Background and purpose: More than 90% of cancer patients are incurable because of drug resistance. Activation of PI3K/Akt/mTOR signaling pathway in breast cancer, as a target for chemotherapy drugs has become a hot topic of breast cancer treatment. This study aimed to investigate the effect and mechanism of XCL1 on the proliferation of drug-resistant breast cancer cell, whether is related with the mTOR signaling pathway. Methods:Established gemcitabine-resistant breast cancer cell lines (MDA-MB-231/Gem). CCK8 to detect the proliferation of MDA-MB-231 and MDA-MB-231/Gem, RT-PCR and ELISA to determine the XCL1 expression level of the two cell lines, Western blot to detect the expression of mTOR. Results:Compared with MDA-MB-231, MDA-MB-231/Gem showed an enhanced proliferative capacity. The expression of XCL1 was increased in the resistant cell lines. Both of protein level and phosphorylation level of mTOR increased in drug-resistant cell lines. The MDA-MB-231 added exogenous XCL1 for 24 h, showed an enhanced cell proliferation. Adding anti-XCL1 antibodies in MDA-MB-231/Gem could reduce cell proliferation and treating MDA-MB-231/Gem with the mTOR inhibitor could also reduce cell proliferation, as well as the XCL1 expression level. Conclusion:XCL1 promotes the proliferation of drug-resistant breast cancer cells mediated by activation of the mTOR pathway.

Objective To investigate the effect of tadpole extract(T871 3) on tumor cells and its mechanism. Methods We studied the effects of T871 3 on proliferation, differentiation and apoptosis of HL 60 cells by cytomorphological observation, cytochemistry and TUNEL method. We also examined gene expression during the induction of apoptosis and differentiation in tadpole extract treated HL 60 cells by in situ hybridization and intact cell mRNA dot blot techniques. Results 1 T871 3 was able to inhibite HL 60 cells proliferation. 2 T871 3 was able to induce HL 60 cells to differentiate along monocyte macrophage lineage at low concentration, and apoptosis at higher concentration. 3 The differentiation of HL 60 cells was accompanied by downregulations of c myc,c myb gene expression, The apoptosis of HL 60 cells was accompanied by downregulations of c\|myc bcl 2 gene expression, suggesting that these genes may be involved in the apoptosis and differentiation process. Conclusion Tadpole extract may have effects on HL 60 cells through changing the oncogene expression. [

Objective: Our purpose was to investigate the changes of adrenocorticotropin (ACTH), Cortisol (Cs) and prolactin (PRL) levels in peripheral blood of patients with active systemie lupus erythematosus(SLE). Methods: Radioimmunoassy. Results: Active SLE patients had a low level of ACTH,Cs in peak phase and a high level of PRL. Consequentaly, the ratio of PRL/Cs increased, which was positively correlative to active index of SLE. Conclusion: Endogenetic secretion in sufficiency of glucocorticalsteroid and increase of PRL have important function in the initiation and development of SLE.

OBJECTIVE To investigate the relationship between gastric polyps and Helicobacter pylori infection.METHODS The patients with gastric polyps were taken by gastroendoscopy in 2005.The tissues from their antrums were examined for presence of H.pylori.We collected and analyzed all of their general information and the data about their gastric polyps and H.pylori infection condition.RESULTS In the 95 gastric polyps patients,76 cases(80.0%) had inflammatory polyps and 19 cases(20.0%) had H.polyps.The total H.pylori infection rate was 33.7%.The H.pylori infection rate in the inflammatory polyps patients and H.pylori patients were 38.2% and 15.8%,respectively.CONCLUSIONS H.pylori infection promotes the formation of gastric inflammatory polyps. The examination and treatment for H.pylori is necessary for the gastric polyps patients.

Objective To evaluate the correlation and difference of reversed phase high performance liquid chromatography (RP-HPLC) and fluorescence polarization immunoassay (FPIA) on determining serum concentration of carbamazepine. Methods Fifty serum samples were collected, both RP-HPLC and FPIA methods were employed to determine the concentration of carbamazepine. The results were analyzed by paired t test,Bland-Altman and Deming regression methods, respectively. Results The results of measuring 50 samples by the two methods showed that FPIA datas were significantly higher than RP-HPLC datas, and there was statistically significant difference(P<0.05) and poorer consistency between two methods; There was good correlation between carbamazepine concentrations determined by the two methods.Deming regression equation was CFPIA=1.195 3 CRP-HPLC-0.144 0,and Pearson correlation coefficient was 0.968 5.Conclusion Clinicians should pay more attention to the difference of carbamazepine concentration determination by different methods when carbamazepine individualized dosage regimen was adjusted according to therapeutic drug monitoring.

Objective:To study the stability of compound Kushen mixed liquid to provide reliable evidence for safe clinical drug application. Methods:Compound Kushen injection was mixed respectively with 0.9% sodium chloride injection and 5% glucose injection, and then placed under the different conditions for 0,1,2,4,8,12,24 and 48 h.HPLC was conducted to determine the content changes of four components in compound Kushen mixed liquid, and the appearance,pH and insoluble particles were observed as well. Results:The mixed liquid of compound Kushen with 0.9% sodium chloride injection was stable in 48 h without the influence of light and temperature. However, the mixed liquid of compound Kushen with 5% glucose injection had poorer stability with storage time shorter than 12 h at room temperature and 2 h at high temperature. Conclusion:The stability of the mixed liquid of compound Kushen is closely related to the pH value of solvent.0.9% Sodium chloride injection is recommended as the solvent, and the mixed liquid should be used up in 48 h.

Polycyclic aromatic hydrocarbons ( PAHs) are ubiquitous environmental pollutants, whose carcinogenicity is determinated. The mechanism of their carcinogenicity: PAHs are able to combine with aryl hydrocarbon hydrocarbon receptor ( AhR ) , resulting in some toxicity and carcinogenicity.AhR is a ligand-dependent activation transcription factor, which is activated by a large variety of ligands, regulating the expression of a series of gene involved in metabolism, and participating in important biological processes, such as singal transduction, cell proliferation, differentiation and apoptosis, and so on.Besides, it's closely related with the tumor development. Thus, it will provide a new approach for cancer prevention and treatment to study the role of PAHs and AhR in the development of tumor.

This paper was aimed to study the method of rapid determination of total flavonoids in Chrysanthemum of different processing methods by near infrared spectroscopy (NIRS). The Chrysanthemum was dried by three different processes. The three methods were directly drying, drying after steamed and drying after fry, respectively. The determination of total flavonoids in Chrysanthemum by different processing methods was produced by using UV-Vis spectrophotometry. Collecting the NIRS spectra of Chrysanthemum, the quantitative analysis model of total flavonoids content in Chrysanthemum of different processing methods was established by partial least square (PLS) and the model was validated. The correlation coefficient (R2), the root-mean-square error of calibration (RMSEC) and the root-mean-square error of prediction (RMSEP) were 0.996 19, 0.104 and 0.168, respectively. The correlation coefficient of predication (r) was 0.979 3 which state that the prediction was accurate. The method of NIRS had the advantage of fast determination, simple operation and high accuracy of prediction, and could be used for rapid determination of total flavonoids content in Chrysanthemum of different processing methods.
