1.The effect of transmembrane tumor necrosis factor-alpha on human cervical carcinoma HeLa cells

Objective To study the effect of transmembrane tumor necrosis factor-alpha (TM-TNF-αt) on human cervical carcinoma HeLa cells,and explore the correlations with silencer of death domain (SODD).Methods The expression of proliferating cell nuclear antigen (PCNA) was detected by immunocytochemistry.The expression of SODD in human cervical carcinoma HeLa cells undisposed and after disposed with TM-TNF-α was detected by reverse transcription-polymerase chain reaction,and the influence of TM-TNF-α on human cervical carcinoma HeLa cells was analyzed.Results The positive rate of PCNA in undisposed human cervical carcinoma HeLa cells was 80.3% (155/193),in human cervical carcinoma HeLa cells disposed with TM-TNF-α was 46.7% (85/182),there was statistical difference (P < 0.05).The cycle index of polymerase chain reaction in human cervical carcinoma HeLa cells was 28 times,the amplification product was disposed by 2% agarose gel electrophoresis,the gray scale disposed by TM-TNF-α and undisposed human cervical carcinoma HeLa cells were 1.377 ± 0.170 and 0.815 ± 0.040,there was statistical difference (P < 0.05).Conclusion TM-TNF-α has obvious cytotoxic effect on human cervical carcinoma HeLa cells in vitro which may due to its up-regulating the expression of SODD.
2.Correlation of neutrophils to lymphocyte ratio and platelet to lymphocyte ratio with prognosis of hepatocellular carcinoma patients treated with transarterial chemoembo-lization

Objective:To identify the correlation between neutrophil to lymphocyte ratio (NLR), platelet to lymphocyte ratio (PLR), and hepatocellular carcinoma (HCC) patients who underwent transarterial chemoembolization (TACE). Methods: We retrospectively re-viewed 216 patients who were diagnosed with HCC and treated with TACE between January 2007 and June 2015 at the Department of Hepatobiliary Oncology, Cancer Center, Sun Yat-sen University. The patients were stratified into two groups using NLR and PLR cutoff values. NLR and PLR were analyzed and compared through the area under receiver operating characteristic curves at different time points. Univariate and multivariate analyses were applied to evaluate the correlation between NLR, PLR, and HCC patients who were treated with TACE. Results:The median follow-up period was 431.11 d. The estimated 1-, 2-, and 3-year overall survival (OS) rates were 61.3%, 44.2%, and 40.5%for the entire study cohort, respectively. The median OS was 410.5 d. The estimated 1-, 2-, and 3-year OS rates for NLR<1.77 group and NLR≥1.77 group were 81.6%, 63.0%, and 45.7%;and 43.1%, 27.0%, and 19.3%, respectively. The dif-ference was significant (P<0.001). The estimated 1-, 2-, and 3-year OS rates for PLR<94.62 group and PLR≥94.62 group were 62.7%, 47.0%, and 37.0%;and 46.8%, 29.0%, and 18.5%, respectively. The difference was significant (P=0.002). In a multivariate analysis, NLR≥1.77 was a risk factor associated with poor outcome for patients with HCC who received TACE therapy. Conclusion:NLR level be-fore TACE is an indicator of systemic inflammation and is a risk factor associated with the prognosis of HCC patients who were treated with TACE.

4.Efficacy of Facial Muscle Exercise on Peripheral Facial Paralysis: 45 Cases Report

Objective To investigate the efficacy of facial muscle exercise on peripheral facial paralysis. Methods 45 patients with peripheral facial paralysis were treated with acupuncture, physical therapy and facial muscle exercise. Results The effective rate was 100% and 44.4% were cured after 2 courses of treatment. Conclusion The facial muscle exercise is effective on peripheral facial paralysis.
5.Effect of Moxibustion with Warming Needles Combined with Joint Mobilization on Scapulohumeral Periarthritis

Objective To investigate the efficacy and analgesia of moxibustion with warming needles combined with joint mobilizationon scapulohumeral periarthritis. Methods 126 subjects were randomly divided into 2 groups, the observation group received moxibustionwith warming needles combined with joint mobilization; the control group received moxibustion with warming needles only. Short-form ofMcGill pain questionnaire and effect were assessed. Results The effective rate was 98.4% and 95.2% for the 2 groups respectively(P>0.05);but there was significant differences between the 2 groups in the visual analogue scale (VAS), present pain intensity (PPI) and pain rating index(PRI) (P<0.05). Conclusion The moxibustion with warming needles combined with joint mobilization is a good effective therapy forscapulohumeral periarthritis.
6.Influencing factors on thrombin time assay by coagulation specimen insufficient centrifugation

Objective To investigate the influencing factors on thrombin time (TT) assay to ensure the accuracy of the detection results.Methods Totally 3 out-and in-patients from January to July 2015 with normal results of prothrombin time (PT),activated partial clotting enzyme live time (APTT) and fibrinogen (FIB) were selected,whose detection results were not satisfactory.The specimen underwent re-centrifugation,and then Beckman ACL TOP700 automatic coagulation analyzer was used to detect four coagulation indexes.The detection results were compared with those before re-centrifugation,and the differences between the results were analyzed statically.Results The values of PT,APTT and FIB of the three patients before and after re-centrifugation were (10.8,11.5,9.7 s),(29.5,32.7,25.2 s),(2.49,3.12,2.85 g/L) and (10.9,11.3,10.0 s),(30.4,31.5,25.9 s),(2.31,3.28,2.67 g/L) respectively,and the differences between the results were not significant (with t ranging from 0.627 to 1.719 and P>0.05).The values of TT of the three patients before and after re-centrifugation were (66.51,127.3,89.62 s) and (12.2,15.7,13.8 s) respectively,and there were obvious differences between the values (with t ranging from 51.743 to 79.167 and P<0.001).Conclusion Insufficient centrifugation has high influences on the detection result of TT except other coagulation indexes,and re-centrifugation is necessary for the accuracy of TT value to eliminate misdiagnosis.
7.The performance evaluation of emergency immunological parameters in Johnson Vitros 5600 fully automatic biochemical analyzer

Objective:To have the performance verification of Johnson Vitros 5600 fully automatic biochemical analyzer (hereinafter referred to as Vitros 5600 analyzer) emergency immunological parameter is to provide more accurate and more efficient service for the patients in emergency inspection items.Methods: According to the clinical laboratory standards institute (CLSI) evaluation criterion and Johnson validation plan, we verified the precision, accuracy, linear range, reference range, and carry pollution rate detection indexes of Myoglobin, Troponin I (TropI), N-terminal brain natriuretic titanium (NTproBNP) and total beta human chorionic gonadotropin (TβHCG) about the Vitros 5600 analyzer emergency projects evaluated the performance of the analyzer.Results: The batch precision CV of Myoglobin, TropI, NTproBNP and TβHCG tested by Vitros 5600 analyzer is 0.58~2.02%, < 2.5%, and the inter assay CV is 1.43~3.15%, < 5.0%. Accuracy relative bias is -3.52~4.21%. Linearity relationship is good within the scope of testing,r2 is 0.9836~0.9998, and the slope (a) is 0.9865~1.0207. Carry pollution rate is 0.096~0.180%. Reference range verification coincidence rate is R≥95%.Conclusion: The detection index performance of Vitros 5600 analyzer detecting immune emergency projects is good. It can meet the demand of emergency immune inspection work.
8.To Compare the Content of the Volatile Oil from Curcuma Aromatica Salisb. of Different Growth Periods

[Objective] To study the effects of different growth periods on the contents of volatile oil and the curcumol in Curcuma aromatica Salisb. samples,which provided theoretical bases to control the quality of the crude drug of Curcuma aromatica Salisb.[Method]The contents of volatile oil in Curcuma aromatica Salisb. samples were determined by the method of Pharmacopeia,and HPLC method for determination of curcumol in different samples was established. [Result]The contents of volatile oil in different samples lied in the range of 0.820%~0.471%(g/g),and the contents of curcumol in different volatile oil samples were between 0.862%(g/g) and 1.380%(g/g). [Conclusion]Compared with the Curcuma aromatica Salisb. of a shorter growing period,the contents of longer growth period were higher.Different growing time had effects on the accumulating of volatile oil and the curcumol in Curcuma aromatica Salisb.
9.Anterior elbow approach and internal fixation for the treatment of capitulum humeri fractures

Objective To evaluate the clinical result of anterior elbow approach and internal fixation for the treatment of capitulum humeri fractures. Methods From January 1997 to December 2003,11 patients of 9 males and 2 females with capitulum humeri fractures were chosen anterorredial approaches of elbow. The fractures were terated with under direct vision and were fixed with screws. The left arms in 7, right in 4, the age of the group were from 14 to 18 years old, with the average of 15 years. 7 cases of them were treated with close reduction but failed. The operative time was from 3 days to 7 days after injury. They were fresh and close fractures. According to wangyicong's classification system all were TypeⅠ. Results All operations were successful, no nerves and blood vessels injury, the incisions were all healed by first intention. The X-ray films shows that the fractures were anatomic reduction. The healing time was 6 weeks in average and all patients were followed up with an average 3.5 years, no postoperative complications such as:myositis ossificans, post-traumatic arthritis, avascular necrosis of capitulum humeri. According to Cassebanm and the percentage of elbow joints pain was measured with patient visual pain index function score system: 7 caese of excellent, 4 case of good. Conclusion Displace of capitulum-trochlea hemeri does not link with muscle and capsula articularis, blood carries supply. Without avascular necrosis probably occurs at far stage. The close manipulation of capitulum humeri fractures hardly succeeds. The treatment with accurate open reduction associated with strong fixation can decrease the rate of avascular necrosis in the capitulum humeri fractures. The capitulum humeri fractures are suitable for opening reduced direct vision and fixing with screws through anteroeedial approaches of the elbow. The patients were encouraged to proceed for active rehabilitation early such as the elbow flexion-extension. Its short-term clinical outcomes is more excellent.
10.Expression changes of SHIP1 and the possible mechanism in the chronic mylogenous leukemia

Objective To explore the expression changes in SHIP1 gene in patients with chronic granulocytic leukemia (CGL), healthy volunteers and the K562 cells (one of human CGL cell lines), and to explore the effects of BCR/ABL expression in K562 cells on the SHIP1 gene by blocking the BCR/ABL expression in K562 cells with a specific siRNA. Methods The expression levels of SHIP1 mRNA in leukocytes of the patients with CGL, healthy controls and K562 cells were assessed with RQ-PCR. In addition, K562 cells were divided into three groups: control group (untreated), non-specific interference group (treated with non-specific siRNA), and specific interference group (treated with specific siRNA to block BCR/ABL gene). RQ-PCR and Western blotting were employed to examine the expression level of mRNA and protein (P210 and P145) of BCR/ABL and SHIP1 genes. Results The mRNA expression of SHIP1 in CGL patients and K562 cells was significantly lower than that of the healthy controls. The expression of BCR/ABL gene was down-regulated, while SHIP1 gene up-regulated, in the specific interference group. No statistical significant difference was found between the non-specific interference group and the control group. Conclusions The mRNA expression of SHIP1 is lower in patients with CGL than in the healthy control. The specific siRNA can block the expression of BCR-ABL and further suppress the expression of SHIP1.
