2.Effects of PM2.5 on Expression of Pigment Epithelium-derived Factor in Bronchial Epithelial Cells

Objective To explore the effect of particulate matter (PM) 2.5 on the expression of pigment epithelium-derived factor (PEDF) protein in bronchial epithelial cells (BEAS-2B). Methods Human BEAS-2B were subcultivated, followed by low, medium and high concentrations of PM2.5 (25μg/ml, 50μg/ml, 100μg/ml) stimulation for 24 hours. The expression of PEDF protein in supernatant was ana-lyzed by enzyme-linked immunosorbent assay (ELISA), and the expression in BEAS-2B cells was detected by Western blotting. Results Compared with the control group, the expression of PEDF protein in supernatant and BEAS-2B cells induced by PM2.5 (25 μg/ml) in-creased, but no significance was found (t=-0.730, t=-1.840, P>0.05), and the expression induced by PM2.5 with the concentrations of 50μg/ml and 100μg/ml significantly increased (t>5.798, P<0.05). Conclusion PM2.5 with the concentrations of 50μg/ml and 100μg/ml could increase the expression of PEDF protein in a concentration-dependent manner both in supernatant and BEAS-2B cells.
3.Clinical and endoscopic features of pediatric eosinophilic gastroenteritis in 49 cases

Objective To explore the clinical features and endoscopic manifestations of eosinophilic gastroente-ritis(EG)in children.Methods A retrospective analysis was conducted of the clinical manifestation,laboratory examination,endoscopy(upper and/or colonoscopy)performance,diagnosis and treatment of 49 cases of patients who were diagnosed as EG in Children's Hospital of Nanjing Medical University from July 2013 to July 2015.Results The common clinical manifestations of EG in children were hematochezia(23 cases),diarrhea(20 cases),vomiting(18 cases)and abdominal pain(15 cases).The ages of children admitted to hospital for the first time ranged from 1 month and 7 days old to 13 years and 7 months old,and the mean age was 59.4 months old,in which most patients were younger than 1 year old,accounting for 38.77%(19 cases)of all.Peripheral blood eosinophilia was present in 20 cases(40.82%)of the patients,and only 6/18 cases(33.33%)of the patients had elevated serum IgE.Upon endoscopic analysis,the lesions involved esophagus(4 cases),stomach(14 cases),duodenum(20 cases),small intestine(15 cases),colon(28 cases)and rectum(6 cases),and the most common manifestation under gastroscopy was mucosal hyperemia edema(27 cases)and erosion(9 cases),while the most common manifestation under colonoscopy was mucosal hyperemia edema(25 cases)and nodular hyperplasia(24 cases).All patients improved with food restriction,in which 8 cases were treated with glucocorticoid while 9 cases with oral Singulair and 9 cases with oral Loratadine.All children with symptoms were alleviated somewhat,but 5 cases of them relapsed after drug withdrawal.Conclusions The clinical manifestations of EG in children varied and were mainly hematochezia,vomiting,diarrhea and abdominal pain.Some patients had the elevated peripheral blood eosinophilia and serum IgE.The most common manifestations under gastroscopy were mucosal hyperemia edema and erosion while the most common manifestations under colonoscopy were mucosal hyperemia edema and nodular hyperplasia.

5.Investigation on the Situation of Self-management in 206 Diabetic Patients

OBJECTIVE: To investigate the impact of self-management of diabetic patients on diabetes mellitus and its complication in Chongqing. METHODS: Hospitalized diabetic patients were surveyed randomly using questionnaire modified and simplified from diabetes care profile(DCP)and diabetes knowledge test(DKT)designed by Diabetes Research and Training Center of Michigan University. RESULTS: Most of diabetic patients showed a low level of self-management. The self-management of young and middle aged diabetic patients was poorer than that of aged patients. The glucose control in patients who received diabetic education was superior to those without receiving diabetic education. The glucose control in patients with regular outpatient follow-up was better than patients followed up by other methods. A large number of diabetic patients combined with cardiovascular and cerebrovascular diseases got the bad habits of drinking and smoking. CONCLUSION: Diabetic education is a important method to improve the self-management of diabetic patients. The conception of diabetic patients on diabetic education in Chongqing should be improved.
6.Clinical observation on acupoint catgut embedding therapy for treatment of ulcerative colitis

Objective To observe therapeutic effects of acupoint catgut embedding therapy on ulcerative colitis.Methods 55 cases of ulcerative colitis were randomly recruited into a treatment group and a control group. The treatment group was treated with catgut embedding, and the control group was treated with oral administration of salicylazosul fapyridine. Results The therapeutic effect of the treatment group was better than those of the control group (P=0.002<0.05). Conclusion Acupoint catgut embedding therapy has a better therapeutic effect on ulcerative colitis.
7.Effects of Acupuncture on Blood Lipids in Hyperlipidemia Mice

Objective To observe the effects of acupuncture on blood lipids of hyperlipidemia mice, and explore the mechanism of acupuncture against endothelial dysfunction. Methods 40 ApoE (-/- ) mice were randomly divided into control group (C), acupuncture at non-acupoint group (D), acupuncture at acupoint group (E), and simvastatin group (F) with 10 cases in each group. After 8 weeks, total cholesterol (TC), the plasma angiotensin II (Ang II), endothelin-1 (ET-1) and nitric oxide (NO) were measured with enzyme-linked immuno sorbent assay (ELISA). The mice myocardial angiotensin II type 1 receptor (AT1R), and endothelin-1 type A receptor (ETAR) protein were detected with Western blotting. Results The blood lipid, the content of plasma Ang II and ET-1, and the level of AT1R and ETAR in the heart tissue were significantly lower, and the content of NO was significantly higher in groups E and F than in group C (P<0.05). Conclusion Acupuncture can inhibit the level of blood lipids in ApoE(-/-) mice, reduce the levels of Ang II and ET-1, increase the level of NO in peripheral blood, and inhibit the expression of AT1R, ETAR in heart tissue.

Objective To investigate blood plasma melatonin level in night-shift nurses and explore the relationship of blood plasma melatonin level with nervous system symptom (insomnia、anxiety、depression). Methods ELISA were used for detection of blood plasma melatonin level in 80 night-shift nurses of different age group. Results Blood plasma melatonin level of shift work nurses (36to40、41to45 yearold) were significant lower than the corresponding age group of the control group, the nervous system symptom of these age group night-shift nurses correlated to melatonin level of melatonin. Conclusions Blood plasma level of melatonin have a close relation to nervous system symptom(insomnia、anxietydepression).

Objective To investigate the effect of thymosin ?1 (T?1) on cellular immune function in gastric cancer patients through observing its treatment on the differentiation of T-lymphocyte subsets from screened peripheral blood mononuclear cells (PBMCs). Methods PBMCs were obtained by centrifugation of blood samples from 18 healthy subjects and 32 patients with gastric cancer,and then cultured in the presence of culture medium with addition of T?1 at 50,10 and 1 ?g/ml for 2 d. T lymphocyte subsets (such as CD4+,CD8+ and CD4+ CD25+ Foxp3+ T cells) and Th1/Th2 multiplex cytokines were detected by flow cytometry (FCM). Results After PBMCs isolated from healthy people and patients were incubated with or without T?1,there was no significant change in percentage of CD4+,CD8+ peripheral lymphocyte subsets and ratio of CD4+/CD8+. Objective:To identify the effective concentrations of Ropivacaine in the modified fascial iliac compartment block(FICB)that would not affect the movement of the affected limb but would offer effective pain relief after total knee arthroplasty(TKA)in elderly patients.In addition, adverse reactions within 24 hours of FICB were examined.Methods:This study was a prospective, single-arm sequential trial.Forty-five elderly patients treated with TKA at the First Affiliated Hospital of Soochow University between September 2021 and March 2022 were selected, with an American Society of Anesthesiologists(ASA)score of Ⅰ or Ⅱ.All patients were given ultrasound-guided FICB on the surgical side under general anesthesia and 10 minutes before the operation, and the injection volume was 30 ml.According to preliminary experiments and relevant literature, the initial concentration of Ropivacaine was 0.1%, and the concentration for the next patient was determined using a modified Dixon sequential method.If the quadriceps femoris muscle strength score of the first patient was ≥4, there was no resting pain[visual analogue scale(VAS)score ≤1], and the VAS score during activity was ≤3, the concentration for the next patient would be reduced.Conversely, the concentration would be increased.The Ropivacaine concentration was increased or decreased by 0.01% each time and the trial would be stopped after 12 reentries.The median effective concentration(ED50), 95% effective concentration(ED95)and corresponding 95% confidence interval(CI)of ropivacaine were calculated using the probit model.Meanwhile, adverse reactions within 24 hours of FICB were monitored.Results:Of 43 elderly patients who completed the trial, the intervention was effective in 23 and ineffective in 20.While ensuring that postoperative limb movement in elderly patients was not affected, a single injection of 30 ml ropivacaine through FICB was able to effectively inhibit postoperative pain, and the ED50 and ED95 of ropivacaine were 0.072%(95% CI: 0.065%-0.078%)and 0.093%(95% CI: 0.084%-0.124%), respectively.Within 24 hours of FICB, 2 patients had lower limb weakness and could not get out of bed and walk, and 5 patients had severe pain and needed additional analgesics.No other adverse reactions were found. Conclusions:The effective ED50 and ED95 of Ropivacaine for postoperative pain relief in elderly patients after TKA are 0.072% and 0.093%, respectively.And the incidence of adverse reactions is low.
