1.THE PROGRESS OF RADIOTHERAPY IN LUNG CANCER

Lung cancer is the most common cause of the death from cancers for both male and female in the world. Surgery remains the most effective method for controlling the primary tumour in most cases, but only 20% primary lesions can be resected. 80% patients should be treated by non-surgical methods. In recent years, radiotherapy has a fast development in the equipment and especially in the theory of radiobiology, but the curability of lung cancer still poor. In this paper we discussed the radiotherapy of lung cancer and how to choose the energe of X-ray, design the fields; the proceeding of irradiation; and it is necessary to find out a new regulation for stages that is available to indicate non-surgical treatment.
2.Adverse Drug Reactions of Defibrase

OBJECTIVE:To discuss the general pattern and features of adverse drug reactions(ADRs) of defibrase.MET-HODS:The cases of ADRs of defibrase reported in the national medical journals during 1999 and 2003 were collected and analyzed.RESULTS:ADRs of defibrase mainly included bleeding,urticaria,high fever,positive cutantest,liver function injury,ecchymoma,purpura haemorrhagica,incision hemorrhea,and arterial embolism.CONCLUSION:Great importance should be attached to ADRs of defribrase in clinic in order to ensure the safety of drug use.
3.The effect of calorie restriction at early age on their islet β cell mass in adulthood of rats

Objective To study the effect of calorie restriction at early age of rats on their islet β cell mass in adulthood.Methods Sixteen 8-week-old male SD rats were randomized to control group (n=7) and calorie restricted group (n =9).The rats in control group took food freely,while the ones in calorie restricted group were given 70% calorie of the control group.After 24 week calorie restriction,cholesterol (TC),triglyceride (TG),high density lipoprotein-cholesterol (HDL-C) and low density lipoprotein-cholesterol (LDL-C) were tested.The β cell mass was measured by immunohistochefistry and the activity of superoxide dismutase (SOD) as well as the level of malondialdehyde (MDA) in pancreas homogenate were determined by ELISA.Results The increase of the body weights(45 g vs.184 g)and the level of TG [(0.61±0.15)mmol/L vs.(0.78±0.14)mmol/ L]of the rats in the calorie restricted group were lower than those of the control group (P<0.05),while the β cell mass [(43.6±9.8)mg vs.(31.9± 11.6)mg],β cell mass of every milligram pancreas tissue[(89.7 ± 7.4) μg/mg vs.(44.8g ± 14.1) μg/mg] and β cell mass per body weight[(11.5±2.5) × 10-5 vs.(6.3 ±2.3) × 10-5]of the rats in the calorie restricted group were higher than those of the control group (P<0.05).There were no differences in the SOD activity [(0.91±0.30)nmol/ mg protein vs.(0.68±0.14)nmol/ mg protein]and MDA level [4.97± 0.65)U /mg protein vs.(6.05 ±2.14)U/mg protein] in pancreas homogenate between the two groups (P>0.05).Conclusions Calorie restriction at early age of rats may increase the islet β cell mass in their adulthood.
4.Effect of caloric restriction at early age of rats on their isletβcell function in adulthood

Eleven-week-old SD rats were randomized into control and calorie restriction group. The pancreatic β cell function and oxidative stress indexes in the two groups were compared after 24-week intervention. The results showed that calorie restriction, which started from young age, improved the early insulin secretion after glucose loading and alleviated the oxidative stress in adult rats, which wag related to the reduction of body weight.
5.Treatment of intercondylar humeral comminuted fractures

[Objective]To evaluate surgical treatment and curative effect of intercondylar humeral comminuted fractures.[Method]Twenty-one cases of intercondylar humeral comminuted fractures were collected from February 2001 to December 2005. The fractures were classified to 5 cases of C1,9 cases of C2 and 7 cases of C3 according to the AO classification.Seventeen fractures were internal fixed by open-reduction, and 4 fractures were treated by total elbow replacement. After olecranon osteotomy, capitate eminence and trochlea humerus were reducted and fixed with bone screw,while shaft of humerus and the distal end of humerus were fixed with AO double plate or Y-shaped plate in internal fixed group. Early physiotherapy were started as soon as possible. The elbow replacement group were replaced by semi-restriction elbow prosthesis and fixed with cemente following physiotherapy after 4 to 7 days.[Result]forteen of 17 cases were postoperatively followed-up in internal fixed group from 12 to 44 months, and all cases had been visited for 14 to 36 months in elbow replacement group. According to modified Cassebaum scoring system, 71.4% were good in internal fixed group and 100% in elbow replacement group.[Conclusion]It has good curative effect in intercondylar humeral comminuted fractures with olecranon osteotomy, the AO double plate or Y-shaped plate internal fixation and early physiotherapy. With severe injury of the distal end of humerus, obviously osteoporosis and older than 50 years, the total elbow arthroplasty and replacement are preferred.
6.Vertebroplasty for severe vertebral compression fractures:imaging evaluation

BACKGROUND:Recent reports only concern vertebroplasty for mild to moderate vertebral compression fractures, but seldom address vertebroplasty for severe vertebral compression fractures such as vertebral colapse. OBJECTIVE:To evaluate the efficacy and imaging features of percutaneous vertebroplasty for severe vertebral compression fractures. METHODS:A total of 25 patients underwent single-level vertebroplasty for vertebral compression fractures. Imaging features were then analyzed including location, pattern of compression, extent of colapse, pre- and post kyphotic angle and adjacent disc height before and after vertebroplasty. RESULTS AND CONCLUSION:60% (16/25) patients involved the thoracolumbar junction. The height of colapsed vertebral body was 14%-30% of original vertebral body. The average height of colapse was 5.17 mm or 22% of original vertebral body. Kyphotic angle before vertebroplasty ranged from 0-33° (averagely 16°) with an average correction of 12° after vertebroplasty. Average disc height before vertebroplasty was 7.3 mm above and 7.7 mm below. Visual analogue scale score was significantly higher preoperatively than that postoperatively, showing significant difference in pain improvement (P< 0.015). These data suggest that percutaneous vertebroplasty is safe and effective in the treatment of single level severe vertebral compression fractures.
7.Comparative experimental study of different administration methods for steroid-induced femoral head nec-rosis in rabbits

Objective The method of administration is one of the important factors influencing steroid-induced femoral head necrosis ( FHN) .From the perspective of administration compliance, time and persons needed, and pharmacodynamics, this study compared different administration methods for steroid-induced FHN in rabbits aiming to provide some experimental evidence for selec-ting correct methods of administration. Methods The steroid-induced femoral head necrosis rabbits ( New Zealand, male) were ran-domly divided into five groups according to the different administration methods of intervention:deep oral administration group (n=10), in-tragastric gavage administration group ( n =10 ) , free-drinking drug group (n=10) and model control group (n=5), blank control group (n=5), the administration compliance, administration time, mortal-ity, pharmacodynamic index of lipid content and empty lacunae rate were compared among deep oral administration group, intragastric ga-vage administration group, free-drinking drug group. Results Compliance effect size of deep oral administration group ( 1.78 ± 0.64) lower than intragastric gavage administration group (4.04 ±0.87) and free-drinking drug group (8.94 ±1.05) (P<0.01). Administration time among deep oral administration group ([0.94 ±0.02]min), intragastric gavage administration group ([9.47 ± 0.31]min) and free-drinking drug group ([889.50 ±235.38]min) overall comparison gradually increased (P=0.000), the differ-ence between the two groups was statistically significant (P<0.05).At the 2 and 4 weeks, cholesterol, triglycerides and low-density lipoprotein of deep oral administration group, intragastric gavage administration group, free-drinking drug group compared with model control group and blank control group the difference was statistically significant ( P<0.05) , and deep oral administration group com-pared with free-drinking drug group, the difference was also statistically significant ( P<0.05) .At the 2 weeks, empty lacunae rate of deep oral administration group ([15.44 ±2.68]%), intragastric gavage administration group ([15.02 ±3.34])%), free-drinking drug group ([16.72 ±4.06]%) compared with model control group ([18.59 ±3.12]%) and blank control group ([10.82 ± 2.76]%, the difference was statistically significant (P<0.05).At the 4 weeks, empty lacunae rate of deep oral administration group ([18.53 ±3.26]%), intragastric gavage administration group ([18.85 ±3.17]%), free-drinking drug group ([20.41 ±4.18]%) compared with model control group ([24.66 ±3.74]%) and blank control group ([11.37 ±2.23]%), the difference was also sta-tistically significant ( P<0.05 ) . Conclusion Compared with traditional methods of administration, deep oral administration has better compliance, shorter administration time, and similar to intragastric gavage administration in pharmacodynamics, but more effec-tive than free-drinking drug administration, and it is a new and effective method of administration.
8.Construction of human telomerase reverse transcriptase-immortalized rat bone marrow mesenchemal stem cell strains

BACKGROUND:Because of convenient source, multi-lineage differentiation and low immunogenicity, bone marrow mesenchymal stem cels are the ideal cel type to serve as vectors of transgenic cels in pain management. However, the replicative senescence and smal amount of cels obtained from the bone marrow restrict the application of bone marrow mesenchymal stem cels in pain research. OBJECTIVE:To construct human telomerase reverse transcriptase (hTERT)-immortalized rat bone marrow mesenchymal stem cels as transgenic celular vectors for pain therapy. METHODS:Bone marrow mesenchymal stem cels were obtained from whole rat bone marrow, and then transfected with a lentivirus containing the hTERT (pLV-Puro-EF1α-hTERT) folowed by puromycin selection. hTERT expression and telomerase activity in these transfected cels were determined by RT-PCR and TRAP. Morphological changes, capacity of cel growth and multi-lineage differentiation, chromosome karyotype and tumorigenicity were observed in vitro. Moreover, the expression of cel surface molecule, Nestin, MHC-I and MHC-II in transfected cels were also detected by flow cytometry and immunocytochemistry. RESULTS AND CONCLUSION: The bone marrow mesenchymal stem cels geneticaly modified by hTERT could be cultured and passaged through 30 generations in vitro. Compared to the primary and negative transfected cels, the hTERT-modified bone marrow mesenchymal stem cels showed higher expression of hTERT mRNA, telomerase activity and cel proliferation. Most of transfected cels stayed at G2/M and S stages. The proliferation index of the transfected cels were increased dramaticaly. The positive rates of CD29, CD44 and CD90 were over 70%, but the positive rates of CD34 and CD45 were less than 5%. Transfected cels were positive for Nestin in the cytoplasm, but negative for MHC-1 and MHC-11. In addition, this cel line continued to exhibit the characteristics of fibroblastic bone marrow mesenchymal stem cels, including phenotype, differentiation into osteoblasts, adipocytes and neuron-like cels. No chromosome abnormality and tumor formation were observed in this experiment. Taken together, these data suggests that the rat bone marrow mesenchymal stem cels immortalized by hTERT gene are constructed successfuly and stil maintain major stem cels characteristics, which provide safe and stable cel vectors as research base for pain therapy.
9.The changes of ultrastructural microcirculation of small intestine in canines with portal hypertension

Objective To examine the changes of ultrastructural microcirculation of small intestines of portal hypertension (PHT) canines. Methods PHT canine models were established by coarcating a half main portal vein with silk line chronic emboliztion. The ultrastructural changes of small intestine epithelium, mucous membrane and submucosa microcirculation were examined. Results The characteristics of ultrastructural changes of small intestine epithelium, mucous membrane and submucosa microcirculation were as follows: the number of blood vessels was increased and the diameter of them was expanded significantly; the lumen of arteriole was decreased, and the wall was thickened; arteriole collagen fibers were hyperplastic and confused; the lumen of venule was increased, the wall was thinned; basement membrane was damaged; microcirculatary endothelial cell was damaged generally; leukocytes was infiltrated; epithelial cells and basement membrane of intestinal mucosa were damaged; smooth muscle cell nucleus of ileum were deformed. Conclusion Small intestine epithelium, mucous membrane and submucosa microcirculatary ultrastructral showed obvious changes in PHT canines.
10.Observation on effect of Livin on VSV-induced apoptosis of A549 cells

Objective To investigate the effect of Livin expression on VSV-induced apoptsis of A549 cells.Methods The expression of Livin of A549 cells was inhibited by RNA interference.VSV-induced apoptosis of A549 cells was observed by Tunel assay.Protein Level of livin was detected by Western blot.Caspase-3 activity was detected by the fluorescence-based quantitative method.Results Livin downregulation VSV-induced apoptosis of A549 cells.Inhibited the expression of Livin of A549 cells had increased Caspase-3 activity.Conclusion The effect of Livin on VSV-induced apoptotic of A549 cells could be increased by RNA interference.
