1.Performance of scaffold materials for construction of tissue-engineered vessels

Vascular construction is an important content in tissue engineering domain,in vitro implanting seed cells on absorbable scaffold materials to form multi-layered cellular layer and establish autoblood vessels along the scaffold absorption. The tissue-engineered vessels are characterized by high biocompatibility and plasticity,less foreign body reaction,and no thrombus production and infection; in addition,the vessels may finally become autoblood vessels in the complete significance. In this article,artificial vascular materials,scaffold processing methods,and vascular culture of seed cells were compared to explore an ideal method to make artificial vessels.
2.Progress in medical ultrasound image restoration based on blind deconvolution

Medical ultrasound imaging suffers from poor resolution due to its inherit imaging mechanism.In order to obtain the important diagnostic image in detail,restoration procedure needs to be performed.However,in practical ultrasound imaging,it's difficult to demonstrate the degradation process in detail.Therefore,blind image deconvolution is applied to estimate the original image from the degraded image with unknown point spread function or little available prior knowledge.In this paper,the main algorithms of medical ultrasound image restoration based on blind deconvolution are reviewed and classified into two categories,priori or joint with the image,according to the identification methods,and the basic theories and improved methods are analyzed and discussed.Finally,the future directions in blind restoration for medical ultrasound images are proposed.
3.Effects of mild hypothermia on endothelial cell of cerebral vessels in patients with moderate or severe cerebral infarction

Objective To study the effects of mild hypothermia on endothelial cell of cerebral vessels in patients with moderate or severe cerebral infarction. Methods Sixty patients with moderate or severe cerebral infar-ction were divided into 2 groups: a mild hypothermia group (30 cases) and a non-mild hypothermia group (30 cases). Endothelin (ET), endothelial nitric oxide synthases(eNOS) and von Willebrand factor(vWF) were tested with RIA or ELISA. All the patients were treated with routine medication, in addition, those in mild hypothermia group were also treated with mild hypothermia. 4.Determination of Phenolic Environmental Estrogens in Food Packing Materials by Nonaqueous Capillary Electrophoresis-Chemiluminescence

Nonaqueous capillary electrophoresis-chemiluminescence (NACE) method was developed for the determination of five phenolic environmental estrogens including bisphenol A (BPA),4-nonylphenol(4-NP) ,4-tert-octylphenol(4-tOP),4-tert-butylphenol(4-tBP) and tetrabromobisphenol A(TBBPA) in food packaging material. Target compounds in soaking solution of food packaging material samples were separated by NACE-CL after derivatization with 4-(4,5-diphenyl-lH-imidazol-2-yl) benzoyl chloride ( DIB-Cl) ,reacted with per-oxyoxalate chemiluminescence system,and then detected through magnifying light signal with photomultiplier. With 17β-E_2 as internal standard,qualitative and quantitative analysis of the target compounds were performed by relative migration time and the ratio of relative chemiluminescence intensity,respectively. Several influence factors on separation with NACE,such as the composition and proportion of organic solvent,the concentration of electrolyte,temperature surrounding of the capillary,the concentration of acetic acid and work voltage,were investigated. And the peroxyoxalate chemiluminescence system was optimized. Under these conditions,4-tBP,BPA,4-OP,4-NP and TBBPA were all separated and showed good linearities in the ranges of 0.0095-3.0 mg/L,0.0087-3.0 mg/L,0.0085 -3.0 mg/L,0.011 -3.0 mg/L,0.009-3.0 mg/L,respectively,with correlation coefficients over 0. 9947. The RSDs of relative migration time and relative chemiluminescence intensity ratio were 0.9% -3.0% and 6.5% - 8.6%. Recoveries for the spiked samples ranged from 86.7% to 98. 8%. Five common food packaging material samples were analyzed. The proposed method is simple and sensitive for the quantitative determination of phenolic environmental estrogens in food packaging material samples.
5.Status analysis and countermeasure research of the establishment of the Ministry of Education innovation team in independent medical colleges

Up to now,there are 21 ministry of education innovation teams established by 37 independent medical colleges and universities,which has become an important part of the medical education system innovation team building.From the perspective of SWOT (strengths,weaknesses,opportunities,threats) analysis,in the process of ministry of education innovation team establishment in the independent medical colleges,there are unique advantages,followed by obvious disadvantages,and there are favorable outside opportunities,accompanied with unprecedented challenges.Independent medical colleges shall keep up with the development trend,leverage the national regional development policy,give full play to the advantages,endeavor to overcome weaknesses,seize the opportunities and face the challenges actively,make the right strategic decisions,in order to better play an irreplaceable role in higher medical education system.

7.Influence of tamoxifen on the level of cellular proliferation on cultured human pituitary adenoma cells and its mechanism

Aim To investigate the effects of tamoxifen on the proliferation and DNA synthesis of cultured human pituitary adenoma cells and make a further investigation of the mechanism of the inhibitory effect of tamoxifen on the proliferation of pituitary adenoma cells. Methods The techniques of MTT colorimetry, 3H-TdR, flow cytometry, PKC activity detection and cAMP/ cGMP levels detection were used to detect or observe the effects of tamoxifen on proliferation, DNA synthesis, cell cycle, PKC activity and cAMP/ cGMP levels of cultured human pituitary adenoma cells, respectively.Results ①Tamoxifen (0.1,1 and 10 ?mol?L -1) inhibited the proliferation and DNA synthesis of cultured human pituitary adenoma cells in a dose-dependent manner.② tamoxifen (1,10 and 20 ?mol?L -1) increased the ratio of G_1 phase of pituitary adenoma cells, and decreased the ratio of S and G_2 phase markedly;②compared with control, PMA, a PKC activator, increased the activity of membrane and total PKC in human pituitary adenoma cells. However, after a 15-min treatment with tamoxifen (10 ?mol?L -1),a significant reduction of the activity of cytoplasm, membrane and total PKC in human pituitary adenoma cells was observed;③tamoxifen (1 and 10 ?mol?L -1) increased the amount of cAMP in the cytoplasm of human pituitary adenoma cells, but had no effect on that of cGMP. Conclusion These data provide an important clue to explore the molecular mechanisms of the inhibitory effect of tamoxifen on the proliferation of pituitary adenoma cells, and suggest that the modulating effect of tamoxifen on the proliferation of pituitary adenoma cells results from interactions of several cellular signaling pathways.

The effect of recombinant human phospholipase D2(rhPLD2)in vivo was investigated on the secretion of serum glycosyl phosphatidylinositol-specific phospholipase D(GPI-PLD)in guinea pigs of chronic asthma.Ater treating the guinea pigs attacked by chronic asthma with rhPLD2,the GPI-PLD activity detection was canrried out by phase separation of human placental alkaline phosphatase in Triton X-114.Compared with the healthy guinea pigs(NS group),the serum GPI-PLD in the guinea pigs of chronic asthma are much higher than that of control groups,P≤0.01.Our results showed that rhPLD2 could significantly reduce the secretion of GPI-PLD when the guinea pigs were attacked by chronic asthma.
9.Characteristic of salvianolic acid binding to bovine serum albumin and structure-performance relationship

Objective To study the characteristic of salvianolic acid(SA) binding to bovine serum albumin(BSA) and the structure-performance relationship.Methods The interactions between BSA and four natural SA(SAⅠ,rosmaric acid;SAⅡ,lithosperic acid;SAⅢ,salvianolic acid A;and SAⅣ,salvianolic acid B) were investigated by fluorescence and ultraviolet spectroscopy.Results The intrinsic fluorescence of BSA was quenched by SA via forming SA-BSA complexes and non-radiation energy transfer.The parameters of SA-BSA binding process,such as the static apparent association constant KA,the number of binding site n,the efficiency of energy transfer E,the spatial distance r were obtained,and the thermodynamic constants ?G,?H,and ?S were calculated.Conclusion The results indicate that SAⅢ bounds to BSA mainly through a hydrophobic force and the other three SA-BSA interactions are mainly driven by hydrogen bond and Van der Waals' force.Y,a comprehensive binding parameter constructing from the equation Y=lg (KA?E?n/r),could be used to reflect the interaction extent of SA-BSA system.The Y value changes with the number of free phenolic hydroxyl and molecule volume and decreases in the order of SAⅢ→SAⅣ→SAⅡ→SAⅠ.The results from correlation analysis indicate that it could be possible to estimate SA-BSA binding extent from hydrophobic parameter clogP and topo-polar surface area per unit of molecular weight tPSA/Mr of SA molecule.

The research of structural biology is closely related to the field of biotechnology.Carrying out the second class of structural biology will be helpful in learning theoretical knowledge of biotechnology and in improving the teaching quality.Based on our experiences of carrying out the second classroom activities among students majoring in biotechnology,we believe that the selected teaching contents,reasonable classroom design,sufficient preparation before the class,flexible teaching methods and objective after-school summaries are essential for improving the teaching quality.
