RNA interference (RNAi), as a new technology of gene therapy, has been used in the studies of many diseases in vitro, however, targeting delivery of small interference RNA (siRNA) is still a bottleneck for clinical therapy of siRNA agents. Aptamer is a group of oligonucleotides with high affinity and targeting, and is becoming another important means of delivery for siRNA. In this review, we summarized siRNA delivery obstacles in vivo and recent attractive developments increatively using cell-internalizing aptamers to deliver siRNAs to target cells.

The period of perimenopausal causes an sudden change of internal environment and disorder of many systems. But the body has a self-adjusting ability, which can help the body to adjust and maintain the body,s state of homeostasis. If this ability too weak or environmental changes in the body is too severe, so that the organism is difficult to reach a new equilibrium, the physiological stress in this time will turn into pathological stress, leading to menopause and menopause syndrome prone to the disease. Heat shock protein in the cell is the stress induced in the broad and non-specific proteins to protect the cells, as a ‘chaperone’ involved in a number of physiological and pathological process of the body. As a result of heat shock protein is induced by stress response in peirmenopausal period and it has an intinate relationship with many links during peirmenopausal period such as neutroendocrine, immunity, antioxidation, estrogen receptor, and insulin receptor, so discussing the function of HSP will benefit the prevention of peirmenopausal syndrome.

OBJECTIVE:To explore the effects of umbilical therapy by Rheum plamatum combined with early colonic irriga-tion on the quality of life in elderly patients with colon stoma. METHODS:80 patients underwent radical resection of abdominoperi-neal rectal carcinoma were randomly divided into control group and observation, with 40 cases in each group. Control group was given early colonic irrigation;observation group was additionally given umbilical therapy by R. plamatum. Treatment course of 2 groups lasted for one month, and both were followed up for 6 months after treatment. The quality by life in 2 groups was evaluated by using QLQ-C30,and therapy costs were calculated before and after treatment. RESULTS:Compared with before treatment, function score and total health score of 2 groups improved significantly in 1,3,6 months after treatment, while symptom score and single item score decreased significantly, with statistical significance(P<0.05). In 1,3,6 months after treatment, role func-tion,social function, cognitive function, emotional function and total health score of observation group were significantly higher than those of control group, and insomnia and pain score were significantly lower than control group, with statistical significance (P<0.05). 3 and 6 months after treatment, diarrhea and economic difficulty score of observation group were significantly lower than those of control group, with statistical significance(P<0.05). 1 months after treatment, loss of appetite score of observation group were significantly lower than those of control group, with statistical significance (P<0.05). 6 months after treatment, fa-tigue,nausea and vomiting score of observation group were significantly lower than those of control group, with statistical signifi-cance (P<0.05). 6 months after treatment, nursing cost, drug cost and total cost of observation group were significantly lower than those of control group, with statistical significance(P<0.05). CONCLUSIONS:The umbilical therapy by R. plamatum com-bined with early colonic irrigation can improve the quality of life in elderly patients with colon stoma significantly and reduce treat-ment cost.

BACKGROUND: Learning and memory is a very complex biological phenomenon. Although quite a few researches on the substances participate in learning and memory and the brain areas related with learning and memory, its mechanism is still not completely clarified.OBJECTIVE:ro study the difference of antioxidase activity at different brain area in rats with different ability of learning and memory to reveal the relationship between the ability of learning and memory and the activity of antioxidase in specific brain area.DESIGN: A randomized controlled trial based on the experimental animals.SETTING: Department of Biochemistry in Jining Medical College.MATERIALS: The study was conducted in the Laboratory of Biochemistry of Jining Medical College and Suzbeu Medical College between March 2001 and January 2004. Forty 15-month old male Wistar rats with a body mass between 580 g and 650 g were selected.INTERVENTIONS: The detection of learning memory ability was carried out in MG-2 trisection radiation maze. Correction response was that the rats escaped to safe area after electric shock. Standard of master was that the rats had 9 out of 10 times continuously of correction. Observatory indicators included times of response required reaching the standard and the correct response rate. Good learning. ability meant 40 or less than 40 times of responses to reach standard; otherwise, poor learning ability was considered. Detection was repeated after 24 hours to observe the memory. Good memory meant 3 times continuously of correct response; otherwise, poor memory was considered. Rats with good learning ability and memory were included into group 1 (n= 10) and the rats with poor learning ability and memory were included into group 2( n = 12). The rest rats were washed out.MAIN OUTCOME MEASURES: Activities of superoxide dismutase (SOD), catalase(CAT) and glutathione-peroxidase(GSH-Px) of five brain areas including cerebral cortex, cerebella, striatum, hippocampus and hypothalamus in rats of two groups.RESULTS: To compare the rats with poor learning and memory ability with rats with good learning and memory ability: SOD activity in cortex, hippocampus and striatum significantly reduced ( t = 3.82, 4. 50, 6. 76, P <0.01); CAT activity in cortex, hippocampus, striatum and hypothalamus significantly reduced(t =4.75, 7.06, 10. 88, 17.28, P<0.001); and GSH-Px activity was similar in each brain area without statistical significance.CONCLUSION: Hippocampus, cortex, striatum and hypothalamus all might participate in learning memory process, and the activities of antioxidases in these areas are closely related with learning memory.

Objective To explore the effect of glucose regulated protein (GRP)78 on the neuronal apoptosis after cerebral ischemia reperfusion injury in rats. Methods Middle cerebral artery ischemia reperfusion rat's models were used with the modified filament method. The expression of GRP78 in the ischemic penumbra tissue was detected by RT-PCR, Western blot and immunohistochemistry at different time points. Primary cultured rat's neurons were exposed to hypoxia and subsequently reoxygenation. Western blot was used to investigate the expression of GRP78. The changes of the neuronal apoptosis after overexpression of GRP78 induced by 2-deoxyglucose were detected. Results The expression of GRP78 in the ischemic penumbra tissue in model group was significantly increased (mRNA : 0.7367±0.0651, F= 477.160, P < 0.01 ; Protein : 0. 8129±0. 0748, F=39.857, P < 0.01). The neuronal survival status was increased after overpression of GRP78 (increased by 39.22% ± 0. 44%, t=46.374, P < 0.01) while the neuronal apoptosis was decreased (decreased by 16.60±1.02, t=7.530, P <0.01). Conclusion Focal cerebral ischemia reperfusion can induce endoplasmic reticulum stress which plays a role in the neuronal apoptosis. The increased expression of GRP78 may protect the ischemic tissue from neuronal apoptosis.
