1.Construction and expression of IL-24 in eukaryotic expression vector and study of its antitumor effects

Objective:To construct an eukaryotic expressing plasmid of IL-24,and investigate its inhibitory effects on the growth of tumor cells in vitro and in vivo.Methods:The eukaryotic expressing plasmid of IL-24(pEGFP-C1-IL-24) was constructed by DNA recombination technique.The recombination plasmid and empty vector were transfected into HIC cells,respectively with Lipofectamine 2000 Reagent and the expressions were determined by LSCM;the proliferation of HIC cells was measured by MTT assay;and apoptosis rate and cell-cycle distribution of HIC cells were measured with Flow Cytometry.Mice were inoculated with B16 cells,which were transfected with pEGFP-C1-IL-24 or empty vector,and the tumor size in mice was detected.The inhibitory effect of IL-24 gene transfection in mice solid tumor was observed and measured.Results:The expression of pEGFP-C1-IL-24 in HIC cells was determined by LSCM after the transfection of pEGFP-C1-IL-24.Comparing to the control group,the proliferation of HIC cells was inhibited by transfection with pEGFP-C1-IL-24 and the G2-M phase of the transfected cells was also increased.Moreover the percentage of mice with detectable tumor was significantly decreased after inoculated with B16 cells transfected with pEGFP-C1-IL-24.Growth rate of tumor in mice model was also significantly inhibited in IL-24 gene therapy group as compared with the control grouop(P
2.Clinical Efficacy of Docetaxel Combined with Fluorouracil in the Treatment of Advanced Gastric Carcino-ma

Objective:To observe the clinical efficacy and safety of docetaxel combined with 5-fluorouracil in the treatment of pa-tients with advanced gastric cancer. Methods:Totally 84 cases of gastric cancer patients were randomly divided into group A and group B with 42 ones in each. Group A was given chemotherapy of docetaxel combined with 5-fluorouracil, and group B was treated with chemotherapy of Tegafur combined with docetaxel. One cycle was 21 days, and the patients were given at least two cycles of chemother-apy. The recent clinical curative effect, physical state improvement and adverse reactions in the two groups after the treatment were ob-served. Results:The total effective rate of the two groups showed no significant difference (P<0. 05), however, the physical state improvement rate of group A (40. 48%) was significantly higher than that of group B (20. 24%) with statistical significance (P<0. 05). The incidence of thrombocytopenia and neutropenia decrease of group A was significantly lower than that of group B with statis-tically significant difference (P<0. 05). Conclusion:The treatment of advanced gastric cancer patients with docetaxel combined with 5-fluorouracil can achieve significant clinical efficacy with relatively mild toxicity,it can significantly improve the physical state of pa-tient.
3.Clinical significance of expressions of bcl-2 and BCRP in diffuse large B cell lymphoma

Objective To explore the expression of b-cell lymphoma/leukemia-2 (bcl-2) and breast cancer drug-resistant protein (BCRP) in diffuse large B cell lymphoma (DLBCL) and their correlation. Methods Using flow cytometry (FCM) and semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) assay the expressions of BCRP gene was analysed in 40 DLBCL samples. bcl-2 protein were measured by immunohistochemistry on paraffin-embedded slices in 40 cases of DLBCL. Results The positive expression rate of bcl-2 and BCRP protein were 60.0 % (24/40) and 37.5 % (15/40) in 40 patients with DLBCL respectively. There was significant differences between positive group and negative groups (x2 = 5.7618,P <0.05; x2 = 6.5541,P <0.05). There was no significant correlation between Bcl-2 and BCRP gene in effective treatment group and therapies group or progress group (x2 = 2.0263,P >0.05). Conclusion BCRP may play an important role in DLBCL primary much medicine drug-resistant,thus it may be a useful prediction of chemotherapeutic treatment and risk of relapse. The level bcl-2 expression was closely related with the grade of malignancy and the prognosis of DLBCL. There was no significant correlation between bcl-2 and BCRP gene. The detection of bcl-2 and BCRP gene played an important role in evaluating DLBCL outcome.
4.Specific Chromatogram of Fufang Shexiang Injection and Simultaneous Determination of Five Components by GC

Objective:To establish the specific chromatogram of Fufang Shexiang injection and simultaneously determine five com-ponents( menthol,borneol,patchouli alcohol,muscone,α-asarone) in the injection by GC. Methods:The sample solution was extracted by a solid-phase extraction method. The separation was performed on a DB-WAX GC capillary column with an FID detection. Results:The specific chromatogram had six common peaks, and the similarity was above 0. 91. The five chemical components(menthol,borne-ol,patchouli alcohol,muscone,α-asarone) were well separated and with good linear relationship, the recoveries were between 95. 1 and 102. 6%, and RSD was less than 3. 0%. Conclusion:The method is simple, accurate and reproducible. The specific chromatogram combined with multi-component determination can reflect the overall quality of Fufang Shexiang injection, which can be used for the quality control of the preparation.
5.The influence of combined treatment of interleukin-2 and chemotherapy on T lymphocyte subgroup in acute myeloid leukemia

Objective To investigate the influence and the clinical significance of interleukin-2 (IL-2) on T lymphocyte subgroup after chemotherapy in acute myeloid leukemia patients. Methods Fiftyfour acute myeloid leukemia patients were divided into treatment group (chemotherapy combined with IL-2,28 cases) and control group (pure chemotherapy,26 cases) by radom digits table. In treatment group, IL-2400 000 U was dripped after chemotherapy for 14 days. Then the T lymphocyte subgroup change before and after treatment was examined. Results After 14 days' treatment, the levels of CD3 (0.6026±0.2275 ),CD4(0.4972±0.1224),CD56(0.3016±0.1053 ) in treatment group were significantly higher than those before treatment(0.3926±0.2010,0.2264±0.1190,0.1729±0.1226) and in control group (0.4352±0.1930,0.2738±0.1362,0.1937±0.1268)(P< 0.05). Conclusion IL-2 treatment can obviously raise the levels of T lymphocyte subgroup and can raise the immunologic function.
6.Evaluating the value of CEA and CA153 during follow-up in HER-2 overexpressing breast cancer u-sing ROC curves

Objective To explore the value of carcinoembryonic antigen ( CEA) and cancer antigen 153 ( CA153 ) in detecting tumor recurrences and to establish the optimal operating point of diagnosis of recurrences in HER-2 overexpressing breast cancer using ROC curves .Methods A total of 127 HER -2 overexpressing breast cancer patients was enrolled into this study and subdivided into two groups ,the recurrence group and the non-recurrence group .We created a ROC curve and found out the optimal operating point .Then we compared the advantage of detecting recurrences between optimal operating point in the ROC curve and the diagnostic point that we usually used .Results The recurrence groups showed significantly higher CEA and CA 153 levels than the non-recurrence groups(P<0.0001).The area under the curve(AUC)of the CEA and CA153 were 0.752 and 0.820 respectively and the optimal operating point were 3.5 ng/mL and 17.89 U/mL,respectively.The sensitivi-ties of the optimal operating point of CEA and CA 153 were 46.15%and 65.38%respectively,and the specifici-ties were 97.33%and 89.33%, respectively.Conclusion CEA and CA153 process medium value for detecting recurrences in HER-2 overexpressing breast cancer .

目的探讨后外侧小切口行全髋置换术(THA)后患者康复护理。方法后外侧入路小切口患者45例48髋,经精心护理后与常规THA患者在手术切口、围手术期出血量、术后疼痛、住院天数、早期活动方面比较。结果在相同条件下,小切口手术患者减少围手术期出血量、术后疼痛、住院天数及术后早期康复快,缩短住院时间。结论康复护理有助于THA患者的恢复。
8.Plasma adenomedullin, calcitonin gene-related peptide and c-type natriuretic peptide in rats with hypoxic pulmonary hypertension

Objective To explore the effects of hypoxia on the syntheses and secretion of adrenomedullin (AM), calcitonin gene related peptide (CGRP) and c-type natriuretic peptide (CNP) and the relationship between these peptides. Methods Rat models were established with hypoxia for 10, 20 and 30 d respectively and rats under normal altitude were served as control. Pulmonary artery pressure and the maximum increasing speed of right ventricle (RVdp/dtmax) were measured in every group. The dynamic changes of AM, CGRP and CNP concentrations in plasma were studied with radioimmunoassay. Results During hypoxia, pulmonary artery pressure and RVdp/dtmax were enhanced. Plasma AM and CNP concentrations were increased while CGRP was decreased significantly. The plasma level of AM had positive correlation with that of CNP, but negatively correlated with that of CGRP. Conclusion Results indicate that hypoxia may cause pulmonary artery pressure change and right ventricle has compensatory reaction to hypoxic pulmonary hypertension. Dynamic changes of plasma AM, CGRP and CNP concentrations can be regarded as indexes for condition of illness.
9.Investigation and study of depression status and influencing factors of rural widowed elderly with different genders

Objective To understand the depression status and its influencing factors of Chenzhou rural widowed elderly with different genders.Methods A random sample of 342 widowed elderly from 30 villages was investigated using the Geriatric Depression Scale and a self-designed questionnaire.Results Widowed elderly depression detection rate was 29.8%,depressive symptoms among elderly women detection rate was 34.7%,higher than men's depressive symptoms detection rate,which was 22.9%.Binary Logistic regression analysis showed:Alone,decreased self-care ability was the common risk factors of depressive symptoms of widowed elderly.The impact on men was bigger than on female.Age,chronic diseases were the main factors affecting depression status of elderly men,and economic security was the main factors affecting the depression status of elderly women.Conclusions The incidence of depressive symptoms in rural widowed elderly is high,but there are differences between widowed elderly with different genders in depression status and influencing factors.Therefore the community workers need to take targeted interventions to promote mental health for the elderly.
10.Analysis of the Result of Clinical Pharmacists' Investigation of Medical Advice

Objective:To explore the methods and the role of clinical pharmacists in the medical advice audit. Methods: The questionnaire was designed to analyze the results of the medical advice investigated by the clinical pharmacists from November 2011 to October 2012. Results:Totally 3 232 patients were in the use of antimicrobial drugs in a investigation of 14 675 patients in 178 724 medical advices, accounting for 22. 0% of the total number of the investigation;the use of antimicrobial agents in patients with medical advices of antimicrobial were 5 387, accounting for 3. 0% of the total number of the investigation;597 errors were detected and inter-vened, accounting for 0. 33% of the total number of the investigation;the main errors were dose errors, usage errors and repeated med-ication errors. Indication error and interaction error were more in the critically ill patients monitored by the clinical pharmacists than that in the general ward patients. Conclusion:Clinical pharmacists and ward pharmacists to participate medical advice review, partic-ularly to concern about the patients with medical advices of antimicrobial drugs, and to intervene reasonably in the dose errors, usage errors and repeated medication errors can improve the overall quality of the hospital pharmacist team, make sustainable development of rational use of drugs and reduce the risk of medication.
