1.Clinical Study of Acupuncture Intervention in Gastroscopy Side Reaction by Superficial Needling at Point Fenglong

Objective To investigate the clinical intervention effect of superficial needling at point Fenglong on gastroscopy side reaction.Methods Eighty-two gastroscopy patients were randomly allocated to treatment and control groups, 40 cases each. The treatment group received gastroscopy and superficial needling at point Fenglong, and the control group, gastroscopy and sham acupuncture at point Fenglong. The pain score and the incidence of nausea and vomiting were observed in the two groups during gastroscopy.Results There was a statistically significant difference in the pain score between the two groups (P<0.05). The incidence of nausea and vomiting was 31.7% in the treatment group and 65.9% in the control group; there was a statistically significant difference between the two groups (P<0.05).Conclusion Superficial needling at point Fenglong can effectively reduce the incidence of gastroscopy side reaction.
2.Meta-analysis on the relationship between MTHFR gene polymorphisms and colorectal cancer

Objective To evaluate the relationship between 5,10-Methylenetetrahydrofolate reductase(MTHFR) gene polymorphisms and colorectal cancer(CRC).Methods Studies were selected based on the criteria for inclusion.The Meta-analysis software,REVMAN 4.2,was applied for checking the heterogeneity across the studies and calculating the pooled OR.The results were evaluated by the analyses of publication bias and sensitivity.Results A total of 9 787 cases and 12 986 controls from 18 studies for C677T and a total of 4 422 cases and 5 819 controls from 9 studies for A1 298C were included.No heterogeneity among the studies was found.For codon 677,the frequencies of CC,CT and TT genotypes were 46.48%,43.81% and 9.71% in cases,and 45.03%,43.08% and 11.89% in controls,respectively.The pooled OR of TT vs.CT+CC was 0.80(95%CI 0.74~0.87).For codon 1 298,the frequencies of AA,AC and CC genotypes were 53.60%,39.39% and 7.01% in cases,and 53.31%,38.67% and 8.03% in controls,respectively.The pooled OR of CC vs.AC+AA was 0.84(95%CI 0.72~0.97).Conclusions MTHFR 677TT is at lower risk of developing CRC and 1 298CC genotypes might be associated with the decreased risk of developing CRC.
3.The feasibility of endothelial reconstruction in the injured arterial wall by autologus endothelial cells transplantation

Objective To investigate the feasibility of endothelial reconstruction in the injuried arterial wall by autologous endothelial cells transplantation. Methods New Zealand white rabbits (n=20) were subjected to bilateral iliofemoral artery balloon injury, Cultured autologous venous endothelial cells were immediately transplanted by balloon catheter into one vessel, whereas the contralateral artery received the medium only. In 10 rabbits, vessels were harvested 4 hours and 4 days after transplantation for analysis of endothelial coverage by scanning electron microscope (SEM); In 5 rabbits, the cultured endothelial cells were labeled with a fluorescent tracer before their introduction into the injured vessel, and 4 days after transplantation, vessels were harvested to obtain fluorescent imaging of the seeded endothelial cells; Another 5 rabbits were sacrificed 4 days after transplantation for Evans blue staining. Results Four hours after the operation, SEM demostrated that the endothelial layer in control vessels were denuded completely, whereas some round endothelial cells had adhered into the aterial wall in cell transplantation group; four days after cell transplantation, the transplanted cells had attached and spread in the injuried aterial wall by SEM, a number of fluorescent labeling endothelial cells were observed in the endothelial denuded aterial wall, the vessels that received the medium only were stained nearly completely by Evans blue, whereas in those vessels that received cell transplantation, about 60% area were not stained. Conclusion Autologus endothelial cells can be effectively transplanted into the injuried arterial wall by balloon catheter.
4.The role of microRNA-219-2-3p in Gastric Cancer and the underlying mechanism

Objective To evaluate the role of miR219‐2‐3p in gastric cancer and the underlying mechanisms .Methods Real time RT‐PCR was employed to quantify the expression level of miR219‐2‐3p in matched tissues of 82 patients .In vitro cell proliferation , the expression of tumorgenesis related protein ERK1/2 was performed by overexpressing miR‐219‐2‐3p in gastric cancer cell line MGC803 .Results The expression level of miR‐219‐2‐3p significantly decreased in late stage gastric cancer (P< 0 .05) .The cell proliferation rate was significantly reduced in gastric cancer cell lines overexpressing miR‐219‐2‐3p .Furthermore ,the expression level of p‐ERK that activated form of ERK was significantly decreased whereas the total level of ERK was unchanged after overex‐pressing of miR‐219‐2‐3p in MGC803 cell lines .The expression level of p‐ERK was also increased in gastric cancer tissues .Conclu‐sion miR‐219‐2‐3p may acts as a tumor suppressor in gastric cancer by decreasing the activity of ERK 1/2 signaling pathway .
5.The express and research of chemerin and CRP in 45 patients with early-onset preeclampsia

Objective The purpose of this study was to compare chemerin and CRP levels between women with and without ear‐ly‐onset preeclampsia based on maternal body mass index (BMI) .Methods 45 patients with early onset preeclampsia (gestation week is less than 34 weeks) ,30 normal pregnancy women were enrolled in the study .The serums levels of chemerin and CRP were measured in these pregnancy women .Results The chemerin [(379 .18 ± 52 .09)ng/L ,(481 .27 ± 63 .12)ng/L] and CRP [(7 .59 ± 1 .41)mg/L ,(11 .74 ± 1 .61)mg/L] levels in MPE and SPE were significantly higher than those [(316 .08 ± 51 .90)ng/L ,(3 .47 ± 1 .27)mg/L]in normal pregnancy women(P<0 .05) .While the chemerin(424 .64 ± 46 .61)ng/L and CRP (8 .32 ± 3 .82)mg/L levels increased with maternal BMI in patients overweight and obese women levels compared with normal weight women [(331 .66 ± 51 .77)ng/L ,(5 .93 ± 2 .95)mg/L] ,the difference are significant(P<0 .05) .The correlation analysis showed that the chemerin were positively correlated with BMI ,systolic blood pressure ,diastolic blood pressure ,triglycerides ,and CRP were positively correlated with BMI .Conclusion The expression of chemerin and CRP were associated with severity of preeclampsia and BMI .
6.The effects of different training regimens on motor function recovery and nerve-muscle morphology after spinal cord injury in rats

Objective To investigate the effects of three different motor training regimens on motor function improvement after spinal cord injury (SCI) in rats.Methods Forty-five healthy,adult Sprague-Dawley rats (female),weight 260-300 g,were included.Six rats were selected as the normal group.A model of incomplete SCI at the T9 level was induced in the others using a modification of Allen's method.Nine rats died after the injury and were excluded.The other 30 rats were randomly divided into 5 groups:a 7 d control group,a 35 d control group,a bodyweight-supported-treadmill-training (BWSTT) group,a swimming training group and a wheel running group,with 6 rats in each group.The three training groups began exercising at the 8th day post surgery,30 min per day,5 days a week for 4 weeks.Locomotor function was evaluated by inclined plane tests,modified Tarlov scores,and the BassoBeattie-Bresnahan (BBB) scale before the operation and on the 1st,7th,14th,21st,28th,and 35th day post surgery.Histomorphological changes of the T9 level spinal cord and the gastrocnemius muscle were observed with light microscopy and electron microscopy,and the cross sectional areas and diameters of the gastrocnemius muscle fibers were calculated.Results ①In the BWSTT group and the swimming training group,locomotor function scores increased significantly at all time points compared with the two control groups.There was no significant difference in rehabilitative effect between the BWSTT group and the swimming training group.But compared with both control groups,improvement in the wheel running group was not significant.②After 4 weeks of training,histomorphological observation of the injured T9 spinal cord tissue and gastrocnemius muscle fibers showed that the improvement in the BWSTT group was the most significant.The edema of the injured T9 level spinal cord tissue had decreased,cell vascular degeneration had lessened,the morphology of the neurons and glial cells trended to perfect recovery and nerve fibers had proliferated significantly.③Comparing the cross sectional areas and diameters of gastrocnemius muscle fibers showed that the improved amyotrophy in the BWSTT group was the most significant.The average cross sectional area (55.34 + 14.46 μm2) and diameter (8.32 + 0.99 μm) were close to the normal group (55.49±13.84 μm2 and 8.37 +1.13 μm).The swimming training group also had great improvement (46.05±8.50 μm2 and 7.68 + 0.76 μm) compared with the 35 d control group,while improvement in the wheel running group was not significant.Conclusions All three motor training regimens can improve locomotor and neurologic function,but the effects of BWSTT and swimming are better than that of wheel running.

AIM:To investigate the therapeutic protective effects and function regulations of conventional antidiabetic treatment plus piperazine ferulate tablet on traumatic vascular endothelial cells in type 2 diabetes mellitus.METHODS:80 patients were randomly divided into the treating group(n=37)and the control group(n=43).Conventional antidiabetic treatment plus piperazine ferulate was given to the treating group,and the control group was treated with conventional antidiabetic treatment plus vitamin C every day,and all the patients were treated with 4 weeks.The levels of fasting blood glucose(FBG),glycosylated hemoglobin A1C(Hb A1C),circulating endothelial cell(CEC),serum lipid peroxide(LPO),endothelin(ET)、nitric oxide(NO),tissue-plasminogen activator(t-PA)were determined during the therapeutic period.RESULTS:After 4 weeks of treatment,the levels of serum CEC,LPO and ET were lower in the treating group than those in the control group(P

Objective To investigate the feasibility of using clustered regularly interspaced short palindromic repeats ( CRISPR)-mediated genome editing to downregulate the expression of programmed cell death protein 1 (PD-1) on primary T cells by using a lentivirus delivery system. Methods Lentivirus vec-tors pLentiCRISPR A1-A6 containing different PD-1 genomic DNA sequences as single guide RNA ( sgRNA) for Cas9 targeting were constructed individually. The lentivirus vectors were tranduced into primary CD4 T cells. Flow cytometry analysis was performed to detect the expression of PD-1 for evaluating the knockout ef-ficiency. Results The lentivirus vectors pLentiCRISPR A1-A6 carrying six different target sites were con-structed and respectively tranduced into primary CD4 T cells. The expression of PD-1 accompanied with the activation of T cells. Co-expression of CD25 and PD-1 was observed on activated T cells. All of the six sites could be targeted by Cas9, of which A2 and A6 sites were more efficient in knocking out the gene encoding PD-1 with a rate of 19% and 29%, respectively. Conclusion This study suggests that it is feasible to knock out the expression of PD-1 on primary T cells by using CRISPR.
9.Research on self-assembly micelles of N-(4-methylimidazole)-hydroxyethyl-chitosan loading quercetin

To improve the solubility of quercetin ( QT) , one of flavonoids that can inhibit the proliferation of vari-ous types of cancer cells, the novel amphiphilic polymer N-( 4-methylimidazole)-hydroxyethyl-chitosan ( MHC) , synthetized by chemical derivatization from chitosan, was used as the self-assembly micelles of QT. The formed polymer was characterized by 1 H NMR, elemental analysis and pyrene fluorescence spectrometry. The formulation of MHC micelles loading quercetin was optimized through single factor experiment. Then the optimized formulation was obtained as follows:the concentration of MHC was 0. 67% and the ratio of drug and carrier was 1 ∶10. The micelles particle size was ( 99. 21 ± 1. 71) nm, Zeta potential was +( 20. 01 ± 0. 72) mV and drug loading was ( 5. 42 ± 0. 32 )%. The in vitro release curve was investigated and was found to conform to Higuchi equation of Q=0. 1101 t1/2 -0. 064. The results of in vivo experiment showed that the mean rentention time and bioavail-ability of the MHC-QT micelles were 21. 42 h and 57. 49 μg h/mL, respectively, compared to 0. 30 h and 2. 50 μg h/mL of the free QT solution. These indicated that the MHC micelles could significantly improve the solubility of QT, the drug sustained-release effect and bioavailability, which would used as carrier for the anti-tumor drugs.
10.Characteristics of Biochemical Markers in Patients With Pulmonary Hypertension Related to Left Heart Disease

Objective: To investigate the differences of biochemical markers between the patients with pulmonary hypertension related to left heart disease (PH-LHD) and LHD; to explore the sensitive bio markers which may predict PH in LHD patients. Methods: A total of 355 LHD patients admitted to our hospital from 2014-01 to 2015-05 were enrolled. According to 2009 ESC/ERS guidelines, PH was deifned by pulmonary artery systolic pressure (PASP)>50 mmHg and patients were divided into 2 groups: LHD group,n=224 and PH-LHD group,n=131. The basic information with blood levels of biomarkers was recorded and their accuracy for predicting PH was analyzed. Results: The pathogenesis of LHD included 184 (51.83%) patients of coronary heart disease, 90 (25.35%) of dilated cardiomyopathy and 81 (22.81%) of cardiac valve heart disease. Compared with LHD group, PH-LHD group had increased ratio of NYHA III and IV degree (89.31% vs 45.54%), decreased LVEF [42.0 (33.0, 59.0) % vs 60.0 (42.0, 65.0) %], all P<0.001; PH-LHD group presented elevated blood levels of BNP, bilirubin, red cell distribution width (RDW), uric acid and cystatin C, while reduced lipoprotein (HDL), allP<0.001. PASP was positively related to biomarkers as BNP, bilirubin, RDW, uric acid and cystatin C, while negatively related to HDL. With the combination of BNP, direct bilirubin and RDW, the predictive value for PH-LHD under ROC curve was 0.828 with the sensitivity at 0.813, speciifcity at 0.708. Conclusion: Blood levels of biochemical markers were statistically different between the patients of PH-LHD and LHD; the combination of BNP, direct bilirubin and RDW showed the higher accuracy for predicting PH occurrence in LHD patients.
