Chinese Journal of Plastic Surgery 2015;31(6):450-455
OBJECTIVETo explore the effect of recombinant human parathyroid hormone 1-34 [rhPTH(1-34)] on bone regeneration rabbit mandible during distraction osteogenesis (DO).
METHODS40 Japanese white rabbit (weight 2.0-2.5 kg) were randomly divided into control group and groups. The experimental groups were divided inito 12.5, 25 and 50 µg/kg group according to the dosage of rhPTH (1-34) in each group. Each group involved 10 rabbits, and unilateral DO models were established at the right mandible of the rabbits. From the first day of distraction to the day of execution, the rabbits in the experimental groups were injected subcutaneously rhPTH (1-34) of the corresponding dose respectively, and the rabbits in the control group were injected subcutaneously 2% heat inactivated rabbit serum 1 ml respectively.. Five rabbits in each group were executed respectively at 1 week and 3 weeks after completion of distraction, and the specimens of DO were harvested. The gross observation, X-ray examination, and histological study were performed.
RESULTSGross appearance: At the first week of consolidation, the dense and opaque white tissue was seen in the distraction gap of the 50 µg/kg group, and the white translucent tissue was seen in the distraction gaps of the rest groups. At the third week of consolidation, the greyish white tissue was seen in the distraction gap of the control group, while the cartilage-like tissue was seen in the buccal side of the distraction gap of the 12.5 µg/kg group, the color of new-formed tissues was close to that of normal bone tissue in the lingual side. The buccal tissue at the edge of the distraction gap of the 25 µg/kg group fitted together with the primary bone tissue in its two sides. It was difficult to distinguish the boundaries between the distraction gap and the bone tissues in its two sides in the 50 µg/kg group. X-ray findings: At the first week of consolidation, a sparse opaque image was seen in the distraction gap of the 50 µg/kg group, and a low-density image was seen in the distraction gap of the rest groups. At the third week of consolidation, a sparse bone image was seen in the control group, and the edge of the bone was not continuous. With the increase of the dose in the experimental groups, the image of the distraction gap became more and more opaque, and the image of the distraction gap in the 50 µg/kg group was close to that of the normal bone tissue. HISTOLOGICAL FINDINGS: At the first week of consolidation, few osteoblasts were present at the edge of the distraction gap of the control group. A large number of bone cells and bone trabecular were present in the distraction gap of the 12.5 µg/kg group, the network of the bone trabecula was present in the 25 µg/kg group, and a few new bones were found in the 50 µg/kg group. At the third week of consolidation, the network of the trabecular bone was present in the distraction gap of the control group, while the network of the bone trabecula was present in the 12.5 µg/kg group, a lot of bone-like tissues in the 25 µg/kg group, and near-mature bone in the 50 µg/kg group.
CONCLUSIONSrhPTH(1-34) can promote the formation of new bone in the distracted gap during mandibular DO in rabbits.

Chinese Journal of Experimental Ophthalmology 2014;32(4):303-307
Background Minocycline possesses neuroprotective effect in a variety of animal models and clinical trials of central nervous system, but whether it works on optic nerve injury remains unclear. Objective This study aimed to observe the protective effects of minocycline on retinal ganglion cells (RGCs) in the early stage of optic nerve crush and explore its mechanism. Methods One hundred and thirty-six clean C57BL/6J mice were randomly divided into normal control group, normal saline solution group and minocycline group. The optic nerve crush injury models were induced in the left eyes of the mice in the normal saline solution group and minocycline group by a cross-action forceps for 3 seconds.Minocycline was injected intraperitoneally in the minocycline group firstly 45 mg/kg(0.4 ml) and followed by 22.5 mg/kg per day after 24 hours until sacrifice of the animals, and the equivalent volume of normal saline solution was injected in the same way in the normal saline solution group. The mice were euthanized at 4,7,11,14 days postoperatively and the left eyeballs were collected.Retinal flat mounts and DAPI staining was used to observe and compare the change of RGCs density among different groups and various time points.Apoptosis of mice RGCs were assessed by TdT-mediated dUTP nick end labeling (TUNEL).Real-time polymerase chain reaction (real-time PCR) was used to detect the expression of CD11b mRNA in retinal microglials. Results DAPI staining in retinal flat mounts showed that the average RGCs density was (77.50±2.38)/0.01 mm2 and (70.00±2.94) /0.01 mm2 in the 4th and 7th day after modeling in the normal saline solution group, and those in the minocycline group were (88.75 ± 2.36) /0.01 mm2 and (81.00 ± 3.92)/0.01 mm2,with significant differences between the two groups (t4d =-6.708,P<0.01 ;t7d =--4.491,P<0.01).The apoptotic RGCs were (12±1)/mm and (4±1)/mm in the normal saline solution, which were significantly more than (4±1)/mm and (1±0)/mm in the minocycline group (t4 d =12.832,P<0.01 ; t7d =3.455,P =0.026).However,no significant difference was found in apoptotic RGCs in postoperative 11 days and 14 days between the normal saline solution group and the minocycline group (P =0.708,0.777).The expressing levels of CD11 b mRNA in the retinal microglials were significantly higher in the 4th and 7th day in the normal saline solution group than those in the minocycline group (t4 d =8.312,P<0.01 ;t7d=5.407,P<0.01),but were not significantly different in the 11st and 14th day after modeling between the two groups (P=0.055,0.170).Conclusions Minocycline can play a neuroprotective effect on RGCs in the early stage of optic nerve crush in mice by inhibiting microglia activation and decreasing RGCs apoptosis.

Chinese Journal of Tissue Engineering Research 2013;(42):7389-7395
BACKGROUND:Studies addressing reconstruction of tooth tissue engineering have shown that tooth structure can be constructed using tissue engineering technology. Tooth root and its periodontal attachment are critical for tooth survival and functions, based on which, whether we can target root tissues with simple structure for tissue engineering construction by bypassing a complex dental tissue engineering concept with the structural integrity?
OBJECTIVE:To construct a tissue-engineered dental root by seeding dental papil a cells, as seed cells, into poly(lactic-co-glycolic acid)/sodium alginate hydrogel.
METHODS:Rabbit dental papil a cells were isolated and cultured. The cells were then mixed with 1%sodium alginate hydrogel at a final density of 6×109/L. The cellsuspension was seeded into a poly(lactic-co-glycolic acid) scaffold with predetermined shape of human tooth and solidified with calcium chloride. Final y, the cel-scaffold composites were subcutaneously implanted into the back of nude mice. The specimens were harvested after 4 and 8 weeks respectively and processed for gross inspection, X-ray and CT examination and histological observation.
RESULTS AND CONCLUSION:The newly formed tissue kept the original shape of human dental root 4 and 8 weeks post-implantation. After 4 weeks of implantation, the specimen density was low;the root implants appeared to be incompletely mineralized, alginate hydrogels were degraded, but the copolymer scaffold was not degraded;a number of dentin-like structure appeared, and a fibrous membrane structure was visible on the surface of specimens paral el to the root surface, but the structure was not continuous, and no pulp cavity formed. After 8 weeks, the newly formed tissue was highly mineralized close to root tissue of the nature tooth;the copolymer scaffold was mostly degraded;specimens appeared to have a large number of mature dentin-like structure, and form continuous fibers membrane on the surface paral el to the root surface, below which, cementum-like structure formed. Artificial dental root with biological y similar structures of human dental roots can be constructed using the method of tissue engineering.
