Objective To construct survivin-targeting siRNA-expressing plasmid. Methods DNA sequence correspond to siRNA targeting survivin was designed and synthesized, and cloned into plasmid pRNAT-U6.1/Neo to produce surviving-targeting plasmid. Two oligos in the template with cohesive BamHⅠ and HindⅢ sites were prepared and annealled to form the insert fragment for siRNA vector. The vector was cut with BamHⅠ and HindⅢ and ligated with the insert fragment using T4 ligase. The recombinant vector was confirmed by restriction digestion and DNA sequencing, and then was transfected into T24 cells with Lipofectamine TM2000 and the expression of survivin was detected by real-time quantitive PCR.Results DNA sequencing for the PCR product showed that the recombinant vector pRNAT-U6.1/Neo-survivin was successfully constructed without any base pair mutation. The plasmid pRNAT-U6.1/Neo-survivin could efficiently reduce the expression of survivin and confer G-418 resistance in T24 cells. Conclusion The siRNA-expressing plasmid which were successfully constructed and transfected into T24 cells in this study may facilitate the application of RNA interference technique, and lay foundation for further studies on the function of survivin.

It is necessary to build a three-dimensional human body model to display the position and track the route of a micro device inside the human body exactly. The key is to construct a three-dimensional human body model according to the patient characteristic. This paper gives a simple way to build the personal geometric model based on the OpenGL programming in the Visual Basic environment. It is proved that the method contributes to the realtime interactive operation to change the model character according to the individual parameter.

Objective To evaluate contrast-enhanced ultrasonography (CEUS) in diagnosis of small hepatocellular carcinoma (SHCC) ( ≤ 2. 0 cm) with liver cirrhosis after contrast-enhanced CT (CECT) examination. Methods Forty five patients with liver cirrhosis received CECT and CEUS examinations before operation or needle biopsy and the diagnosis was confirmed by pathological examination.CEUS and CECT findings of 51 liver space-occupying lesions from 45 patients were retrospectively analyzed. Results Among all 51 lesions detected CEUS and CECT found 49 and 35, respectively. The typical characteristics of SHCC were "fast-in and fast-out" and "fast-in and slow-out". The sensitivity of CEUS and CECT in diagnosis of SHCC was 88. 9% (32/36) and 69. 4 % (25/36) respectively ( x2 = 3. 02, P =0. 08);the diagnostic accuracy was 84. 3 % (43/51 ) and 56. 9% (29/51 ) respectively ( x2 = 1.46, P =0. 22). Among 16 lesions missed by CECT, 12 were detected by CEUS. Conclusions CEUS and CECT show the similar diagnostic rate for typical SHCC ,however, CEUS is more sensitive for atypical lesions. With high time resolution, CEUS have advantages for follow-up study of benign liver lesions.

Objective To study the relationship between handgrip level and characteristics of surface electromyogram(sEMG)signal from forearm muscles. Methods Nine healthy volunteers were asked to do a specific grip test with three force levels of maximum volunteer contraction(MVC),80% MVC and 40% MVC.sEMG signal was recorded from flex digitorum superficial(FDS)and extensor carpi radialis longus(ECRL)simultaneously. The amplitude characteristics of sEMG signal were calculated by the means of root mean square(RMS)and wavelet analysis.Furthermore,to compare the relationship of sEMG and force level between different participants,SPSS was applied to analyze the relative value that was defined as the ratio between sEMG characteristics of MVC condition and that of different grip levels. Results There existed positive correlation between grip level and sEMG activity, stronger grip corresponded the greater sEMG characteristics for every participant, and this correlation revealed significant coherence between different volunteers. Conclusion There is significant correlation between grip force and forearm muscle activities represented as sEMG characteristics. The result suggests that sEMG can be used to predict grip force noninvasively for measuring motor function and assessing the motor function rehabilitation.

Objective To explore the correlation between the moving angle of elbow joint and surface electromyogram signal(sEMG)activities of biceps and triceps. Methods The eigenvalue of sEMG during elbow joint moving under constant load were calculated with means of root mean square(RMS)and integrated electromyogram(iEMG)in time domain. Results The experimental results showed that both the chavacteristics of RMS and iEMG for biceps differed from those of triceps. The sEMG activities of biceps increased with the increment of elbow joint angle, while those from triceps reached to the maximum values when elbow joint angle was 90?.Furthermore,the sEMG activities of triceps at the elbow angle of 45? was similar with that when elbow angle was 135?.Conclusion The sEMG from biceps and triceps can reflect the moving angle of elbow joint under constant load.
10. Co-transplantation of adipose mesenchymal stem cells and endothelial progenitor cells in ulcerative colitis mice
Chinese Journal of Tissue Engineering Research 2020;24(25):3981-3987
BACKGROUND: Mesenchymal stem cell transplantation has been proved to be effective for ulcerative colitis, while the effect of endothelial progenitor cells in ulcerative colitis treatment is still unknown. They are both promising cells for tissue engineering, which can be used for cell transplantation. OBJECTIVE: To explore whether co-transplantation of endothelial progenitor cells and adipose mesenchymal stem cells can relieve ulcerative colitis in a mouse model. METHODS: C57BL/6J mice were randomly divided into six groups (n=24 per group): co-transplantation group, mesenchymal stem cell transplantation group, endothelial progenitor cell transplantation group, glucocorticoid treatment group, transplantation control group and normal control group. Murine ulcerative colitis model was established in all groups except for the normal control group. At 7 and 10 days after modeling, transplantation groups were respectively injected via tail vein with adipose mesenchymal stem cells and/or endothelial progenitor cells, glucocorticoid or PBS. Mice were sacrificed at 12 days after modeling. Colon length, disease activity index, histological score and the serum level of tumor necrosis factor-α were compared between groups. RESULTS AND CONCLUSION: Treatment with glucocorticoid was significantly effective for ulcerative colitis relative to the transplantation control group (P < 0.05). Adipose mesenchymal stem cells were proved to have better effects than glucocorticoids in the murine ulcerative colitis model (P < 0.05). No significant differences were found between endothelial progenitor cell transplantation group and transplantation control group (P > 0.05). Co-transplantation of adipose mesenchymal stem cells and endothelial progenitor cells was better than the other treatments, which significantly improved the shortening of the colon, disease activity index, histological score, and serum level of tumor necrosis factor-α.
