Objective: To set a quantified diagnostic standard for large intestinal cancer of spleen qi deficiency syndrome. Methods: The spleen qi deficiency syndrome was identified by experts on the basis of clinical epidemiological investigation of 311 patients suffering from large intestinal cancer. Corresponding points were assigned to the correlative factors (traditional Chinese medicine symptoms) on the basis of symptom differences between spleen qi deficiency syndrome and non-spleen-qi-deficiency syndrome. The best threshold was determined by receiver operating characteristic curve (ROC) according to syndrome differentiation from expert team, and the quantified diagnostic standard was established. The syndrome identification from the expert team which was regarded as golden standard was tested retrospectively. Results: All the traditional Chinese medicine symptoms possibly related to spleen qi deficiency syndrome were analyzed based on the opinions of experts, and 28 symptoms were confirmed as candidate correlative factors. The occurrence of 11 symptoms between spleen qi deficiency syndrome and non-spleen-qi-deficiency syndrome showed statistical differences by means of crosstabs analysis (P<0.05). The 11 symptoms were filtered by logistic regression analysis, and tiredness, fatigue, loose stool, and poor appetite were finally determined as the symptoms relative to large intestinal cancer. These four symptoms were analyzed with conditional probability conversion and endowed with 16, 11, 4 and 8 points respectively. The diagnostic standard of spleen qi deficiency syndrome of large intestinal cancer was over 13 points. The sensitivity, specificity and accuracy of retrospective examination were all above 80%, and its positive likelihood ratio was 9.89. Conclusion: The quantified diagnostic standard for spleen qi deficiency syndrome of large intestinal cancer is in accordance with clinical characteristics of large intestine cancer and the characteristics of TCM syndrome diagnosis.

Objective: To investigate the relationship of nm23 and VEGF expression with hilar lymph node micrometastasis and the prognosis of stage Ⅰ non-small cell lung cancer (NSCLC). Methods: Immunohisto-chemistry was used to detect nm23 and VEGF protein expression in primary cancer tissue and cytokeratins in 86 hilar lymph nodes from 40 patients with stage Ⅰ NSCLC. Kaplan-meier method and Log rank test were used to analyze the 5-year survival. Results: The rate of positive hilar lymph node micrometastasis was 12.5% for stage Ⅰ NSCLC. Lymph node micrometastasis was not statistically correlated with gender, age, histologic type, differentiation, primary tumor size or VEGF protein expression (P>0.05). But it was reversely associated with nm23 protein expression in primary cancer tissue of NSCLC (P<0.05). The 5-year overall survival of pa-tients with well-differentiated NSCLC, positive nm23 expression and negative lymph node micrometastasis was better than those with moderately and poorly differentiated NSCLC, negative nm23 expression and posi-tive lymph node micrometastasis (P<0.05). Lymph node micrometastasis and nm23 protein expression were identified as two independent prognostic factors for stage Ⅰ NSCLC by univariate Cox regression analysis. Conclusion: nm23 protein expression in pdmary cancer tissue of stage Ⅰ NSCLC is closely associated with hi-lar lymph node micrometastasis, nm23 protein and hilar lymph node micrometastasis are two independent prognostic factors for stage Ⅰ NSCLC. Patients with nm23 protein deletion and positive lymph node microme-tastasis have a poor prognosis.

Objective To study the effects of low dose sodium arsenite to human bone marrow mesenchymal stem cells(hBMSCs) differentiation during establishing of arsenic-resistant cell model. Methods hBMSCs were prepared in conventional method and continuously exposed to 1μmol/L sodium arsenite for ≥12weeks inv vitro. Forty-eight hours acute arsenite toxicity test was drived to assay if the cells acquired arsenic-resistance. The proliferation capacity of CAsE-hBMSCs was observed by the rate of colony formation. The expression of Oct-4 in CAsE-hBMSCs was assayed by RT-PCR and immunocytochemistry. The expression of ABCG2 in CAsE-hBMSCs was analyzed by RT-PCR. Results hBMSCs continuously exposed to 1μmol/L sodium asenite for ≥12 weeks exhibited dramatic resistance to acute arsenite toxicity. The LC 50 for acute arsenite exposure in CAsE-hBMSCs was 35.59μmol/L versus 18.04μmol/L in control cells. Compared to control cells, the CAsE-hBMSCs didn't show malignant proliferation ability. Expression of Oct-4 gene was positive in 4th, 18th passage hBMSCs and the hBMSCs induced by arsenite for 4 weeks but negative in CAsE-hBMSCs. The expression of Oct-4 protein was positive and weakly positive in 4th passage hBMSCs and CAsE-hBMSCs respectively, and the positive granules of Oct-4 distributed in cytoplasm. The expression of ABCG2 gene in CAsE-hBMSCs was obviously lower than that in control cells ( P <0.001). Conclusion Human bone marrow mesenchymal stem cells could be induced to differentiation by low dose sodium arsenite.

Journal of Central South University(Medical Sciences) 2013;38(11):1177-1187
Despite seminal studies in the 1920s by Warburg showing a characteristic metabolic pattern for tumors, cancer bioenergetics has otfen been relegated to the backwaters of cancer biology. Recent studies have shown that metabolism in the tumor tissue is far more complicated than we previously knew. Despite vigorous glycolysis, in fact, the tumor tissue still retains mitochondrial aerobic metabolism. Mitochondria is one of the main sites of the biosynthesis process of tumor cells. Recent studies revealed that abnormal fatty acid metabolism. Amino acid metabolism plays a key role in the tumorigenesis. The metabolic network in tumor cells was reprogrammed, leading to nutrition lfux reorganization and re-direction. Metabolism reprogramming in tumor cells facilitates the balance between the needs of energy supply and the synthesis of biological macromolecules. Targeting cell metabolism is not intended to interfere the energy supply of tumor cells but to affect the synthesis of metabolic rate, thereby inhibiting the proliferation of the tumor. hTe review focuses on the importance of metabolic reprogramming in tumor development and cancer therapy. We summarize what is currently known about metabolic reprogramming and establish a framework to understanding its contribution to the altered metabolism of cancer cells.
