1.Effects of exercise preconditioning on expression of STAT3 and Caspase-3 in the heart of rats with exhaustive exercise injury

BACKGROUND:Signal transducer and activator of transcription 3 (STAT3) and Caspase-3 are the important factors in JAK2/STAT3 signaling pathway. However, there are few studies on their role in the exercise preconditioning for myocardial protection. OBJECTIVE: To analyze the effects of exercise preconditioning on the expression of STAT3 and Caspase-3 in the heart of rats and investigate the cardioprotective mechanism of action. METHODS: Eighty Sprague-Dawley rats were randomly divided into control, exhaustive, exercise preconditioning, and exercise preconditioning+AG490 groups. The exercise preconditioning rat models were established by subjecting to intermittent treadmil exercise for 3 successive days. Rats in the control group were fed routinely for 3 days. Rats in the exhaustive group underwent treadmil exercise at 30 m/min until exhaustion after 3 days of routine raise. Rats in the exercise preconditioning group underwent exercise preconditioning for 3 days, and 24 hours later, they underwent treadmil exercises at the speed of 30 m/min until exhaustion. 2.Wireless human body communication technology.

The Wireless Body Area Network (WBAN) is a key part of the wearable monitoring technologies, which has many communication technologies to choose from, like Bluetooth, ZigBee, Ultra Wideband, and Wireless Human Body Communication (WHBC). As for the WHBC developed in recent years, it is worthy to be further studied. The WHBC has a strong momentum of growth and a natural advantage in the formation of WBAN. In this paper, we first briefly describe the technical background of WHBC, then introduce theoretical model of human-channel communication and digital transmission machine based on human channel. And finally we analyze various of the interference of the WHBC and show the AFH (Adaptive Frequency Hopping) technology which can effectively deal with the interference.

3.Clinical study of human papillomavirus 16 integrated role in the pathogenesis of cervical cancer

Objective To explore the effect of the integration of human papillomavirus 16 on the cervical cancer patients.Methods Outpatient colposcopy cervical liquid-based cytology examined (TCT)information in Kailuan General Hospital and Linxi Hospital of Kailuan General Hospital from January 2014 to June 2015 were selected.The Results of TCT were positive in 180 patients with as case group,were negative in 128 patients as control group.The effect of the integration of human papillomavirus 16 was analyzed.Results In the case group,there were 86 cases with HPV-positive,37 cases with HPV16-positive,10 cases with HPV16-free,10 cases with HPV16-concomitant,17 cases with HPV16-integration,and the integrated rate was 46.0%.While in the control group,there were 20 cases with HPV-positive,3 cases with HPV16-positive and all of them were HPV16-free.The integration degree was significantly higher than the control group (P =0.000).Conclusion The integration of human papillomavirusl6 has much significant influence on the cervical cancer.
4.Observation of the effect of acarbose tablets combined with insulin in the treatment of elderly patients with type 2 diabetes

Objective To evaluate the efficacy and safety of acarbose tablets combined with protamine zinc recombinant lai pulp mixing insulin injection in the treatment of elderly patients with type 2 diabetes.Methods 170 elderly patients with type 2 diabetes were randomly divided into the observation group and the control group.85 cases in the observation group were given acarbose tablets combined with protamine zinc recombinant lai pulp mixing insulin injection,85 cases in the control group received aspart 30.The patients were followed up for 12 weeks,the blood glu cose levels,blood glucose fluctuation,glycosylated hemoglobin (HbAlc),the incidence of hypoglycemia and insulin dosage were observed.Results' The level of fasting plasma glucose (FPG) (t =9.92,P < 0.05),2h postprandial blood glucose(2hPG) (t =18.18,P <0.05),the mean blood glucose within 4h(t =13.62,P <0.05),days of average blood glucose fluctuations (t =14.56,P < 0.05) of observation group dropped significantly compared with that before treatment;HbAlc achieved rate(6.5%)of the observation group and control group were 35.29%,28.23%,re spectively,HbAlc achieved rate (7.0%) of observation group and control group were 57.64%,52.94%,the differences were not statistically significant(all P > 0.05).The dose of insulin was significantly lower in the observation group(20.11 ±3.36) u/d than(35.78 ±4.68) u/d in the control group(t =25.08,P <0.01).There were 2 cases of hypoglycemic events and 3 cases in the control group.Conclusion Protamine zinc recombinant insulin lispro injection combined with acarbose has reliable clinical efficacy and safety.
5.JAK2 expression in myocardial tissue of rats after exercise preconditioning and exhaustive exercise injury

BACKGROUND:Several studies have demonstrated that JAK2 kinase plays an important role in myocardial protection by ischemic preconditioning and medicine preconditioning. OBJECTIVE:To investigate the effects of exercise preconditioning on JAK2 mRNA and protein expression in the rat heart and the underlying mechanism of action. METHODS:SD rats were randomly divided into control, exhaustive, exercise preconditioning, and exercise preconditioning + AG490 groups. Rats in the control and exhaustive groups were fed routinely for 3 days. Rats in the exercise preconditioning and exercise preconditioning + AG490 groups were subjected to intermittent treadmil exercise for 3 successive days to establish exercise preconditioning animal models. Rats in the exercise preconditioning + AG490 group were intraperitonealy injected with JAK2 inhibitor AG490 at 10 minutes before exercise preconditioning. After 3 days, rats in the exhaustive, exercise preconditioning, and exercise preconditioning + AG490 groups were subjected to exhaustive treadmil exercise. Serum levels of lactate dehydrogenase and creatine kinase isoenzyme were determined. JAK2 mRNA and protein expression in the rat myocardial tissue was detected by real-time fluorescent quantitative PCR and immunohistochemistry.RESULTS AND CONCLUSION:Compared with the control group, serum levels of actate dehydrogenase and creatine kinase isoenzyme were increased, and JAK2 mRNA and protein expression in the myocardial tissue was significantly increased in the exhaustive group (P < 0.05). Compared with the exhaustive group, serum levels of actate dehydrogenase and creatine kinase isoenzyme were decreased (P < 0.05), while JAK2 mRNA and protein expression in the myocardial tissue was increased in the exercise preconditioning group (P < 0.05). Compared with the exercise preconditioning group, serum levels of actate dehydrogenase and creatine kinase isoenzyme were increased (P < 0.05), while JAK2 mRNA and protein expression in the myocardial tissue was decreased in the exercise preconditioning + AG490 group (P < 0.05). The results confirm that exercise preconditioning plays its cardioprotective role through activating JAK/STAT signaling pathway, up-regulating the expression of JAK2 mRNA and protein in the cardiac tissue and aleviating myocardial ischemia injury.
6.Effect of Exercise Preconditioning on the Expression of Calcitonin Gene-Related Peptide in the Dorsal Root Ganglion of Rats

Objective The goal of this study was to explore the effect of exercise preconditioning on the expression of calcitonin gene-related peptide(CGRP)in the dorsal root ganglion of rats.Methods Fifty healthy male SD rats were randomly divided into control group (C) and exercise preconditioning group(EP).Group EP performed intervial treadmill exercise for 3 weeks for establishing exercise preconditioning animal model.The expression of CGRP mRNA in dorsal root ganglion was investigated by real-time fluorescence quantitative PCR.The immunoreaetion of CGRP in dorsal root ganglion was shown by immunohistochemistry.Results There was no significant difference in the expression of CGRP mR.NA in two groups(P>0.05).As compared with the group C,the immunoreaction of CGRP was increased in group EP,and the positive area and mean optical density of CGRP immunoreaction in group PE were significant higher than that in group C(P<0.05).Conclusion Exercise preconditioning does not change the expression of CGRP mRNA in dorsal root ganglion,but enhances the expression of CGRP in dorsal root ganglion to promote the reserve and release of CGRP in peripheral nerve endings and has the same endogenous protection as ischemie preconditioning.
7.Effect of Exercise Intensity on Cardiac Calcitonin Gene-Related Peptide in Rats

To study the effect of exercise on cardiac calcitonin gene-related peptide (CGRP) using immunnohistochemistry and the technique of computer image analysis, the expression and mechanism of cardiac CGRP on the animal model trained with different exercise intensities were investigated. The result showed that long-term low intensity of exercise was not able to induce obvious change in cardiac CGRP. After long-term moderate intensity of exercise, the expression of cardiac CGRP increased so as to improve blood supply and protect myocardium. Long-term high intensity of exercise decreased expression of cardiac CGRP and weakened the protection of myocardium which could be a chief cause of myocardial ischemia.
8.Comparison of biological characterisitics between adult and fetal bone marrow mesenchymal stem cells

BACKGROUND:At present,mesenchymal stem cells(MSCs) is mainly obtained from stem cells.But the quantity of human bone marrow mesenchymal stem cells(BMSCs) is lower.Fetal MSCs possess weak immunogenicity and good differentiation potential.OBJECTIVE:To compare the biological difference in adult and fetal BMSCs.DESIGN,TIME AND SETTING:The cytology in vitro controlled study was performed at the Central Laboratory,Hospital Affiliated to Medical College,Qingdao University from February to October 2008.MATERIALS:Three samples of adult bone marrow blood were obtained from Hospital Affiliated to Medical College,Qingdao University.Three samples of fetal bone marrows were supplied by Qingdao Municipal Hospital.METHODS:Human and fetal BMSCs were harvested in vitro by the Percoll method,and incubated in LG-DMEM containing 10% volume fraction fetal bovine serum.Till cells were 90% confluent,cells were digested and passaged.The same passage of adult and fetal BMSCs were collected,and treated with KGla suspension and DMEM containing fetal bovine serum at 37 ℃ in 0.05 volume fraction CO2 for 2 hours.Non-adherent cells were removed.At the third passage,BMSCs at a density of 1?106 were incubated.When cells were 60%-80% confluent,cells were incubated in osteoblast medium for 16 days.MAIN OUTCOME MEASURES:Cell morphological changes were measured.Growth curve was detected by MTT assay.Cell adherent rate in vitro was calculated.Alkaline phosphatase staining was used to detect the differentiational results of osteoblasts.RESULTS:Adult and fetal BMSCs showed spindle-shape,but the fetal BMSCs were small.The doubling time of them was 50 hours and 30 hours,respectively.Compared with the adult BMSCs,cell adherent rate was significantly lower in the fetal BMSCs(t=4.22,P

Bone marrow mesenchymal stem cells is a new approach for diabetes treatment, it can transdifferentiate into insulin cells in vitro, that may overcome the shortage of human donor islets, avoid the immunological rejection and moral principle problems which are involved into the research of embryonic stem cells.Studies have shown that insulin gene and its other necessary gene can introduce into primary cells or other somatic stem cells directly in diabetes mellitus patients, such as hepatic cells, intestines epidermis K cells, fibroblast cells and liver tumor cells and so on.Thereby, it can be translated into insulin-secreting cells.At present, the main study direction internationally is how to import insulin gene into target cells effectively, and make it express in a physical model.
10.Adeno-associated Virus Serotype 2 Mediated Transduction and Expression of The Human β-Globin Gene in Human Early Fetal Liver Hematopoietic Cells

β-Thalassemia is an inheritance anaemia disease due to the defect in β- globin gene. Gene therapy is considered to be the only method which could cure this disease. Adeno-associated virus type 2 (AAV2) has benn gaining more attention as a vector in human gene therapy for its non pathogenic character and broad host range. Although, the efficacy of recombinant AAV2 (rAAV2) in transducing human hematopoietic stem cells has been investigated by researchers, the results were varied from different laboratory. The view was proposed recently that it may be resulted from helper virus in their packaging system. Respecting this, the packaging system without helper virus was used to produce rAAV2. Human early fetal liver hematopoietic cells not only possess many superior peculiarity compared to hematopoietic cells of bone marrow or cord blood, but also the inherent β-globin gene in the cells is not expressed. Studies on the AAV2 transduction of human fetal liver hematopoietic cells and mediated expression of β- globin gene in vivo were performed and the potential role of AAV2 in β-thalassemia gene therapy was analyzed. The rAAV2 containing a normal human β-globin gene (rAAV2-β-globin) without helper virus contamination were produced. The viral titer, purity and the ability of mediating expression of β-globin gene were detected in vitro. Then, human early fetal liver hematopoietic cells were isolated and were further transducted with the rAAV2-β-globin, followed by transplantation into sublethally irradiated BALB/C nude mice to analyze the β-globin gene expression. The results showed that the high titer and purity of rAAV2-β-globin had the ability of mediating β-globin gene expression in vitro. In 8 recipient BALB/C nude mice, the β-globin gene expression were detected in the 2 mice marrow by RT-PCR. The results suggested that rAAV2 could transduce human fetal liver hematopoietic cells and mediate the β-globin gene expression in BALB/C nude mice, meanwhile the expression level of the gene was still rather low. It is necessary to perform further research on AAV2 biology before applying in β-thalassemia gene therapy.
