1.Research progress on the relationship between FOXM1 and neo-plasm metabolism
Chinese Journal of Clinical Oncology 2015;42(22):1108-1111
FOXM1 is a member of the FOX family of transcription factors and has been confirmed to participate in cell prolifera-tion and cell cycle progress. FOXM1 has been reported to play an important role in more than 20 types of human neoplasms, and its up-regulation promotes tumor cell proliferation, invasion, metastasis, chemotherapy drug resistance, and resistance to apoptosis. In recent years, various studies have shown that FOXM1 also plays an important role in tumor metabolism. The metabolism of cancer cells deter-mines the growth and survival of cancer, and metabolic alteration is a key feature of tumor cells. The connection between metabolic ab-normalities and tumorigenesis has been reported. The O-linkedβ-N-acetylglucosamine transferase regulates the expression of FOXM1, which helps tumor cells survive without damage caused by oxidative stress. This review aims to summarize the known relationships of FOXM1 and tumor cell metabolism, as well as its role in the potential pathways of tumor metabolism.

Objective To explore the anti-infection mechanism of carboxyamidotriazole (CAI) through studying the effects of CAI on the proliferation, apoptosis and degranulation of RBL-2H3 mass cells. Methods Compound 48/80 (C48/80) was used to induce the model of activation and degranulation in RBL-2H3 cells. The morphological change of cell degranulation was observed by neutral red staining. The release levels of histamine and β-hexosaminidase were measured by ELISA method and chromogenic assay, respectively. The cell activity was determined by CCK-8 method. And cell apoptosis was detected by Hoechst 33342 fluorescent staining. Results Compared with the control group, 10, 20, 40 μmol/L CAI inhibited C48/80-induced degranulation of RBL-2H3 cells in different degrees.CAI (20, 40 μmol/L) reduced the histamine release (P<0.01), and CAI (40 μmol/L) decreased the β-hexosaminidase release (P<0.01).In addition, the viability and apoptosis of RBL-2H3 cells were not affected at the concentrations of CAI used. Conclusions CAI can effectively inhibit the activation and degranulation of RBL-2H3 mast cells, and this effect is not through cytotoxicity. The anti-infection effect of CAI may partially due to the down-regulation of mast cell activity.

BACKGROUND:Donor selection for high-risk acute leukemia is still controversial.OBJECTIVE:To compare the therapeutic efficacy of the unrelated donor peripheral blood and matched sibling allogeneic hematopoietic stem cell transplantation for high-risk acute leukemia.METHODS:Total 65 patients with high-risk acute leukemia treated during January 2008 to January 2016 were included,in which 30 patients chose the unrelated donor peripheral blood stem cell transplantation (UD), and other 35 chose the matched sibling allogeneic hematopoietic stem cell transplantation (MS) according to the wishes of patients and their own situation. After treatment, the chi-square test, Kaplan-Meier survival analysis method, and other methods were used to compare the implanted and hematopoietic reconstitution, the occurrence of graft-versus-host disease, relapse mortality and long-term survival between the two groups.RESULTS AND CONCLUSION:The implantation rate, platelet hematopoietic reconstitution time, the incidence of acute and chronic graft-versus-host disease, and its type exhibited no significant differences between the two groups (P > 0.05).The relapse rate, total death rate, and transplant-related mortality rates were 10.0%, 50.0%, and 40.0% in the UD group and 20.0%, 48.6%, and 25.7% in the MS groups, respectively, and the intergroup difference was insignificant (P > 0.05).The expected 2-year cumulative disease-free free survival and overall survival rates were (49.4±9.2)% and (52.6±9.2)% in the UD group and (53.9±8.5)% and (53.9±8.5)% in the MS group, respectively, and the intergroup difference was also insignificant (P > 0.05). Our experimental findings show that unrelated donor peripheral blood stem cell transplantation can be used as an effective alternative in the absence of sibling donors.

Biopartitioning micellar chromatography (BMC) is a potentially high throughput and low cost alternative for in vitro prediction of drug absorption, which can mimic the drug partitioning process in biological systems. In this paper, a data set of 56 compounds representing acidic, basic, neutral and amphoteric drugs from various structure classes with human oral absorption (HOA) data available were employed to show the effect of acidity of drugs in oral absorption prediction. HOA was reciprocally correlated to the negative value of the capacity factor (kBMC) determined by BMC at pH 7.4 and 6.5. The relationships between kBMC and the corresponding HOA values of all compounds were rather poor, but the correlations were improved when the acidity of drugs was taken into consideration. Moreover, the proposed models allowed obtaining of good predictive values for both highly and poorly absorbed compounds. It is demonstrated that the constructed models derived from compounds with the same kind of charge property are of more practically meaningful and rigorous.
