1.Effect of estradiol on the gene expression of peroxisome proliferator-activated receptor gamma-2 in rat bone marrow stromal cells

BACKGROUND: Whehter estradiol (E2) can change the expression of peroxisome proliferator-activated receptor gamma-2(PPAR-γ2) during differentiation of bone marrow stroma cells should be studied further.OBJECTIVE: To investigate the effects of 17β-estradiol (E2) on the gene expression of PPAR-γ2 mRNA and PPAR-γ2 protein.DESIGN: Contrast observational trial.SETTING: Laboratory Department of People's Hospital of Deyang City; Laboratory Center of Chengdu Military Command General Hospital and Chengdu Bai'ao Biol-Tech Limited Company.MATERTALS: A 3-month-old female SD rat (200±20) g used to isolate bone marrow stromal cells was obtained from Animal center of Chengdu Chinese Medicine University (Medical Experimental Animal Number 11). Dulbecco's mimimum essential medium (DMEM) was obtained from Hyctone Compnay. 1α, 25(OH)2D3, dexamethasome (DEX) and E2 were purchased from Sigma Company. Total RNA kits were obtained from Omga. One step RNA PCR kit (AMV) was obtained from Takara Shuzo Co, Ltd. Northern direct HRP labeling and detection kit was purchased from PIERCE. Western blotting luminol reagent was obtained from Santa cruz.METHODS: The experiment was carried out from April 2001 to July 2002 in the Laboratory Department of People's Hospital of Deyang City, Laboratory Center of Chengdu Military Command General Hospital and Chengdu Bai'ao Biol-Tech (0, 0.1, 10, 1 000 n) was used to interfere cell differentiation for 3 days. Cultured cells were crushed with Tris-Triton X-100 PBS; activity of alkaline phosphatase was detected with Beckman CX-7 biochemical analytical device; effect of 100 g/L formalin, stained with Weigert-hematoxylin for 10 minutes, rinsed with water, differentiated with 5 g/L hydrochloric ethanol, stained with Van Gieson, desiccated with ethanol of the fractional volume of 0.95, cleared with dimethylbenzene with RT-PCR, Northern blot and Western blot during cell differentiation.protein.proliferated within 24-72 hours. Cells shaped as triangle, multiple angles and fusiform. Three days later, volume of adherent cells was increased and colony, and 10 days later, they confluenced. Bone marrow stroma cells in many generalight red to yellow). Red plasma presented synthesis of collagen. The deeper the red was, the more the collagens were.pression of PPAR-γ2 mRNA was (4.0±0.4)%, (1.7±0.2)% and (2.8±0.2)% (t=6.1, 7.2, 11.5, P< 0.01), which was higher crease the expression of PPAR-γ2 protein. When concentration of E2 was 0.1, 10 and 1 000 nmol/L, expression of PPAR-γ2 protein was (2.2±0.2)%, (2.6±0.2)% and (4.1 ±0.2)%, which was higher than that in 0 nmol/L E2 group [(1.2±0.10)%, t=6.6, 8.5, 13.2, P<0.01].CONCLUSTON: E2 can inhibit expression of alkaline phosphatase and promote differentiation of bone marrow stromal cells and expression of PPAR-γ2 mRNA and PPAR-γ2 protein.
2.Qualitative research on the demands of nursing staff on nursing repository

Objective To explore the demands of nursing staff on nursing repository and provide reference for the develop-ment of nursing repository. Methods In-depth interview was conducted on 21 nursing staff by qualitative research. The themes were formed by category analysis. Results There were four themes about the demands of nursing staff on nursing repository:necessity to develop nursing repository ,contents of the repository ,forms of the repository and prospect of the reposi-tory. Conclusions Nursing staff need a nursing repository. They hope that the repository can provide comprehensive,concrete and practical knowledge,and provide a good interface with digitization. The design of repository should be consistent with in-ternational standards.
3.Effects of rhein on renal tubular epithelial cells transdifferentiation in diabetic rats

Objective To study the effect of rhein on the process of tubular epithelial-mesenchymat transformation in kidney of diabetic rats. Methods Wistar male rats were randomly assigned to 3 groups: Control group (N group, n=12),diabetic group(D group, n=12), rhein treatment group(R group, n=12).The rats of rhein treatment group were treated with daily intragastric administration of periment. The excretion of urinary protein and serum creatine were measured. Histological changes of renal tissue were observed by HE and MASSON stain. Immunohistochemistry was performed to investigate the expression of E-cadherin, α-SMA,FN and TGF-β1 in kidney. Results Compared with the control group, the tubulointerstitial injury and the accumulation of extraeellular matrix protein in diabetic models were obvious(P<0.01).Compared with the control group, the expression of E-cadherin was decreased significantly and the expression of α-SMA,FN and TGF-β1 was increased significantly in diabetic group. E-cadherin was negatively correlated with TGF-β1(rs=-0.60,P<0.05),α-SMA and FN was positively correlated with TGF-β1(rs=0.88,P<0.05;rs:0.91,P<0.01).In comparison with diabetic group,rhein could up-regulate the expression of E-cad and down-regulate the expression of α-SMA and FN in renal tubular epithelial cells(P<0.01).Conclusion Rhein could protect kidney by ameliorating interstitial fibrosis in diabetic rats. The mechanism may be depend on down-regulating the expression of TGF-β1 and suppressing tubular epithelial-mesenchymal transformation.
4.Epidemiological characteristics of coronavirus disease 2019 in Huzhou

Objective:
To learn the epidemiological characteristics of coronavirus disease 2019(COVID-19)in Huzhou,so as to provide reference for prevention and control of COVID-19.
Methods:
All the confirmed cases of COVID-19 in Huzhou,diagnosed according to the COVID-19 Diagnosis and Treatment Plan(Sixth Version Trial)and reported from January 25 to February 7,2020,were recruited. The process of diagnosis and treatment,clinical manifestation,exposure history and close contacts were collected to analyze the epidemiological characteristics.
Results:
On January 25,the first confirmed cases of COVID-19 in Huzhou was reported. By February 7,totally 10 confirmed cases were reported and no asymptomatic infection was found. They were all imported,including three Wuhan residents,two with a trip to Wuhan,three with a trip to Suizhou,one with a trip to Hangzhou and one with a trip to Thailand(two Wuhan passengers on the same flight). The ratio of male to female cases was 1∶1. The median age was 32 years old. Seven cases were found when they went to a doctor by themselves,and three cases were found during the quarantine. The main clinical manifestations were fever,dry cough and fatigue. The median time from onset to diagnosis was 3 days. By March 3,all the cases were discharged,with median course of 24 days. There were 312 close contacts,and all of them were released after 14 days of quarantine.
Conclusions
To prevent imported cases from outside and stop spread inside taken by Huzhou government was proved to be effective. All the COVID-19 cases in Huzhou were imported,mostly from Wuhan. No local cases were reported.
5.Clinical efficacy and safety of chidamide in treatment of peripheral T-cell lymphoma

Objective:To investigate the clinical efficacy and safety of chidamide monotherapy or its combination of chemotherapy in the treatment of peripheral T-cell lymphoma (PTCL).Methods:The clinical data of 40 cases PTCL patients (26 cases newly diagnosed PTCL and 14 cases relapsed/refractory PTCL) treated with chidamide between December 2015 and April 2019 in Shanxi Provincial Cancer Hospital were retrospectively analyzed. The overall response rate (ORR), progression-free survival (PFS), overall survival (OS) of the patients were observed, and the adverse reactions were analyzed.Results:ORR of all patients was 70.0% (28/40), ORR of the newly diagnosed group was 80.8% (21/26), ORR of the relapsed/refractory group ORR was 50.0% (7/14). The short-term efficacy of the newly diagnosed group was better than that of the relapsed/refractory group ( P = 0.049). The ORR of prognostic index for PTCL (PIT) 0-1 score group was 83.3% (10/12), PIT 2-4 score group was 78.6% (11/14); the therapeutic efficacy of PIT 0-1 score group was better than that of PIT 2-4 score group, and the difference between the two groups was not statistically significant ( P = 0.578). ORR of angioimmunoblastic T-cell lymphoma (AITL) and PTCL not otherwise specified (PTCL-NOS) subtype in the newly diagnosed group was 90.0% (9/10). The median follow-up time was 14.5 months. The median PFS time was 12 months in the newly diagnosed group, 1-year PFS rate and OS rate was 49.6% and 84.2%, 2-year PFS rate and OS rate was 35.9% and 57.4%. The median PFS time was 7 months in the relapsed/refractory group, 1-year PFS rate was 28.6%, 1-year OS rate was 49.0%. There was no Ⅲ-Ⅳ neutropenia and Ⅲ-Ⅳ gastrointestinal reaction in the chidamide monotherapy group; the incidence of Ⅲ-Ⅳdegree neutropenia was 39.4% (13/33) in the chidamide combined with chemotherapy. The incidence of Ⅲ-Ⅳ degree gastrointestinal response rate was 27.3% (9/33), and there was no Ⅲ-Ⅳ degree of liver and kidney dysfunction. Conclusion:Chidamide has good short-term efficacy in newly treated or relapsed/refractory PTCL patients. All patients are well tolerated with chidamide monotherapy or its combination of chemotherapy.
6.Study on clinical management of HPV+/Pap-during cervical cancer screening

Objective To study the clinical management way for HPV+/papanicolaou (Pap)-during cervical cancer screening.Methods To analyze retrospectively the data from the patients who had loop electrical excision procedure (LEEP) for biopsy confirmed cervical intraepithelial neoplasia (CIN)Ⅱ in Peking University People's Hospital from Jan.2010 to Dec.2014.Results (1) For biopsy confirmed CINⅡ,HPV positive rate was 98.5% (135/137),Pap test positive [≥atypical squamous cell of undetermined significance (ASCUS)] rate was 69.3% (95/137),there was significant difference between them (x5=43.32,P<0.01).(2) For the 42 patients with HPV+/Pap-,whose cytology slides were reviewed again.Among them,the interpretations of there were 16 cases confirmed as the same before,while 26 cases were changed to abnormal (≥ASCUS).Cytology be misdiagnosed was 19.0% (26/137) at the first review.Among the 26 cases,13 (50.0%) cases were missed for the little amount of abnormal cells,8 (30.8%) cases for mild atypical morphology changed;the other 5 (19.2%) cases missed for stain problems.(3) For the cervical LEEP samples,37 cases of the pathology diagnosis were upgrade to CIN Ⅲ+,among them,2 cases of microinvasive cervical carcinoma,1 case of invasive cancer,34 cases of CIN Ⅲ;37 cases were CINⅠ or no lesion found;63 cases were still CIN Ⅱ.Four to six months later after LEEP,the cytology abnormal rate was 11.7% (16/137),and the HR-HPV positive rate was 34.3% (47/137).Conclusions Compared with cytology alone,cytology combined with HPV testing increase the sensitivity of cervical high grade lesion.For the cases of HPV+/Pap-cases,the cytology slides should be reviewed.The quality control of cervical exfoliate sample collection and interpretation should be strengthened.LEEP procedure is not only a treatment method,but also it could provide samples to confirm the diagnosis.
