1.Research progress in T-cell lymphoblastic lymphoma

T-lymphoblastic lymphoma (T-LBL) is a rare form of non-Hodgkin lymphoma (NHL),which is biologically similar to T-acute lymphoblastic leukemia(ALL),and occurs most frequently in childhood and adolescence.T-LBL progresses fast.The treatment of LBL with conventional chemotherapy regimens for NHL has shown relatively low rates of complete remission and of disease-free survival.Now,it is reasonable to treat patients with LBL with the current ALL-type protocols,and hematopoietic stem cell transplantation may yield a more favorable overall survival (OS) in T-LBL patients,especial in relapsed or refractory LBL patients.

Bone marrow infiltration in lymphoma is crucial to the disease diagnosis,staging,treatment options and prognostic evaluation.Although bone marrow biopsy is the main detecting method,it has high false negative rate.At present,18F-fluorodeoxyglucose positron emission tomography-computed tomography (18 F-FDG PET-CT) technology which is integrating morphological characteristics with metabolic function provides a new way for the diagnosis,staging and prognostic evaluation of bone marrow infiltration in lymphoma due to its advantages of high sensitivity,high specificity and functional image.
4.Study on Inhibition of C.Chrysantha on DEN Induction of Murine Liver Cancer

Objective Using a short-term in vivo laboratory model of murine liver cancer induced with diethylnitrosamine (DEN),we inspected the impact of 5% C.Chrysantha leaves and 5% concentrations on induction of liver cancer with DEN.Methods Laboratory Winstar rats,all being male and SPF grade,were administered with the liver cancer initiator DEN(200?g/kg,ip,once only),then with murine food containing 0.015% 2-AAF (2-Acetaminoflueren) for 2 weeks,following an excision of the augmented parts as a selective promoting program that speeded up the proliferation of precancerous foci from the initiated hepatocytes.On the beginning of this experiment,5% C.Chrysantha leaves and 5% concentration-contained food were given to the trail rats; in contrast,normal food to the control.Rats were killed 3 days after feeding of 2-AAF-contained food.Targeting part of liver were collected for ?-glutamyltranspeptidase (?-GT) staining,and numbers and areas of?-GT positive foci per cm2 of the liver area were calculated using a software designed by us.Results Numbers and areas of?-GT positive foci of the trail are significantly less than those of the control.Conclusion 5% C.Chrysantha leaves and 5% concentrations inhibit the DEN induction to liver cancer.
5.Effect of dendritic cells co-cultured with cytokine induced killer cells on cytotoxicity against non-Hodgkin's lymphoma cell

Objective To study proliferation,secreted cytokines,immune phenotypes and cytotoxicity on Raji cells by cytokine induced killer (CIK) cells co-cultured with dendritic cells (DC).Methods The mononuclear cells from peripheral blood of healthy individuals were extracted,then cultured the cells under 5 % CO2 at 37 ℃ for 2 hours.DC were induced from suspended cells,and CIK cells were from adherent cells.After 9 days of nurturing,two types of cells were mixed.CIK cells were cultured alone as the control.The cytotoxic activity of CIK and DC-CIK cells were detected by MTT assay.The morphologies,proliferation,secreted cytokines,and immune phenotypes of the two cells in day 0,3,6,9,12,15 in culture were monitored.Results In day 12 in culture,comparing with CIK cells,DC-CIK cells significantly enhanced the proliferation rate [(42.44±2.68) fold vs (30.01±2.05) fold] (t =11.64,P < 0.05) and had increased IL-2,IFN-γ,IL-12 and TNF-α secretion [(124.34±12.57) ng/L vs (56.32±6.58) ng/L,(496.60±95.32) ng/L vs (247.80± 69.45) ng/L,(84.92±6.07) ng/L vs (24.18±3.31) ng/L,(380.6±45.95) ng/L vs (196.61±24.19) ng/L] (t =15.16,P < 0.05; t =6.67,P < 0.05; t =27.78,P < 0.05; t =11.20,P < 0.05),and there were more CD3+ CD8+ cells and CD3+ CD56+ cells in the co-culture [(71.79±1.73) % vs (60.37±3.24) %,(48.54±3.30) % vs (33.07±2.22) %](t =9.83,P < 0.05; t =12.30,P < 0.05),and DC-CIK cells had a significandy increased cytotoxicity on Raji cells in vitro at the same ratio of effector cells to target cells.Conclusion CIK cells have higher proliferation rate and cytotoxicity against Raji cells when co-cultured with DC.
6.Effects of triptolide on proliferation and apoptosis in rat mesangial cells induced by transforming growth factor β1 and related mechanisms

Objective To investigate the effects of triptolide on proliferation,apoptosis and the changes of Ski,Smad3,Smad7 and collagen type I(ColI) in cultured rat mesangial cells induced by transforming growth factor (TGF)-β1.Methods Cultured HBZY-1 rat mesangial cells were divided into 5 groups:(1)normal control group; (2)TGF-β1 group (10 μg/L); (3)-(5)triptolide (0.4,2,10 μg/L)+TGF-β1 (10 μg/L) groups.The cell proliferation was detected by MTT.Apoptosis of mesangial cells was detected by TUNEL assay.The expressions of Ski,Smad3,Smad7 mRNA were examined by real-time quantitative PCR.The expressions of Ski,Smad3,Smad7 and ColI protein were detected by Western blotting.The localizations of Ski and Smad3 protein were detected by laser confocal fluorescence microscope.Results Compared with the normal control,TGF-β1 (10 μg/L) significantly stimulated mesangial cells proliferation,while decreased apoptosis.The mRNA and protein expressions of Ski,Smad7,Smad3 and ColI protein expression in TGF-β1 group were increased (P >0.05).In comparison with TGF-β1 group,triptolide could significantly inhibit TGF-β1-induced mesangial cells proliferation in dose-dependent manner,and promote the apoptosis of mesangial cells.In TGF-β1 group,mRNA and protein expresscons of Ski and Smad7 were increased (P<0.05),Smad3 mRNA and protein were decreased (P >0.05),and ColI protein was decreased (P<0.01).In comparison with TGF-β1 group,fluorescence intensity of Ski,Smad3 proteins was significantly increased in cytoplasm,while decreased in nucleus.Conclusions Triptolide can inhibit TGF-β1-induced mesangial cells proliferation through regulating the expressions of Ski,Smad7 mRNA and protein,inhibiting Ski.Smad7 translocation to the nucleus,and down-regulating Smad3 mRNA and protein expression.Triptolide can promote apoptosis of mesangial cells.
7.Observations on the Efficacy of Pricking-cupping Bloodletting in Treating Cervical Spondylotic Radiculopathy

Objective To investigate the clinical efficacy of Yang’s pricking-cupping bloodletting in treating cervical spondylotic radiculopathy and its effect on trapezius tension.Methods Seventy-eight patients with cervical spondylotic radiculopathy were randomly allocated to treatment and control groups, 39 cases each. The treatment group received pricking-cupping bloodletting and the control group, conventional acupuncture plus cupping. The nerve function was scored, the visual analogue scale (VAS) score was recorded and trapezius tension was measured in the two groups before and after treatment. The clinical therapeutic effects were compared between the two groups.Results There were statistically significant pre-/post-treatment differences in the nerve function score, the VAS score and trapezius tension in the two groups (P<0.01). There were statistically significant post-treatment differences in the VAS score and trapezius tension between the treatment and control groups (P<0.05). The total efficacy rate was 94.9% in the treatment group and 89.7% in the control group. There was a statistically significant difference in theclinical therapeutic effect between the two groups (P>0.05).Conclusions Yang’s pricking-cupping bloodletting can effectively improve pain symptoms and trapezius tension in patients with cervical spondylotic radiculopathy.
8.The association study on the polymorphisms of estrogen receptor α and β genes and systemic lupus erythematosus in Chinese Han cohort of Yunnan Province

Objective To analyze the association of polymorphisms of estrogen receptor (ER) α and β genes with systemic lupus erythematosus (SLE) in Chinese Han cohort of Yunnan Province.Methods XbaⅠ and Pvu Ⅱ of ERα gene,Rsa Ⅰ and Alu Ⅰ of ERβ gene were typed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in 697 SLE patients and 638 healthy controls.The frequency distribution of the alleles and genotypes were analyzed by Hardy-Weinberg equilibrium test and x2 test.Results ① For ERα gene,the frequency of minor allele of Pvu Ⅱ C in SLE patients was significantly higher than healthy controls (x2=15.427,P=0.001);the allele frequencies of XbaⅠ in SLE patients showed no significant difference compared with healthy controls (P>0.05).The frequency of minor genotype of Pvu Ⅱ CC in SLE patients was significantly higher than healthy controls (x2=17.371,P=0.011).The frequency of two locus haplotype AATT in SLE patients was significantly lower than healthy controls (x2=6.333,P=0.012);the frequency of the two locus haplotype AACC in SLE patients was significantly higher than healthy controls (x2=7.771,P=0.038).② For ERβ gene,the frequency of minor allele RsaⅠ A in SLE patients was significantly lower than healthy controls (x2=12.595,P=0.013);the allele frequencies of Alu Ⅰ in SLE patients showed no significant differences compared with the healthy controls (P>0.05).The frequency of minor genotype AA of Rsa Ⅰ in SLE patients was significantly higher than healthy controls (x2=41.456,P=0.000).The frequency of two locus haplotype AAGG in SLE patients was significantly higher than healthy controls (x2=37.063,P=0.000).The frequency of the two locus haplotype AAGA in SLE patients was significantly lower than healthy controls(x2=21.086,P=0.001).③ Pvu Ⅱ C was related with splenomegaly (x2=4.212,P<0.05).The two locus haplotype AGTC of Xba Ⅰ and Pvu Ⅱ was related with edema (x2=7.898,P<0.05).Conclusion There are associations between the polymorphisms of ERα and ERβ genes and SLE.The ERα and ERβ genes may be the susceptible genes for SLE in Yunnan Han Chinese Cohort.
9.Clinical Research of Acute Stroke Treatment Using Acupuncture

Objective: To study the clinical effects of acupuncture therapy for acute stroke. Methods: 150 patients suffered from acute stroke were randomly divided into two groups: drug group, acupuncture group. To apply routine drug treatment of Neurology for drug group: pisacetam 250mL, Salvia injection 20mL added into normal sodium 250mL for intravenous drip, once a day. For acupuncture group applied calm the liver and suppress yang acupuncture therapy beside drugs. Modified Scandinavian Stroke Scale (SSS) and activities of daily living (ADL) scale (Barthel Index) were used for evaluation, as well as biochemistry\hemorheology determination. Results: The scale showed significant difference as compared with the drug group, biochemistry index had non-significant difference, and hemorheological index had partly improved. Conclusion: The scale could synchronously reflect the clinical symptom and changes of patient with stroke.Acupuncture therapy has a significant improvement of acute stroke treatment.
10.Clinical Study of the Effect of Electroacupuncture on Rehabilitation and TCD in Convalescent Stroke Patients

Objective To investigate the clinical therapeutic effect of electroacupuncture on ischemic stroke by recording the therapeutic effect score and performing transcranial Doppler (TCD) detection in electroacupuncture-treated convalescent stroke patients before and after treatment.Methods Sixty convalescent patients with ischemic stroke were randomly allocated to treatment and control groups, 30 cases each. The control group received routine medication and the treatment group, electroacupuncture in addition. In the two groups, pre- and post-treatment neurological defect scores were recorded and the clinical therapeutic effects were observed after four weeks of treatment. Pre- and post-treatment middle cerebral artery (MCA) pulsation indices (RI), resistance indices (RI) and ratios of systolic to end-diastolic (S/D) blood flow velocity detected by TCD were compared between the two groups. Results The total efficacy rate was 80.0% in the treatment group and 50.0% in the control group; there was a statistically significant difference between the two groups (P<0.01). There was a statistically significant pre-/post-treatment difference in the MESSS score in the two groups (P<0.01). There was a statistically significant post-treatment difference in the MESSS score between the treatment and control groups (P<0.05). There were statistically significant pre-/post-treatment differences in TCD indices (PI, RI and S/D) on the affected side in the two groups (P<0.05). There were statistically significant post-treatment differences in TCD indices on the affected side between the treatment and control groups (P<0.05).Conclusion Electro- acupuncture can promote rehabilitation in convalescent stroke patients.
