It has been confirmed that hepatocellular carcinoma belongs to gene related disease.Biological characteristic of hepatocyte can be infected by abnormal expression of related genes,resulting in normal cell transformed to carcinoma cell,therefore,we should do further research to find new methods for diagnosis and treatment of HCC.As a new technique for gene knockdown,RNAi has been applied more in the fields of gene function and gene therapy.This paper is a review about recent advance in HCC gene therapy with RNAi.

3.Study on Optimization of Ultrasonic-assisted Extraction Technology for Total Flavonoids from Tibetan Scindapsus aureus by Response Surface Method

OBJECTIVE:To optimize the ultrasonic-assisted extraction technology of total flavonoids from Tibetan Scindapsus aureu. METHODS:The single-factor test was adopted to investigate the effects of extraction time,extraction temperature,the vol-ume fraction of ethanol,the ratio of liquid to solid and power on the extraction rate of total flavonoids from Tibetan Scindapsus au-reu in the process of ultrasonic-assisted extraction,based on which,four-factor and five-level Box-Behnken central component de-sign was performed with unchanged ultrasonic power,and then response surface method was adopted to analyze the effects of the 4 factors and their interaction on the extraction rate of total flavonoids to determine the optimal factor and level,and verification tests were conducted.RESULTS:The optimal ultrasonic-assisted extraction technology was as follows as extraction time of 29 min,tem-perature of 44 ℃,volume fraction of ethanol of 78%,ratio of liquid to solid of 13∶1(ml/g). In three verification tests,the aver-age extraction rate of total flavonoids was 2.130%,with an relative error of 0.005% compared to the theoretical value. CONCLU-SIONS:The optimal technology is reasonable and feasible,and can be used for theultrasonic-assisted extraction of total flavonoids from Tibetan Scindapsus aureu.

Alpha-fetoprotein is an important molecular marker for clinical diagnosis, evaluation of curative effect and prognosis of cancer for long time, especially for hepatocellular carcinoma. But for physiological function of alpha-fetoprotein, we know less than its clinical significance. So here this article presents a review about physiological function of alpha-fetoprotein and its application in molecular therapy of hepatocellular carcinoma.
6.Research on medical equipment reservation system based on hospital information platform

Objective: To further optimize the hospital inspection process, shorten the patient waiting time of examination and improve the overall utilization of medical equipment. Methods:HIS and PACS to realize the information system integration by using hospital information platform, and then develop a set of medical equipment reservation system. The doctor workstation through the system could real-time help patient appoint check items, and eliminate patients to check the front desk or reservation center reservation registration steps. Results:The successful application of medical equipment reservation system not only optimize hospital inspection process, shorten the patient waiting in line inspection time, but also increase the utilization rate of MR equipment by 35%. Conclusion: The use of medical equipment reservation system saves the patient queuing registration, waiting for the examination of the time, and improve the utilization rate of hospital medical equipment, create economic benefits for the hospital and brought widespread popularity and reputation for the hospital.
7.Carrageenan oligosaccharides inhibit growth-factor binding and heparanase activity.

This study is designed to investigate the anti-tumor and anti-angiogenesis mechanism of carrageenan oligosaccharides. The effects of carrageenan oligosaccharides on basic fibroblast growth factor (bFGF) induced cell proliferation, heparanase activity and bFGF binding ability were evaluated in human cervical cancer cells (HeLa) and human umbilical vein endothelial cells (HUVEC). Results indicate that, at rational concentrations, carrageenan oligosaccharides showed low cytotoxic effect. At relatively low concentrations (0.2-200 microg x mL(-1)), these oligosaccharides could competitively bind bFGF and inhibit bFGF induced cell proliferation. In these samples, oligo-lambda-carrageenans (dp2-8) were the most potent bFGF antagonists. At concentration of 20 microg x mL(-1), their inhibitory ratio reached to 30%. The heparanase enzyme assay revealed that three kinds of carrageenan oligosaccharides showed different inhibitory activities to two cell lines. For HeLa cell, oligo-lambda-carrageenans showed highest inhibitory effect, but for HUVEC, oligo-kappa-carrageenans (dp9-17) were the best inhibitors. Current observations demonstrated that the biological activities of carrageenan oligosaccharides are closely related to the molecular weight, carbohydrate structure and the content and linking position of sulfur groups. Carrageenan oligosaccharides with high sulfate fraction, 2-8 units saccharide size and suitable molecular structure are able to achieve potent heparin sulfate-like compounds.
8.Analysis of consistency between clinical and pathological TNM staging for gastric cancer

Objective To explore the consistency between preoperative clinical TNM stage and postoperative pathological TNM stage of gastric cancer and look for the underlying reason.Methods Sixty-four patients of gastric cancer with complete clinical data from December 2010 to December 2011 in Gansu Provincial Hospital were enrolled into this study.All cases were performed postoperative pathological TNM staging according to the TNM stage criteria of gastric cancer from JGCA(the 14th edition,2010),and the results were compared with preoperative TNM staging,then we studied the consistency of T staging,N staging and M staging using SPSS 18.0.Results The Kappa value from T staging of preoperative and postoperative was 0.729,showing a good consistency.The N stage' s consistency was lower,Kappa value being only 0.216.For M staging,Kappa value 0.743,having a good consistency.Conclusions According to the General Rules for Gastric Cancer Treatment,a accurate TNM staging and individual treatment plan can be made for gastric cancer patients in clinic.But the overestimate and underestimate still existed,especially in N staging.Therefore,making a accurate judgment for N staging according to all kinds of preoperative clinical data and images is the key to improvement of preoperative TNM staging accuracy and treatment.
9.Microsurgical Management of Pigmented Villonodular Synovitis of the Knee

Objective To investigate the value of microsurgical management for the diagnosis and treatment of pigmented villonodular synovitis(PVNS)of the knee.Methods Thirty-eight patients with PVNS of the knee that were confirmed by arthroscopy were treated in our unit between January 2002 and April 2006.Among the patients,5 were referred from other hospitals because of postoperative recurrence.Villusectomy was performed in each of the patients by using arthroscopy.In the cases with larage extra-articular lesions,small auxiliary incision was made for complete resection.All the resected masses were examined pathologically after the surgery,and the patients received intra-anticular drainage,local ice therapy,and functional exercises afterwards.Results Unilateral surgery was performed on this group of patients.Under an arthroscope,the synovium showed characteristic yellow-brown color.Patholgocial examination confirmed PVNS in all the cases,including 20 cases of local PVNS and 18 diffused lesions.Open surgery was performed on 6 patients owing to large lesion at the popliteal fossa.According to the International Knee Documentation Committee(IKDC)Scoring Forms,the mean IKDC score increased from 60.4?6.1 to 89.3?7.2 in one year after the operation.The 38 patients were followed up for 1 to 4 years(mean 2.1 years),none of them had recurrence during this period.Two of the patients,who had been treated by open surgery and then were referred to our hospital,could not flex their knees to 90? after the microsurgery.Whereas,all the others achieved normal knee flexion.Conclusions Arthroscopy is reliable for the diagnosis and treatment of PVNS.For large extra-articular lesions,open surgery using small auxiliary incision is necessary for complete resection and avoidance of recurrence.
10.Expression changes of SHIP1 and the possible mechanism in the chronic mylogenous leukemia

Objective To explore the expression changes in SHIP1 gene in patients with chronic granulocytic leukemia (CGL), healthy volunteers and the K562 cells (one of human CGL cell lines), and to explore the effects of BCR/ABL expression in K562 cells on the SHIP1 gene by blocking the BCR/ABL expression in K562 cells with a specific siRNA. Methods The expression levels of SHIP1 mRNA in leukocytes of the patients with CGL, healthy controls and K562 cells were assessed with RQ-PCR. In addition, K562 cells were divided into three groups: control group (untreated), non-specific interference group (treated with non-specific siRNA), and specific interference group (treated with specific siRNA to block BCR/ABL gene). RQ-PCR and Western blotting were employed to examine the expression level of mRNA and protein (P210 and P145) of BCR/ABL and SHIP1 genes. Results The mRNA expression of SHIP1 in CGL patients and K562 cells was significantly lower than that of the healthy controls. The expression of BCR/ABL gene was down-regulated, while SHIP1 gene up-regulated, in the specific interference group. No statistical significant difference was found between the non-specific interference group and the control group. Conclusions The mRNA expression of SHIP1 is lower in patients with CGL than in the healthy control. The specific siRNA can block the expression of BCR-ABL and further suppress the expression of SHIP1.
