Acta Anatomica Sinica 2002;0(05):-
Objective To research pacemaker cells outside conductive system of rat and mouse heart. Methods Paraffin and frozen serial sections (stained with HE, Masson, cholinesterase technique), and ultrastructural sections, from atrial tissue of 10 rats and 15 mice were observed by light and electron microscopy to find latent pacemaker-like cells. Results Some round or elliptical cells were scattered and irregularly detected in right and left atrial tissue of rat and mouse. Their nuclei were larger, plasma was clearer and cellular organelles were fewer than those of working myocyte. These cells were similar to pacemaker cells in sinoatrial node. Conclusion There are some pacemaker-like cells in right and left atrial tissue of rat and mouse. The finding can provide a morphological basis for ectopic beat of atrium.

Chinese Pharmacological Bulletin 2014;(4):550-553,554
Aim To investigate the effect of rapamycin on proliferation, migration, and invasion of squamous cell carcinoma A431 cells in vitro. Methods Human squamous cell carcinoma A431 cells were cultured in vitro. MTT assay was used to investigate proliferation of A431 cells treated by rapamycin at different concentra-tions of (5,10,20 nmol·L-1). Wound healing and transwell assay were performed to detect migration and invasion of A431 cells treated by rapamycin, respec-tively. Reverse transcription PCR ( RT-PCR ) and Western blot were used to determine the expression of the osteopontin ( OPN ) in A431 cells at mRNA level and protein level, respectively. Results Rapamycin significantly increased the inhibitory rates of prolifera-tion and inhibited the migration and invasion of A431 cells in vitro. Furthermore, rapamycin treatment re-duced the expression of OPN in A431 cells. Conclu-sions Rapamycin can inhibit the migration and inva-sion of A431 . The intrinsic mechanism of rapamycin might be related to the down-regulation of the OPN ex-pression.

Chinese Journal of Radiology 2016;(2):110-113
Objective To study the imaging findings, clinical and pathological characteristics of langerhans cell histiocytosis(LCH)in limbs of children, and to improve the imaging diagnostic accuracy of LCH . Methods Imaging findings and clinical data of 12 patients with limbs LCH were analyzed retrospectively. X-ray examination was performed in 10 cases, while 9 cases underwent CT and 7 cases underwent MRI examination. All imaging findings were analyzed. Results Seven lesions were solitary, including 4 lesions in the tibia, 2 in the radius and 1in the ulna. Five cases had limbs lesion in multiple sites, which included 12 bone lesions in total, with 6 lesions in the femur, 3 in the humerus (1 case involved the epiphysis), 2 in the tibia and 1in the radius. The X-ray or CT images of the lesions showed osteolytic destruction and swelling of the bone, which was commonly associated with the reaction of periosteum. Eleven cases were with peripheral soft tissue swelling, and one case had soft tissue mass. The lesions showed low signal on T1WI and high signal or mixed signal on T2WI. Six cases were diagnosed as LCH at the first time and the other 6 cases were diagnosed as infectious lesions. Twelve cases were regularly followed up after the treatment, and the prognosis of the patients with single lesion was better. Conclusions LCH of limbs usually involves diaphyseal and metaphyseal bone. The imaging features include osteolytic destruction and swelling of the bone, with peripheral soft tissue swelling or mass of the soft tissue. Although LCH of limbs has some specific imaging features, it is difficult to differentiate it from malignant bone tumors. Multi-imaging modalities such as radiography, CT and MRI imaging may suggest the diagnosis and provide important information for evaluating prognosis.

Journal of Chinese Physician 2015;17(5):671-674
Objective To investigate the role of p38 MAPK in the trypsin-induced injury in human esophageal epithelial cells. Methods Primary cultured human esophageal epithelial cells were stimulated with trypsin (20,40,and 80 μg/ml) for4 hours, phosphorylation of p38 MAPK was evaluated by Western blotting.Primary cultured human esophageal epithelial cells were stimulated with trypsin (40 μg/ml) and treated with p38 MAPK inhibitor (SB203580,1 and 10 μmol/L) simultaneously.Four hours later, the cells were collected for analysis. Results Western blotting results revealed that stimulation with trypsin enhanced phosphorylation of p38 MAPK,indicating that trypsin activated p38 MAPK in esophageal epithelial cells.SB203580 treatment suppressed trypsin-induced expression of pro-inflammatory cytokines including interleukin-8 (IL-8),cyclooxygenase 2 (COX2),and tumor necrosis factor alpha (TNFcα).Finally,SB203580 treatment suppressed trypsin-induced upregulation of protein expression of inducible nitric oxide synthase (iNOS),and subsequently reduced nitric oxide (NO) levels. Conclusions The regulation of p38 MAPK was involved in the trypsin-induced injury in esophageal epithelial cells.

The free radical scavenging effect of flaxseed was screened by HPLC-DPPH ( 2 , 2-diphenyl-1-picrylhydrazyl-high performance liquid chromatography assay ) and colorimetric DPPH methods. To test the effectiveness of the approach, three Lignans ( secoisolariciresinol diglucoside ( SDG ) , secoisolariciresinol ( SECO) and enterodiol( ED) ) with antioxidative properties were investigated both in monomer and mixture. HPLC conditions were optimized using following methods: Waters XBridge C18 was used as stationary phase, acetonil/H2 O was used as mobile phase and detective wavelength was set at 280 nm. Antioxidant activity of standards was investigated by reaction with or without DPPH radical for 20 min as sample and control, respectively. Both of them were analyzed by high performance liquid chromatography. According to the changes of amount of sample and control, the antioxidant activities of standards were calculated as following order:SDG>SECO>ED. Based on above DPPH-HPLC assay and UPLC-Q-TOF-MS, antioxidants extracted from flaxseed were separated, identified and screened. The radical scavenging activities were in the following order:SDG isomer (5)>SDG (4)>7α-[(β-D-glupyranosyl) oxy]-1-methoxyisolariciresinol (1)>(6R,7R, 8S)-1-methoxyisolariciresinol (2)>herbacetindiglucoside (3). It indicated that the HPLC-DPPH assay could be successfully used for the antioxidant activity screening of complex flaxseed extract.
