1.Effect observation of conical stone needle on foot combined with bian anvil for insomnia.

OBJECTIVETo observe the effect of conical atone needle on foot combined with Nan anvil at Yintang (CV 29) for insomnia.
METHODSSeventy patients were randomly divided into a ibian technique group and an acupuncture group,35 cases in each one. In the Sian technique group, conical stone needle on foot combined with bias anvil at Yintang(GV 29) were used,and the therapy was applied at Yongquan(KI 1), Taichong(LR 3) and Gongsun(SP 4), etc. In the acupuncture group, common acupuncture was implemented at Sanyinjiao(SP 6), Shenmen (HT 7) and Sishencong(EX-HN 1), etc. In the two groups, treatment was given for 2 weeks,once a day. There were two days to have a rest between 5-time treatment. The change of Pittsburgh sleep quality index(PSQI) before and after treatment was recorded and the clinical effect was observed after 10-time treatment and in two weeks aftet treatment.
RESULTSAfter treatment,the total scores of PSQI were declined significantly compared with those before treatment in the two groups(both P<0. 01). The total PSQI scores after treatment,the curative and effective rates and total effective rates when followed up bad no statistical significance(all P>0. 05).
CONCLUSIONConical stone needle on foot combined with Sian anvil at Yintang(GV 29) treating insomnia is safe and effective,and this treatment acquires similar efficacy with less suffering.

2.Construction of prokaryotic expression vector of Id3 and expression of fusion protein in E. coli

Objective: To express and purify the inhibitor of differentiation(Id3) in E.coli.Methods: The DNA fragment in the coding region of the human Id3 gene was amplified by PCR and cloned into the pGEM-T Easy vector for sequencing.The Id3 cDNA fragment was then subcloned into the prokaryotic expression vector(pET-32a)(+) and transformed into E.coli BL 21(DE3).The expression of the histidine-tagged(His-Tag) fusion protein was induced with isopropy-?-D-thiogalactoside(IPTG),confirmed by Western blot and purified by the immobilized Ni2+ absorption chromatographic column.Results: The prokaryotic expression vector of Id3 was successfully constructed.Western blot confirmed the expression of the His-Tag fusion protein in E.coli BL 21(DE3) and that of the Id3 fusion protein with the relative molecular mass size of 33 000 after purified by the affinity chromatographic column. Conclusion: Recombinant Id3 can be expressed in E.coli BL 21(DE3) and the obtained fusion protein can be purified by the Ni-affinity chromatography column.
3.Curative effects of hepatocyte growth factor in treatment of infant hepatitis syndrome caused by cytomega lovirus with ganciclovir

Objective To compare the curative effect of hepatocyte growth factor(HGF) in treatment of infant hepatitis syndrome (IHS) caused by cytomegalovirus with ganciclovir.Methods Sixty-eight cases with infant hepatitis syndrome caused by cytomegalovirus from neonatology department of our hospital from September 2010 to September 2014 were randomly divided into two groups, the observation group (32 cases)took HGF and ganciclovir therapy and the control group (36 cases) only received ganciclovir treatment Both groups were given conventional guard liver therapy.After one month of treatment, liver function, cytomegalovirus DNA content,extinction time of jaundice, treatment efficiency and the adverse reaction rate were evaluated for two groups.Results Liver function of children in observation group improved significantly after treatment compared with the control group, CMV-DNA quantitatively lower than the control group, the extinction time of jaundice was earlier than the latter,and the overall treatment efficiency was higher in observation group (P <0.05);The adverse reaction has no significant difference between two groups (P > 0.05).Conclusion HGF combined with ganciclovir treatment can significantly improve liver function in patients of IHS caused by cytomegalovirus infection, and can collaboratively reduce viral load.Symptoms subside quickly with fewer adverse reactions, which is worthy of clinical promotion.
4.An update of RNA interference in tumor therapy

Tumor is a reciprocity result of various genes and factors,and its incidence and mortality remain high.Such traditional strategies as surgery,radiotherapy and chemotherapy cannot completely restrain and reverse the growth of tumor cells.So,new and more effective therapeutic methods are being expected.This article updates the development of RNA interfering(RANi) technique in the targeting therapy of the tumor gene.
5.Detection and Susceptibility to Antibiotics of Mycoplasma in Genitourinary Tract

OBJECTIVE To investigate the infection status and antibiotic susceptibility of Mycoplasma in our area,so as to instruct the rational use of antibiotics.METHODS Genitourinary secretions were collected with swabs.They were cultured with the diagnostic kit of Mycoplasma(Biomerieux Company).Meanwhile the susceptibility of Mycoplasma against 9 antimicrobial agents was tested with the same kit.According to the instrnction of the kit,the results were read.The data were statistically analyzed.RESULTS A total of 382 samples were collected,and the positive rate was 44.8%.which were composed by Uu 69.6%,Mh2.9%,and Uu+Mh infection 8.8% and low titer Uu+Mh infection 18.7%.The result of drug sensitivity showed that sensitivity of Mycoplasma to pristinamycin PRI,86.5% was the highest,then was to JOS(84.8%)and CIP(26.3%).CONCLUSIONS Ureaplasma urealyticum is the major cause of infection in genitourinary system.It shows serious resistance to commonly used antibiotics.Sensitive antibiotics should be selected based on the results of bacterial culture and drug sensitivity tests so as to raise the clinical curative effects.

Inhibitors of differentiation(Id) are negative regulators of basic helix-loop-helix(bHLH) type transcription factor.There are four related members of the Id family called Id-1,Id-2,Id-3 and(Id-4) in mammalian cells.Since the identification of Id proteins in 1990,the roles of Id in cell proliferation,differentiation,development,maturation,apoptosis and tumorigenesis have been widely investigated.This article focuses on the role of Id in tumorigenesis.
7.CD40-ligand down-regulates inhibitor of differentiation 3 expression in WEHI-231 B lymphoma cells

Objective: To study mouse Id3(mId3) protein expression in WEHI-231 B lymphoma cells after CD40 ligand(CD40L) treatment. Methods: The mId3 cDNA was amplified from total RNA of WEHI-231 cells by RT-PCR and introduced into the p3FLAG-CMV-10 vector.The 3FLAG-Id3 fusion gene was amplified from p3FLAG-Id3-CMV-10 vector and subcloned into retroviral vector pMX-CITE/IRES-EGFP.Recombinant retrovirus was produced by transient transfection of the Phoenix-ecotropic packaging cell line with pMX-3FLAG-Id3-EGFP vector using FuGene-6 transfection reagent.(WEHI)-231 cells were transduced with recombinant retrovirus.EGFP expression was monitored by flow cytometry.Two cell lines stably expressing 3FLAG-Id3 fusion protein were generated. Results: The proportion of EGFP-positive cells in the population transduced with pMX-3FLAG-EGFP remained constant up to 2 days after transduction,whereas the proportion in the population of pMX-3FLAG-Id3-EGFP-transduced cells decreased with time.After coculturing with NIH3T3 cells or medium alone,the proportion of EGFP-positive cells in pMX-3FLAG-Id3-EGFP-transduced WEHI-231 cells decreased with time,whereas after coculturing with CD40L-NIH3T3 cells,the proportion of EGFP-positive cells in the transduced cells remained constant up to 2 days,suggesting that CD40L could rescue WEHI-231 from Id3 overexpressioninduced growth arrest.After coculturing with CD40L/NIH3T3 or NIH3T3 cells,3FLAG-Id3 fusion protein stably expressing WEHI-231 cells were harvested,lysed,and immunoblotted by using anti-FLAG antibody.The results showed that treatment via CD40 ligation induced significant downregulation of Id3 expression in pMX3FLAG-Id3-EGFP tansduced WEHI 231 cells. Conclusion:CD40 ligation pvevents the Id3 induced cell growth avvest by the dousn-regulation of Id3 expression.
8.A Simple Method for Dispelling Motion Artifacts in MRI Examination of Brain

Objective To explore a method that restrain and dispel motion artifacts of MRI examination for better image quality and higher working efficiency.Methods The sponge package of CT tube was used to design and make sponge cushions in types of 2 cm?7.5 cm?22 cm,3 cm?7.5 cm?22 cm and 4 cm?7.5 cm?22 cm,which were used to MRI examination by quadrature coil of brain.Results The method of"adding cushion" was adopted to examine brain of MRI in 2 638 cases.Among them,2 633 cases were successfully scanned,which accounts for 99.81%;5 cases failed,accounts for 0.19%.For the failed cases,4 cases were successfully scanned in the second time by using medical bandage in cooperation,while 1 case failed.Conclusion The method of"adding cushion" in routine MRI examination of brain can restrain and dispel motion artifacts,improving image quality and work efficiency.It is an easy,reliable,ideal method for restraining and dispelling motion artifacts in brain MRI examination.
9.Progress in The Relation Between Inhibitors of Differentiation and Cancer

The ubiquitously expressed family of Id(inhibitor of differentiation) helix-loop-helix(HLH) proteins function as dominant negative regulators of basic HLH (bHLH) transcriptional regulators. In eukaryotic organisms, Id proteins have been implicated in development, cell differentiation, proliferation, angiogenesis, invasion and migration. Recent studies revealed that Id proteins are not only significantly correlated with cancer progression and prognosis, but also exploited as a tumor therapeutic target. The roles of Id proteins in tumorigenesis were reviewed and the opportunities of Id proteins in cancer targeted therapy were discussed.
10.Prosthetic infection after tension-free hernioplasty in 12 cases

Objective To evaluate the prevention and treatment of prosthetic infections after tension free hernioplasty. Methods The clinical date of 12 prosthetic infections admitted from May 2005 to May 2008 were analyzed retrospectively. Patch was inserted under prefascial retromuscular site in 8 cases, between fascial edges in 2, Mesh plug and patch Lichentstein in one each. As for the material used there were flat knitted polypropylene in 7 cases, polytef in 1 case and composite prosthesis in 4 cases. Treatments were based on prothetic materials and levels infected. Infectious patchs were removed completely in 8 cases and partially removed in 2 cases, Open dress change with patchs left in situ in 2 cases. 3 cases in 10 cases of removed patchs were given immediate suturation with closed drainage. Results Infection was cured in all cases without mortality. There was no hemorrhage and injury of bladder during the procedures. No retention of urine, pain and other complications during postoperation. Change of dressing was made from 7 days to 6 months( median 2 weeks). Selected antibiotic was used from 3 to 7 days (average 4.5 days). All patients were followed up from 18 to 78 months (average 38.5 months) without hernia recurrence and infection. Conclusion It was most important to prevent infections after tension-free hernioplasty. Management should be individualized according to different type of patch used and the surgical procedure in hernia repair.
