1.Simultaneous Determinations of Four Ingredients in Yiqi Hewei Capsules by HPLC

Objective To establish a method for the quantitative determinations of the ingredients in yiqi hewei capsules. Methods RP-HPLC method was used. The separation was performed on a Thermo C18 column with mobile phase consisting of acetonitrile-water (20: 80) (pH was adjusted to 3 with glacial acetic acid) at the flow rate of 1. 0 mL·min-1. The detection wavelength was 283 nm. Results The linear ranges were ( 0. 5735 -5. 7360) × 10 -2 mg·mL-1 for baicalin, (0. 1940-1. 9395) × 10 -2 mg·mL-1 for hesperidin, (0. 2051 -2. 0510) × 10 -3 mg·mL-1 for naringin and (0. 2020 -2.0200) ×10 -2 mg·mL-1 for neohesperidin;RSD =0.28%,0.54%,0.54%,0.31%(n =5),respectively. The average recoveries were 103. 94%, 98. 06%, 103. 09%, 95. 67%, respectively. Conclusion The established method is simple, sensitive and accurate,which can be used for the quality control of yiqi hewei capsules.
2.Observations on the Therapeutic Effect of Herbal Cake Moxibustion on Chronic Renal Failure

Objective To investigate the clinical efficacy of herbal cake moxibustion in treating chronic renal failure. Methods Sixty CRF patients were randomly allocated to treatment and control groups, 30 cases each. The control group received medication and diet control and the treatment group, herbal cake moxibustion on Shenshu, Pishu or Ganshu in addition. Pre-/post-treatment differences in serum creatinine (Scr), blood urea nitrogen (BUN), endogenous creatinine clearance rate (Ccr) and 24-h urinary protein quantity were observed in the two groups and the clinical therapeutic effects were compared between the two groups after 3 months of treatment.Results The marked efficacy rate and the total efficacy rate were 46.7% and 86.7%, respectively, in the treatment group and 20.0% and 46.6%, respectively, in the control group, and there were statistically significant differences in both between the two groups (P<0.05). There were statistically significant pre-/post-treatment differences in Scr, BUN, Ccr and 24-h urinary protein quantity in the two groups (P<0.05). There were statistically significant post-treatment differences in Scr, BUN and 24-h urinary protein quantity between the treatment and control groups (P<0.05).Conclusion Herbal cake moxibustion is an effective way to treat chronic renal failure.
4.Effect of selective head cooling therapy on cerebral blood flow and oxygenation in neonates with hypoxic-ischemic brain damage

Objective To study the changes of cerebral blood flow and cerebral oxygenation during selective head cooling (SHC) in neonates with hypoxic-ischemic brain damage (HIBD). Methods Thirty-eight neonates with moderate and severe HIBD were enrolled in two groups: SHC group (n=18) and normal temperature group (n=20). The gestational age of all subjects were more than 38 weeks and Apgar score in 5 minutes after birth was less than 5 scores. Also, nerve system symptoms and electroencephalogram abnormal was found in 6 hours after birth. Neonates in SHC group had nasopharyngeal temperature maintained 34℃ and in normal trmerature guoup had rectum temperature maintained about 36℃. Seven healthy neonates were as control group. Three groups were observed 84 h. Cerebral blood flow velocity (CBFV) of middle cerebral artery (MCA) was measured with transcranial Doppler (TCD) at 6, 12, 24, 48, 72 and 84 hours after birth. Cerebral cytochrome aa3 (Cytaa3 ) was monitored continuously throughout the study using near infrared spectroscopy (NIRS). 5.Construction of rat CnA/EGFP recombinant eukaryotic gene coexpression plasmid

Objective To construct rat calcineurin catalytic subunit ?(CnA)/enhanced green fluorescent protein EGFP gene coexpression plasmid for exploring the effect of calcineurin on the myocardium apoptosis induced by ischemia-reperfusion.Methods Total RNA was isolated from the heart of the adult Wistar rat,and CnA CDS segment of approximate 1 590 bp size was amplified by reverse transcription PCR method.CnA cDNA segment was cloned into pMD18-T Simple vector for sequencing,and the right clone was named T-CnA.CnA cDNA segment excised from plasmid T-CnA was ligated with pShuttle2 which had inserted IRES-EGFP segment into before.The DNA of the recombinant plasmid was extracted and was identified by double digesting with Nhe Ⅰ and Not Ⅰ.The right clone was named pShuttle2-CnA-IRES-EGFP.Results Sequencing result verified that the PCR product of CnA gene was identical to GenBank(NM_017041).1% agarose electrophoresis showed the bands of recombinant plasmid pShuttle2-CnA-IRES-EGFP digested by Nhe Ⅰ and Not Ⅰ were in the right range corresponding with expectation(1 590 bp and 5 240 bp).Conclusion Recombinant eukaryotic expression plasmid carrying rat CnA cDNA as well as a report gene-EGFP gene is successfully constructed in this experiment.
6.Establishment and evaluation on SD rats model with yang-deficiency and constipation

Aim To establish and evaluate Sprague-Dawley (SD) rats model with yang-deficiency and constipation at the direction of traditional Chinese medicine theories. Methods Rats model with yang-deficiency and constipation were induced by orally administration of white vinegar and active carbon-ice water. Changes of model rats in common biological characteristic, the number and time of excretion stool, colonic electromyography, gastrointestinal hormones and intestinal neurotransmitters were observed. Results Compared to the control group: changes of the model group in common biological characteristic including abdominal distension, puffed and withered hair, sluggish response, falling body weight, black and dry stool; Excretion time of the first granule stool became longer; Number of excretion stool within six hours became shorter; Intestine pushing rate decreased, and secretion of gastrointestinal hormones and intestinal neurotransmitters disordered, etc. After being treated with Tongbianling, the syndrome and indexes of model rats markedly improved. After 6 days′ resume, the model group in the experiment of excreting stool had markedly signification in comparison with the control group. Conclusion SD rats model with yang-deficiency and constipation could be stably established by administering white vinegar and active carbon-ice water.
7.Clinical trials with thyroid hormone supplementation in preterm infants with transient hypothyroidism

The morbidity of transient hypothyroxinemia of prematurity (THOP) was high. There is controversial about thyroid hormone supplementation in THOP. Clinical studies suggest that thyroid hormone supplementation can reduce the incidence of patent ductus arteriosus,but no effect on the mortality and the incidence of respiratory disease.Thyroid hormone supplementation can improve the neurodevelopmental outcomes in infants with gestational age less than 28 weeks with THOP, but no effect on infants with gestational age more than 28 weeks. Future research should focus on the normal range of thyroid hormone according to gestational age in preterm infants,and randomized clinical trial welldesigned stratified according to gestational age to study the long-term neurodevelopmental outcome of thyroid hormone replacement therapy in infants with THOP
8.Construction of rat calcineurin A alpha cDNA recombinant adenovirus vector and its identification.

Rat calcineurin (CaN) A alpha isoform (Ppp3ca) cDNA recombinant adenovirus vector was constructed in order to explore the effect of CaN on the myocardium apoptosis induced by ischemia-reperfusion injury. Total RNA was isolated from the heart of the adult Wistar rat, and Ppp3ca CDS segment of approximate 1.59 kb size was amplified by reverse transcriptional PCR method. Ppp3ca cDNA segment was cloned into pMD18-T Simple vector for sequencing, and the right clone was named T-Ppp3ca. Ppp3ca cDNA segment obtained from T-Ppp3ca was ligated with pShuttle2-IRES-EGFP to construct a recombinant plasmid pShuttle2-Ppp3ca-IRES-EGFP. Ppp3ca-IRES-EGFP expression cassette containing CMV, Ppp3ca-IRES-EGFP and SV40 polyA DNA fragment (3.97 kb) obtained from pShuttle2-Ppp3ca-IRES-EGFP was connected with pAdeno-X backbone sequence to construct a recombinant plasmid pAdeno-Ppp3ca. After being identified by PCR and enzyme digestion, recombinant plasmid pAdeno-Ppp3ca was packaged in HEK293 cells. Supernatant of adenovirus from HEK293 cells was collected after a visible cytopathic effect (CPE) appeared. The DNA of the recombinant adenovirus was extracted with the standard method. The presence of the recombinant adenovirus was verified by PCR. The results showed that sequencing results verified that the PCR product of Ppp3ca gene was identical to GenBank. Agarose electrophoresis showed the bands of recombined plasmid pAdeno-Ppp3ca and the recombinant adenovirus identified by enzyme digestion and PCR were in the right range corresponding with expectation. It was concluded that the recombinant adenovirus carrying rat calcineurin A alpha (Ppp3ca) cDNA as well as a report gene-enhancer green fluorescent protein gene was successfully constructed in this experiment.

9.HISTOCHEMICAL OBSERVATIONS ON THE RENAL TUBULO-TOXICITY OF GOSSYPOL

Gossypol has been shown to cause a side effect hypokalemia. This study was designed to evalulated the possible role of gossypol in relation to renal potassium loss by using histochemical method for comparative analysis of gossypol and gentamicin, a well known tubulo-toxic agent, as positive control with respect to nephrotoxic demage in rats and guinea pigs. The results indicated that gossypol had obvious effect on reabsorption function by inducing transitional decrease of membrane enzymes activities of renal tubular cells but had no effect as comparable with gentamicin to cause structureal damage of tubular cells.

Objective To retrospectively analyze the result of hemoglobin(Hb)test by using full-automatic Hb electrophoresis and evaluate the its significance in hemoglobinopathy.Methods The data of patients who underwent Hb electrophoresis test and regular blood tests in the hospital from January 2011 to December 2013 were included in the study.The test results were recorded including mean corpuscular volume(MCV),mean corpuscular hemoglobin(MCH)and results of Hb electrophoresis test.Final diag-nosis were made for suspected patients by using genetic testing,then disease detection rates and gene coincidence rates and constitu-ent ratios were calculated.Results 12 898 cases were included in the study,after statistical analysis the MCV was(85.32±13.61) fL,MCH was(29.87±6.44)pg.By using automatic hemoglobin electrophoresis,1 315 cases were found to be positive,in which 568 were male,747 were female,the detection rate was 10.19%.In the 1 315 patients,there were 761 cases suspectedα-thalassemia,ac-counted for 5.90%.There were 495 cases of suspectedβ-thalassemia,accounted for 3.84%,11 patients with HbJ(0.08%),15 pa-tients with HbK(0.12%),9 patients with HbG(0.07%),3 patients with HbD(0.02%),21 patients with HbE(0.16%).The sus-pected case′s final diagnosis were made by using genetic testing,α-thalassemia gene′s coincidence rate was 80.55%,β-thalassemia gene′s coincidence rate was 96.77%.Conclusion Automatic hemoglobin electrophoresis detection is of great significance for the di-agnosis of hemoglobinopathy.
