1.Protective effects of EPA and DHA on lipopolysaccharide-stimulated rat mesangial cells

AIM: To investigate the effects of EPA and DHA on oxidative stress of lipopolysaccharide-stimulated rat mesangial cells. METHODS: The glomerular mesangial cells (GMCs) were stimulated by lipopolysaccharide (LPS) and incubated with EPA (10 μmol/L or 100 μmol/L) and DHA (10 μmol/L or 100 μmol/L) for 24 h, 48 h and 72 h. The activity of SOD, GSH-Px and the level of MDA was measured. The protein and mRNA expressions of MCP-1 and TGF-β_1 were detected by immunocytochemistry and real-time PCR method, respectively. RESULTS: The activities of SOD and GSH-Px were decreased and the concentration of MDA was increased when stimulated with LPS. EPA and DHA increased the activities of SOD and GSH-Px and decreased the concentration of MDA significantly. Meanwhile, the protein and mRNA expressions of MCP-1 and TGF-β_1 stimulated by LPS were decreased. DHA was more effective than EPA at the same concentration. CONCLUSION: EPA and DHA enhance the activities of antioxidant enzymes, decrease the concentration of MDA and inhibit the expression of TGF-β_1 and MCP-1, suggesting that the protective effect of EPA and DHA on kidney is related to the antioxidation and the inhibition of TGF-β_1 and MCP-1 expression.
2.Cortistatin expression in renal tissue of humans and IgA nephropathy patients

Objective To explore the condition of cortistatin (CST)expression in human renal tissue and the changes in the level of CST in IgA nephropathy (IgAN)of different degrees.Methods Ten tumor adjacent normal renal tissue samples were collected.The mRNA and protein expressions of CST in human renal tissue were detected by reverse transcription-polymerase chain reaction (RT-PCR)and Western blotting,respectively.Immunohistochemisty (IHC)was performed to locate the expression of CST in renal tissue.According to the grading system of Lee et al,IgAN was divided into three groups:grade Ⅰ -Ⅱ (group A),grade Ⅲ -Ⅳ (group B),and grade Ⅴ (group C),and ten renal biopsy tissue samples were collected for each group.IHC was performed to detect the change in the level of CST in normal and IgAN renal tissue of different degrees.The effect of clinical indices on the level of CST in IgAN renal tissue was assessed by multiple linear regression analysis.Results RT-PCR and Western blotting showed that CST was expressed in renal tissue and IHC showed that CST was expressed on renal tubular epithelial cells.In IgAN,the higher the pathological grade was, the higher the expression of CST in renal tubules was.Multiple linear regression analysis showed that the pathological grade was associated with the expression of CST in renal tissue (r =0.875,P <0.01).Conclusion CST may participate in the inflammatory reaction of IgAN pathological injury and exert anti-inflammation effects.
3.Establishment of sensitivity indexes system for estimating nursing quality

Objective To establishthe sensitivity indexes system for estimating nursing quality which conforms to China's national conditions and have the certain representative among the whole country. Methods After usingStructure-process-outcome as the theoretical framework and surveying the literatures domestic and abroad,a total of 42 nursing management experts from 30 provinces were consulted by the Delphi method,including symposium and two rounds of questionnaire consultation. Results The authority coefficient of the two rounds consultation were 0.89 and 0.91,familiarity coefficient were 0.86 and 0.88 respectively,and the coordination coefficient of expert opinion were 0.20 and 0.25. The sensitivity indexes system for estimating nursing qualityconsists of two level index systems,which were divided into three first level indexes and seven second level indexes. The first level index in-cluded nursing manpower allocation,nursing safety and nursing events,while the second level index includes nurse-inpatient ratio,accuracy rate of identification,the number of medication error events,the number of blood transfusion error events,the number of specimen error events,the number of drug extravasation events and high-risk patient falls (falling bed) incidence. Conclusion The results of this study are reliable. There is a clear definition and interpreta-tion of sensitivity indexes for estimating nursing quality and index monitoring is operable.
4.Quality control of multicenter nursing investigation research

Quality management of scientific research is a systematic work,quality control of multicenter nursing research depends on perfect and effective quality management system,measures and procedures,as well as rigorous scientific attitude in project team. To establish and improve quality management system of scientific research,to for-mulate standard quality control procedures,to promote the usage of electronic data acquisition system in multicenter nursing researches,and to improve scientific,standard,and refined quality management of research are effective mea-sures to guarantee quality control of multicenter nursing research. This paper summarized quality control methods for multicenter investigation,to provide more experience for nursing researchers as references.
5.Design and application of the electronic data capture system in multi-center nursing study

We designed and applied the electronic data capture system (EDC) in order to strengthen the re-search quality and improve the efficiency of data collection of multi-center nursing research. According to the needs of research,we reasonably designed the EDC structure and functions,including data collection,verification, summary and export. Further improvement was performed after EDC was inputted into using. By using EDC,resear-chers from sub-centers completed the data collection smoothly,and the data quality met with research requirements. The design and application of EDC could effectively guarantee the data quality and improve the research efficiency.
7. Application of horn shaped flap pedicled with the angular artery perforator for midface reconstruction

Objective:
To evaluate the effect of the horn shaped flap pedicled with the angular artery perforator for midface reconstruction.
Methods:
From March 2014 to April 2017, twenty patients underwent operations for the reconstruction of midface defect, defect due to trauma, tumors, moles, scar and other soft tissue defects after resection. Various horn shaped flap pedicled with the angular artery perforator, perforator flap was rotated and pushed to repair the defect area.The maximum width of the perforator flap was equal to the width of the defect, and the length is about 2.5 to 3.0 times the length of the defect.
Results:
All flaps survived.After 1 month to 1 year follow up on 20 cases, the flap is significantly improved in color、shape and function.The patients were satisfied with the final aesthetic and functional results.
Conclusions
The flap has flexibility and simply with reliable blood supply.The donor sites could be closed directly without skin graft.it is a simple and fast method for the reconstruction of midface defect.
8. Mutational analysis of RNA splicing machinery genes SF3B1, U2AF1 and SRSF2 in 118 patients with myelodysplastic syndromes and related diseases

Objective:
To investigate the incidence, molecular features and clinical significance of RNA splicing machinery genes mutation in myelodysplastic syndromes (MDS) and related diseases.
Methods:
Mutational analysis of splicing factor 3B subunit 1 (SF3B1) (K700E) , U2 small nuclear RNA auxiliary factor 1 (U2AF1) (S34, Q157P) and serine/arginine-rich splicing factor 2 (SRSF2) (P95) in 118, de novo MDS and related diseases were separately performed by using polymerase chain reaction (PCR) followed by sequence analysis.
Results:
Of 118 MDS patients, 76 males and 42 females, the median age was 53.5 (13-84) years old. 19.49% (23/118) had SF3B1 (K700E) mutation. As compared with those with wild type SF3B1, patients with SF3B1 K700E were of older[58 (32-78) years
9.A Pilot Study of Clinical Evaluation and Formation Mechanism of Irritable Bowel Syndrome-like Symptoms in Inflammatory Bowel Disease Patients in Remission

Background/Aims:
Some inflammatory bowel disease (IBD) patients in remission suffer from irritable bowel syndrome (IBS)-like symptoms (IBD-IBS). The pathogenesis has not yet been elucidated. The study aim is to evaluate relationships among quality of life (QOL), psychological status, and visceral sensitivity, and explore the formation mechanism of IBD-IBS.
Methods:
Forty-seven patients with Crohn’s disease in remission, 24 ulcerative colitis in remission, 26 IBS, and 20 healthy controls were included in the study. The abdominal pain, QOL, anxiety, and depression were evaluated through questionnaires. Visceral sensitivity was measured by rectal balloon distension. The serum levels of 5-hydroxytryptamine (5-HT) and nerve growth factor (NGF) were measured by enzyme-linked immunosorbent assay. The expressions of tryptase, 5-HT, NGF, and related receptors in colonic tissues were detected by immunohistochemistry and western blot.
Results:
Prevalence of IBS-like symptoms in Crohn’s disease and ulcerative colitis patients in clinical remission was 29.8% and 50.0%, respectively. The QOL was lower, the anxiety/depression scores were higher in IBD-IBS patients than those without IBS-like symptoms. Additionally, patients with IBD-IBS existed visceral hypersensitivity. Besides, abdominal pain was associated with poor QOL, visceral hypersensitivity, anxiety, and depression in IBD-IBS patients. The number of mast cells (MCs) and expressions of 5-HT, NGF, and related receptors were higher in IBD-IBS patients than those with no such symptoms. The serum levels of 5-HT and NGF positively correlated with abdominal pain and visceral hypersensitivity.
Conclusion
IBD-IBS patients may have low QOL and psychological abnormalities, as wells as visceral hypersensitivity which may be related to increased 5-HT and NGF levels released from activated mast cells.
