AIM: To establish the HPLC fingerprint of Genti ana straminea Maxim. METHODS: It was adopted gradient with Methanol and 0.4% phospho ric acid, the detection wavelength was at 230nm and recording time. And it w as compared with a software published by Zhejiang University. RESULTS: The constituents of Gentiana straminea Maxim. were well se parated by HPLC, and the similarity was more than 0.90 . CONCLUSION: The method can be used as the fingerprint of G entiana straminea Maxim.
2.Determination of protopine and tetrahydropalmatine in Corydalis Decumbens (Thunb.) Pers. by HPLC

Objective:To establish the content determination method of the main alkaloids protopine and tetrahydropalmatine in Corydalis decumbens (Thunb.) Pers. Methods: HPLC was used to determinate the main alkaloids protopine and tetrahydropalmatine in Corydalis decumbene (Thunb.) Pers. Supelcosil LC 18 DB Column was used, mobile phase consisted of Methol NaAc/HAc(75∶25 pH5.0) and detection wavelength at UV 285 nm. Results: Linearity of this method was good with the average recoveries, 98.04% and 99.63%. Conclusion: This method is accurate, reliable with good separability and reproducibility, it can be applied as standard for the control of Corydalis decumbens (Thunb.) Pers.
3.Effect of percutaneous vertebroplasty and percutaneous kyphoplasty on adjacent intervertebral disc degeneration and mechanism

BACKGROUND:Compared with conservative treatment, percutaneous vertebroplasty (PVP) and percutaneous kyphoplasty (PKP) can rapidly relieve the pain caused by the compression of osteoporotic vertebral body. However, the perfusion of bone cement can not only restore the rigidity of fractured vertebrae, but also theoretical y alter the stress distribution of fractured vertebrae and increase the stress load of adjacent vertebrae, thus causing adjacent intervertebral disc degeneration. <br> OBJECTIVE:To observe the effects of PVP and PKP on the adjacent intervertebral disc degeneration and explore the possible mechanism. <br> METHODS:A total of 85 patients with osteoporotic vertebral compression fracture were divided into three groups:conservative treatment group (n=23), PVP group (n=42) and PKP group (n=23). ODI scores, DHI, MRII, and peripheral interleukin-1β, interleukin-6, tumor necrosis factor-αlevels were col ected and analyzed at different time points. <br> RESULTS AND CONCLUSION:The results showed that, PVP group and PKP group relieved pain effectively, reduced the time in bed, and had a better treatment satisfaction, but there were no significant differences in the scores of ODI scores between PVP group and PKP group at 24 months after discharge (P>0.05). At 24 months after discharge, MRII in the PVP group and PKP group were lower than that in conservative treatment group and before treatment (P<0.05), while peripheral interleukin-1, interleukin-6, tumor necrosis factor-αlevels were higher (P<0.05), there were significant differences between PVP group and PKP group (P<0.05). The disc degeneration adjacent to the fractured vertebral body can be accelerated by PVP and PKP procedures, and the latter is more serious. Interleukin-1, interleukin-6, tumor necrosis factor-αlevels are closely related to the degree of intervertebral disc degeneration.
5.Perioperative protective procedures of myocardium in patients undergoing heart-valve replacement

The objective of this retrospective study was to evaluate the myocardial protection of different periop erative procedures in 100 cases with heart-valve replacement (HVR) between 1980 and 1992. Before Feburary. 1989. HVRs of 48 cases (group Ⅰ) were undertaken with hypothermia (28C=30C) and cold cardioplegic solution (4C. 500ml). under morphine-diazepam-pancuronium anesthesia: following cardiac resuscitation, the procedures were performed including blood transfusion, diuresis, supplemental KCI. inotropic support with dopomine, norepincphrine and aramine. and continuous mechnical ventilation. Since 1989, the procedures were increased to be conducted in 52 cases (group Ⅱ) as followed: A. preoperative managements for achiving the optimum status of cardiac function: 1. oxygen therapy by mask. 2. antirhenmatic treatment for patients aged less than 35 years. 3. oral isosorbide and captopril for pulmonary hypertension, 4, infusion with fructose 1.6-diphusphatc and glucose-insulin-KC1 mixture. 5. clinical dose of digitora and diuretics for cardiac failure; B. methods during surgery: 1. anesthesia was induced with fentanyl, midazolam and pancuronium. and maintained with inhalation of nitrous oxide and infusion of fentanyl at 30-35?g/kg/min, 2, hy pothermia (24C-27C) and cold cardioplegic solution(600-1000ml) 3. immediately before the blockage of aorta was overcome. the artificial perfusion blood pressure was reduced; afterwards, it was gradually recovered, in order to prevent myocardium from injury of reperfusion. 4. intra-and extracoporal circulations worked together over half of blokage time of arota, to improve myoardial oxygen delivery; C. treatments in early postoperative stage: 1. continuous positive end-expiratory pressure ventilation, 2. infusion with magnesium-potassium aspartyl: and the others were similar to those before 1989. 6.Effects of Carvedilol on Blood Pressure in Hypertensive Patients and Vascular Endothelial Function

OBJECTIVE: To investigate the effects of carvedilol on blood pressure in hypertension patients and vascular endothelial function. METHODS: 100 inpatients with primary hypertension were collected from our hospital from Jan. 2008 to Jan. 2010 and randomly divided into treatment group and control group(n=50). Treatment group were given initial oral dose of carvedilol tablets 6.25 mg twice a day for 7~14 days if it was tolerated; the dose of carvedilol tablets increased to 12.5 mg or 25 mg twice a day according to blood pressure at trough concentration if it was necessary. Control group were given carvedilol tablets 6.25 mg twice a day if it was tolerated. After 1 week, control group received double dose of metoprolol with maximum dose of carvedilol of 50 mg. The treatment course was 3 months. The contents of NO and ET in serum of 2 groups were detected. Antihypertensive effect and effects of carvedilol on vascular endothelial dilatation and systolic function were observed and compared between 2 groups. 7.The influence of mass screening for prostate cancer on the diagnostic status of the clinical prostate cancer

Objective To investigate the influence of t he mass screening on the diagnostic status of the conventional ordinary prostati c cancer (CaP). Methods Sixty seven cases of prostate ca ncer detected by mass screening (January 1996~December 2001) and 358 cases diag n osed at the outpatient of various hospitals in Changchun (January 1986~December 2001) were compared in terms of age distribution,clinical stages,level of serum prostate special antigen(PSA),pathological grade and treatment modalities,with a SPSS analysis system. Results The annual average incide nce of CaP during 1999 to 2001 increased 4.72 times when compared to the 1986 to 1989’s.In the screening group,the cases with clinical stage A and B accounted for 58.2%. In the clinical group, it was only 27.9%,and mostly detected incident ally.Furthermore,in the screening group the percentage of the cases beyond 20 ng /ml PSA and of poorly differentiated carcinoma were much lower compared to the c linical group.These differences were statistically significant.The cases of radi cal prostatectomy in the screening group increased 15.3% compared with that of t he clinical group. Conclusions Only the mass screening c an really reveal the actual prevalence of prostate cancer.It can markedly raise the number of prostate cancer,especially the cancer in the early stage.The mass screening is inevitable to detect the curable early stage prostate cancer.
8.Construction of recombinant plasmid pCMV-Myc-PIASx? and its protein expression

Objective To construct the recombinant plasmid pCMV-Myc-PIASX? and express the fusion protein in mammalian cells.Methods PIASx? fragment was digested from the original vector pGADT7 with SalⅠand NotⅠ,and then was inserted into the targeted pCMV-Myc vector by the recombinant DNA technique.After identification,the recombinant plasmid was transfected into CHO cells.The expression of recombinant Myc-PIASx? fusion protein was detected by Western blotting.Results By the restriction enzyme digestion,fragment purification,ligation and transformation,the recombinant plasmid was obtained.The right recombinant plasmid pCMV-Myc-PIAS3 was identified with enzyme digestion and sequencing.By EcoRⅠ digestion analysis,pCMV-Myc-PIASx? showed a 5641 bp band.By XbaⅠdigestion analysis,pCMV-Myc-PIASx? showed two expected band of 3291 bp and 2349 bp.A specific protein expression band at 68 000(PIASx? fusion protein) was showed in Western blotting,which matched recombinant plasmid.Conclusion The recombinant plasmid of pCMV-Myc-PIASx? is sucssesefully constructed,which provids a good tool for further function study on PIAS family.
9.Three-dimensional finite element analysis of stress on connector of CAD/CAM fixed bridge

BACKGROUND:Artificial teeth computer aided design and computer aided manufacture (CAD/CAM) is a high technology involving various subjects.Many CAD/CAM systems have designed.However,they have some disavantages and a high cost.Therefore,only the system with proprietary intellectual property can solve this problem.OBJECTIVE:To optimize the connector of the CAD/CAM fixed bridge.METHODS:Three-dimensional finite element models of the connector were established which was used to design different styles of connectors.Stress under different vertical loading conditions was compared.RESULTS AND CONCLUSION:The connectors with different appearances displayed varying stress.Stress on the connector with arc transition was less than connector without arc transition.Connector was supposed to design to style with arc transition.
