1.mTOR and glomerular disease
International Journal of Pediatrics 2012;(6):602-605
The mammalian target of rapamycin (mTOR)is a evolutionary conserved serine/threonine kinase that catalytics specific substrates and regulates cellular basic physiological events, such as growth, proliferation, autophagy and protein translation. It plays an important role in the regulation of cancer, diabetes and a variety of kidney diseases. Most of glomerular diseases are resulted from the podocytes and filtration barrier damage.Studies have suggested that there is a close relationship between mTOR and podosytes damage, but specific mechanism of regulation is not clear now. This review discusses the regulatory mechanism of mTOR kinase and its role in the glomerular disease to provide new ideas for understanding of glomerular disease pathogenesis.

International Journal of Pediatrics 2014;41(6):644-648
Objective To explore the role of mammalian target of rapamycin (mTOR) complex in the adriamycin induced the podecytes. Methods 1 μmol/L adriamycin (ADM) was used to induce mice podocytes for 24 hours and detected the apoptosis ratio, podosytes damage markers (Desmin) and the related expression of mTOR,then the role of mTOR was analysized in ADM induced podosytes. Results 1 umol/L ADM induced mice podosytes for 24 hours can make the podocytes damage.Compared with control group, podosytes activity declined(t =28.68,P < 0.001),apoptosis rate increased both early (t =10.24,P =10.24) and late (t =4.26,P =4.26),and Desmin expression increased(t =6.16,P <0.001).mTOR expression decreased in ADM group.Compared with control group, both mTOR and phosphorylation mTOR expression decreased (t =3.57,P =3.57).mTOR downstream target molecular S6K1 and 4ebpl mRNA expression decreased(t =18.28,P < 0.001).Conclusion ADM can make podosytes damage and the mechanism of ADM induced podosytes damage may be related to the decreasing mTOR and the low activity of mTOR.

Objective: To study angiogenesis and its influencing factors in human non-small cell lung cancer(NSCLC). Methods: The values of intratumor microvessel density(IMVD) in 95 cases of NSCLC were determined and its correlation with pathological parameters was analyzed. The relationship between IMVD and expressions of MMP-2,MMP-9,TIMP-2,VEGF,b-FGF,CD44v6,NOS2,NOS3 and E-cad in NSCLC was assessed. Results: There was active tumor angiogenesis of NSCLC, and the values of IMVD were correlated with clinicopathological stages of NSCLC and lymph nodes metastasis. The values of IMVD in positive expressions of VEGF, b-FGF, MMP-2, TIMP-2,CD44v6,NOS2 and NOS3 were significantly higher than that in negative expressions, opposed to the expression of TIMP-2 and E-cad. Conclusion:IMVD is a valuable marker for evaluating stages and metastasis of NSCLC. MMP-2, TIMP-2, VEGF, b-FGF, CD44v6, NOS2, NOS3 and E-cad may take part in the regulation of angiogenesis in NSCLC.

It remains controversial whether tumor necrosis factor (TNF)-α antagonism is effective for asthma. This meta-analysis was performed to evaluate efficacy of TNF-α antagonism in treatment of patients with asthma. MEDLINE, EMBASE, LILACS, and CINAHL databases were searched for English-language studies published through January 3, 2010. Randomized-controlled trials comparing TNF-α antagonism with control therapy were selected. For each report, data were extracted in relation to the outcomes analyzed: asthma exacerbation, asthma quality of life questionnaire scores, and forced expiratory volume in 1 second. Four assessable trials were identified including 641 patients with asthma. TNF-α antagonism therapy was superior to control therapy in preventing exacerbations in asthmatics [pooled odds ratio 0.52 (95% confidence interval 0.29-0.88), P=0.02]; however, there was a nonsignificant reduction in asthma quality of life questionnaire scores [0.23 (0 to 0.47), P=0.05], forced expiratory volume in 1 second [0.03, (-0.14 to 0.10), P=0.74] when analyzed using standardized mean differences. TNF-α antagonism was superior to control chemotherapy in terms of asthma exacerbation, but not asthma quality of life questionnaire scores or forced expiratory volume in 1 second.

Objective: We observed the influence of recombinant human growth hormone(rhGH) on protein metabolism in hepatocirrhotic rats and investigated mechanisms. Methods: Ninety rats were randomized into three groups(n=30 for each group): group A(normal rats), B(cirrhotic rats) and C(cirrhotic rats receiving rhGH). Rats in group C were subcutaneously given rhGH with a dosage of 1 U/(kg?d) for up to 1 week. The plasma concentrations of albumin(ALB), prealbumin(PA), transferrin(TF) and insulin-like growth factor-1(IGF-1) were detected on experimental day 14. And liver biopsy was taken to examine the expression level of albumin mRNA with TR-PCR. Results: The albumin, prealbumin, transferrin and IGF-1 levels in groups A and C were significantly higher than those in group B. The expression level of albumin mRNA in grup C was significantly higher than that in group B. Conclusions: The rhGH can improve the level of IGF-1 in plasma and improve the expression of albumin mRNA. Moreover, it may significantly improve the synthesis of protein and attenuate hypoproteinemia in cirrhotic rats.

Objectives To investigate the effect of matrine (MAT) on adriamycin (ADR) induced podocyte injury in vitro and explore the function of the mammalian target of rapamycin (mTOR) protein signaling pathway during the intervention. Methods ADR (1μmol/L) was used to induce the model of podocyte injury and then the podocytes were intervened by 10, 20 and 40μg/ml MAT for 24 hours. The viability and apoptosis rate of podocytes, mRNA and protein expressions of desmin and mTOR were detected. The effect of different concentrations of MAT on ADR-induced podocytes was analyzed. Results Com-pared with the control group, declined viability of podocytes (P<0.001), increased percentage of apoptotic podocytes (P<0.001), and increased expression of damage marker desmin (P=0.002) were observed in ADR group. In ADR group, after intervention with MAT of 10-40μg/ml, increased viability of podocytes (P<0.05), decreased percentage of apoptotic podocytes (P<0.05), and decreased expression of damage marker Desmin were found (P<0.05). The protein expression of mTOR and phosphorylation state of mTOR (p-mTOR) decreased in ADR induced-podocytes (P<0.05), and after intervention with MAT of 10-40μg/ml, the expression of mTOR and p-mTOR increased (P<0.05). The expression of mTOR downstream target protein s6k1 and 4EBP1 mRNA decreased in ADR group (P=0.071), while increased after intervention with MAT of 10-40μg/ml (P<0.05). Conclusions 10-40μg/ml MAT have protective effects on ADR-induced podocytes in vitro. The protection mechanism may be related to mTOR signaling pathway.
