1.THE RESEARCH ON THE PROLIFERATION AND DIFFERENTIATION OF MOUSE PRIMORDIAL GERM CELLS IN VITRO

Objective To research on the proliferation and differentiation in vitro of primordial germ cells. Methods We got plenty of primordial germ cells(PGCs) by co-culture of PGCs with sertoli cells(SCs).The primary cells formed embryoid bodies(EBs) by suspension culture.furthermore,these EBs were plated on gelatin-coated culture plates for spontaneous differentiation. Results We got plenty of PGCs by coculture.Primary PGCs and passaged PGCs for histochemistry staining with AKP and immunohistochemistry staining for SSEA-1 display positive character.The EBs by suspension culture differentiated into epithelial-like,fibroblast-like and neuronal-like cells in vitro. Conclusion Primordial germ cells can proliferate and keep the character of stem cells by co-culture with SCs.PGCs can form EBs by suspension culture,most of these EBs can differentiate into neuronal-like cells,they also can differentiate into epithelial-like and fibroblast-like cells in vitro.
2.Changes of serum GH/IGF- Ⅰ axis and related clinical significance following traumatic long bone fracture

ObjectiveTo investigate the changes of serum growth hormone (GH)/insulin-like growth factor- Ⅰ ( IGF- Ⅰ ) in patients with traumatic long bone fractures and the related clinical significance.MethodsTwenty-one patients with traumatic long bone fractures from the same trauma care center treated between November 2009 and April 2010 were enrolled in the study..There were 14 males and 7 females,at age range of 22-60 years (mean 45.0 years).Seventeen patients (81%) were with closed trauma and four ( 19% ) with open trauma.Enzyme Linked Immunoadsorbent Assay (ELISA) was used to determine the levels of serum GH and IGF- Ⅰ within 14 days after fractures and the relationship between changes of GH-IGF- Ⅰ axis and injury severity score (ISS) was further analyzed.Eight patients with normal GH/IGF- Ⅰ were set as control group.ResultsThe GH level in the peripheral blood was significantly higher at day 1 after fractures ( P < 0.05 ),and rapidly declined and stabilized at day 5 compared with the control group.The GH level in the patients with ISS≥ 18 points was sigmficantly higher than that in those with ISS<18 points at day 1 after fractures (P<0.05).The IGF- Ⅰ level in the peripheral blood in the patients with 1SS≥ 18 points was significantly higher than that in those with ISS < 18 points at days 5-10 after fractures ( P < 0.05 ).ConclusionThe hypothalamus GH-IGF- Ⅰ axis plays an important role in early metabolism and fracture healing during the acute phase of traumatic long bone fractures.
3.Impact of Symbiotic Probiotic Fermentation on Anti-tumor Effect of Agaricus blazei Polysaccharide

Objective To investigate the impact of symbiotic probiotic fermentation on anti-tumor effect of Agaricus blazei polysaccharide. Methods Probiotic fermentation and non-fermentation of Agaricus blazei polysaccharide samples were divided into large (0.8 g/kg), middle (0.4 g/kg), small (0.2 g/kg) dose groups, and negative control group for a total of 7 group. The anti-tumor effect on tumor-bearing mice was observed. Results The highest inhibition rate of probiotic fermentation Agaricus blazei polysaccharides group was 53.71%, while that of non-fermatation of Agaricus blazei polysaccharide group was 38.90%. Conclusion Inhibition rate of probiotic fermentation samples of Agaricus blazei polysaccharide was significantly higher than non-fermentation samples. Probiotic fermentation can significantly improve the anti-tumor effect of the Agaricus blazei.
4.Effect of oral health instructions in nursing of marsupialized jaw cysts

Objective To evaluate the effect of oral health instructions in nursing of marsupialized jaw cysts. Methods 46 patients with cysts of jaw were divided randomly into two groups (education group and control), 23 in each. In addition of normal nursing processes in both groups, patients in education group were offered more oral health instructions, such as pathogenesis of jaw cysts, mechanism of marsupialization, wear and clean of a cyst plug, and oral hygiene maintenance. 12 months later, compliance, satisfaction, treatment effect and oral hygiene condition of the patients in the two groups were studied and compared. Results 100%of patients in the education group could return to clinical visits on time and 96%of patients did cyst rinsing after every meal, which were significantly higher (χ2=6.9, P<0.05) than in the control (74% and 70% respectively). Patients′satisfaction in the two groups were also significantly different (χ2=9.109,P<0.05). Patients in the education group were much more satisfied with the treatment procedures than control. The treatment effect of the cyst in the education group [good (83%), moderate (17%), not so good (0%)] was significantly (χ2=7.793, P<0.05) better than control (good (43%), moderate (52%), not so good (5%)]. The secondary infection rates in the two groups were similar (χ2=1.022, P>0.05). Oral hygiene condition of patients in the education group was better than control in Debris Index (χ2=9.576, P < 0.05) and Plaque Index (χ2=8.212, P < 0.05). Conclusions Oral health instructions played positive role in improving patients′ compliance, degree of satisfaction, treatment effect and oral hygiene condition in patients with jaw cysts.
5.Effect of square dance exercises on symptom scores in menopausal women with coronary heart disease

Objective: To observe effect of square dance exercises on symptom scores in menopausal women with coronary heart disease (CHD). Methods: A total of 39 menopausal CHD patients participated in square dance exercises, exercise intensity was 47.2%HRmax~58.1%HRmax(60~120 min/d), 5~7 times/week, three months, and they were regard as dance group. Control group consisted of 42 CHD women of same age living in the same district, who didn’t take part in exercise. After enrollment, these two groups received assessments of Seattle angina questionnaire (SAQ) and frequent physical and mental symptoms of menopausal women self-rating scale (Kupperman self-rating scale) within 3d and after three months. Results: There were no significant difference in score of each item and total score of SAQ in first assessment between two groups (P>0.05 all). Compared with first assessment and control group after three months, there were significant increase in scores of most items and total score of SAQ [(66.40±15.16) scores vs. (67.18±13.93) scores vs. (85.15±16.30) scores] in dance exercises group, P<0.01~0.05. There were no significant difference in score of each item and total score of Kupperman scale in first assessment between two groups (P>0.05 all). Compared with first assessment and control group after three months, there were significant decrease in scores of most items and total score of Kupperman scale [(29.40±6.51) scores vs. (29.73±6.26) scores vs. (23.65±6.29) scores] in dance exercises group, P<0.01~0.05. Conclusion: Square dance exercises may significantly decrease angina pectoris symptom and menopausal physical and mental symptoms in menopausal women with coronary heart disease.
7.The inhibition effects of Zyflamend on DMBA-induced oral carcinogenesis

Objective:To study the effects of an unique anti-inflammatory herbal preparation Zyflamend(ZF)on 7,12-dimethyl-benz anthracene (DMBA)-induced oral carcinogenesis in golden Syrian hamsters.Methods:10 hamsters were used as the blank controls.106 hamsters were treated topically on the left cheek pouch with 0.5% DMBA solution,3 times per week for 6 weeks. Then the hamsters in positive control group(n =40)were not treated afterwards,in the other 2 groups were treated by topically ap-plication of ZF of original solution(high dose,n =33)and 1 ∶1 dilution(low dose,n =33)respectively.Hamsters were sacrificed at the 24th week and tissue samples of the left cheek pouch were obtained for the examination of tumor incidence,histopathology and BrdU immunohistochemisty.Results:Gross inspection showed that the incidences of the tumor in positive,low dose and high dose groups were 86.7%,53.6%(P <0.01 )and 50.0%(P <0.01 ),histopathological examination showed that the tumor incidences were 86.7%,78.6% and 64.3%,respectively(P <0.05).High dose of ZF significantly decreased the incidence of hyperplasia , dysplasia and cancer(P <0.05);decreased BrdU-labeling index(P <0.05).Conclusion:Zyflamend may inhibit DMBA induced oral carcinogenesis and such prevention may be related to the effect of the suppression of cell proliferation.
8.Three-dimensional kinematic analysis of the ability in the activities of daily living of hemiplegic patients and normal subjects

Objective To study the kinematic characteristics of normal subjects and hemiplegic patients in activities of daily living (ADL) by three-dimensional (3D) kinematic analysis. Methods A Vicon motion capture system was used to record 3D kinematic data on 15 normal subjects ( the healthy group) and 10 hemiparetic patients (the hemiplegic group) performing 5 ADL tasks: reaching up, combing, drinking, touching the opposite shoulder and touching the back pocket. The movement times and the 3D motion angles of the sternoclavicular joint,the acromioclavicular joint, the shoulder joint, the elbow joint and the wrist joint were recorded. Three-dimensional joint angles and movement times were compared between the normal group and the hemiplegic group. Results Compared with the normal group, the hemiplegic group had significantly more lateral rotation and abduction of the sternoclavicular joint, medial rotation and extension of the acromioclavicular joint and flexion of the elbow joint.The hemiplegics used significantly less medial rotation of the sternoclavicular joint and adduction/abduction, flexion/extension and lateraL/medial rotation of the shoulder joint. Additionally, the hemiplegic group showed significantly longer movement times. Conclusion Hemiplegics use different joint motor patterns from healthy subjects in completing ADL actions.
9.On the legal attributes of self-payment agreement regarding medical expenses

In the macrocosmic perspective,the development trend of the rights and obligations of doctors and patients is regarded as changes in the healthcare mode.In the microcosmic perspective,it is regarded as a clarification and emphasis of the informed consent of the patients as required by laws and regulations.However,medical workers tend to neglect their obligation of informing the patients of the medical expenses to be paid out of their own pocket.In this consideration,informing the patients in writing of their self-payment in medical expenses is obligatory and important,i.e,the self-payment agreement is highly important in clinical work.This paper analyzed the legal at-tributes of self-payment agreement in the aspects of both the informed consent and the signing of the medical service contract,with discussions of the tining of signature,in the hope of inspiring and helping medical workers in their clinical practice.
10.Linear measurement accuracy using cone-beam computed tomography for human skull

BACKGROUND:Cone-beam computed tomography has been widely applied in the dentistry due to irreplaceable advantages. Among those advantages, linear measurement is one of the important ones. Further studies are suggest to explore the accuracy and repeatability of linear measurement for the whole skul , as wel as the effect of different resolution and scanning thickness on the linear measurement results.
OBJECTIVE:To evaluate the accuracy of linear measurement using cone-beam computed tomography (Newtom VG) on the human skul , with two different voxel sizes.
METHODS:In this study, 22 anatomic landmarks in four dry human skul s were marked and 11 linear measurements were obtained. These were considered to be the gold standard (real measurement). The skul s were scanned by cone-beam computed tomography (Newtom VG) at two voxel sizes:0.3 mm and 0.15 mm. Linear measurement was performed in axial and coronal planes. SPSS software version 17 was used for data analysis between radiographic measurement and real measurement.
RESULTS AND CONCLUSION:The mean differences of real and radiographic measurements were-0.27 to 0.14 in four different sections, and statistical analysis showed no significant difference between linear measurements and gold standard (P>0.05). There was no significant between two different voxel sizes (P>0.05). Cone-beam computed tomography (Newtom VG) is highly accurate and reproducible in linear measurements in the axial and coronal planes. A cone-beam computed tomography scan with a larger voxel size (0.3 mm) is recommended, resulting in lower radiation dose and faster scan time.
