1.An preliminary study of the relationship between chronic prostatitis and IgA nephropathy

Objective To study the role of chronic prostatitis(CP)in the pathogenesis and development of IgA nephropathy(IgAN). Methods The incidence rate of CP was compared between the patients with IgAN and the patients with non-IgA mesangial proliferative glomerulonephritis. Furthermore, the clinical and pathological features were compared between the IgAN patients without CP and those with CP. The therapeutic effect of anti-inflammatory treatment in IgAN patients with CP was evaluated. Results Incidence of CP in IgAN patients was 44.7% and that in non-IgA mesangial proliferative glomerulonephritis patients was 21.6%. There was significant difference in the incidence of chronic prostatitis between the two groups. The incidence of glomerular sclerosis and the deposition of immune complexes in vascular wall in IgAN patients with CP were higher than that in patients without CP. The treatment of CP may be beneficial in ameliorating IgAN in some patients. Conclusion Chronic prostatitis is one of the probable pathogenetic factors in IgAN.
2.Three ceramides from gorgonian Echinogorgia sp.

Objective To find new compound from gorgonian.Methods Three ceramides have been isolated from the South China Sea gorgonian Echinogorgia sp.by silica gel column chromatography.Results Their structures were established as(2S,3S,4R)-N-[2-(1,3,4-trihydroxyicosan-2-yl)]-hexadecanamide(1),(2S,3S,4R)-N-[2-(1,3,4-trihydroxyicosan-2-yl)]-heptadecanamide(2),and(2S,3S,4R)-N-[2-(1,3,4-trihydroxyicosan-2-yl]-octadecanamide(3) by spectroscopic methods and chemical conversion.Conclusion It is the first time to report the three chemical compounds from coral Echinogorgia sp.and compound 2 is a new compound.
3.Diagnostic value of multiple tumor marker protein biochip detective system for patients with lung cancer

Objective To evaluate diagnostic values of multi-tumor markers protein biochip detective system for lung cancer. Methods Patients with lung cancer group(78 cases),benign pulmonary diseases group (118 cases) and control group (68 cases) were enrolled.Twelve tumor markers in serum (AFP,CEA,NSE,CA125,CA153,CA242,CA199,PSA,f-PSA,FER,?-HCG and HGH) were measured by the multi-tumor markers protein biochip detective system. Result The serum levels of CEA,CA125,Fer and CA242 in the lung cancer group were much higher than those of the other two groups.There were no significant differences of serum levels and positive rates of other tumor markers among three groups.The sensitivity of CEA,CA125,Fer and CA242 was 41.0%,35.9%,26.9% and 17.9% respectively,the specificity 89.8%,86.0%,86.6% and 95.7% respectively。 Conclusions The multi-tumor markers protein biochip detective system has diagnostic values in lung cancer.When more than three markers are positive simultaneously,the sensitivity was 24.4%,and the specificity was 96.8%.The result could help to detect the character of tumor.
5.Clinical and genetic analysis of two patients with congenital neutropenia caused by ELANE gene mutation.

OBJECTIVE:
To explore the clinical characteristics of congenital neutropenia caused by ELANE gene mutations.
METHODS:
Clinical manifestations, absolute blood neutrophil count, high-throughput exome sequencing for mutation screening, suspected locus Sanger sequencing verification, processes of diagnosis and treatment of two patients with congenital neutropenia caused by ELANE gene mutation were retrospectively analyzed.
RESULTS:
High-throughput sequencing has found that proband 1 has carried a heterozygous c.170C>T (p.Ala57Val) missense mutation in exon 2 of the ELANE gene, which was known to be pathological, and a heterozygous c.251T>G (p.Leu84Arg) mutation in exon 3 of proband 2, which was unreported previously. Sanger sequencing confirmed that neither mutation was inherited from their parents.
CONCLUSION
ELANE mutation is an important cause for congenital neutropenia. Detection of new pathogenic variants has enriched the mutation spectrum of the ELANE gene.
6.Identification of genes with consistent expression alteration pattern in ACC-2 and ACC-M cells by cDNA array.

OBJECTIVETo investigate the molecular events and metastasis-related genes in ACC-2 and ACC-M cells of adenoid cystic carcinoma.
METHODSAdenoid cystic carcinoma cell line ACC-2 and a sample of adenoic cystic carcinoma cell clones highly metastatic to the lung (ACC-M) were investigated. ACC-2 and ACC-M cells were cultured and collected. Total RNA was extracted using standard Trizol RNA isolation protocol. The poly A mRNA was purified and labeled in reverse transcription using M-MLV reverse transcriptase in the presence of Easytides deoxyadenosin 5' triphosphate [alpha-(33)p]. A cDNA array was assembled with 7675 EST clones which represented the same number of independent single genes. Prepared nylon membranes were hybridized with the [alpha (33)p]-dATP labeled mRNA from ACC-2 and ACC-M cells. Membranes were exposed to phosphor screen.
RESULTSThe high-through put analysis of gene expression pattern was obtained from ACC-2 and ACC-M cells by the hybridization of the cDNA array. The difference of parallel gene expression was analyzed. Genes were clustered according to their expression level in the ACC-M compared with ACC-2 cells. According to each gene's ratio of expression level, there were 17 genes which were upregulated with ratios over 3.0, and there were 12 genes which were downregulated with ratios below 0.33 (1/3.0 = 0.33).
CONCLUSIONSSignificantly different expression patterns between ACC-2 and ACC-M by cDNA array were observed. The differences lie in signal pathways, tumor antigens, immune molecular and some unknown genes.

7.Preparation and characterization of paclitaxel-loaded poly lactic acid-co-glycolic acid coating tracheal stent.

BACKGROUNDIn-stent restenosis caused by airway granulation poses a challenge due to the high incidence of recurrence after treatment. Weekly applications of anti-proliferative drugs have potential value in delaying the recurrence of airway obstruction. However, it is not practical to subject patients to repeated bronchoscopy and topical drug applications. We fabricated novel pacilitaxel-eluting tracheal stents with sustained and slow pacilitaxel release, which could inhibit the formation of granulation tissue. And we assessed the quality and drug release behaviors of drug-eluting stents (DESs) in vitro.
METHODSStents were dipped vertically into a coating solution prepared by dissolving 0.5 g (2% w/v) of poly lactic acid-coglycolic acid (PLGA) and 0.025 g (0.1% w/v) of pacilitaxel in 25 ml of dichloromethane. DES morphology was examined by scanning electron microscopy (SEM). Pacilitaxel release kinetics from these DESs was investigated in vitro by shaking in PBS buffer followed by high performance liquid chromatography (HPLC).
RESULTSUsing an orthogonal experimental design, we fabricated numerous pacilitaxel/PLGA eluting tracheal stents to assess optimum coating proportions. The optimum coating proportion was 0.1% (w/v) pacilitaxel and 2% (w/v) PLGA, which resulted in total pacilitaxel loading of (16.380 6 ± 0.002 1) mg/stent. By SEM the coating was very smooth and uniform. Pacilitaxel released from DES was at (0.376 3 ± 0.003 8) mg/d, which is a therapeutic level. There was a prolonged, sustained release of pacilitaxel of >40 days.
CONCLUSIONSPaclitaxel-loaded PLGA coating tracheal stents were successfully developed and evaluated. Quality assessments demonstrated favorable surface morphology as well as sustained and effective drug release behavior, which provides an experimental reference for clinical practitioners.

8.Evaluation of three-dimensional CT reconstruction on the anatomic variation of inferior mesenteric artery and left colic artery.

OBJECTIVETo demonstrate the clinical applicability of three-dimensional CT angiography by evaluating the anatomic features and variation of inferior mesenteric artery(IMA) and left colic artery(LCA) in order to provide reference to vessel ligation strategy in laparoscopic rectal cancer surgery.
METHODSClinical and image data of 123 patients receiving abdominal multislice CT at The Sixth Affiliated Hospital from 2014 to 2015 were retrospectively analyzed. The images were 3D-reconstructed with computer 3D CT angiography and arterial enhancement phase images were chosen for analysis. Linear distances from IMA root to abdominal aortic bifurcation and from LCA at IMA root level to IMA root were measured. Branch types of IMA, coursing pattern of LCA, and association between LCA and inferior mesenteric vein (IMV) site were summarized.
RESULTSOf 123 cases, 80 were males and 43 were females, mean age was (46.8±16.6) years, body weight was (57.7±10.4) kg, and BMI was (21.3±3.6) kg/m. The average distance from IMA root to abdominal aortic bifurcation was (42.5±7.9) mm, and this distance was closely associated with body weight (OR=4.771, 95%CI: 1.398 to 16.283, P=0.013). Longer distance tended to appear in the heavier patients. LCA and sigmoid artery (SA) originating from same single IMA was found in 61(49.6%) cases; LCA and SA forking at same point in 35(28.5%) cases; LCA and SA coursing together and forking afterwards in 24(19.5%) cases, and LCA disappearing in 3(2.4%) cases. In 71(57.7%) patients, LCA ascended medial to the lateral border of left kidney, while in 16(13.0%) patients, LCA arranged below the inferior border of left kidney. When the LCA site was higher and the distance from LCA to IMA root was closer [distance from LCA to IMA root level was (24.2±9.9) mm, (30.0±15.2) mm and (66.6±12.3) mm, F=83.2, P<0.001]. At the level of IMA root, LCA located medial to IMV in 21(17.1%) cases, located just lateral to IMV in 54(43.9%) cases, and located lateral and ascended far away from IMV in 48(39.0%) cases.
CONCLUSION3D-CT angiography is non-invasive, efficient and accurate in evaluating coursing features and variation of IMA and its branches, which can provide important reference to the surgeons, promising laparoscopic surgery smooth and safe.
