1.Protective Effects and Mechanisms of Longyanshen on Acute Chemical Liver Injury in Mice

OBJECTIVE:To study the protective effects and mechanisms of Longyanshen on chemical liver injury in mice.METHODS:The models of CCl 4 ,TAA and acetaminophen induced acute liver injury in mice were used to study the pro?tective effects of Longyanshen,and relevant parameters were detected and compared.RESULTS:Compared with model group,in Longyanshen group,activity of ALT markedly decreased,the contents of hepatic cyt P 450 ,cyt b 5 increased,the content of MDA in liver tissue significantly decreased,the activities of aniline hydroxylase and aminopurine-N-demethylase increased,the activity of Ca 2+ -ATPase in hepatic cell microsome markedly decreased and the activities of GSH-PX and the content of GSH increased.CONCLUSION:Longyanshen has protective effect on chemical liver injury in mice and the mechanisms may be related to the antioxidative action,inhibiting effect on lipid peroxidation,inducing the activities of hepatic microsomal enzymes and blocking the calcium channel.

AIM:To evaluate the ability of dendritic cells (DCs) loaded with tumor lysate to initiate cell-mediated immune responses by stimulating naive T cells, and the efficiency of activated T cells to kill autologous tumor cells in vitro. METHODS: The peripheral blood lymphocytes and monocytes were obtained from the advanced renal cell carcinoma patient by conglutination method. The immature dendritic cells were generated in the presence of interleukin-4(IL-4) and granulocyte/macrophage colony-stimulating factor(GM-CSF) from monocytes of healthy individuals. These cells were pulsed with tumor lysate or not. Induction of tumor-specific cytotoxic T lymphocytes (CTLs) response by mature dendritic cells (mDCs) was evaluated by the CD95(Fas) expression assay through FCM and the cytotoxic assay against autologous human tumor cells. RESULTS: Human immature dendritic cells and T cells obtained from healthy donors were stimulated with tumor-pulsed dendritic cells. The immature dendritic cells were applied to the cytotoxicity assay against target autologous tumor cells. Antigen-specific DCs vaccine can induce T cells activation and proliferation, thus we can obtain higher proportion of tumor specific cytotoxic T cells(CTLs), and enhance the CTLs to secret IFN-γ and TNF-α. CONCLUSION: Our results indicate that monocyte-derived human dendritic cells pulsed with tumor lysate could in duce the specific antitumor effect against autologons tumors. This in vitro model offers a new and simple approach to the development of DC + CTL - based immunotherapy.
7.Dengue 2 virus induces molecular alteration in the coagulant and fibrinolytic pathways of human umbilical vein endothelial cells

Objective To study the changes of tissue plasminogen activator (t-PA), plasminogen activator inhibitor 1 (PAI-1), and soluble thrombomodulin (sTM) induced by dengue 2 virus (D 2V) infection of vascular endothelial cells. Methods The effects of D 2V infection on the production of t-PA, PAI-1, and sTM of human umbilical cord vein cells were studied. Results D 2V infection significantly induced the secretion of sTM and t-PA but showed no such effects on PAI-1 of human endothelial cells. Antibody against IL-6 inhibited D 2V-induced t-PA production of endothelial cells. A close correlation between serum levels of IL-6 and t-PA was found in dengue hemorrhagic fever (DHF) but not in dengue fever (DF) patients. Conclusion IL-6 can regulate D 2V-induced t-PA production of endothelial cells, suggesting that endothelial cells can be the target for D 2V infection and that D 2V-induced t-PA, TM, and IL-6 production of endothelial cells may contribute to the pathogenic development of dengue haemorrhagic fever/dengue shock syndrome (DHF/DSS).
8.Curative effects of percutaneous vertebroplasty with poly(methyl methacrylate) versus conservative treatment on fresh osteoporotic thoracolumbar vertebral compression fractures

BACKGROUND:Percutaneous vertebroplasty for vertebral fractures can effectively relieve acute pain and has the advantages of smal trauma, good curative effect and less complications, but for patients with osteoporotic compression fractures, there were varying degrees of osteoporosis after surgery, which have a longer course of disease and cannot be easy to cure. So the effectiveness of percutaneous vertebroplasty cannot be ful y evaluated based on the pain relief. OBJECTIVE:To study the curative effect of percutaneous vertebroplasty for patients with fresh osteoporotic thoracolumbar vertebral compression fractures. METHODS:We selected 24 patients undergoing percutaneous vertebroplasty and 24 patients receiving conservative treatment at the same time who had fresh osteoporotic compression fractures as research objects;and compared pain degree, vertebral body height and the kyphosis Cobb angle, function activity of the lower lumbar before and after treatment, the quality of life and clinical incidence of complications within 6 months after treatment in the two groups. RESULTS AND CONCLUSION:The degree of pain, the vertebral body height, kyphosis Cobb angle, function activity of the lower lumbar were al improved in the two groups after treatment (P<0.05), and these indexes in the percutaneous vertebroplasty group were better than those in the conservative treatment group (P<0.05). The quality of life and incidence of complications within 6 months after treatment were improved better in the percutaneous vertebroplasty group than the conservative treatment group (both P<0.05). These results suggest that the percutaneous vertebroplasty for fresh osteoporotic thoracolumbar vertebral compression fractures can effectively reduce the pain of patients, improve vertebral deformity and activities of the lower lumbar, and has obvious role in promoting the postoperative quality of life of patients.
9.LPS increases plasminogen activator inhibitor type-1 expression in human endothelial cells

AIM: The aim of this study is to elucidate the effects of lipopolysaccharide (LPS) on tissue plasminogen activator(tPA) and plasminogen activator inhibitor type-1(PAI-1) expression and secretion in endothelial cells. METHODS: Cultured human umbilical vein endothelial cells (HUVECs) were induced by LPS for different times. Cell viability was then determined by cell counting kit-8. tPA and PAI-1 activities in the media were assayed by fibrin overlay and reverse fibrin autograph, respectively. Cytoplasmic RNA was prepared using the Trizol method and was assayed for PAI-I and tPA mRNA levels by reverse transcript-polymerase chain reaction(RT-PCR). RESULTS: LPS(10 mg/L) did not produce cell toxicity according to LDH determination in culture media. PAI-1 activity in LPS group was high (P0.05). CONCLUSION: LPS (10 mg/L) did not show signs of cell toxicity, but promoted the expression of PAI-1 mRNA and induced an increase activity of PAI-1. However, LPS (10 mg/L) did not change tPA mRNA expression. The time-dependent increase in PAI-1 mRNA expression and activity shifts the local balance toword increased anti-fibrinolytic capacity, which can amplify the extent of acute thrombosis after plaque rupture. This is one of the possible reasons that cause thrombus,blood coagulation and disseminated intravascular coagulation (DIC) during septicemia.
10.Imaging features and pathological findings of alveolar soft part sarcoma

Objective To explore the correlation between imaging features and pathological findings of alveolar soft part sarcoma (ASPS).Methods The clinical data,radiographic images and pathological characteristics of seven patients with histopathologically proved ASPS from 2003 to 2009 were retrospectively reviewed.Digital radiography (DR),plain CT,pre and post-contrast MR and DSA were performed in all patients.HE staining and periodic acid-Schiff (PAS) staining were used in pathological examination and CD34 were marked in immunehistochemical study.Results All patients were females.Their age ranged from 16 to 37 years with median of 24 years.Two tumors were located in pelvics,three were located in thighs,and two were located in legs.DR and CT plain scan features of ASPS were non-specific.MR demonstrated the tumors with large diameter and well defined margin locating in the deep tissues.Tumors were hyperintense on T2-weighted and T1-weighted images with many flow void vessels and persistent remarkable enhancement.There were numerous vessels with arteriovenous fistula on DSA.Pathological examination revealed solid nests of several polygonal tumor cells arranged in an alveolar pattern and separated by thinwalled,sinusoidal vascular channels.Tumor cell were noted on periodic acid-Schiff (PAS) stains.And the endothelial cells of sinusoidal vascular channels were positive on CD34.Conclusions As a malignant tumor which predominantly developed in deep location of trunk or extremities in young females,numerous void signal and hyperintensity on T1-weighted were the features of ASPS on MR.Numerous vessels could also be confirmed on DSA.These image findings were correlated closely with the pathological manifestations.
