.Surgery and postoperative pathological study were performed in all. Results Postoperative pathology showed adrenocortical adenoma in 27 cases, adrenal metastatic carcinoma in 7,adrenal pheochromocytoma in 5,adrenal cyst in 4,primary aldosteronoma in 3,adrenal neurinoma in 2,adrenocortical carcinoma in 1,retroperitoneal neuroblastoma in 1,retroperitoneal primary lymphoma in 1 and adrenal ganglionic fibroma in 1.Diagnoses based on the results of imaging examinations and laboratory tests were correct in 14 cases (27%).Of the 10 patients with malignancies,5 with primary diseases died 6-10 months after chemotherapy and 5 were lost to follow-up.The remaining 42 patients were followed up for 6 months to 9 years with no evidence of recurrence. Conclusions Multidisciplinary approaches are required to identify and differentiate adrenal incidentalomas, and early surgical treatment is suggested.

Objective:To investigate the relation between the AT 1 receptor gene polymorphism and some biochemical indices of essential hypertension patients. Methods:Sixty-four patients of mild-moderate essential hypertension were not given any anti-hypertension drugs for 5 half lives, then used the PCR-RFLP to detect the AT 1 receptor gene type and measured the basic blood pressure at the same time. After that, fasting serum glucose , serum total cholesterol(TC) , triglyceride(TG) , high-density lipoprotein cholesterol(HDL-C) , low-density lipoprotein cholesterol(LDL-C) , blood urea nitrogen(BUN) , creatinine(Cr) were measured in all subjects. Results:①The frequency of AC gene type in these essential hypertension patients and the C1166 allete of AT 1 receptor gene was 23.4% and 11.7%. ②There was a remarkable difference on TG between two gene groups. ③There were obvious difference on Cr and UA between two gene groups .④There were no obvious difference on Glu, TC ,HDL-C ,LDL-C ,BUN between AC and AA gene type. Conclusion:①The concentration of TG of AC gene type is lower than that of AA , there may be relativity between two gene type. ② A1166/C gene polymorphism may be associated with renal function. AA gene type may have poorer renal function. ③A1166/C gene polymorphism may be not relative with Glu,TCH ,HDL-C ,LDL-C ,BUN.

Objective:To study the effect of NF-?B “decoy” oligodeoxynucleotides on nitric oxide synthase expression in LPS-induced mouse macrophages. Methods: Mouse macrophage cell lines J774.1 were cultured with LPS and liposome-mediated oligodeoxynucleotides, and the level of nitric oxide and inducible nitric oxide synthase were measured in the different culture fluids. RNA was extracted from macrophages, and the mRNA expression of macrophages inducible nitric oxide synthase was observed by RT-PCR. Results: NF-?B “decoy” oligodeoxynucleotides decreased the activity of inducible nitric oxide synthase in LPS-induced macrophage, and inhibited the generation of nitric oxide. The level of nitric oxide and inducible nitric oxide synthase did not change in control group. Conclusion: NF-?B “decoy” oligodeoxynucleotides inhibit the expression of inducible nitric oxide synthase in LPS-induced macrophages, reducing the amount of nitric oxide, which is probably due to specific inhibition of the binding sites of activated nuclear factor-kappa B.
