1.A preliminary study of serum vitamin D level in Uighur patients with hypertension in Kashgar prefecture

ObjectiveTo determine the serum vitamin D levels of patients with primary hypertension to provide a basis for prevention and control of the hypertension in Kashgar prefecture.Methods The clinical data of 200 patients with hypertension in Kashgar prefecture admitted to the Second People's Hospital from January 2014 to December 2015 were retrospectively analyzed. According to risk factors, the 200 patients with primary hypertension were divided into three groups: 60 patients were selected as low risk group (hypertension 1 grade and no risk factors), 64 patients were arranged into an intermediate risk group (hypertension 2 grade or hypertension 1 grade accompanied by 1-2 risk factors) and 76 patients were assigned in a high risk group (hypertension 3 grade or hypertension 1-2 grade accompanied by ≥3 risk factors or hypertension of any grade accompanied by 1 target organ damage or 1 kind of clinical disease). In the same period, 66 healthy subjects having taken physical systemic medical examination with normal blood pressure in this hospital were grouped into a healthy control group. The levels of blood pressure and vitamin D in serum were measured, and the correlations between vitamin D and systolic blood pressure (SBP), diastolic blood pressure (DBP) were analyzed by pearson correlation analysis in the four groups.Results The SBP and DBP were significantly higher in low, intermediate and high risk group than those in healthy control group [SBP (mmHg, 1 mmHg = 0.133 kPa): 142±6, 161±5, 173±12 vs. 112±12, DBP (mmHg): 89±7, 101±4, 103±11 vs. 74±8], and their levels of vitamin D were lower in low, intermediate, high risk group than the level in healthy control group (μg/L: 24±6, 26±5, 20±4 vs. 30±7, allP < 0.05). Correlation analysis showed that the level of vitamin D was negatively correlated with SBP, DBP (r value was -0.373, -0.324, allP < 0.01).Conclusions The serum level of vitamin D is decreased in Uighur patients with blood hypertension in Kashgar prefecture, especially in the patients with high risk factors, the descent of the serum level is more significant. It is suggested to appropriately use vitamin D supplement to prevent blood hypertension.

Quantum dots (QDs) are novel photo-stable semiconductor nanocrystals with wide excitation spectra and narrow, symmetrical emission spectra. QDs can be used as molecular probes by conjugating to a wide range of biological targets, including proteins, peptides and nucleic acids. It has been widely used in bio-labeling, fast detection and biological imaging. In this review, we focus on the applications of QDs in tissue engineering and its potential bio-safety issues.

3.Effects of blood pressure by intravenous infusion of different doses of oxytocin in cesarean section

Objective To observe the effects of blood pressure by intravenous infusion of different doses ofoxytocin in cesarean section.Methods Sixty full-term pregnant women undergoing cesarean section with continuous epidural anesthesia were divided into three groups by random digits table method with 20 cases each:group A,B and C.Three groups were injected 10 U oxytocin in uterine muscle after infant delivery.Group A,B and C received 5,10 and 20 U oxytocin (sodium lactate ringer,500 ml) continuous intravenous infusion at the speed of 10 ml/min.If happened uterine contractions bad,they were sublingual administering 0.2 mg misoprostol.If happened severe hypotension,they were intravenous injected 5 mg ephedrine.The change of mean arterial pressure (MAP) and heat rate before anesthesia (T0),after fetal childbirth (T1),5 m in (T2),10 min (T3),30 min (T4) after infusion of oxytocin and the dosage of ephedrine and misoprostol were recorded.Results There were no significant differences in MAP and heart rate at every time point between group A and B (P> 0.05).MAP decreased and heart rate increased in group C at T2,T3 compared with those in group A and B,and there were significant differences (P< 0.05).The number of cases of sublingual misoprostol were increased in group A (7 cases) compared with that in group B (2 cases) and group C(1 case).The 8 patients injected ephedrine in group C were more than group A(1 case) and group B (3 cases).Conclusion Cesarean section after the delivery of the fetus in the uterus muscle injection of oxytocin 10 U,after 10 U of oxytocin added 500 ml sodium lactate ringer injection at the speed of 10 ml/min intravenous infusion has little effect on the blood pressure and heart rate,and has good uterine contractions.
4.Analysis of risk factors of lymph node metastasis and clinical significance of deep submucosal invasion in 64 cases of T1 stage colorectal cancer

Objective:To investigate the risk factors of lymph node metastasis and the clinical significance of deep submucosal invasion in patients with T1 stage colorectal cancer.Methods:From January 30, 2010 to December 31, 2019, at Shandong Provincial Hospital Affiliated to Shandong First Medical University, among patients with T1 stage colorectal cancer, 41 patients underwent radical surgery for colorectal cancer (surgery group) and 23 patients received endoscopic submucosal dissection (ESD) (ESD group) were enrolled. The tumor gross type, maximum diameter, histologically poorly differentiated components, degree of invasion (the type of mucosal muscle destruction, the width and depth of invasion), the budding grade of tumor, and whether with vascular tumor thrombus were recorded. The additional treatment and prognosis of patients were collected by telephone follow-up. The risk factors of lymph node metastasis in stage T1 colorectal cancer, the correlation between the complete muscularis mucosa destruction and the width and depth of invasion in the ESD group, and the effects of additional treatment after operation on the prognosis of patients were analyzed. Independent sample t test and chi-square test were used for statistical analysis. Results:The rate of lymph node metastasis in patients with poorly differentiated components or vascular tumor thrombus was higher than that in patients without poorly differentiated components or vascular tumor thrombus (3/6 vs. 12.1%, 7/58; 3/4 vs. 11.7%, 7/60), and the differences were statistically significant ( χ2=5.934 and 11.409, both P<0.05). All patients in the surgery group had complete muscularis mucosa destruction. In ESD group, the width of tumor invasion was ≥ 2 mm in 16 cases, including complete destruction of muscularis mucosa in 15 cases and partial destruction in one case; the width of tumor invasion was <2 mm in seven cases, including complete destruction of muscularis mucoa in two cases and partial destruction in five cases; the depth of infiltration was ≥ 2 000 μm in 14 cases, including complete destruction of muscularis mucosa in 13 cases and partial destruction in one case; the depth of infiltration was <2 000 μm in nine cases, including complete destruction of muscularis mucosa in four cases and partial destruction in five cases. The complete muscularis mucosa destruction was related with tumor of invasion width ≥ 2 mm and invasion depth ≥ 2 000 μm (15/16 vs.2/7, 13/14 vs. 4/7), and the differences were statistically significant ( χ2=10.729, 6.659, both P<0.05). Among the 64 patients with T1 stage colorectal cancer in this study, six cases (9.4%) had poor prognosis; five cases (7.8%) died, and three of them (4.7%) were tumor-related deaths. Adjuvant therapy was added in 10 cases in surgery group and 10 cases in ESD group, and there were no poor prognosis in those patients. There were no significant difference in the incidences of poor prognosis of patients without additional treatment and patients with additional treatment of the two groups (9.7% (3/31) vs. 0 (0/10) and 23.1% (3/13) vs. 0 (0/10)) (both P>0.05). Conclusion:When T1 stage colorectal cancer with tumor submucosal invasion, clinicians should comprehensively evaluate the prognostic risk based on various pathological characteristics such as the degree of tumor differentiation, vascular tumor thrombus and mucosal muscle destruction.
5.Research on Classifed Punishments on Manufacturers of Unconformity Products in Supervision and Sampling Inspection of Medical Devices.

Theories of classified management which are based on risk governance are introduced into the newly revised , but the punishments on the manufacturers of unconformity products are not classified based on the severity and risks caused by the test items. This article analysed the disadvantages of current punishment measures on the manufacturers of unconformity products and the theoretical basis of classified punishments. The feasibility of classified punishments had also been studied and some basis of discretionary punishments was provided under the current regulations.

6.Pressurizing and Stretching Experiment in Arterialized Veins of Rabbits

Objective To establish the model of rabbit vena arterialization, so as to investigate the difference of mechanical parameters between arteries and veins as well as before and after arterialization. Methods Twenty-four rabbits were randomly divided into experimental group (n=12) and control group (n=12). By establishing the rabbit vena arterialization model for experimental group, the arterial blood could flow into the veins. After model creation, the vein would be removed 4 weeks after surgery. In the meantime, the external jugular veins and cephalic arteries extracted from control group were acquired. Compressive pressurizing and stretching tests on all vessels were conducted at the same time(including arteries, veins and arterialized veins). Observation was supported by HE staining and immune tissue chemical techniques. Results There were no deaths among the 24 rabbits, with unobstructed blood flow in veins. With the increase of intravascular pressure, the outer diameter of veins changed at first and then stabilized at a fixed value. The elasticity of veins was worse than that of arteries. The external diameter of veins increased rapidly with internal pressure of veins increasing and reached its extreme elasticity. Comparatively, the elasticity of arteries increased slowly. HE staining results showed that thickness of the vascular wall was thinner, while it became thicker after vena arterialization. After vena arterialization, proliferating cell nuclear antigen(PCNA) and α-actin showed positive results. It further proved that proliferation existed among smooth muscle cells, and veins showed the tendency of restenosis again. The elasticity of veins after transplantation into the arterial system was improved compared with that before transplantation. Conclusions Accompanied by the increasing pressure, the vein could reach its elasticity extremity faster than the artery. Under such a long-term high pressure, vein intima was vulnerable. After vena arterialization, with the gradual thickening of vein intima, the tendency of vessel restenosis was obvious, and the elasticity of veins has been improved after transplantation.
7.Mechanism of Dendrobium huoshanense Polysaccharide Against Neuroinflammation in Parkinson's Disease Model: Based on NLRP3 Inflammasome

ObjectiveTo explore the mechanism of Dendrobium huoshanense polysaccharide (DHP) against inflammatory damage of neurons in Parkinson's disease (PD) model. MethodSH-SY5Y cells were randomized into blank group, model group, and DHP group. The survival rate of cells was measured by thiazole blue(MTT) assay, and the levels of lactate dehydrogenase (LDH), reactive oxygen species (ROS), malondialdehyde (MDA), and superoxide dismutase (SOD) were measured by colorimetric analysis. BV-2 microglia were classified into blank group, model group, DHP group, and MCC950 group (positive control group), and enzyme-linked immunosorbent assay (ELISA) was applied to detect the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-18 (IL-18). The expression of NOD-like receptor protein 3 (NLRP3), adaptor protein apoptosis-associated dot protein (ASC), cysteine aspartic protease-1 (Caspase-1), and IL-1β was measured by Western blot. A total of 50 C57BL/6 mice were randomized into blank group, model group, DHP low-dose (100 mg·kg-1) group, DHP equivalent-dose (350 mg·kg-1) group, and MCC950 group (positive control group), 10 mice in each group. The motor balance and coordination of C57BL/6 mice were observed by beam walking test, tail suspension test and rotarod test. The levels of Iba-1 and tyrosine hydroxylase (TH) were detected by immunofluorescence staining. The damage of dopaminergic neurons in the substantia nigra was detected by FJB staining. The levels of inflammatory factors such as IL-1β, IL-18, and TNF-α in mouse midbrain tissues were detected by ELISA and the protein levels of NLRP3, ASC, Caspase-1, and IL-1β protein were measured by Western blot. ResultCompared with the blank group, the SH-SY5Y model group showed decreased cell survival, increased levels of LDH, ROS, and MDA (P<0.05), and decreased levels of SOD (P<0.05). Compared with the model group, the DHP group demonstrated increased cell survival, decreased levels of LDH, ROS, and MDA (P<0.01), and increased level of SOD (P<0.01). Compared with the blank group, BV-2 model group had high levels of IL-1β, IL-18, and TNF-α (P<0.05) and high protein expression of NLRP3, Caspase-1, IL-1β, and ASC (P<0.05). Compared with the model group, DHP and MCC950 groups demonstrated low levels of IL-1β, IL-18, and TNF-α (P<0.01) and low protein expression of NLRP3, Caspase-1, IL-1β, and ASC (P<0.01). Compared with the blank group, the C57BL/6 model group displayed long time to pass the balance wood (P<0.05), short time spent on the rod in the rotarod test (P<0.05), high levels of IL-1β, IL-18, and TNF-α (P<0.05) and expression of Iba-1 in the midbrain substantia nigra (P<0.05), low TH expression (P<0.05), more positive neurons in the FJB staining (P<0.05), and high expression of NLRP3, Caspase-1, ASC, and IL-1β proteins (P < 0.05). Compared with the model group, the mice in the DHP and MCC950 groups had short time to pass the balance beam (P<0.01), long time spent on the rod (P<0.01), low levels of IL-1β, IL-18, and TNF-α (P<0.01), low Iba-1 expression in midbrain substantia nigra (P<0.01), high TH expression (P<0.01), and small number of positive neurons in the midbrain substantia nigra (P<0.01). The expression of NLRP3, ASC, and IL-1β proteins was lower in the MCC950 group (P<0.01), and the expression of NLRP3, ASC, Caspase-1 and IL-1β proteins was lower in the DHP equivalent-dose group (P<0.01) than in the model group. ConclusionDHP has anti-oxidative stress effect. It regulates the expression of NLRP3 inflammasome and inhibits the overactivation of microglia, thereby alleviating the neuroinflammatory injury in PD and exerting the neuroprotective effect.
