1.Nutritional Risk Screening 2002 (NRS 2002) as a predictor of outcome during ward based non-invasive ventilation in COPD with respiratory failure

Objective To determine whether Nutritional Risk Screening 2002 (NRS2002) of admission independently predicts outcome of non-invasive positive pressure ventilation (NIPPV) in chronic obstructive pulmonary disease (COPD) patients during hypercapnic respiratory failure.Methods Patients with COPD presenting with hypercapnic respiratory failure at Sichuan University Hospital between December 2010 and May 2012 and receiving NIPPV were studied prospectively.NRS2002 was measured before NIV administration.233 patients met the inclusion criteria,with NIPPV failed in 71 cases and succeed in 162 cases.The patients were followed up till they were discharged.Results After multivariate Logistic regression,the items such as baseline PaCO2,NRS2002 score could predicted 76.4% of the failure outcome.The area under the curve was 0.767.The positive predictive value was 35.23%.The negative predictive value was 84.21%.Conclusions We can predict the risk failure of NIPPV in patients with COPD and hypercapnic respiratory failure with NRS2002 score and baseline PaCO2,and adjust the treatment project according to the evaluation result.NRS2002 supply non-invasive and portable method for predicting the failure of NIPPV.
2.A Preliminary Study on Preservation Methods of Blastocystis hominis

Objective To observe the effect of different cryoprotective agents and temperature factors on the viability of Blastocystis hominis so as to explore the ideal method for preservation of B.hominis.Methods B.hominis agents were obtained from a patient's fecal specimen.Having washed by normal saline and divided into tubes,the samples were cryopreserved in-20 ℃ refrigerator or in-l96 ℃ liquid nitrogen with 10% DMSO,40% glycerol and 15% ethylene glycol respectively.The thawed B.hominis agents were then used for culture.By trypan blue staining and microscopy,the viability and proliferation of those resuscitative cells were investigated.Results B.hominis survived for 3 weeks at 18 ℃-20 ℃ while less than 1 week at 4 ℃-6 ℃.When stored in-20 ℃ refrigerator or liquid nitrogen with cryoprotective agents,they survived for more than 3 months.The cryopreservation with 40% glycerol at-196 ℃ for 6 months resulted in 41.7% viability of the revivified cells.Cleavage cells were easily observed after culturing for 72 hours.Conclusion Preserving B.hominis in liquid nitrogen with 40% glycerol is an optimal cryopreservation protocol.
3.Study on Quality Standard of Anti- virus Oral Liquid

Objective To improve the quality standard of Anti- virus Oral Liquid. Methods Rhizoma Anemarrhenae and Rhizoma Acori tatrinowii in Anti- virus Oral Liquid were identified by TLC. Forsythin in Fructus Forsythiae was determined by HPLC. Results Rhizoma Anemarrhenae and Rhizoma Acori tatrinowii can be identified by TCL. The linearity of forsythin in the range of 0.1~ 0.6 ? g/? L was good, r=0.9998. The average recovery is 99.87 % , RSD=1.93 % . Conclusion The methods is accurate, reliable and specific and can be used for the quality control of Anti- virus Oral Liquid.
4.Cloning, Expression and Identification of Surface Antigen SAG4 of Toxoplasma gondii

Objective To clone and express surface antigen SAG4 gene of Toxoplasma gondii, and analyze its immunoreactivity. Methods Specific primers were designed based on the reported SAG4 gene of T. gondii RH strain (GenBank Accession No: AF340224.1). Using genomic DNA from T. gondii as templates, SAG4 gene was amplified by PCR. The PCR product was cloned into pMD19-T vector and identified by digestion with restriction enzyme and PCR. Then the target fragment was subcloned into pET28a(+) vector, transformed into E. coli BL21 and followed by expression of the protein induced by IPTG. The protein was identified by Western blotting. Results The target gene was amplified with the length of 537 bp. Sequence analysis showed that the predicted amino acid sequence was identical with that of SAG4 as a membrane protein in T. gondii. After induced by IPTG, the recombinant SAG4 protein existed in an inclusion body form. The recombinant SAG4 (Mr 18 740) was recognized by serum of infected mice. Conclusion SAG4 has been expressed and shows certain immuno-response activity.
5.Effect of ketamine on expression of hippocampal microRNA-206 in mentally depressed rats

Objective To evaluate the effect of ketamine on the expression of hippocampal microRNA-206 (miR-206) in the mentally depressed rats.Methods Eighty healthy female Sprague-Dawley rats,weighing 200-300 g,aged 2-3 months,were randomly divided into 4 groups (n=20 each) using a random number table:normal control group (C group),ketamine group (K group),depression group (D group) and depression + ketamine group (D+K group).Mental depression was induced by exposing the animal to chronic unpredictable mild stress.At 1 day after successful establishment of the model,ketamine 15 mg/kg was injected intraperitoneally in K and D+K groups,while the equal volume of normal saline was given in C and D groups.Before establishment of thc model,at 7,14,21,28 and 35 days after beginning of establishment of the model,and at 1,3,5 and 7 days after administration,sucrose preference test was performed.Open field test was carried out at 1 day after administration.Forced swimming test was performed at 2 days after administration.Five rats were sacrificed at 6 h after administration,and hippocampal tissues were isolated for determination of the expression of hippocampal miR-206 by quantitative real-time polymerase chain reaction.At 2 days after administration,5 rats were sacrificed,and hippocampal brain-derived neurotrophic factor (BDNF) expression was detected by Western blot.Results Compared with C group,the volume of sucrose intake at 21-35 days after beginning of establishment of the model and at each time point after administration and the score of open field test were significantly decreased,the immobility time in forced swimming test was significantly prolonged,the expression of miR206 was up-regulated,and the expression of BDNF was down-regulated in D group,the immobility time in forced swimming test was shortened,the expression of miR-206 was down-regulated,and the expression of BDNF was up-regulated in group K,and the score of open field test and volume of sucrose intake at 21-35days after beginning of establishment of the model and the score of open field test were significantly decreased in D+K group (P<0.05).Compared with D group,the volume of sucrose intake at each time point after administration was significantly increased,the immobility time in forced swimming test was shortened,the expression of miR-206 was down-regulated,and the expression of BDNF was up-regulated in D+K group (P<0.05).Conclusion The mechanism by which ketamine exerts antidepressant effects is related to down-regulation of miR-206 expression and up-regulation of BDNF expression in the hippocampus of rats.
6.Effects of different dental implant systems on the peri-implant bone absorption

BACKGROUND:The peri-implant bone absorption is closely related to the repair effect. OBJECTIVE:To compare the effects of three kinds of dental implant systems on the peri-implant bone absorption. METHODS:116 patients who underwent the dental implant systems were col ected, including 46 cases with 3I implant system, 40 cases with ITI implant system and 30 cases with BLB implant system. The peri-implant bone absorption, sulcus bleeding index and periodontal probing depth of three groups were detected at 1, 3, 6, 9 and 12 months after implantation, respectively. RESULTS AND CONCLUSION:The peri-implant bone absorption of three groups within 1 year after implantation was in a rise, and the bone absorption of BLB group was significantly higher than that of ITI and 3I groups at 3 and 12 months after implantation (P<0.05). Compared with the natural teeth, the gingival sulcus bleeding index of three groups were al increased at different time points after implantation;the gingival sulcus bleeding index of BLB group was significantly higher than that of natural teeth at 6 months after implantation (P<0.05);the gingival sulcus bleeding index of three groups were significantly higher than that of natural teeth at 9 months after implantation (P<0.05). The periodontal probing depth of three groups showed an ascending trend at 6 months after implantation;the periodontal probing depth of three groups was higher than that of natural teeth at different time points after implantation, which exhibited significant differences at 6 and 9 months after implantation (P<0.05). In conclusion, three kinds of dental implant systems exhibit differet effects on the peri-implant bone absorption, but al achieve excel ent clinical efficacies.
7.The construction of permanent colostomy patients with rectal cancer in archives information module content

Objective To construct the permanent colostomy patients with rectal cancer in archives information module content. Methods Using expert meeting,semi- structured interview, literature review, Delphi expert consultation method, establish the system of permanent rectal cancer patients file information specific module. Results In two- round consultation authority coefficient was 0.847. Kendall coordination coefficient (Kendall′s W) was 0.195-0.331, the difference had statistical significance (P<0.01); through the expert consultation method to construct the file information module for permanent colostomy patients with rectal cancer, which contained 4 first-level indicators (colostomy patient information module, stoma nursing modules, stoma patients transitional care modules, colostomy patients with network platform module), 11 secondary indicators, 36 third grade indicators. Conclusions This study established file information module content for permanent stoma patients with rectal cancer. It has good scientificity and reliability, and can provide a theoretical basis for the clinical development of a file information system network for transitional care of permanent stoma patients with rectal cancer.
8.Quality standard of Fuke Yangkun Pill

AIM:To improve the quality standard of Fuke Yangkun Pill. METHODS: Fructus Viticis was(identified) by micrology,Radix Angelica sinensis,Rhizoma Chuanxiong,Rhizoma Cyperi,Radix Aucklandiae,Rhizoma Corydalis,Radix Rehmanniae,Radix Rehmanniae preparata,Radix Paeoniae alba,Radix et Rhizoma Glycyrrhizae,Fructus Amomi were identified by TLC,baicalin content was determined by HPLC. RESULTS: The TLC spots were fairly clear and distinguishable,the linear range of baicalin was within 3.28-131.2 ?g/mL.The average recovery was 100.4% with RSD 1.0%(n=6). CONCLUSION: The method is simple,and specific,and can be used for quality control of Fuke Yangkun Pill.

Objective To investigate the expression and clinical significance of T-cell receptor (TCR) Ⅴβ subfamily in hepatitis B virus (HBV)-related acute-on-chronic liver failure (HBV-ACLF) patients.Methods Twenty-eight patients with HBV-ACLF (HBV-ACLF group) and 32 patients with chronic hepatitis B flare (CHB-F group),who were treated in The Second People's Hospital from Oct.2010 to Mar.2012,and 20 healthy controls (HC group) were included in the study.Reverse transcriptase-polymerase chain reaction was used to detect the levels of TCR Ⅴβ subfamily and enzymelinked immunosorbent assay was used to detect the levels of serum cytokines [interleukin (IL)-2,IL-4,IL-6,IL-10,interferon (IFN)-γ and tumor necrosis factor (TNF)-α)] in the three groups.The comparison among three groups was done by one-way analysis of variance and the comparison between two groups was done by LSD-t test or rank sum test.Results The three groups had similar gender and age distribution (all P>0.05).The HBV-ACLF group had significant different profiles of total bilirubin,albumin,prothrombin activity,international normalized ratio and cholesterol tatol compared with the CHB-F group (all P<0.05).For patients in the HBV-ACLF group,the serum IL-2,IL-4,and IL-10 levels were lower(all P=0.000),and the IL-6 and IFN γ levels were higher than those of the HC group (all P=0.000).The IL-4,IL-10,and TNF-α levels in the CHB-F group were also significantly lower than those of the HC group (all P=0.000).Compared with the CHB-F group,the HBV-ACLF group had significantly lower IL-2,IL-10,and TNF-α levels (P=0.003,0.002,0.004),and higher IL-6 and IFN-γ levels (P=0.015,0.006).By one-way analysis of variance,there were significantly differences of △Ct1,△Ct5,△Ct7,△Ct12,△Ct15,△Ct20,△Ct22,and △Ct23 among the three groups (H=20.368,14.368,19.500,31.532,19.985,19.116,41.752 and 20.649,all P<0.05).Conclusion The expression levels of TCR Ⅴβ subfamily and cytokines are changed in HBV-ACLF patients.
10.Association between autoimmune diseases and aging

The pathogenesis of autoimmune diseases(AID)involves the inflammatory response of the human body, and inflammatory pathways are also shared by mechanisms of aging.Clinical observations of AID have revealed evidence of aging-related diseases in younger adults.This article reviews the literature related to research progress on AID combined with aging-related diseases, such as cardiovascular diseases, diabetes and osteoporosis, and briefly explains the underlying mechanisms, in order to promote interdisciplinary research between the two fields.
