1.Effect of Sijunzi Decoction on Mitochondrial DNA Missing of the Aging Model Mice

Objective To observe the effect of Sijunzi Decoction (SD) on the mitochondrial DNA deletion of the aging model mice, and to explore its anti-aging biomolecular mechanism. Methods Kunming mice were divided into 6 groups according to the body weight: normal control group, .model control group, high-, middle-and low-dose SD groups (at the dose of 20, 10, 5 g·kg-1 respectively), and vitamin E group (250 mg·kg-1), 20 mice in each group.The mice models of aging were induced by D-galactose. Water maze test was used to evaluate the learning and memory ability of the aging model mice. The effect of SD on the mitochondrial DNA deletion of the aging model mice was ob-served. Results In the hippocampal mtDNA of model control group and low-dose SD group, there were about 3.87 kb fragment missing. But there was no missing pieces in high-and middle-dose SD groups. Conclusion The anti-aging biomolecular mechanism of SD is probably related to the preventive effect on the mutation and deletion of the mitochondrial DNA in the aging model mice.
2.Evaluation of the effect of preemptive analgesia receiving tramadol combined with local anesthesia in neck surgery

Objective: To evaluate the preemptive analgesia of intravenous tramadol preoperatively in neck-surgery. Methods: Eighty patients, ASAⅠ-Ⅱ,were scheduled for neck-surgery under local anesthesia. Patients were randomly divided into 2 groups (40 cases/group).In group P intravenous tramadol was administered as a bolus dose of 3mg/kg 15min prior to skin incision. Group C received saline as a placebo. The patients-controlled analgesia(PCA) devices were used in all patients after operation. We observed BP, HR,VAS score, the time of first PCA activation, the valid number of press PCA and the total consumption of analgesic after operation. 3.Quality standard for prepared pieces of Rhizoma Panacis Majoris

AIM: To establish quality standard for prepared pieces of Rhizoma Panacis Majoris. METHODS: Traditional identification and TLC were used for qualitative analysis of prepared pieces. The contents of ginsenoside Ro and chikusetsusaponin IVa were determined by HPLC. Water,ash and acid-insoluble ash were also detected. RESULTS: The average content of ginsenoside Ro was 7. 19% ,chikusetsusaponin IVa was 3. 16% from different habitants. CONCLUSION: The established method has been proved to be simple,stable,accurate,and can be applied to the determination of ginsenoside Ro and chikusetsusaponin IVa in Rhizoma Panacis Majoris. The quality control standards for Rhizoma Panacis Majoris is normative and the result is accurate.
4.Effect of Sijunzi Tang and Yougui Yin on Learning-Memory Ability and Free Radicals in Aging Rat Model

Objective To compare the effect of Sijunzi Tang and Yougui Yin on learning-memory ability and free radicals in aging rat model.Methods The aging rat model was induced by intraperitoneal injection of D-galactose.The swimming time and error times in water maze test were observed and the free radicals SOD,MDA,NO,NOS in rat brain tissue were measured.Results Sijunzi Tang could shorten the rat swimming time in water maze test.Yougui Yin could increase SOD level and decrease MDA level in brain tissue of model rat,while Sijunzi Tang could decrease NO and NOS level.Conclusion Sijunzi Tang could improve the rat spatial learning and memory ability,and Yougui Yin could relieve the damage of free radical in brain tissue of the aging rat model.
5.Application of Quality Control Circle in Improving the Completion Rate of Bed Shifting of Duty

Objective:To improve the the completion rate of bed shifting of duty via application of quality con-trol circle( QCC) , and to build a harmonious relationship between doctors and nurses and improve nursing moral standards. Methods:After organizing QCC group, taking improving the completion rate of bed shifting of duty as the theme and analyzing the effect, according to the ten steps of QCC to control the quality. Results:The comple-tion rate of bed shifting of duty was improved from 69% to 69%, the patients' satisfaction was improved from an av-erage of 96 to 100 after the implementation of QCC. Conclusion: QCC can improve the completion rate of bed shifting of duty and patients' satisfaction, and thus worth of promoting in routine nursing.
6.Effect of valsartan on Notch pathway and extracellular matrix in glomeruli of diabetic mice

Purpose To investigate effect of valsartan on expression of Notch pathway and production of extracellular matrix. Methods Urine protein and production of extracellular matrix were measured after diabetic mice were treated with valsartan. The levels of Jag-ged1, Notch1, Notch intracellular domain 1 (NICD1), transforming growth factor-β1 (TGF-β1), TypeⅣcollagen and Laminin were determinated by immunohistochemistry, Western blot and real-time PCR. Results Valsartan suppressed urine protein and production of extracellular matrix in diabetic mice (P<0. 05). Valsartan also inhibits overexpression of Jagged1, Notch1, NICD1, TGF-β1, Type Ⅳ collagen and Laminin in glomerular tissues of diabetic mice (P<0. 05 or P<0. 01). Conclusion Valsartan inhibits activa-tion of Notch pathway in glomerular tissues of diabetic mice, inhibits production of extracellular matrix in glomerulus and delays glomer-ulosclerosis.
7.Study on expression level of proto-oncogene Wip1 in endometrial carcinoma tissue

Objective To investigate the related factors of clinical stage and prognosis in the patients with endometrial carcinoma and their relation with proto-oncogene Wip1 expression level.Methods The paraffin samples of resected endometrial carcinoma in 120 cases of endometrial carcinoma in our hospital from January 2002 to January 2012 were collected as the experimental group,the samples were verified by pathology.Contemporaneous 120 samples of biopsy normal endometrial tissue served as the control group.The expression leve of Wipl were detected in turo groups.Results (1) In the Wip1 immunohistochemical staining results:Wip1 immunohistochemical staining was negative or weak in normal endometrial tissue cells,while showed pale yellow to yellowish-brown in endometrial cancer tissue.The positive expression rate of Wip1 protein in endometrial carcinoma tissue was 77.5%(93/120),which was higher than 22.5%(27/120) in normal endometrial tissue,the difference was statistically significant (P<0.05).(2)In the Western blot results of Wip1 protein in endometrial cancer tissue and normal endometrial tissue:the relative amount of Wip1 protein in endometrial carcinoma tissue was 0.635±0.023,which was significantly higher than 0.325±0.018 in normal endometrial tissue,the difference between the two groups was statistically significant (P<0.05).(3)In the real time quantitative qRT-PCR results of various samples:Wip1 mRNA expression level was higher than that in normal endometrial tissue,which were 0.628±0.053 and 0.191±0.009 respectively,the difference between the two groups was statistically significant(P<0.05).(4) The expression level of Wip1 had no correlation with age,estrogen and progesterone status,HER2,lymph node status and TNM stage,but had correlation with P53 expression level.Conclusion (1) The Wip1 expression amount is high in endometrial carcinoma and low in normal endometrial tissue.(2)The Wip1 expression level has no relation with age,estrogen and progesterone status,HER2,lymph node status and TNM stage,while has association with P53 expression level.
8.Treating allergic conjunctivitis combined with frequently winking in children using olopatadine

Objective To observe curative effect of olopatadine in treatment of allergic conjunctivitis combined with frequently winking in children. Design Prospective case series. Participants Ninety seven children with frequently winking aged 3-12 years in Shanghai Eye and ENT Hospital. Methods According to the factor in causation of frequently winking (FW), 97 children were divided into allergic conjunctivitis (AC) group, trichiasis combined AC group, AC with tics group, trichiasis group, tics group and unknown cau-sation group after clinical examination. The first 3 groups with AC were treated with olopatadine 0.1% ophthalmic solution b.I.d.(n=35) and emedastine 0.05% ophthalmic solution was used q.I.d, in AC group as control(n=32). The patients were followed after 3 weeks and the extent of symptoms of FW was recorded. Main Outcome Measures Extent of the symptoms of FW. Result The effective rate of olopatadine in treating FW caused by AC or AC combined with other diseases was 80.43%. The effective rate in FW children caused mainly by AC was 82.86% which was higher than that of control drug emedastine (75.0%) (P=0.02). Seven cure cases were all in olopatadine treating group. Conclusion AC is the most common and main causative factor in FW children. Early use of olopatadine ophthalmic drop can effectively alleviate FW symptoms. (Ophthalmol CHN, 2009, 18: 272-274)
9.Screening of target genes in esophageal squamous cell cancer in Kazakh by oligonucleotide microarray

Objective To screen the differentially expressed genes in esophageal squamous cell cancer (ESCC) and normal tissue of esophageal mucosa in Kazakh. Methods RNA was extracted from the ESCC sections in Kazakh patients, and was amplified to obtain cRNA. The gene expression profiles in ESCC and normal tissue of esophageal mucosa were detected by HG-U133 Plus 2.0 gene chip. The results were analyzed by bioinfor-matics. Results One hundred and seventy differentially expressed genes in ESCC and normal tissue of esophageal mucosa were found, with a difference of more than 10 times in expression levels. Of the 170 genes, 39 were up-regulated (signal log ratio > 3 ) and 131 down-regulated (signal log ratio < - 3). These factors such as cell cycle regulation, apoptosis, cytoskeleton; extracellular matrix, intracellular signal transduction, protein translation and synthesis, and immunological functions were correlated with the genes with abnormal expression. Conclusion The use of oligonucleotide microarray accurately and efficiently screen the 170 target genes in ESCC in Kazakh. It is suggested that these genes may be related to the carcinogenesis and development of ESCC in Kazakh.
