[ ABSTRACT] AIM:To explore the effects of romidepsin ( FK228) , a novel histone deacetylase inhibitor, on the effector and regulatory T cells in vitro.METHODS:As the reactive cells, lymphocytes, CD4 +T cells and CD8 +T cells were labelled with CFSE, and stimulated with anti-CD3 and anti-CD28 mAbs in the presence and absence of different levels of romidepsin (experimental group and positive control group), or PBS (placebo group). After 72 h, the proliferation of the cells was detected in different groups. The lymphocytes were stimulated with anti-CD3 and anti-CD28 mAbs in the pres-ence and absence of different levels of romidepsin ( experimental group and positive control group ) , or PBS ( placebo group) .After 72 h, the percentage of CD4+Foxp3 +T cells and the levels of related cytokines were detected in different groups.RESULTS:The proliferation of CFSE-labelled lymphocytes, CD4 +T cells and CD8+T cells triggered by anti-CD3 and anti-CD28 mAbs all were inhibited when cultured with romidepsin at concentrations of 1 μmol/L, 3 μmol/L and 5 μmol/L in a dose-dependent manner (P<0.05).Compared with placebo group, in the presence of anti-CD3 and anti-CD28 mAbs, 1 μmol/L romidepsin did not increase the percentage of CD4 +Foxp3 +T cells ( P>0.05 ) .When cultured with romidepsin at concentrations of 3 μmol/L and 5 μmol/L, the percentage of CD4 + Foxp3 +T cells was enhanced markedly ( P<0.05) .The levels of IL-10 and TNF-αin the supernatant were markedly increased in positive control group
and 3 experimental groups ( P<0.05 ) , and the levels of cytokines in different experimental groups were gradually de-creased with the elevation of FK228 concentration (P<0.05).The level of TGF-βwas slightly increased in positive con-trol group with no significant difference compared with placebo group (P>0.05).With the increase in the concentration of FK228 in different experimental groups, the TGF-βlevel was increased in a dose-dependent manner and there were signifi-cant differences in the 3 experimental groups.Meanwhile, significant differences existed between experimental groups and placebo group and between experimental groups and positive control group (P<0.05).CONCLUSION:Romidepsin in-hibits the proliferation of CD4 +and CD8 +effector T cells and increases the percentage of CD4 +Foxp3 +regulatory T cells. It may be related to the increased level of TGF-β, but independent of IL-10.

Xiaojun MA ; Min LIU ; Derong XU
China Biotechnology 2006;0(05):-
Taxoid 14?-hydroxylase directs a side-route of taxol pathway that leading to a large number of 14?-hydroxy taxoid side products. A RNAi vector was used to suppress the expression of taxoid 14?-hydroxylase (14OH) gene in Taxus?media suspension cells. The dsRNA expression unit was introduced into Taxus?media cells via Agrobacterium tumefaciens-mediated transformation approach. PCR-Southern blot analysis of transgenic cells showed that the dsRNA unit was integrated into the genome of Taxus?media. The levels of transcripts for 14OH gene in transformed and the control cell lines were determined by using RT-PCR method. In addition, the yield of taxoids were analyzed by HPLC. The transcription of 14OH gene was suppressed and the total amount of three C-14 oxygenated taxoids (yunnanxane, sinenxan A,sinenxan C) in Taxus?media transgenic cell lines reduced significantly as compared to those in the non-transgenic control. Inhibition of the 14OH gene via RNAi might provide a new way to enhance the production of Taxol in Taxus cells.

Clinical Medicine of China 2011;27(2):213-215
Objective To assess the effect of Ambroxol combined with Naloxone on respiratory insufficiency on premature infants. Methods A total of 168 premature infants treated from 2006 to 2009 in our hospital were divided into two groups randomly. Additionally to the conventional treatment, Ambroxol (5 mg pertime) combined with Naloxone (0.1mg pertime) intravenously injected 2 to 3 times one day for 3 to 5 days in the treatment group(86 cases). The control group (82 cases) was treated by routine Aminophylline injection,3- 5 mg/kg pertime, 2 to 3 times one day. Respiratory rates, regulately or not, breathholding, cyanosis and apnea, mechanically ventilated time, time of oxygen inhaling, time of hospital stay and the overall response rate were measured. Results The overall response rate was 80% (69/86) in the treatment group and 56% (46/82)in the control group, with significant difference between the two groups ( P < 0. 05 ). Mechanically ventilated time, time of oxygen inhaling and hospital stay in the treatment group was (110.5±45.1) hrs, (169.3±58.6) hrs and ( 12±3) days, which were all significantly lower than that in the control group ( 181.1±81.2) hrs, (236.6±87.8)hrs and (16±5)days,respectively(P<0.01 or 0.05). Conclusion Ambroxol and Naloxobe additional to conventional treatment could improve the respiratory function in premature infants with respiratory insufficiency safely with less side effects. Moreover, breathing machine could be removed to some extent, which made it more applicable in primary hospitals.
