1.Development and verification of a quantitative real-time PCR method for determination of host cell DNA residues in inactivated SARS-CoV-2 vaccine(Vero cells)

Objective To develop and verify a quantitative real-time PCR method for determination of the content of host cell DNA residues in severe acute respiratory symptom coronavirus 2(SARS-CoV-2) inactivated vaccine(Vero cells),in order to better control the safety of products.Methods DNA was extracted from inactivated SARS-CoV-2 vaccine(Vero cells) bulk by magnetic bead separation method,and the DNA residues of host cells were quantitatively analyzed by probetype PCR.The linear range,repeatability,intermediate precision,quantitative limit,specificity,durability and accuracy of the developed method were verified,and the host cell DNA re sidues of 5 batches of inactivated SARS-CoV-2 vaccine(Vero cells)were determined by this method.Results DNA standard curve showed good linearity in the range of 300~0.003 pg/μL(each R~2> 0.99);Relative standard deviations(RSD) of repeatability and intermediate precision verification were less than 20%;The quantitative limit was 0.001 pg/μL;Sample dilution and purified liquid dilution had no interference to detection;The results of 60 min incubation at 53,55,57 ℃ and 56,60,64 min incubation at 55 ℃ showed no significant difference;The recoveries of accuracy verification were 79%~83%,RSD <5%.This method had good adaptability in detecting DNA residues in the bulk of inactivated SARS-CoV-2 vaccine(Vero cells).Conclusion The quantitative realtime PCR method for determination of host cell DNA residues in inactivated SARS-CoV-2 vaccine(Vero cells) has been successfully developed,of which the linearity and range,repeatability,intermediate precision,quantitative limit,specificity,durability and accuracy meet the acceptable standards,and are suitable for the detection and quality control of host cell DNA residues in inactivated SARS-CoV-2 vaccine(Vero cells).

3.Practice and Experience of Pharmaceutical Care for Hemophilia Children by Clinical Pharmacists

OBJECTIVE:To explore appropriate pharmaceutical care methods for hemophilia children,and to improve rational drug use. METHODS:Drug consultation hemophilia records collected from our hospital during Nov. 2014 to Jul. 2015 were summa-rized in respects of personal structure,form and content;key cases were analyzed specially. RESULTS:During Nov. 2014-Jul. 2015,clinical pharmacists received 130 times/cases of medication consultation that related to hemophilia in our hospital;patient’s family members occupied the main part (70.0%);most of drug consultation form was information platform for consultation (58.5%);consultation content mainly covered drug combination and auxiliary medication (77.7%). Clinical pharmacists have re-duced the risk of bleeding,improved the quality of life,reduced the economic burden of families and strengthened the awareness of medical staff and patient’s family member on safe drug use through finding out medication error timely,paying attention to the component of compound drug,guiding medication direction and optimizing therapy plan. CONCLUSIONS:Clinical pharmacists ac-tively provide pharmaceutical care and promote rational drug use during hemophilia treatment from medication consultation,ratio-nal drug use,etc.
4.Complications in laparoscopic gynecologic surgery

Objective To investigate the complications of gynecological laparoscopies and its related fac-tors. Methods The clinical data of 8700 cases with laparoscopic surgeries from August 2003 to August 2008 were retrospectively analyzed. Results Complications occurred in 259 cases,with a overall complication rate of 2.98%, the serious complication occurred in 37 cases(0.43%), and surgical treatment was needed in 21 cases. The compli-cations included: 129 complications related to puncture and pneumoperitoneum(including subcutaneous emphysema and abdominal wall congestion) ,20 related to bleedings, 17 related to injuries ,26 related to postoperative infections. Conclusion Operative gynecologic laparoscopy-induced complications were associated with operation difficulty, so great attention should be paid. In order to reduce the risks of laparoscopic surgery, indications should be selected cor-rectly,and procedures should be operated carefully.
5.Exploration of bilingual teaching mode in gynecology and obstetrics of medical college

6.Screening out nasopharyngeal carcinoma by two-stage ELISA for EB virus

Objective:Screening out NPC from susceptable crowd by serological test for EB virus.Methods:Checking antibody of EB virus by ELISA. At first, EBNA1 IgA was detected for susceptable crowd as a primary screening index, secondary , EBNA1 IgG and Zta IgG were detected in the EBNA1 IgA positive group.Results:The sensitivity and specificity of EBNA1 IgA were 91.8% and 91.4%, both were higher than those of EBNA1 IgG and Zta IgG; the sencondary detection of EBNA1 IgG and Zta IgG raised the specificity of NPC screening to 96.5% and also helped to divide the crowd into 3 groups as high, median, and low risk. And the high risk group accounted for 0.39%.Conclusion:Two-stage screening of NPC raise the sensitivity and specificity of NPC detection. It also helps to divide the crowd into 3 groups of risk.
7.Changes of inflammatory cytokines in leukocyte-depleted PCs and decreased FNHTR by leukoreduced PCs transfusion

Objective To investigate the changes of leukocyte derived cytokines,IL 1?,IL 6,IL 8,and TNF ?,in platelets concentrates (PCs) during storage,and to study the effect of leukodepletion on cytokine concentration in PCs and the incidence rate of febrile non hemolytic transfusion reaction (FNHTR).Methods 8 bags of apheresed PCs from 8 voluntary donors were each divided into two identical portions (A and B).Group A underwent leukocyte filtration,while group B did not.Then both groups were stored at 22℃ for 5 days.The levels of cytokines (pg/ml),white blood cell (WBC) count and platelet count in both groups were tested on the day 0,3 and 5 during storage.In clinic,240 patients were randomly divided into two groups with 120 in each group to receive non filtered or filtered PCs (1 bag/patient).The incidence of FNHTR was investigated.Paired t test and ? 2 test were used in data analysis. Results There was a significant positive correlation between the content of WBC in PCs and the level of cytokines.On the 5th day of storage,residual WBC in leukoreduced PCs were less than 1?10 6/PC,the levels of cytokines remained the same as those on 0;however,WBC in non filtered PCs was (351?81)?10 6/PC,and the levels of IL 1??IL 6?IL 8 and TNF ? were 105.0pg/ml,269.0pg/ml,1840pg/ml and 42.0pg/ml respectively and increased 18 ( P

Objective Seeking for a new therapy for effectively correc ting hypoxemia of severe hepatitis cases in terms of oxygen-enriched liquids in f usion, for a rapid boost of the pressure of blood oxygen bypassing the pathologi cally damaged alveolus of such patients.Method Intravenous drip with oxygen -enriched liquids for 23 cases who suffered from Severe hepatitis in courses of tre atment varying between 14 to 30 days, 500-700 mL per day.Result For all the 23 c ases, their hypoxemia was gradually corrected to normal in 20 minutes' or 7 hour s du ring intravenous drip with oxygen-enriched liquids and TNF ?IL-6?IL-8 were dec r eased after the treatment courses.The death rate of treatment group was l ess.Conclusion Intravenous drip the oxygen-enriched liquids has effectively helped to corre ct the hypoxemia of severe hepatitis cases bypassing the malady alveolus throug h which the oxygen failed in by conventional oxygen halation. Death rate hase been decreased after the courses .This therapy of oxygen-enriched liquid infusion can be quite useful to save their life in the combined treatment for severe hepatitis.
9.Clinical analysis of 1272 cases of pelvic endometriosis

Objective To assess the surgical effectiveness of pelvic endometriosis (EMS) by Laparoscopic. Methods Retrospective analysis was performed in 1272 pelvic EMS patients underwent laparoscopic surgical treatment. All patients were classified into four groups according to the R-AFS,63 patients of Ⅰ stage,44 patients of Ⅱ stage,475 patients of Ⅲ stage and 690 patients of Ⅳ stage. Symptoms of EMS,surgical effect and follow-up outcomes were compared. Results In all stages, there were no significant differences on moderate to severe dysmenorrhea,increase in CA125 and infertility (Ps > 0. 05 ). In the EMS patients combined with AM or/and DIE, the rates of the moderate to severe dysmenorrhea, CA125 increase and infertility were 84.6% (11/13 ) ,92.3 %(12/13) and 53.8% (7/13)in stage Ⅰ ,81.8% (9/11) ,90. 9% ( 10/11 )and 54.5% (6/11) in stage Ⅱ ,84.8%(173/204) ,93. 1% (190/204) and 47.1% (96/204) in stage Ⅲ ,and 81.6% (213/261) ,91.2% (238/261) and 46.4% ( 121/261 ) in stage Ⅳ, respectively. The rates in non-consolidated AM or/and DIE patients were 46.0%(23/50),62.0% (31/50) and 22.0% ( 1 1/50) in stage Ⅰ ,45.5 % ( 15/33 ),54.5% ( 18/33 ) and 18.2% (6/33)in stage Ⅱ ,41.7% ( 113/271 ) ,62.7% ( 165/271 ) and 23.6% (64/271) in stage Ⅲ ,and 47.3% (203/429),60.1% (248/429) and 21.7% (93/429). The difference between the two groups with and without consolidated AM or/and DIE had statistical significance( P < 0.05 or P < 0.01 ). After the treatment, the scores of life's energy, ache,emotion,sleep,social activity and body movement of NHP were significantly superior than those before treatment in all patients ( P < 0.01 ). The surgery time-consuming of Ⅲ and Ⅳ stage patients were (64.5 ± 18.4) min and (61.7 ± 17.1 ) min respectively, which were significantly higher than that of Ⅰ and Ⅱ stage ( ( 31.9 ± 12.3 ) min and (40.3 ± 10. 6) main ] ( t = 20.25, P < 0.01 ). The massive hemorrhages and the internal damage organs occurred in Ⅲ and Ⅳ stage surgery [2.9% and 3.5% ,respectively] ,whereas much less in Ⅰ and Ⅱ stage (0.8% and 1.4% ) ,with no significant difference(P >0. 05). After treatment,the rate of symptoms recurrence of Ⅲ and Ⅳ stage patients respectively were 21.4% ,which were higher than that of Ⅰ and Ⅱ stage patients ( 2.3% ) (P < 0.05 )The rate of pregnancy in Ⅲ and Ⅳ stage patients ( 15.4% ) were lower than that of Ⅰ and Ⅱ stage patients (69. 6% ) ( P < 0. 01 ). Conclusions Pelvic EMS decreased the quality of life. EMS patients combined with adenomyosis or/and deep infiltrating EMS have more severe pelvic pain symptoms, less surgical effectiveness and more serious complications. Ⅲ and Ⅳ stage patients are more liable to symptoms recurrence and lower pregnancy rate.
10.Influence of contrast injection methods in interstitial magnetic resonance lymphography of breast cancer

Objective To study the influence of different contrast injection skills in interstitial magnetic resonance lymphography( IMRL) of breast cancer, and standardize the IMRL contrast injection process. Methods A total of 36 patients with invasive breast cancer were ran-domly divided into four groups. The imaging time of different contrast agent dose and local massage time on sentinel lymph node ( SLN) were analyzed. According to the results, the best contrast injection methods technology program was choosed. Results For the same massage time, different contrast agent doses showed no significant difference in the SLN imaging time (P>0. 05);For same contrast agent dose, dif-ferent massage time showed significant statistically different SLN imaging time(P<0. 05). Conclusion In clinical work, IMRL with a con-trast agent injection dose of 0. 5 mL/person, local massage 1 min after injection can successfully achieve the lymphatic system image of breast cancer.
