2.Clinical analysis of nineteen elderly patients with rheumatoid arthritis

Objective To investigate the clinical features of rheumatoid arthritis (RA) in the aged.Methods The clinical features and inflammatory index of 19 aged patients with RA were retrospectively analyzed and compared with those of 103 young patients with RA. Results The acute onset was ocurred in 11 cases in EORA group(11/19,57.9%) ,32 cases in MRA group (32/103,31.1%), there was sighicant difference two groups (P <0. 05). There were 10 cases with intertitial lung disense in EORA group(52.6%) ,27 cases(26.2%) in MRA group.There were no statistical differenes between two groups in erythrocyte sedimentation rate, rheumatoid factor, c-reactive,EORA incidence rate in male and female. Conclusions In comparison with MRA group,the onset of disease in patients of EORA group was more acute than that of MRA,it had higher degree of disease activity. The commonly seen extra-joint manifestation were feeble and interstitial disease lung. Early diagnosis and treatmentof EORA was favor for the prevention of deterioration of this disease.
3.Mosapride citrate in reflux esophagitis treatment and intervention effect analysis

Objective To explore the use of mosapride citrate in treating reflux esophagitis and nursing intervention spitley effect. Methods In 40 cases of reflux esophagitis in patients with routine therapy, and classified as the control group, the other 40 patients in the control group based on the use of mosapride citrate. And classified as the observation group, two groups of patients were in our hospital from June 2015 to January 2017. Results The study in the treatment of the two groups of patients with effective rate of 95.0%, the observation group was significantly higher than the control group 80.0%, between the two groups showed significant differences (P<0.05). The adverse effects of more than two groups found, two groups of patients with adverse reaction incidence was no significant difference, that shows no significant difference. Conclusion Mosapride citrate in reflux esophagitis treatment and intervention effect analysis found that the compared with conventional therapy, can be in At the same time, it can improve the curative effect of the patients, and will not increase the possibility of adverse reactions, so it is worthy of clinical reference.
4.Mutation analysis of a Chinese pedigree of Leber hereditary optic neuropathy with only male patients

Background Leber hereditary optic neuropathy (LHON) is a maternally inherited disorder characterized by a bilateral acute or subacute painless central visual loss in young adults,predominately in males.So far no one theory can completely explain all clinical manifestations of LHON.Objective This study was to investigate whether there is a linkage between X-chromosomal and mitochondrial mutation in the inheritance of a Chinese LHON pedigree with only male patients.Methods This study was approved by Ethic Committee of Affiliated First Hospital of Zhengzhou University and followed by Declaration of Helsinki.A Chinese LHON pedigree was included in Anyang city from January 2008 to August 2016.Periphery blood of 5-10 ml was collected from 4 sufferers,13 maternal members and 10 non-maternal members for DNA extraction and PCR sequencing.The gene scanning and genotyping analysis were performed by ABI-PRISM 3100 genetic analyzer and Genotyper 3.7 software,and linkage analysis was carried out with Linkage software for the calculation of logarithm of odds (LOD).Mitochondrial DNA (mtDNA) sequence,fluorescence-based Genescan for X-chromosomal sequence were analyzed in the propositus and haplotype was evaluated.Results A total of 5 generations and 71 families were included in the pedigree,with 6 male sufferers,30 maternal members and 41 non-maternal members.The visual acuity was ≤0.10,and the central visual field defection,the optic nerve flushing was found in the acute phase,different levels of the optic nerve fibers atrophy were found in the chronic phase;visual evoked potential (VEP) amplitude was low and peak latency were found in the male patients,and no any ocular abnormality was seen in the maternal members,meeting a maternally inherited characteristics,with the penetranee of 20%.The three primnary mutations were not been found in this family bv PCR sequencing,mtDNA sequencing appeared 31 variation of loci in the proband,including a known G3635A mutation,as well as an unknown ND5 A12340G missense mutation and ND4 T11809C synonymous mutation as well as 28 polymorphism of locus,and the proband was mitochondrial haplotype F1.The maternal families were mutation carriers of G3635A and AI2340G loci,while the same mutation was not found in the normal family members and 107 controls.The maximum two point parametric LOD score was 1.46(θ=0.0) for marker DXS1060,1oeated at Xp22.3,and the two-point and multipoint non-parametric linkage analysis were significant (all at P>0.05).Conclusions The ND5 A12340G and ND1 G3635A mutations coexist in this LHON family,and the ND5 A12340G mutation is a newly reported mutation.There is no evidence for an X-linked modifiers loci in this Chinese LHON family.

6.Mutations of androgen receptor gene in prostate cancer tissues

Objective To study the relationship between the mutation of androgen receptor(AR) gene and the development of human prostat e cancer. Methods All exons of AR gene except trineucl eic acid repeat polymorphism were amplified by PCR and selected by single strand conformation polymorphism( SSCP) in 27 paraffin-embedded slides of human pros tate cancer. The abnormal mobility shifts were further detected by direct DNA c ycle sequencing. Results Three abnormal mobility shifts respectively in exon A.D.G were found by SSCP in three prostate cance r tissues. One point mutation was found in 296 amino acid (C 966→A,Ser29 6Arg) from exon A.This point mutation has not been reported before. Conclusions This mutation might change the transcriptive activity of AR, and it might be correlated to PCa progression.

Objective To study the short term and long term response and the safety of the different doses of recombinant interferon alfa 1b(IFN? 1b) for the treatment of the patients with chronic hepatitis B and the predictive factors of the response. Methods 146 Chinese patients with chronic hepatitis B entered randomly into four groups. Three groups were given with IFN ? 1b 3 million unit (MU), 5 MU and 10 MU respectively. The control group was given placebo. The short term(ETR) and long term(SR 12) responses and side effect were observed. Selected nineteen items (host related, drugs related and virus related), which affected the response of IFN alfa 1b probably (host, virus and drugs, et al) were analyzed. Results The end treatment response (ETR) of the IFN ? 1b 5 MU therapy was 55.3%, and 10 MU was 58.1%. 8.Effects of pulmonary rehabilitation exercise on mood and subjective and objective sleep quality of elderly patients with chronic obstructive pulmonary disease

Objective:To investigate the effects of pulmonary rehabilitation on anxiety/depression and subjective/objective sleep quality of elderly patients with stable chronic obstructive pulmonary disease(COPD).Methods:From February 2018 to February 2019, 120 elderly patients with stable COPD were selected and randomly divided into the experimental group (pulmonary rehabilitation exercise combined with conventional COPD treatment) and the control group (simple COPD conventional treatment). Sixty cases in each group were intervened for 8 weeks. Before and after treatment, Hamilton anxiety scale (HAMA) was used to evaluate anxiety, Hamilton depression scale(HAMD)was used to evaluate depression, Pittsburgh sleep quality index(PSQI)and sleep log were used to evaluate subjective sleep quality, and objective sleep quality was monitored by multi-channel sleep monitor.SPSS 21.0 software was used to analyze and process the data. Chi square test, independent sample t test and paired t test were used for statistical analysis. Results:After 8 weeks of intervention, the HAMA and HAMD scores of the experimental group were lower than those of the control group (HAMA: (7.57±3.19) vs (10.15±4.89), t=-3.428, P=0.001; HAMD: (8.22±4.73) vs (10.60±6.49), t=-2.300, P=0.023). COPD patients with anxiety decreased (χ 2=7.566, P=0.006). After treatment, the subjective sleep latency of the experimental group was shorter than that of the control group ((42.00±9.88)min vs (47.25±10.27)min, t=-2.854, P=0.005). The subjective sleep efficiency was higher than that of the control group ((76.00±4.50)% vs (74.00±5.20)%, t=2.272, P=0.025), and the objective sleep latency was shorter than that of the control group ((28.02±5.59)min vs (32.95±6.21)min, t=-4.575, P<0.05). Conclusion:Pulmonary rehabilitation exercise can improve the anxiety and depression of elderly patients with stable COPD, and improve the subjective and objective sleep quality.
9.Study on Antimicrobial Effectiveness of Zhenzhu Mingmu Eye Drops

Objective:To evaluate the antimicrobial effectiveness of Zhenzhu Mingmu eye drops according to the requirements de-scribed in Chinese pharmacopoeia (2010 edition). Methods:Totally 7 batches of Zhenzhu Mingmu eye drops from 7 enterprises were evaluated. All the samples were tested according to the requirements described in Chinese pharmacopoeia, and the testing points of 6h and 24h were added. Results:As for bacteria, the reduction value of staphylococcus aureus in 7 days treated with Zhenzhu Mingmu eye drops from enterprise e was 0. 61g, and that in 14 days was 1. 01g. That in 7 days treated with the eye drops from the other enter-prises was all above 1. 01g, that in 14 days was above 3. 01g, and that in 14-28 days kept stable. As for fungi, the number was stable from the beginning with the treatment of the eye drops from the seven enterprises. Conclusion:Although the type and content of bacte-riostatic agent in domastic Zhenzhu Mingmu eye drops are the same, there is difference in the antimicrobial effectiveness of the product from different enterprise. The eye drops from one enterprise can't meet the requirements for type I product described in Chinese pharma-copoeia,and the others meet the requirements.
10.Use of dentritic cells pulsed with HLA-A2-restricted MAGE-A1 peptide to generate cytotoxic T lymphocytes against malignant glioma.

This study developed a novel approach of targeting malignant glioma with pMAGE-A1(278-286)-specific cytotoxic T lymphocytes (CTLs) induced from the peripheral blood mononuclear cells of healthy donors by multiple stimulations with human leukocyte antigen (HLA)-A2-restricted pMAGE-A1(278-286) peptide-pulsed dentritic cells. Cytotoxic assays were performed by the colorimetric CytoTox 96 assay to analyze cytotoxic activity of the induced CTLs against various target cells. The induced CTLs showed approximately 45% specific lysis against T2pMAGE-A1(278-286) (pMAGE-A1(278-286) peptide pulsed T2 cells) and U251 (HLA-A2(+), MAGE-A1(+)) at an effector:target ratio of 40:1, and approximately 5% cytolysis against T2pHIV, A172 (HLA-A2(-), MAGE-A1(+)), K562 and T2 cells without being pulsed with peptide at any effector:target ratio. The specific killing activity of the induced CTLs against T2pMAGE-A1(278-286) and U251 was much more obvious than in any other control group (P<0.05). The cytotoxic activity against the T2pMAGE-A1(278-286) and U251 was significantly eliminated by anti-HLA class I mAb W6/32. These results suggest that pMAGE-A1(278-286) epitope may serve as a surrogate tumor antigen target of specific immunotherapy for treating HLA-A2 patients with malignant glioma.
