1.Human tumor-related protein hyrdC: prokaryotic expression,antibody preparation and its preliminary detection in gastric cancer

Objective:To prepare pure prokaryotically expressed protein of hyrdC and the monoclonal antibody against hyrdC,and to detect the hyrdC protein in gastric cancer with the prepared monoclonal antibody.Methods: The target DNA sequence of hyrdC was obtained by RT-PCR and subsequently inserted into prokaryotic expression vector pET32a.The TRX-hyrdC fusion protein was purified and was used to immunize 6-week-old BALB/c female mice to prepare monoclonal antibodies.The supernants of hybridoma cells were screened by enzyme-linked immunoabsorbent assay(ELISA) and identified by Western blot.The prepared monoclonal antibody was used to detect the expression of hyrdC protein in gastric cancer and adjacent normal tissues by immunohistochemical method(SP).Results: The sequence of hyrdC ORF was conformed to the reported in GenBank.Two lines of hybrids secreting monoclonal antibodies against hyrdC were established,both belonging to IgG2a subtype.Western blot showed that the prepared monoclonal antibodies specifically combined with human hyrdC protein.Immunohistochemistry(SP) results showed that the expression of hyrdC protein in gastric cancer was significantly higher than that in normal gastric mucous membrane(P

3.Imaging diagnosis of solid-pseudopapillary tumor of the pancreas: A report of 3 cases and literature review

Objective: To investigate the CT and MRI findings of solid-pseudopapillary tumors of the pancreas(SPTP),and to improve the diagnosis of the disease.Methods: Three female SPTP patients received CT examination,1 of them underwent MRI in addition,and all the cases were surgically and pathologically confirmed later.Results: CT images revealed huge solid-cystic mass in the pancreas of the 3 patients,1 with calcification in the mass and all without dilation of the bile duct.MRI exhibited mixed signal intensity on T1WI and T2WI.Both CT and MRI showed obvious enhancement of the cystic wall and the solid part of the mass.Conclusion: SPTP has its characteristic imaging manifestations,which may contribute a lot to the diagnosis of the disease.
4.Clinical Observation of Cinobufacini Capsules Combined with Paclitaxel and Cisplatin in the Treatment of Middle and Advanced Cervical Cancer

OBJECTIVE:To observe therapeutic efficacy and safety of cinobufacini capsules combined with paclitaxel and cisplatin in the treatment of middle and advanced cervical cancer.METHODS:A total of 92 patients with middle and advanced cervical cancer were randomly divided into observation group (46 cases) and control group (46 cases).Both groups received pelvic intensity-modulated radiotherapy (IMRT)+interstitial brachytherapy.Control group was additionally given Paclitaxel injection 135 mg/m2,d1+Cisplatin injection 75 mg/m2,d1,21 d as a treatment course,and received chemotherapy for 2 cycles since the fnrst day of radiotherapy.Observation group was additionally given Cinobufacini capsules 0.5 g orally since the first day of radiotherapy,3 times a day,until the end of radiotherapy.Clinical efficacies as well as platelet count,KPS score,body weight,pain relief and the recovery of platelet abnormality were observed in 2 groups,and the occurrence of toxic reaction was recorded.RESULTS:The complete remission rate,the rate of platelet count abnormality recovery as well as remission rate and total remission rate of pain after 3 weeks of treatment in observation group were significantly higher than control group,with statistical significance (P<0.05).There was no statistical significance in the total response rate and remission rate after 4,5 weeks of treatment between 2 groups (P>0.05).After treatment,platelet count of 2 groups were significantly lower than before,and the observation group was significantly lower than the control group;KPS score of 2 groups and body weight of observation group were significantly higher than before treatment;body weight of control group was significantly lower than before,and the observation group was significantly higher than the control group,with statistical significance (P<0.05).The incidence of grade Ⅲ-Ⅳ neutropenia,nausea and vomiting,grade Ⅰ-Ⅱ diarrhea in observation group were significantly lower than control group,with statistical significance (P<0.05).CONCLUSIONS:Based on conventional treatment,cinobufacini capsules combined with paclitaxel and cisplatin show significantly therapeutic efficacy for middle and advanced cervical cancer,improve blood hypercoagulation and survival quality,relieve pain and reduce the occurrence of toxic reaction.
5.Laparoscopy-assisted anterior resection with prolapsing technique for low rectal cancer

Nineteen patients with low rectal cancer underwent laparoscopy-assisted anterior resection with prolapsing technique for sphincter preservation.The clinical data were retrospectively reviewed.Fecal continence was preserved successfully in all patients.There was no perioperative mortality,only two cases with anastomotic leak and one case with local recurrences.Laparoscopy-assisted anterior resection with a prolapsing technique for low rectal cancer is safe procedure and can be an alternative sphincter-preserving operation.
6.An analysis of the cancer-spectrum of the elderly in Chongqing from 1991 to 1998

Objective To study the urban elder people's cancer-spectrum in Chongqing from 1991 to 1998 in order to obtain the first-hand data and references for the prevention and the treatment of carcinoma in the seniles. Methods The data of population and death causes were collected with cluster sampling method in the communities of business, culture and industry in Chongqing. Their mortality rate, disease-specific mortality rate and the sequence of the death causes were analyzed. Results Their first death causes was cancer (mortality rate 811.95/105 persons, accounted for 21.25% of all death causes). The mortality rate of five cancers was as follows: lung cancer (282.45/105), esophagus cancer(119.47/105), liver cancer(108.41/105), stomach cancer (73.75/105) and intestine cancer (55.31/105). The prevalence of different cancer between male and female at different communiteis was different. Conclusions It is very important to strengthen the protection of the environment and advocate the healthy life mode in order to lower the mortality rate by cancer in urban seniles.
7.Comparison on effects of bipolar femoral head replacement and total hip rep lacement in treatment of femoral neck fractures in the elderly

Objective To compare the effects of bipolar femor al head replacement (BFHR) and total hip replacement (THR) in the treatment of femoral neck fractures (FNF) in the elderly. Methods Between August 1993 and December 2001, BFHR or THR were performed in 48 old patients (at age of 60-85 years, mean 70.8 years) with subcapital and transcervical types of FNF. Of all, 18 cases were tre ated with BFHR (Group BFHR) and 30 with THR (Group THR). Results OF all, two patients died (not related to operation), one received no f ollow up and 45 were followed up for average 5 years and 2 months with no wound infection or hip dislocation. Occurrence rate of hip and thigh pains in the Grou p THR was 3% (1/29), lower than that in the Group BFHR (4/16). Conclus ions BFHR is only fit for senile cases with poor physical con dition. However, THR can be used for most old patients with FNF.
8.Evaluation of three dimensional reconstruction CT angiography in interventional treatment of epigastric tumors

Objective To assess the usefulness of the MSCTA with 3D reconstruction to epigastric tumors in interventional treatment.Methods MSCTA were performed in 27 cases of epigastric tumors before and after interventional treatment.3D reconstruction were taken for blood vessels with technique of MIP(maximum intensity projection)and VRT(volume rendering technique).Results 42 times MSCTA and 3D reconstruction were performed in 27 cases of epigastric tumors.We discovered 5 feeding arteries with mutation in abdominal cavity,7 collateral branches formed after embolization and 2 abnormal courser of celiac artery.The display of vasculative with MSCTA was better than that of DSA.Conclusion 3D-MSCTA for epigastric tumors not only can show directly and clearly the stereo-anotomic images of the tumor feeding vessels but also provide guidance for planning the treatment.

10.Construction of eukarytic expression vector of enhanced green fluorescence protein driven by telomerase catalytic subunit promoter and its expression targeted in human lung cancer cells

Objective:To construct an eukaryotic expression vector of enhanced green fluorescence protein(EGFP) gene driven by telomerase catalytic subunit(hTERT) gene promoter and observe the specific expression of EGFP in lung cancer cell lines.Methods:The 1100bp promoter fragment was obtained by enzyme digestion from a recombinant plasmid of pGL3-hTERTp containing the hTERT promoter.The hTERT promoter was then subcloned into the upstream of the report gene EGFP of pEGFP-1 without promoter.The expression vector pEGFP-hTERTp was successfully constructed.The vector pEGFP-N1 containing cytomegalovirus(CMV) promoter was used as a positive control.The vector pEGFP-1 without promoter was used as a negative control.The vectors were transfected into human lung cancer cell lines 95D,NCI-H446,A2,A549,LTEP-a-2,YTMLC and normal MRC-5 through lipofectamine respectively.EGFP expression was detected under the fluorescence microscope.Results:pEGFP-hTERTp was confirmed by enzyme digestion with correct result.That the EGFP expression was detected in all of eight lung cancer cells transfected with pEGFP-hTERTp,but not in MRC cells.By contrast,high intensity EGFP expression was observed in both lung tumor cells and normal cells,which were transfected with pEGFP-N1.Conclusion:The EGFP controlled by hTERT promoter can be expressed specifically in lung cancer cell lines.hTERT promoter may be used as an excellent regulation element in tumor-targeting gene therapy.
